Application of TCT to Research on Value in Screening for Cervical Cancer in Postmenopausal Women

Objective:To study the screening for cervical cancer in postmenopausal women in liquid-based cell test(TCT)application value.Methods:From January 2023 to March 2024 hospital check-ups and outpatient reached 400 cases of postmenopausal women,and they were TCT and HPV detection,during the study period to pathological diagnosis of numerical results as the standard,analysis of TCT detection application of screening for cervical cancer in postmenopausal women and the effect.Results:After the TCT detection,the inflammation group and normal group by use of HPV testing positive rate higher than other groups(P<0.05).Hospital after pathological diagnosis and research,after the screening system found CINⅠlevel and above cases about 39 cases,the TCT and HPV detection rate of positive difference is not obvious,and there is no statistical significance(P>0.05).Joint test analysis,the sensitivity and specificity were higher than that of TCT and HPV testing positive(P<0.05).Conclusion:TCT detection screening for cervical cancer in postmenopausal women is of great importance to the application,if you can join HPV testing,help to improve the detection sensitivity,avoid missed diagnosis of problems,and then for treatment and later restore to lay a solid foundation. 展开更多

Objective:Large volume radiological text data have been accumulated since the incorporation of electronic health record(EHR)systems in clinical practice.We aimed to determine whether deep natural language processing algorithms could aid radiologists in improving thyroid cancer diagnosis.Methods:Sonographic EHR data were obtained from the EHR database.Pathological reports were used as the gold standard for diagnosing thyroid cancer.We developed thyroid cancer diagnosis based on natural language processing(THCaDxNLP)to interpret unstructured sonographic text reports for thyroid cancer diagnosis.We used the area under the receiver operating characteristic curve(AUROC)as the primary metric to measure the performance of the THCaDxNLP.We compared the performance of thyroid ultrasound radiologists aided with THCaDxNLP vs.those without THCaDxNLP using 5 independent test sets.Results:We obtained a total number of 788,129 sonographic radiological reports.The number of thyroid sonographic data points was 132,277,18,400 of which were thyroid cancer patients.Among the 5 test sets,the numbers of patients per set were 439,186,82,343,and 171.THCaDxNLP achieved high performance in identifying thyroid cancer patients(the AUROC ranged from 0.857–0.932).Thyroid ultrasound radiologists aided with THCaDxNLP achieved significantly higher performances than those without THCaDxNLP in terms of accuracy(93.8%vs.87.2%;one-sided t-test,adjusted P=0.003),precision(92.5%vs.86.0%;P=0.018),and F1 metric(94.2%vs.86.4%;P=0.007).Conclusions:THCaDxNLP achieved a high AUROC for the identification of thyroid cancer,and improved the accuracy,sensitivity,and precision of thyroid ultrasound radiologists.This warrants further investigation of THCaDxNLP in prospective clinical trials. 展开更多

Objective Focused cardiac ultrasound(FCU)and lung ultrasound(LU)are increasingly being used in critically ill patients.This study aimed to investigate the effect of FCU in combination with LU on these patients and to determine if the timing of ultrasound examination was associated with treatment change.Methods This is a multicenter cross-sectional observational study.Consecutive patients admitted to the intensive care unit(ICU)were screened for enrollment.FCU and LU were performed within the first 24 h,and treatment change was proposed by the performer based on the ultrasound results and other clinical conditions.Results Among the 992 patients included,502 were examined within 6 h of ICU admission(early phase group),and 490 were examined after 6 h of admission(later phase group).The early phase group and the later phase group had similar proportions of treatment change(48.8%vs.49.0%,χ^(2)=0.003,P=0.956).In the multivariable analysis,admission for respiratory failure was an independent variable associated with treatment change,with an odds ratio(OR)of 2.357[95%confidence interval(CI):1.284-4.326,P=0.006];the timing of examination was not associated with treatment change(OR=0.725,95%CI:0.407-1.291,P=0.275).Conclusions FCU in combination with LU,whether performed during the early phase or later phase,had a significant impact on the treatment of critically ill patients.Patients with respiratory failure were more likely to experience treatment change after the ultrasound examination. 展开更多

The tool is named Mapreduce and BSP based Graphmining tool (MBGM). The core of this mining system are four sets of parallel graphmining algorithms programmed in the BSP parallel model and one set of data extractiontransformationload ing (ETE) algorithms implemented in MapReduce. To invoke these algorithm sets, we designed a workflow engine which optimized for cloud computing. Finally, a welldesigned data management function enables users to view, delete and input data in the Ha doop distributed file system (HDFS). Most previous works on cloud platforms only focus on the parallel algorithms for structured data. In this paper, we focus on the parallel implementation of web-mining algorithms and develop a parallel web-mining system that includes parallel web crawler; parallel text extract, transform and load (ETL) and modeling; and parallel text mining and application subsystems. The short-range repulsive interactions of any force field must be modified to be applicable for high energy atomic collisions because of extremely far from equilibrium state when used in molecular dynamics(MD)simulations.In this work,the short-range repulsive interaction of a reactive force field(ReaxFF),describing Fe-Ni-Al alloy system,is well modified by adding a tabulated function form based on Ziegler-Biersack-Littmark(ZBL)potential.The modified interaction covers three ranges,including short range,smooth range,and primordial range.The short range is totally predominated by ZBL potential.The primordial range means the interactions in this range is the as-is ReaxFF with no changes.The smooth range links the short-range ZBL and primordial-range ReaxFF potentials with a taper function.Both energies and forces are guaranteed to be continuous,and qualified to the consistent requirement in LAMMPS.This modified force field is applicable for simulations of energetic particle bombardments and reproducing point defects'booming and recombination effectively. 展开更多

Peanut root invasion by Bradyrhizobia is through a crack entry, which is different from many other legumes applying an infection thread entry in root hair. Understanding the role of root hair in the crack entry of Bradyrhizobia invasion of peanut root and subsequent peanut nodulation would facilitate improvement of biological nitrogen fixation in cultivated peanut. The objective of this study was to investigate the involvement of root hair in Bradyrhizobial invasion of peanut. Seedling roots of a nodulating peanut cultivar were observed for root hair emergence, its life span, and nodule formation at the base of the lateral roots with and without rhizobia inoculation for 14 days after germination (DAG). Scanning electron microscopy (SEM) was utilized to observe rhizobia accumulation at lateral roots at 24 hours after inoculation (HAI) before the emergence of root hair. Root hair emerged at 7 DAG with or without rhizobia inoculation. Two variations of rosette hair (RoH) were observed, the transient-thin RoH had life span of 3 days after root hair emergence and the thick and densely distributed RoH type stayed till the time of nodule emergence (9 days after inoculation). The lateral root devoid of root hair at the top 2 cm region was found to produce nodules. The SEM observation of seedling roots at 24 HAI showed that Bradyrhizobia invaded the roots at epidermis, protoplasm of cortical cell, and cortical cells of the main root near the newly emerged lateral root in the absence of RoH. The observations validated that root hair is not required in the Bradyrhizobia invasion of peanut root in the crack entry mode. Results from this study provided important morphological information for the hypothesis of close relationship between RoH and peanut nodulation for further genetic study of crack entry mechanism and signaling pathway of symbiosis between Bradyrhizobia and peanut. 展开更多

The 3-ketoacyl-CoA thiolase is the rate-limiting enzyme for linear dicarboxylic acids production.However,the promiscuous substrate specificity and suboptimal catalytic performance have restricted its application.Here we present both biochemical and structural analyses of a high-efficiency 3-ketoacyl-CoA thiolase Tfu_0875.Notably,Tfu_0875 displayed heightened activity and substrate specificity for succinyl-CoA,a key precursor in adipic acid production.To enhance its performance,a deep learning approach(DLKcat)was employed to identify effective mutants,and a computational strategy,known as the greedy accumulated strategy for protein engineering(GRAPE),was used to accumulate these effective mutants.Among the mutants,Tfu_0875N249W/L163H/E217L exhibited the highest specific activity(320%of wild-type Tfu_0875),the greatest catalytic efficiency(kcat/KM=1.00 min-1mM-1),the highest succinyl-CoA specificity(KM=0.59 mM,28.1%of Tfu_0875)and dramatically reduced substrate binding energy(-30.25 kcal mol^(-1)v.s.-15.94 kcal mol^(-1)).A structural comparison between Tfu_0875N249W/L163H/E217L and the wild type Tfu_0875 revealed that the increased interaction between the enzyme and succinyl-CoA was the primary reason for the enhanced enzyme activity.This interaction facilitated rapid substrate anchoring and stabilization.Furthermore,a reduced binding pocket volume improved substrate specificity by enhancing the complementarity between the binding pocket and the substrate in stereo conformation.Finally,our rationally designed mutant,Tfu_0875N249W/L163H/E217L,increased the adipic acid titer by 1.35-fold compared to the wild type Tfu_0875 in shake flask.The demonstrated enzymatic methods provide a promising enzyme variant for the adipic acid production.The above effective substrate binding pocket engineering strategy can be beneficial for the production of other industrially competitive biobased chemicals when be applied to other thiolases. 展开更多

In the absence of any new drugs, renal replacement therapy (RRT) is the most important treatment option. Randomized controlled trials have concluded that in critically ill patients without an emergency indication for RRT, a watchful waiting strategy is safe;however, further delays in RRT did not seem to confer any benefit, rather was associated with potential harm. During this process, balancing the risks of complications due to an unnecessary intervention with the risk of not correcting a potentially life-threatening complication remains a challenge. Dynamic renal function assessment, especially dynamic assessment of renal demand-capacity matching, combined with renal biomarkers such as neutrophil gelatinase-associated lipocalin and furosemide stress test, is helpful to identify which patients and when the patients may benefit from RRT. 展开更多

BackgroundSeveral prognostic biomarkers have been validated for acute myeloid leukemia(AML),a heterogeneous hematopoietic malignancy.However,the factors associated with the cumulative incidence of relapse(CIR)and leukemia-free survival(LFS)in real-world patients with AML have not been well defined.MethodsThis study examined clinical and mutational data of 246 patients with newly diagnosed AML who received the traditional“3+7”regimen in PLA General Hospital from January 2008 to August 2020.Factors associated with CIR and LFS in patients newly diagnosed with AML were analyzed using next-generation sequencing.ResultsAdditional sex combs-like 1(ASXL1)and Serine/arginine-rich splicing factor 2(SRSF2)mutations were found to be associated with an increased risk of CIR and a reduced LFS in univariate analysis,while only SRSF2 mutations were associated with these factors in the multivariate analysis.Hyperleukocytosis maintained an independent effect on LFS in the multivariate analysis.Hematopoietic stem cell transplantation conferred a significant prognostic benefit on both CIR and LFS in our cohort.Furthermore,we validated the risk classification of patients with AML receiving traditional induction regimens across a broad age range.Based on next-generation sequencing results,we concluded that SRSF2 mutations were predictive of an increased risk of relapse,inferior LFS rates,and non-relapse mortality in patients with newly diagnosed AML.ConclusionThese findings indicate that patients with SRSF2 mutations might not benefit from the conventional“3+7”regimen.Our results may help in developing molecular stratification strategies and could guide treatment decisions for patients with newly diagnosed AML. 展开更多

Transmission of an anisotropic metasurface is analyzed in a polar base relying on the Jones calculus, and polarization conversion from the spatial uniform polarization to the spatial nonuniform polarization is explored.Simple and compact polarization converters based on rectangular holes or cross holes etched in silver film are designed, and polarization conversions from the linear and circular polarization to the radial and azimuthal polarization are realized. Numerical simulations of three designed polarization converters consisting of rectangular holes equivalent to polarizers and quarter-and half-wave plates, exhibit the perfect polarization conversion.The experiment results consistent with the simulations verify theoretic predictions. This study is helpful for designing metasurface polarization converters and expanding the application of a metasurface in polarization manipulations. 展开更多

This paper studies the global bifurcation and Hopf bifurcation of one kind of complicated financial system with different parameter combinations. Conditions on which bifurcation happens, and the critical system structure when the system transforms from one kind of topological structure to another are studied as well. The criterion for identifying Hopf bifurcation under different parameter combinations is also given. The chaotic character of this system under quasi-periodic force is finally studied. The bifurcation structure graphs are given when two parameters of the combination are fixed while the other parameter varies. The presence and stability of 2 and 3 dimensional torus bifurcation are studied. All of the Lyapunov exponents of the system with different bifurcation parameters and routes leading the system to chaos with different parameter combinations are studied. It is of important theoretical and practical meaning to probe the intrinsic mechanism of such continuous complicated financial system and to find the macro control policies for such kind of system. 展开更多

Contamination of soil and water by arsenic is a global problem. In Australia, the dipping of cattle in arsenic-containing solution to control cattle ticks in last centenary has left many sites heavily contaminated with arsenic and other toxicants. We had previously isolated five soil bacterial strains (CDB 1-5) highly resistant to arsenic. To understand the resistance mechanism, molecular studies have been carried out. Two chromosome-encoded arsenic resistance (ars) gene clusters have been cloned from CDB3 (Bacillus sp.). They both function in Escherichia coli and cluster i exerts a much higher resistance to the toxic metalloid. Cluster 2 is smaller possessing four open reading frames (ORFs) arsRorf2BC, similar to that identified in Bacillus subtilis Skin element. Among the eight ORFs in cluster 1 five are analogs of common ars genes found in other bacteria, however, organized in a unique order arsRBCDA instead of arsRDABC. Three other putative genes are located directly downstream and designated as arsTIP based on the homologies of their theoretical translation sequences respectively to thioredoxin reductases, iron-sulphur cluster proteins and protein phosphatases. The latter two are novel of any known ars operons. The arsD gene from Bacillus species was cloned for the first time and the predict protein differs from the well studied E. coli ArsD by lacking two pairs of C-terminal cysteine residues. Its functional involvement in arsenic resistance has been confirmed by a deletion experiment. There exists also an inverted repeat in the intergenic region between arsC and arsD implying some unknown transcription regulation. 展开更多
