随着全球经济的快速发展与人口的日益膨胀,随之而来的能源消耗与环境污染也日益成为一个严峻的挑战.半导体光催化技术能够将低密度的太阳能转化为高密度的化学能,此外它能够通过产生活性自由基来降解空气或水中的污染物,因此在解决上述... 随着全球经济的快速发展与人口的日益膨胀,随之而来的能源消耗与环境污染也日益成为一个严峻的挑战.半导体光催化技术能够将低密度的太阳能转化为高密度的化学能,此外它能够通过产生活性自由基来降解空气或水中的污染物,因此在解决上述问题中具有巨大潜力,被认为是有着广阔前景的绿色无污染的能源转化和环境修复手段.在过去几十年的研究中,一些光催化剂表现出了较好的光催化活性,如TiO_(2)和ZnO等.然而,由于它们的宽带隙,仅仅在紫外光下具有活性,这极大地限制了其对太阳光的利用.为了尽可能地利用太阳能,研究者们开发了许多具有可见光活性的光催化剂.钨酸铋(Bi_(2)WO_(6))作为一种典型的Aurivillius层状钙钛矿材料,因具有独特的层状结构、良好的可见光催化活性、高的热稳定性和光化学稳定性及环境友好性等特点而备受关注.然而,有限的光吸收和光生载流子的快速复合阻碍了Bi_(2)WO_(6)光催化性能的进一步提高.因此,研究者们进行了大量的研究,致力于进一步增强Bi_(2)WO_(6)光催化剂的活性.本文对Bi_(2)WO_(6)基光催化剂的最新研究进展进行了系统综述.首先介绍了Bi_(2)WO_(6)的晶体结构、光学性质和光催化基本原理.然后,基于Bi_(2)WO_(6)的改性策略,包括形貌控制、原子调控和复合材料制备,重点讨论了Bi_(2)WO_(6)在水分解、污染物处理、空气净化、杀菌消毒、二氧化碳还原、选择性有机合成等领域的光催化应用.最后,对Bi_(2)WO_(6)基光催化剂当前面临的挑战和未来的发展作了展望和总结,提出了Bi_(2)WO_(6)光催化剂未来的一些研究方向,包括(1)大规模、精确可控地合成Bi_(2)WO_(6),特别是高活性晶面、多孔结构和量子点的设计;(2)精确调控原子位置,利用先进的技术手段进一步揭示活性位点上的光催化过程;(3)发展原位表征技术来观察复合光催化剂的界面电荷动力学以及开发新型Bi_(2)WO_(6)基复合体系.(4)通过机械应力、温度梯度以及电场等外场的耦合提高Bi_(2)WO_(6)的光催化性能;(5)进一步深入研究Bi_(2)WO_(6)在不同领域的光催化应用,特别是在肿瘤治疗和太阳能燃料制备方面,一些新的应用如固氮等也值得探索.期望本综述能够为Bi_(2)WO_(6)和其他高效光催化材料的设计提供一些指导和帮助. 展开更多

Collaborative f iltering, as one of the most popular techniques, plays an important role in recommendation systems. However,when the user-item rating matrix is sparse,its performance will be degenerate. Recently,domain-specific recommendation approaches have been developed to address this problem.The basic idea is to partition the users and items into overlapping domains, and then perform recommendation in each domain independently. Here, a domain means a group of users having similar preference to a group of products. However, these domain-specific methods consisting of two sequential steps ignore the mutual benefi t of domain segmentation and recommendation. Hence, a unified framework is presented to simultaneously realize recommendation and make use of the domain information underlying the rating matrix in this paper. Based on matrix factorization,the proposed model learns both user preferences of multiple domains and preference selection vectors to select relevant features for each group of products. Besides, local context information is utilized from the user-item rating matrix to enhance the new framework.Experimental results on two widely used datasets, e.g., Ciao and Epinions, demonstrate the effectiveness of our proposed model. 展开更多

To understand the mechanism of benzothiadiazole (BTH)-induced susceptible rubber clone resistance to powdery mildew on gene level, a differentially expressed cDNA library was constructed by suppression subtractive hybridization (SSH) with rubber Reyan 7-33-97 clone. The constructed cDNA library was high integrity through detection of the critical processes of SSH, such as efficiency of adaptor connection, subtraction and conversion, as well as the type of recombinant genes. The positive rate was 99% after identification with random 400 white spots. The size of the cDNA clone inserted fragments was various but most in 400 bp - 1000 bp. There were 23 cDNA sequences matching the function of energy and basic metabolism, signal transduction, membrane and transport, secondary metabolism and so on after detection of the 42 positive clone sequences selected randomly from the cDNA library and comparison on nucleic acid sequences in Genbank. 7 ESTs were logged in Genbank and accession numbers were GW873071 and GW874604- GW874610. The results implicated that BTH could effectively induced rubber tree resistance to powdery mildew through increasing expresses of defense-related genes in leaves of rubber tree susceptible clone. It should provide a new approach for rubber disease management. 展开更多

低的电荷分离效率以及欠丰富的表面活性位点一直是阻碍高效光催化CO2还原的关键问题.针对这些问题,本文首先利用导向剂辅助的水热反应制备了Bi4Ti3O12薄层纳米片,又利用二次水热还原在其表面引入氧空位,实现了气固体系中高效光催化CO2还... 低的电荷分离效率以及欠丰富的表面活性位点一直是阻碍高效光催化CO2还原的关键问题.针对这些问题,本文首先利用导向剂辅助的水热反应制备了Bi4Ti3O12薄层纳米片,又利用二次水热还原在其表面引入氧空位,实现了气固体系中高效光催化CO2还原,产率(11.7μmol g-1h-1)约是未改性Bi4Ti3O12的3.2倍.选择性光沉积Pt实验表明超薄结构缩短了光生电子的迁移距离,有利于其快速参与CO2还原反应.氧空位极大地改善了超薄纳米片的表面态和电子结构,大幅促进了对CO2的吸附、可见光的吸收和电荷分离.这项工作不仅报道了富含氧空位的超薄光催化材料,而且有望为发展耦合策略共同提高光催化CO2还原性能提供新见解和新思路. 展开更多

In this paper, a new strategy is proposed to realize a hierarchically porous metal-organic framework ([Cu2(OH)(L)]'(DMF)0.8 (FJU-11, H3L=3,5-(4-carboxybenzyloxy)benzoic acid, DMF= N,N-dimethylformamide) via using semi-rigid multi-carboxylic acids. Interestingly, FJU-11 possesses the large adsorption capacities and small isosteric heats toward CO2. The column breakthrough experiment for FJU-11 highlights its potential application in the separation of the flue gas. 展开更多

Naturally occurring interleukin-2(IL-2)is a pleiotropic glycoprotein that regulates immune responses by controlling the differentia-tion and homeostasis of T cells.Non-glycosylated IL-2 has been used in clinical settings for three decades.However,the function of the O-glycan of native IL-2 remains elusive.Herein,to stress this issue,we report a highly efficient semi-synthesis of homogeneous glycosylated IL-2 with various glycoproteoforms on a multi-milligram scale.The glycopeptide fragment was prepared by chemical synthesis and then merged with recombinant fragment via a serine ligation to generate the desired glycoprotein in a single opera-tion.Biological evaluation of the homogenous glycoprotein library reveals that the activity of IL-2 in activating individual T cell subset is glycan dependent,thus highlighting the possibility of further improving current clinical medicine. 展开更多

Preliminary characterization of bound extracellular polymeric substances(bEPS) of cyanobacteria is crucial to obtain a better understanding of the formation mechanism of cyanobacterial bloom. However,the characterization of bEPS can be affected by extraction methods. Five sets(including the control) of bEPS from Microcystis extracted by different methods were characterized using three-dimensional excitation and emission matrix(3DEEM) fluorescence spectroscopy combined chemical spectrophotometry; and the characterization results of bEPS samples were further compared. The agents used for extraction were NaOH,pure water and phosphate buffered saline(PBS) containing cationic exchange resins,and hot water. Extraction methods affected the fluorescence signals and intensities in the bEPS. Five fluorescence peaks were observed in the excitation and emission matrix fluorescence spectra of bEPS samples. Two peaks(peaks T1 and T2) present in all extractions were identified as protein-like fluorophores,two(peaks A and C) as humic-like fluorophores,and one(peak E) as a fulvic-like substance.Among these substances,the humic-like and fulvic-like fluorescences were only seen in the bEPS extracted with hot water. Also,NaOH solution extraction could result in strong fluorescence intensities compared to the other extraction methods. It was suggested that NaOH at pH 10.0 was the most appropriate method to extract bEPS from Microcystis. In addition,dialysis could affect the yields and characteristics of extracted bEPS during the determination process. These results will help us to explore the issues of cyanobacterial blooms. 展开更多

Although widely applied in treating hematopoietic malignancies,transplantation of hematopoietic stem/progenitor cells(HSPCs)is impeded by HSPC shortage.Whether circulating HSPCs(cHSPCs)in steady-state blood could be used as an alternative source remains largely elusive.Here we develop a three-dimensional culture system(3DCS)including arginine,glycine,aspartate,and a series of factors.Fourteen-day culture of peripheral blood mononuclear cells(PBMNCs)in 3DCS led to 125-and 70-fold increase of the frequency and number of CD34+cells.Further,3DCS-expanded cHSPCs exhibited the similar reconstitution rate com-pared to CD34+HSPCs in bone marrow.Mechanistically,3DCS fabricated an immunomodulatory niche,secreting cytokines as TNF to support cHSPC survival and proliferation.Finally,3DCS could also promote the expansion of cHSPCs in patients who failed in HSPC mobilization.Our 3DCS successfully expands rare cHSPCs,providing an alternative source for the HSPC therapy,particularly for the patients/donors who have failed in HSPC mobilization. 展开更多

Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its mechanistic study and development of vaccines and antivirals.Here,two infectious clones for the prototype and a Myc-tagged CVA10 were constructed.Viable CVA10 viruses were harvested by transfecting the viral m RNA into human rhabdomyosarcoma(RD)cells.Rescued CVA10 was further confirmed by next generation sequencing and characterized experimentally.We also constructed the vectors for CVA10 subgenomic replicon with luciferase reporter and viral capsid with EGFP reporter,respectively.Co-transfection of the viral replicon RNA and capsid expresser in human embryonic kidney 293 T(HEK293 T)cells led to the production of single round infectious particles(SRIPs).Based on CVA10 replicon RNA,SRIPs with either the enterovirus A71(EVA71)capsid or the CVA10 capsid were generated.Infection by EVA71 SRIPs required SCARB2,while CVA10 SRIPs did not.Finally,we showed great improvement of the replicon activity and SRIPs production by insertion of a cis-active hammerhead ribozyme(HHRib)before the 50-untranslated region(UTR).In summary,reverse genetic tools for prototype strain of CVA10,including both the infectious clone and the SRIPs system,were successfully established.These tools will facilitate the basic and translational study of CVA10. 展开更多

The coronavirus disease 2019(COVID‐19)is still causing a wide range of infections and deaths due to the high variability of the severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2).Therefore,it is necessary to establish a reliable and convenient pseudovirus‐based neutralization assay to develop drug targeted variants of SARS‐CoV‐2.Based on the HIV‐1 backbone,we generated a high titer luciferase(Luc)‐expressing pseudovirus packaging system.Three dominant S mutant substitution pseudovirus were also established and identified compared to wide type in hACE2‐overexpressingHEK‐293T cells(293T‐ACE2 cells).Compared to serine protease inhibitor camostat mesylate,the cysteine protease inhibitor E‐64d could significantly block all SARS‐CoV‐2 mutant S pseudovirus infection in 293T‐ACE2 cells.Furthermore,the neutralization ability of two antibodies targeted receptor‐binding domain(RBD)of SARS‐CoV‐2 spike protein(S)was evaluated,which showeddifferent inhibition dose–effect curves among four types of S pseudovirus.Overall,we developed a pseudovirus‐based neutralization assay for SARS‐CoV‐2,which would be readily adapted to SARS‐CoV‐2 variants for evaluating antibodies. 展开更多
