采用酶联免疫吸附法(ELISA)测定卵巢处于不同发育期的7龄雌哲罗鲑Hucho taimen血清中雌二醇(E_2)与睾酮(T)的含量,检测下丘脑促性腺释放素(GnRH)、垂体促滤泡激素(FSH)与促黄体激素(LH)及肝脏卵黄蛋白原(Vtg)mRNA表达水平的变化,分析生... 采用酶联免疫吸附法(ELISA)测定卵巢处于不同发育期的7龄雌哲罗鲑Hucho taimen血清中雌二醇(E_2)与睾酮(T)的含量,检测下丘脑促性腺释放素(GnRH)、垂体促滤泡激素(FSH)与促黄体激素(LH)及肝脏卵黄蛋白原(Vtg)mRNA表达水平的变化,分析生殖激素基因、性腺指数(GSI)和性类固醇激素之间的相关性,以深入了解下丘脑-垂体-性腺轴(HPG轴)对哲罗鲑卵巢发育的调控作用及其机制。结果显示,在卵巢发育期内下丘脑GnRH mRNA表达水平在Ⅳ早期时显著升高,至Ⅳ晚期时达到最低,随后显著升高;垂体FSH与LH mRNA表达水平自Ⅲ期显著降低,随后显著升高,至Ⅳ晚期达到峰值后逐渐下降;肝脏Vtg mRNA表达水平自Ⅳ早期显著升高,在Ⅳ期中达峰值,随后下降至Ⅱ期、Ⅲ期水平。相关性分析表明,下丘脑GnRH mRNA表达水平与GSI呈正相关;与FSH、LH mRNA的表达水平呈负相关,而垂体FSH与LH mRNA之间则呈现显著正相关(P<0.05)。血清中E_2与T含量变化趋势相似,均与卵母细胞同步发展。GnRH、FSH与LH变化的规律性确保了HPG轴对Vtg、性类固醇激素(E_2与T)等合成与分泌的调控。本研究可为系统、科学地开展哲罗鲑人工繁育与健康养殖提供理论依据。 展开更多

将孵出30d、平均体质量0.043g的黑斑原(鱼兆)Glyptosternum maculatum仔鱼常规饲养在4个循环水养殖系统(长1.8m×宽0.6m×高0.6m)中,水温保持在9℃、12℃、15℃和18℃下,每个温度组设置3个平行,研究不同温度对黑斑原(鱼兆)仔鱼... 将孵出30d、平均体质量0.043g的黑斑原(鱼兆)Glyptosternum maculatum仔鱼常规饲养在4个循环水养殖系统(长1.8m×宽0.6m×高0.6m)中,水温保持在9℃、12℃、15℃和18℃下,每个温度组设置3个平行,研究不同温度对黑斑原(鱼兆)仔鱼生长及存活率的影响。35d的饲养结果表明:随着温度的升高,黑斑原(鱼兆)仔鱼的生长加快,死亡率增加,其中18℃组黑斑原(鱼兆)最早开始死亡,9℃组黑斑原(鱼兆)存活率最高。在整个饲养期间,水温9℃组与12℃组黑斑原(鱼兆)全长和体质量生长最快,死亡率最低,显著高于其他组(P<0.05)。因此,9~12℃为黑斑原(鱼兆)仔稚鱼的适宜培育温度。 展开更多

为了将传染性胰脏坏死病毒(Infectious pancreatic necrosis virous,IPNV)的主要保护性抗原及IPNV检测和疫苗开发的主要靶基因——VP2蛋白展示于酵母细胞表面,本研究中基于IPNV VP 2基因序列设计引物,以IPNV ChRtm213的RNA为模板进行PC... 为了将传染性胰脏坏死病毒(Infectious pancreatic necrosis virous,IPNV)的主要保护性抗原及IPNV检测和疫苗开发的主要靶基因——VP2蛋白展示于酵母细胞表面,本研究中基于IPNV VP 2基因序列设计引物,以IPNV ChRtm213的RNA为模板进行PCR扩增,然后将扩增产物连接到酵母表面展示载体pYD1上构建了重组质粒pYD1-VP 2,将重组质粒转化至酵母EBY100感受态细胞中,得到含有重组质粒的酵母菌EBY100/pYD1-VP2,再向EBY100/pYD1-VP2中加入半乳糖进行VP2蛋白的诱导表达,并采用蛋白免疫印迹和细胞免疫荧光技术对VP2蛋白的酵母表面展示情况进行了分析。结果表明:经半乳糖诱导后VP2蛋白在酵母细胞表面获得了成功表达;细胞免疫荧光分析显示,重组酵母诱导表达后出现特异性绿色荧光,并且在一定时间内荧光强度与诱导时间成正相关,诱导24、36、48 h组及对照组(48 h)各组之间荧光酵母比例有显著性差异(P<0.05)。研究表明,IPNV VP2蛋白已经被酵母细胞高效表达并且成功展示于酵母细胞表面,本研究结果可为IPNV口服活载体疫苗的研制奠定基础。 展开更多

糖蛋白(Glycoprotein,G)作为鲤春病毒血症病毒(Spring Virernia of Carp Virus,SVCV)主要的抗原蛋白,已成为现阶段SVCV病毒检测、抗体制备以及疫苗研制的热点。为了对其进行酵母表面展示,研究以SVCVshlj1分离株基因组为模板,通过RT-PCR... 糖蛋白(Glycoprotein,G)作为鲤春病毒血症病毒(Spring Virernia of Carp Virus,SVCV)主要的抗原蛋白,已成为现阶段SVCV病毒检测、抗体制备以及疫苗研制的热点。为了对其进行酵母表面展示,研究以SVCVshlj1分离株基因组为模板,通过RT-PCR技术,体外扩增获得SVCV表面糖蛋白的基因开放阅读框(1530 bp)片段,将其克隆至酵母表面展示载体pYD1,构建重组质粒pYD1-G。利用电转化方法将重组质粒pYD1-G导入酿酒酵母EBY100感受态细胞,经YNB选择培养基筛选和菌液PCR的鉴定,挑选出阳性转化子(命名为EBY100-pYD1-G),对其进行2%半乳糖诱导。利用细胞免疫荧光和流式细胞仪检测G蛋白的酵母表面展示情况。细胞免疫荧光结果显示,诱导后的酵母细胞EBY100-pYD1-G能产生特异性红色荧光,且随着诱导时间的增加,红色荧光的酵母细胞所占比例不断增加,各组之间差异显著(P<0.05)。流式细胞仪检测结果显示,酵母细胞的荧光强度与诱导时间呈正比,其中诱导48h与72h的酵母细胞荧光强度不存在显著差异,基本趋于稳定不变的状态。因此,选取诱导48h为酵母表面展示的最佳诱导时间。上述研究结果表明SVCV的G蛋白已经成功展示于酿酒酵母细胞表面,研究为鲤春病毒血症酵母口服疫苗的研发奠定了前期基础。 展开更多

The Effects of Compound Chinese Medicine Immune Additives on Growth Performance and Antioxidant Capacity of Acipenser schrenckii 被引量:4

[ Objective] The aim was to explore the effects of compound Chinese medicine on growth and antioxidant capacity of Acipenser schrenckii, and to provide a foundation for the application of traditional Chinese medicine as immune additives in sturgeon aquaculture production. [Methods] Three kinds of compound Chinese medicines (respectively called A, B and C) were added into basic material feed as feed additives, continuous fed 1 ^+ age Acipenser schrenckii for 35 d, then weight gain rate was determined and superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, total antioxidant capacity (T-AOC) and glutathione peroxidase (GSH-PX) activity in serum and liver were detected. [ Results ] Chinese medicine formulae could increase the superoxide dismutase (SOD) activity, total antioxidant capacity (ToAOC) and glutathione per- oxidase (GSH-PX) in serum and liver of sturgeon. Especially SOD enzyme activity of serum in the three formulae groups was significantly higher ( P 〈 0.05) than control group, SOD enzyme activities of fish liver in group A and C were significantly higher ( P 〈 0.05) than control group; T-AOC activities of serum and liver in the three formulae groups were both incressed, but had no significant differences with that of the control group; only GSH-PX activities of fish serum in group B and C had significant differences (P〈0.05) with that of control group, GSH-PX activities of liver in the three formulae groups had no significant differences with that of control group; and traditional Chinese medicine could significantly reduce malondialdehyde (MDA) content of sturgeon serum, MDA contents of serum and liver in three formulae groups had significant differences (P 〈 0.05) with that of control group. [ Conclusion] The selected three kinds of compound Chinese medicine additives had different degree of ability to improve sturgeon antioxidant function. 展开更多

Molecular Typing of Aeromonas hydrophila Isolated from Common Carp in Northeast China

A total of 59 isolates of Aeromonas hydrophila were collected from common carp suffering from freshwater fish hemorrhage disease in 13 fishing grounds in the northeast China, and their phenotypic and genetic characteristics were investigated. All of the isolates were identified as A. hydrophila by traditional biochemical method and yielded a 686-bp DNA fragment of the 16S rDNA gene in the PCR experiments. Collected strains were also evaluated for their susceptibility to 17 different antibiotics. The isolates showed an even trend of the resistance and sensitivity to drugs, highly sensitive to antibiotics, such as Levofloxacin, PolymyxinB, Ofloxacin and resistant to antibiotics, such as Bristopen, Lincomycin, Ampicillin, Teicoplanin. Evaluation of genetic diversity was performed on all isolates by molecular typing with enterobacterial repetitive intergenic consensus (ERIC-PCR) method. The results showed that three different types, i.e. type I, ]I, and type ~I, were found in 59 isolates and type III accounted for a large proportion of 54.84%. There was no dominant difference between the tendency of the isolates of Heilongjiang Province and Jilin Province in these three types, which showed DI 〉 I 〉 I, while the isolates of Liaoning Province showed III 〉 II 〉 I. The percentage of different types in different provinces varied in each other; however, they didn't show any obvious regional or cluster-specific branches. In conclusion, the ability to distinguish Aeromonas hydrophila strains from diseased common carp with ERIC-PCR would be useful for epidemiological investigation and population genetic analysis of this pathogen in China. 展开更多

The purpose of this study was to establish a method for the rapid detection of infectious pancreatic necrosis virus(IPNV,Jasper serotype)using reverse transcription loop-mediated isothermal amplification(RT-LAMP).Four groups of specific primers were designed,according to the genome sequence of a Chinese IPNV isolate ChRtm213.The results showed that primer set B2 had the best amplification effect.When the final concentration of Mg2+was 6 mmol·L-1,dNTPs were 1 mmol·L-1 and betaine was 0.4 mol·L-1,the reaction could be completed in a 63℃water bath within 60 min.This RT-LAMP assay for the detection of IPNV had no cross-reactivity with infectious hematopoietic necrosis virus,viral hemorrhagic septicemia virus,grass carp reovirus and spring viremia of carp virus.The detection limit was 3.2×10-12 ng·μL-1.The sensitivity of this method was 10-fold higher than that of a previously published RT-LAMP assay for detecting the Spajarup(Sp)serotype of IPNV.This method,aimed at detecting IPNV isolates that were currently prevalent in China,possessed the characteristics of strong specificity,high sensitivity and direct interpretation by the naked eyes.The IPNV RT-LAMP was successfully applied to determine the clinical samples,which indicated the IPNV RT-LAMP assay was suitable for the rapid and large-scale detections of IPNV in China. 展开更多
