[目的]工作频率3.7 GHz的TH2103A型高功率速调管广泛应用在托卡马克实验装置中。为了简化高压供电系统(去掉阳极调制中的电真空四极管),在HL-2A装置的TH2103A型速调管中引入了一种基于分压模式的供电方式。为了保证速调管在该供电模式下安全运行,研制了阳极电流的测量和保护系统。[方法]对该供电模式下的阳极电流特点进行了分析:阳极电流会出现正常、正向过流或反向过流的情况。当阳极电流出现正向过流或反向过流时,代表有大量的电子轰击阳极或大量电子从阳极流出,这些现象对速调管来说都是非常危险的,通常会引起速调管钛泵过流,甚至损坏速调管。因此对阳极电流进行准确的测量以及当阳极电流出现过流时切断高压对速调管进行保护非常重要。文章介绍了阳极电流测量系统的原理和供电模式,阳极保护系统的原理和实施方案,在实验过程中对阳极电流的测量和保护系统的可靠性进行了验证。[结果]证明了该供电模式下阳极电流测量和保护系统的有效性和可靠性。[结论]文章所述供电模式和阳极测量保护系统对基于TH2103A型速调管的运行和维护具有重要参考意义。 展开更多

近年来小扰动振荡事故频发,亟需建立能反映新能源并网系统全工况的在线稳定域。小扰动在线稳定域可以为时变性和波动性下新型电力系统的安全评估和控制优化提供强有力的支撑。因此,提出基于分段仿射阻抗模型的电压源变流器(voltage sour... 近年来小扰动振荡事故频发,亟需建立能反映新能源并网系统全工况的在线稳定域。小扰动在线稳定域可以为时变性和波动性下新型电力系统的安全评估和控制优化提供强有力的支撑。因此,提出基于分段仿射阻抗模型的电压源变流器(voltage source converter,VSC)小扰动稳定域在线构造方法。首先,该方法使用电压、功率和频域因子作为分区变量,离线构建VSC的分段仿射阻抗模型。在线计算时根据运行工况可以快速定位运行分区并确定对应的线性仿射模型,从而极大地提高阻抗计算的效率。其次,提出VSC稳定域边界点的双层优化模型,利用改进的广义奈奎斯特判据和复合形法进行快速求解。然后,通过改变双层优化模型的求解方向搜索边界点,基于分段拟合获取准确的边界解析式。最后,仿真算例和实验结果表明,分段仿射阻抗模型与全阶阻抗模型误差小于1%;该方法可以准确和高效地构建在线稳定域。 展开更多

This study examines the impacts of short-term(6 months) fertilization on the community structure and abundance of ammonia-oxidizing betaproteobacteria(β-AOB) and the potential nitrification rate in sediment colonized by S uaeda heteroptera in a saltmarsh located in Shuangtai estuary, China. The sediment samples were collected from plots treated with different amounts of an N fertilizer(urea supplied at 0.1, 0.2, 0.4, and 0.8 g/kg(nitrogen content in dry sediment)), and with different forms of N fertilizers(urea,(NH4) 2 SO 4, and NH_4NO_3, each supplied at 0.2 g/kg). The fertilizers were applied 1–4 times during the plant-growing season in May, July, August and September of 2013. Untreated plots were included as a control. As revealed in denaturing gradient gel electrophoresis of the 16 S r RNA gene, the β-AOB community responded to both the amount and form of N. Real-time quantitative PCR indicated that both abundance and potential nitrification rate of β-AOB increased after N addition, regardless of concentration and form(except NH_4NO_3). These results provide evidence that short-term N application influences the sediment β-AOB community, β-AOB abundance and potential nitrification rate in a saltmarsh ecosystem. 展开更多

The effects of nitrogen (N) addition on microbial biomass, bacterial abundance, and community composition in sediment colonized by Suaeda heteroptera were examined by chloroform fumigation extraction method, real-time quantitative polymerase chain reaction, and denaturing gradient gel electrophoresis (DGGE) in a salt marsh located in Shuangtai Estuary, China. The sediment samples were collected from plots treated with different amounts of a single N fertilizer (urea supplied at 0.1, 0.2, 0.4 and 0.8 g/kg (nitrogen content in sediment) and different forms of N fertilizers (urea, (NH4)2SO4, and NH4NO3, each supplied at 0.2 g/kg (calculated by nitrogen). The fertilizers were applied 1-4 times during the plant-growing season in May, luly, August, and September of 2013. Untreated plots were included as a control. The results showed that both the amount and form of N positively influenced microbial biomass carbon, microbial biomass nitrogen, and bacterial abundance. The DGGE profiles revealed that the bacterial community composition was also affected by the amount and form of N. Thus, our findings indicate that short-term N amendment increases microbial biomass and bacterial abundance, and alters the structure of bacterial community. 展开更多

为提高现有组合型孔式排种器对不同品种的播量适应性,满足不同地区、不同品种的播种量要求。该文在此排种器的基础上进行优化设计和试验研究。以瓢形形状可变容积型孔的排种轮结构为主体,对型孔的形状、最大截面、深度和体积进行数学模... 为提高现有组合型孔式排种器对不同品种的播量适应性,满足不同地区、不同品种的播种量要求。该文在此排种器的基础上进行优化设计和试验研究。以瓢形形状可变容积型孔的排种轮结构为主体,对型孔的形状、最大截面、深度和体积进行数学模型计算,确定在最小、中间、最大合成型孔位置的长轴尺寸分别为9.2、12.4和15.6 mm,截面积范围为71.31~154.58 mm^2,型孔容积大小调节范围为271.91~485.79 mm^3;在排种轮内部安装步进电机和控制电路,通过蓝牙控制每穴播量在3~10粒之间无级调节。选用常规粳稻秀水134、杂交粳稻花优14、常规籼稻黄华占和杂交籼稻晶两优1212四种具有代表性的直播稻品种,对无级调节型孔进行三因素四水平试验,试验结果表明:粳稻每穴粒数调节范围为5~10粒,籼稻每穴粒数调节范围为3~8粒,当型孔容积调节变化时,每穴排种粒数变化趋势明显,从而能达到播量无级调节的效果,满足不同品种每穴播种量3~10粒的设计目标;分析结果表明:水稻品种、型孔容积和排种轮转速3个因素对播量影响主次为型孔容积、水稻品种、排种轮转速。该文设计的播量无级调节水稻精量排种装置实现了不同水稻品种的播量无级调节作业,调节范围大,适应性较好,无级调节简便快速,具有较好的实际实用价值。 展开更多

基于超高效液相色谱-质谱法的肽段分析中非特异性吸附评估及通用型最小化策略

蛋白质组学技术在多肽和蛋白质类新型治疗药物的开发、临床诊断生物标志物的深入发掘中应用广泛。然而,多肽和蛋白质类大分子的非特异性吸附性质给分析方法的开发带来极大挑战,亟须一种通用型的策略去评估和降低非特异吸附对超高效液相... 蛋白质组学技术在多肽和蛋白质类新型治疗药物的开发、临床诊断生物标志物的深入发掘中应用广泛。然而,多肽和蛋白质类大分子的非特异性吸附性质给分析方法的开发带来极大挑战,亟须一种通用型的策略去评估和降低非特异吸附对超高效液相色谱-质谱(UPLC-MS)大分子检测造成的负面影响。研究以牛血清白蛋白(BSA)为模型,探讨其酶解后多肽组理化性质与吸附程度之间的相关性;根据肽段的响应和吸附程度设计分级策略;针对高响应、强吸附的ClassⅡ类肽段,从样品制备中离心管、进样瓶的选择,乃至液相色谱系统中色谱柱固定相、流速、梯度、柱温、洗针液的选择全过程设计试验,探讨非特异吸附的影响因素及其通用型最小化策略。结果显示,肽段的被吸附程度与其理化参数HPLC指数(HPLC Index)、肽段长度等显著相关(p<0.05),但仅凭上述参数仅能解释30%肽段的被吸附程度。改性的聚丙烯材料可使肽段溶液在储存或前处理过程中获得较高的回收率(24 h内回收率大于80%)。在对液相色谱条件的考察和优化过程中发现,C_(8)填料的色谱柱、高流速、缓梯度以及强洗针液,可使残留量降至最低(降低为原来的1/150)。柱温对残留的影响在肽段间存在较大个体差异,需要对不同的肽段具体分析以得到较少量的残留。研究以详实的数据考察并最小化模型肽段组在分析过程中的非特异吸附,提示了蛋白质类大分子药物分析方法建立中应重点关注的影响因素及其有效的解决方案。 展开更多

北斗星基增强(BDSBAS)系统播发格网电离层改正数和格网电离层完好性参数GIVE,用以提升GNSS系统的服务精度并实现区域电离层活动完好性监视,以满足精密进近(GLS PA)需求。本文在实现BDSBAS格网电离层粗差剔除与改正数计算的基础上,提出... 北斗星基增强(BDSBAS)系统播发格网电离层改正数和格网电离层完好性参数GIVE,用以提升GNSS系统的服务精度并实现区域电离层活动完好性监视,以满足精密进近(GLS PA)需求。本文在实现BDSBAS格网电离层粗差剔除与改正数计算的基础上,提出了一种电离层完好性参数GIVE的优化方法,进而评估了BDSBAS格网电离层的应用精度。BDSBAS格网电离层格网点延迟估计采用平面拟合算法计算,异常数据剔除采用稳健的中值容错算法,GIVE的估计考虑了电离层残差分布的偏度与峰度统计特性,能够实现对电离层异常活动的及时响应。2020年1月实测数据分析结果表明,BDSBAS格网电离层修正精度(RMSE)为2~3 TECU,改正百分比达到75%~79%,GIVE包络率优于99.9%。修正格网电离层后可提升GPS定位精度20%~40%。 展开更多

为分析比较不同来源低聚肽中呈味有机酸含量的差别,建立了可以同时测定鸡肉低聚肽粉和大豆低聚肽粉中4种呈味有机酸含量的高效液相色谱方法.色谱条件:Zorbax Eclipse XDB C18色谱柱,柱温25℃,流动相为体积比5﹕95的甲醇与磷酸二氢钾溶液... 为分析比较不同来源低聚肽中呈味有机酸含量的差别,建立了可以同时测定鸡肉低聚肽粉和大豆低聚肽粉中4种呈味有机酸含量的高效液相色谱方法.色谱条件:Zorbax Eclipse XDB C18色谱柱,柱温25℃,流动相为体积比5﹕95的甲醇与磷酸二氢钾溶液(0.01 mol/L,pH 2.8)的混合溶液,流量1 mL/min,进样量5μL,等度洗脱,紫外检测器检测波长210 nm.在此条件下,苹果酸、乳酸、琥珀酸、柠檬酸这4种呈味有机酸可以进行有效分离,在0.01~5.00 mg/mL线性范围内,相关系数为0.99941~1.00000,检出限为0.4~0.9μg/mL,定量限为2.9~5.4μg/mL,加标回收率为86.25%~94.36%,且相对标准偏差(RSD)均小于5%.结果表明:鸡肉低聚肽粉中乳酸含量最高(4.32%),不含柠檬酸,而大豆低聚肽粉中柠檬酸(1.91%)则是含量最高的呈味有机酸.该高效液相色谱检测方法高效快捷、定量准确、灵敏度高,适用于不同来源低聚肽中4种不同呈味有机酸含量的测定. 展开更多

Bacterial community presumably plays an essential role in inhibiting pathogen colonization and maintaining the health of scallop larvae, but limiting data are available for Yesso scallop(Patinopecten yessoensisis Jay, 1857) larval development stages. The aim of this study was to characterize and compare the bacterial communities associating with Yesso scallop larval development at fertilized egg S1, trochophora S2, D-shaped larvae S3, umbo larvae S4, and juvenile scallop S5 stages by Illumina high-throughput sequencing. Genomic DNA was extracted from the larvae and their associating bactera, and a gene segment covering V3-V4 region of 16 S r RNA gene was amplified and sequenced using an Illumina Miseq sequencer. Overall, 106760 qualified sequences with an average length of 449 bp were obtained. Sequences were compared with those retrieved from 16 S r RNA gene databases, and 4 phyla, 7 classes, 15 orders, 21 families, 31 genera were identified. Proteobacteria was predominant phylum, accounting for more than 99%, at all 5 larval development stages. At genus level, Pseudomonas was dominant at stages S1(80.60%), S2(87.77%) and S5(68.71%), followed by Photobacterium(17.06%) and Aeromonas(1.64%) at stage S1, Serratia(6.94%), Stenotrophomonas(3.08%) and Acinetobacter(1.2%) at stage S2, Shewanella(25.95%) and Pseudoalteromonas(4.57%) at stage S5. Moreover, genus Pseudoalteromonas became dominant at stages S3(44.85%) and S4(56.02%), followed by Photobacterium(29.82%), Pseudomonas(11.86%), Aliivibrio(8.60%) and Shewanella(3.39%) at stage S3, Pseudomonas(18.16%), Aliivibrio(14.29%), Shewanella(4.11%), Psychromonas(4.04%) and Psychrobacter(1.81%) at stage S4. From the results, we concluded that the bacterial community changed significantly at different development stages of Yesso Scallop larvae. 展开更多

A marine bacterium, Pseudoalteromonas sp. BC228 was supplemented to feed in a feeding experiment aiming to determine its ability of enhancing the digestive enzyme activity and immune response of juvenile Apostichopus japonicus. Sea cucumber individuals were fed with the diets containing 0(control), 105, 107 and 109 CFU g-1 diet of BC228 for 45 days. Results showed that intestinal trypsin and lipase activities were significantly enhanced by 107 and 109 CFU g-1 diet of BC228 in comparison with control(P < 0.01). The phagocytic activity in the coelomocytes of sea cucumber fed the diet supplemented with 107 CFU g-1 diet of BC228 was significantly higher than that of those fed control diet(P < 0.05). In addition, 105 and 107 CFU g-1 diet of BC228 significantly enhanced lysozyme and phenoloxidase activities in the coelomic fluid of sea cucumber, respectively, in comparison with other diets(P < 0.01). Sea cucumbers, 10 each diet, were challenged with Vibrio splendidus NB13 after 45 days of feeding. It was found that the cumulative incidence and mortality of sea cucumber fed with BC228 containing diets were lower than those of animals fed control diet. Our findings evidenced that BC228 supplemented in diets improved the digestive enzyme activity of juvenile sea cucumber, stimulated its immune response and enhanced its resistance to the infection of V. splendidus. 展开更多

Protein kinase C (PKC) has a crucial role in signal transduction for a variety of biologically active substances which activate cellular functions and proliferation. We previously isolated the full-length PKC gene from Dunaliella salina (DsPKC) using rapid amplification of cDNA ends (RACE) and RT-PCR methods. And we submitted the mRNA sequence of DsPKC gene to NCBI (Genbank No. JN625213). In the present paper, the DsPKC gene open reading frame obtained by PCR was cloned into pGS-21a vector and transformed into Escherichia coli to generate the fusion protein. Bioinformatics analysis revealed that DsPKC gene was a member of serine/threonine kinase with two conserved domains and highly conserved motifs. The DsPKC was highly expressed upon induction with isopropyl-β-d-thiogalactoside (IPTG) at a final concentration of 0.2 mmol L 1 at 37℃. Under salt stress, the fu- sion protein Green Fluorescent Protein (GFP)-DsPKC was transferred from the cytoplasm to the cell membrane. The expression pat- tern of DsPKC gene was analyzed using real-time quantitative PCR, and indicated that DsPKC gene was up-regulated by 3.0 mol L 1 NaCl at 12 h, which was significantly higher than in control values (P < 0.05). These results suggest that the DsPKC gene plays an important role in response to salt stress in D. salina. 展开更多

We used Illumina high-throughput sequencing of PCR-amplified V3-V4 16 S rRNA gene regions to characterize bacterial communities associated with the adductor muscles, gills, gonads and intestines of the Yesso scallop(Patinopecten yessoensis) from waters around Zhangzidao, Dalian, China. Overall, 421,276 optimized reads were classified as 25 described bacterial phyla and 308 genera. Firmicutes, Proteobacteria, Tenericutes, Bacteroidetes, Chlamydiae and Spirochaetae accounted for > 97% of the total reads in the four organs. The bacterial 16 S rDNA sequences assigned to Firmicutes and Proteobacteria were abundant in the adductor muscles, gills and gonads; while reads from Tenericutes were dominant in the intestines, followed by those from Firmicutes, Chlamydiae, Proteobacteria and Bacteroidetes. At the genus level, the dominant genera in the adductor muscles, gills and gonads appeared to be Bacillus, Enterococcus and Lactococcus, whereas Mycoplasma was dominant in the intestines. The relative abundances of Bacillus, Enterococcus, Lactococcus, Alkaliphilus, Raoultella, Paenibacillus and Oceanobacillus were significantly lower in the intestine than in the other three organs. Cluster analysis and principal coordinates analysis of the operational taxonomy units profile revealed significant differences in the bacterial community structure between the intestine and the other three organs. Taken together, these results suggest that scallops have intestine-specific bacterial communities and the adductor muscles, gills and gonads harbor similar communities. The difference in the bacterial community between organs may relate to unique habitats, surroundings, diet and their respective physiological functions. 展开更多

Viable cell count was used to determine whether Metschnikowia sp.C14 can colonize the intestine of juvenile sea cucumber Apostichopus japonicus.Sea cucumber individuals were divided into two groups,which were fed the control diet for 38 days or the C14-supplemented diet at 105 cells g−1 diet for 28 days,then the control diet from day 29 to day 38.The number of C14 cells in the intestine of sea cucumber fed the C14-supplemented diet significantly increased from day 7 to day 28,and decreased from day 29 to day 38.Sea cucumber fed with the diet containing C14 showed a significant increase in trypsin activity and lipase activity from day 21 to day 33 compared with the control.Feeding C14 significantly improved the phagocytic activity and respiratory burst in coelomocytes from day 21 to day 35 and from day 14 to day 38,respectively.In addition,there was an obvious enhancement in lysozyme activity(from day 21 to day 38 or day 33),phenoloxidase activity(from day 21 to day 28)and total nitric oxide synthase activity(from day 14 to day 38)in coelomic fluid supernatant and/or coelomocyte cell lysate supernatant compared with the control.There were significant positive correlations between the number of C14 cells colonizing the intestine and trypsin activity of the intestine,lysozyme activity of the coelomic fluid supernatant and coelomocyte lysate supernatant from sea cucumber.These data suggested that the number of C14 cells should be maintained at 105 cfu(colony-forming units)g−1 intestine material for the maximum benefit. 展开更多

We investigated the changes in communities of bacteria,ammonia-oxidizing bacteria,and Nitrospira during the operation of a pufferfish Takifugu rubripes recirculating aquaculture system by using high-throughput DNA sequencing.Differences in bacterial communities were observed at days 1-32,47-62 and 78-93 of biofilm development by using 16S rRNA gene pyrosequencing.The relative abundance of Proteobacteria(Gammaproteobacteria)increased,while that of Bacteroidetes(Flavobacteria)decreased.The proportions of Nitrosomonas and Nitrospina ranged from 0.02%to 0.30%and from 0.02%to 0.83%,respectively.Ammonia monooxygenase gene pyrosequencing revealed that the top three operational taxonomic units were related to Nitrosomonas aestuarii(17.5%-61.1%),uncultured beta proteobacterium clone B67S-54(1.9%-45.2%),and uncultured bacterium clone AZPa8(3.6%-24.7%).Nitrite oxidoreductase gene pyrosequencing revealed that the relative abundance of the dominant strain Nitrospira sp.Ecomares 2.1 increased,but that of the abundant species Nitrospira marina decreased.Our results demonstrated that the communities of bacteria,ammonia-oxidizing bacteria,and Nitrospira were changing during the operation of the pufferfish recirculating aquaculture system. 展开更多

目的探索分化型甲状腺癌(DTC)发生肺转移的危险因素及其对131I的疗效。方法纳入我院2013年1月至2019年6月收治入院的1854例确诊为DTC的患者其中肺转移45例,根据患者年龄、性别、病理类型、甲状腺炎、被膜侵犯、有无甲状腺外组织侵犯筛选... 目的探索分化型甲状腺癌(DTC)发生肺转移的危险因素及其对131I的疗效。方法纳入我院2013年1月至2019年6月收治入院的1854例确诊为DTC的患者其中肺转移45例,根据患者年龄、性别、病理类型、甲状腺炎、被膜侵犯、有无甲状腺外组织侵犯筛选出DTC肺转移的危险因素。并根据131I治疗效果,将45例肺转移分为有效和无效两组,比较两组年龄、性别、病理类型、手术方式、显影时机方面存在的差别,分析影响131I治疗DTC肺转移疗效的因素。结果131I治疗DTC伴肺转移有效率为55.56%(25/45)。单因素分析显示DTC病理类型滤泡癌、甲状腺外组织侵犯发生肺转移的风险显著增加(χ^2=41.388,11.9112,P均<0.05);年龄、性别、甲状腺炎对DTC发生肺转移无影响(χ^2=2.65,0.074,0.8290.118,P均>0.05);多因素logistic回归分析得病理类型、甲状腺外组织侵犯是DTC肺转移的独立影响因素(P=0.0000.001,OR=8.409,2.792);进一步对DTC肺转移的患者131I的疗效分析显示,年龄≥55岁患者,131I无效的比例显著高于年龄<55岁组(χ^2=6.252,P=0.012);而显影时机分析显示,首次治疗发现肺转移的患者有效比例为92%,显著高于随访发现肺转移患者比例8%,(χ^2=6.354,P=0.012)。多因素logistic回归分析得年龄、显影时机是131I疗效的独立影响因素。结论滤泡状癌、甲状腺外组织侵犯会增加DTC肺转移的风险且高龄、随访时发现肺转移的患者131I治疗疗效不佳。 展开更多
