Ultrasonography has been proven to be efficient in the clinic to diagnose peripheral nerve injury. The aim of this study was to assess the role of ultrasound in the evaluation of persistent peripheral nerve injuries 1 year after the Wenchuan earthquake. Thirty-four patients with persistent clinical symptoms and neurologic signs of impaired nerve function were evaluated with sonography prior to surgi- cal repair. Among 34 patients, ultrasonography showed that 48 peripheral nerves were entrapped, and 11 peripheral nerves were disrupted. There was one case of misdiagnosis on ultrasonogra- phy. The concordance rate of ultrasonographic findings with those of surgical findings was 98%. A total of 48 involved nerves underwent neurolysis and the symptoms resolved. Only five nerves had scar tissue entrapment. Preoperative and postoperative clinical and ultrasonographic results were concordant, which verified that ultrasonography is useful for preoperative diagnosis and postoperative evaluation of injured peripheral nerves.展开更多
Metal-organic frameworks(MOFs)can serve as prevailing anodes for lithium-ion batteries,due to their multiple redox-active sites and prominent structural compatibility.However,the poor electronic conductivity and infer...Metal-organic frameworks(MOFs)can serve as prevailing anodes for lithium-ion batteries,due to their multiple redox-active sites and prominent structural compatibility.However,the poor electronic conductivity and inferior cyclability hinder their further implementation.Herein,a synthetic methodology for trimetallic Fe-Co-Ni MOFs with nanoframe superstructures architecture(Fe-Co-Ni NFSs)via structural evolution is proposed for versatile anode materials for lithium storage.Ascribed to optimal compositional and structural optimization,the Fe-Co-Ni NFSs achieve exceptional electrochemical performance with superior specific capacity(1030 mAh g^(−1) at 0.1 A g^(−1)),outstanding rate capacity(414 mAh g^(−1) at 2 A g^(−1)),and prolonged cyclability(489 mAh g^(−1) upon 1000 cycles at 1 A g^(−1)).Both experimental and theoretical investigations reveal that the multi-component metal centers could boost electronic conductivity,confer multiple active sites,and energetically favor Li adsorption capability.Additionally,the nanoframe superstructures of Fe-Co-Ni NFSs could facilitate stress-buffering effect on volumetric expansion and prevent electrode pulverization,further improving the lithium storage capability.This work envisions a meticulous protocol for high-performance MOF anode materials for lithium-ion batteries.展开更多

BACKGROUND Diabetes is often associated with gastrointestinal dysfunctions,which can lead to hypoglycemia.Dexmedetomidine(DEX)is a commonly used sedative in perioperative diabetic patients and may affect gastrointesti...BACKGROUND Diabetes is often associated with gastrointestinal dysfunctions,which can lead to hypoglycemia.Dexmedetomidine(DEX)is a commonly used sedative in perioperative diabetic patients and may affect gastrointestinal function.AIM To investigate whether sedative doses of DEX alleviate diabetes-caused intestinal dysfunction.METHODS Sedation/anesthesia scores and vital signs of streptozotocin(STZ)-induced diabetic mice under DEX sedation were observed.Diabetic mice were divided into saline and DEX groups.After injecting sedatives intraperitoneally,tight junctions(TJs)and apoptotic levels were evaluated 24 hours later to assess the intestinal barrier function.The role of DEX was validated using Villin-MMP23B flox/flox mice with intestinal epithelial deletion.In vitro,high glucose and hyperosmolarity were used to culture Caco-2 monolayer cells with STZ intervention.Immunofluorescence techniques were used to monitor the barrier and mitochondrial functions.RESULTS MMP23B protein levels in the intestinal tissue of STZ-induced diabetic mice were significantly higher than those in the intestinal tissue of control mice,with the DEX group displaying decreased MMP23B levels.Diabetes-mediated TJ disruption,increased intestinal mucosal permeability,and systemic inflammation in wild-type mice might be reversed by DEX.In Caco-2 cells,MMP23B was associated with increased reactive oxygen species accumulation,mitochondrial membrane potential depolarization,and TJ disruption.CONCLUSION DEX reduces MMP23B,which may potentially contribute to STZ-induced intestinal barrier dysfunction,affecting TJ modification through mitochondrial dysfunction.展开更多

Little is known until now about behavioral characteri...Anas poecilorhyncha is one of improved variety of mallards, which was the protected bird species listed by International Union for Conservation of Nature (1UCN). Little is known until now about behavioral characteristics of the mallard. The objective of this study was to compare the behavioral differentials between A. poecilorhyncha and domestic duck in order to make a strategy to manage A. poecilorhyncha under the condition of animal welfare. A total of 180 birds were distributed into six groups with 30 birds for each group. They were placed in a room of 3 m ~ 4 m and fed for one year old. Sansui ducks, a kind of native domestic duck (Sansui laying duck) were used for behavioral comparison. Two different rooms in the same building were applied, one room for A. poecilorhyncha and another one for Sansui ducks. All behaviors for A. poecilorhyncha and Sansui ducks were coded using the program The Observer XT 11.5 (Noldus Information Technology, Beijing). The duration of observation was from 8:00 am to 18:00 pm daily and lasted 5 d. The results showed that there were similar behavioral percentages between A. poecilorhyncha and domestic duck. Percentages of standing activity spent for A. poecilorhyncha and Sansui duck were 34.59% and 30.25%, respectively. Accordingly, the activities, such as wing plugging, preening and head stretching, were more than 5.51%. The other activities, including walking, drinking and tail wagging, occupied less proportions (〈 3%). While the specific behaviors, like aggression, alerting, wing dithering, clawing, nodding, pendulum clawing and crawling, took less than 1% of percentage. There were large differences between A. poecilorhyncha and domestic duck when comparing eight behavioral peaks. Lag sequential analysis was used to calculate frequency of transition between a pair of activities. Some parameters were very significant, like the frequency value from foraging converted to drinking in Sansui duck was 369-515, but 37-65 in A. poecilorhyncha. The best explaining could be given that Sansui duck was better domesticated than spot-billed duck. This study provides the basic data to study and develop spot-billed duck.展开更多

In contra...Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. Further evaluation showed that p SM3-CAR effectively caused tumor necrosis, providing new options for improved CAR-T therapy in solid tumors.展开更多
Recognition of CpG dinucleotide DNA in epigenetic information flow plays a pivotal role for cellular differentiation and development.The TET3 CXXC domain binds to CpG DNA,serving a basic epigenetic information reading...Recognition of CpG dinucleotide DNA in epigenetic information flow plays a pivotal role for cellular differentiation and development.The TET3 CXXC domain binds to CpG DNA,serving a basic epigenetic information reading mechanism.During the selective recognition of a CpG motif by a CXXC domain from crowded binding sites in a gene sequence,the protein-DNA interactions are beyond CpG dinu-cleotide.However,the selective binding dynamics of CpG within a long DNA context by epigenetic enzymes have been rarely exploit-ed,which is hard for ensemble methods to probe.Here,we used single-molecule magnetic tweezers to quantitatively examine the dynamics of TET3's CXXC domain on a Hoxa9 promoter DNA.Our single-molecule binding profile revealed that CXXC-DNA interactions involve both CpG motifs and their flanking sequences.The residence time of TET3 CXXC differs by about 1000 times in five distin-guished CpG clusters in the context of a CpG island.Moreover,we performed multi-state hidden Markov modeling analysis on the zip-ping/unzipping dynamics of a CpG hairpin,discovering TET3 CXXC's preference on CpG motifs regarding the-2 to+2 flanking bases.Our results shed light on the selective binding dynamics of a CXXC on a gene sequence,facilitating studies on epigenetic information reading mechanisms.展开更多
ObjectiveTo elucidate the mechanism underlying the inhibitory effect of acupotomy on chondrocyte apoptosis in rabbits with knee osteoarthritis (KOA) through screening, analysis, and verification of apoptosis-related d...ObjectiveTo elucidate the mechanism underlying the inhibitory effect of acupotomy on chondrocyte apoptosis in rabbits with knee osteoarthritis (KOA) through screening, analysis, and verification of apoptosis-related differentially expressed proteins via proteomics.MethodsThirty New Zealand white rabbits were randomly divided into the normal, model, and acupotomy groups, with 10 rabbits in each group. The Vidman method was used to prepare KOA models in the model and acupotomy groups. In the acupotomy group, acupotomy was performed at the midpoints of the patellar ligament and medial and lateral collateral ligaments on the left knees once a week for three weeks. Rabbit articular cartilage protein was quantified in each group, and the proteins with differential expression were profiled via proteomics. The apoptosis-related proteins were further screened, and differentially expressed proteins common among the normal, model, and acupotomy groups were collected for cluster analysis. Pathological changes in chondrocytes were observed via HE staining, and chondrocyte apoptosis was observed under an electron microscope. Quantitative real-time PCR (RT-PCR) and western blot were performed to determine the mRNA and protein expression levels, respectively, of the common apoptosis-related proteins in the three groups.ResultsWestern blot and RT-PCR showed that the protein and mRNA expression levels of TXN, CAP1 and FABP3 were lower (all P < 0.05), whereas that of LTF, SCIN and IL-10 were higher (all P < 0.01) in the acupotomy group than that in the model group.ConclusionThe antiapoptotic effect of acupotomy on chondrocytes in KOA rabbits may be achieved by modulating the protein and mRNA expression of differentially expressed apoptosis-related proteins (TXN, CAP1, FABP3, LTS, SCIN, and IL-10).展开更多
Human growth hormone(GH)is a classical pituitary endocrine hormone that is essential for normal postnatal growth and has pleiotropic effects across multiple physiological systems.GH is also expressed in extrapituitary...Human growth hormone(GH)is a classical pituitary endocrine hormone that is essential for normal postnatal growth and has pleiotropic effects across multiple physiological systems.GH is also expressed in extrapituitary tissues and has localized autocrine/paracrine effects at these sites.In adults,hypersecretion of GH causes acromegaly,and strategies that block the release of GH or that inhibit GH receptor(GHR)activation are the primary forms of medical therapy for this disease.Overproduction of GH has also been linked to cancer and the microvascular complications that are associated with diabetes.However,studies to investigate the therapeutic potential of GHR antagonism in these diseases have been limited,most likely due to difficulty in accessing therapeutic tools to study the pharmacology of the receptor in vivo.This review will discuss current and emerging strategies for antagonizing GH function and the potential disease indications.展开更多

文摘ObjectiveTo elucidate the mechanism underlying the inhibitory effect of acupotomy on chondrocyte apoptosis in rabbits with knee osteoarthritis (KOA) through screening, analysis, and verification of apoptosis-related differentially expressed proteins via proteomics.MethodsThirty New Zealand white rabbits were randomly divided into the normal, model, and acupotomy groups, with 10 rabbits in each group. The Vidman method was used to prepare KOA models in the model and acupotomy groups. In the acupotomy group, acupotomy was performed at the midpoints of the patellar ligament and medial and lateral collateral ligaments on the left knees once a week for three weeks. Rabbit articular cartilage protein was quantified in each group, and the proteins with differential expression were profiled via proteomics. The apoptosis-related proteins were further screened, and differentially expressed proteins common among the normal, model, and acupotomy groups were collected for cluster analysis. Pathological changes in chondrocytes were observed via HE staining, and chondrocyte apoptosis was observed under an electron microscope. Quantitative real-time PCR (RT-PCR) and western blot were performed to determine the mRNA and protein expression levels, respectively, of the common apoptosis-related proteins in the three groups.ResultsWestern blot and RT-PCR showed that the protein and mRNA expression levels of TXN, CAP1 and FABP3 were lower (all P < 0.05), whereas that of LTF, SCIN and IL-10 were higher (all P < 0.01) in the acupotomy group than that in the model group.ConclusionThe antiapoptotic effect of acupotomy on chondrocytes in KOA rabbits may be achieved by modulating the protein and mRNA expression of differentially expressed apoptosis-related proteins (TXN, CAP1, FABP3, LTS, SCIN, and IL-10).
文摘Human growth hormone(GH)is a classical pituitary endocrine hormone that is essential for normal postnatal growth and has pleiotropic effects across multiple physiological systems.GH is also expressed in extrapituitary tissues and has localized autocrine/paracrine effects at these sites.In adults,hypersecretion of GH causes acromegaly,and strategies that block the release of GH or that inhibit GH receptor(GHR)activation are the primary forms of medical therapy for this disease.Overproduction of GH has also been linked to cancer and the microvascular complications that are associated with diabetes.However,studies to investigate the therapeutic potential of GHR antagonism in these diseases have been limited,most likely due to difficulty in accessing therapeutic tools to study the pharmacology of the receptor in vivo.This review will discuss current and emerging strategies for antagonizing GH function and the potential disease indications.
