In the past few years,there have been multiple advances in magnetic resonance (MR) instrumentation,in vivo devices,real-time imaging sequences and interventional procedures with new therapies.More recently,interventionists have started to use minimally invasive image-guided procedures and local therapies,which reduce the pain from conventional surgery and increase drug effectiveness,respectively.Local therapy also reduces the systemic dose and eliminates the toxic side effects of some drugs to other organs.The success of MR-guided procedures depends on visualization of the targets in 3D and precise deployment of ablation catheters,local therapies and devices.MR contrast media provide a wealth of tissue contrast and allows 3D and 4D image acquisitions.After the development of fast imaging sequences,the clinical applications of MR contrast media have been substantially expanded to include pre-during-and post-interventions.Prior to intervention,MR contrast media have the potential to localize and delineate pathologic tissues of vital organs,such as the brain,heart,breast,kidney,prostate,liver and uterus.They also offer other options such as labeling therapeutic agents or cells.During intervention,these agents have the capability to map blood vessels and enhance the contrast between the endovascular guidewire/catheters/devices,blood and tissues as well as direct therapies to the target.Furthermore,labeling therapeutic agents or cells aids in visualizing their delivery sites and tracking their tissue distribution.After intervention,MR contrast media have been used for assessing the efficacy of ablation and therapies.It should be noted that most image-guided procedures are under preclinical research and development.It can be concluded that MR contrast media have great value in preclinical and some clinical interventional procedures.Future applications of MR contrast media in image-guided procedures depend on their safety,tolerability,tissue specificity and effectiveness in demonstrating success of the interventions and therapies. 展开更多

Reperfusion injury components and manifestations determined by cardiovascular MR and MDCT imaging

Advances in magnetic resonance(MR) and computed tomography(CT) imaging have improved visualization of acute and scar infarct.Over the past decade,there have been and continues to be many significant technical advancements in cardiac MR and multi-detector computed tomography(MDCT) technologies.The strength of MR imaging relies on a variety of pulse sequences and the ability to noninvasively provide information on myocardial structure,function and perfusion in a single imaging session.The recent technical developments may also allow CT technologies to rise to the forefront for evaluating clinical ischemic heart disease.Components of reperfusion injury including myocardial edema,hemorrhage,calcium deposition and microvascular obstruction(MO) have been demonstrated using MR and CT technologies.MR imaging can be used serially and noninvasively in assessing acute and chronic consequences of reperfusion injury because there is no radiation exposure or administration of radioactive materials.MDCT is better suited for assessing coronary artery stenosis and as an alternative technique for as-sessing viability in patients where MR imaging is contraindicated.Changes in left ventricular(LV) volumes and function measured on cine MR are directly related to infarct size measured on delayed contrast enhanced images.Recent MR studies found that transmural infarct,MO and peri-infarct zone are excellent predictors of poor post-infarct recovery and mortality.Recent MR studies provided ample evidence that growth factor genes and stem cells delivered locally have beneficial effects on myocardial viability,perfusion and function.The significance of deposited calcium in acute infarct detected on MDCT requires further studies.Cardiac MR and MDCT imaging have the potential for assessing reperfusion injury components and manifestations. 展开更多

Reactive Arrays of Colorimetric Sensors for Metabolite and Steroid Identification

The work described herein examines a rapid mix-and-measure method called DETECHIP suitable for screening of steroids and metabolites. The addition of steroids and metabolites to reactive arrays of colorimetric sensors generated characteristic color “fingerprints” that were used to identify the analyte. A color analysis tool was used to identify the analyte pool that now includes biologically relevant analytes. The mix-and-measure arrays allowed the detection of disease metabolites, orotic acid and argininosuccinic acid;and the steroids androsterone, 1,4-androstadiene, testosterone, stanozolol, and estrone. The steroid 1,4-androstadiene was also detected by this method while dissolved in synthetic urine. Some of the steroids, such as androstadiene, stanozolol, and androsterone were co-dissolved with (2-hydroxypropyl)-β-cyclodextrin in order to increase solubility in aqueous buffered solutions. The colorimetric arrays do not intend to eliminate ELISA or mass spectroscopy based screening, but to possibly provide an alternative analytical detection method for steroids and metabolites. 展开更多

Detecting Concentration of Analytes with DETECHIP: A Molecular Sensing Array

DETECHIP is a detection system made of various sensors that has been shown to detect and discriminate between small molecules of interest, including various illicit and over-the-counter drugs. Previously, detection was normalized to a single concentration of analyte. Now this detection assay can detect concentration differences in analytes via red, green, and blue color value changes and shifts in the UV-Vis spectra of the assay. To determine the concentrations differences, the exposed assays were scanned on a flatbed scanner and the images were analyzed for individual RGB values with a custom macro in ImageJ, an image analysis program. Increasing concentrations of the analyte resulted in greater differences in color values between control and analyte wells. These differences showed a linear relationship to concentration change, some with correlation coefficients greater than 98%. This work expands the capability of DETECHIP to give information about the concentration of analyte when the analyte identity is known. 展开更多

The synthesis and characterization of a novel fluorophore(1), with potential application as an optical brightener are reported. This compound was prepared by reacting 4,4-diaminostilbene-2,2-disulfonic acid with cyanuric chloride in the presence of Na2CO3 followed by the addition of trityl aniline. Solution and solid state fluorescence demonstrated a strong blue/purple emission centered at 450 nm. 1H-NMR spectroscopy, mass spectrometry analysis, elemental analysis, and DOSY-NMR were used for the characterization of the fluorophore. 展开更多

Background Cerebral microhaemorrhages are increasingly being recognised as a complication of COVID-19.This observational retrospective study aims to further investigate the potential pathophysiology through assessing the pattern of microhaemorrhage and clinical characteristics of patients with COVID-19 and microhaemorrhage.By comparing with similar patterns of microhaemorrhage in other non-COVID-19 disease,this study aims to propose possible common pathogenic mechanisms.Methods A retrospective observational case series was performed identifying all patients with COVID-19 complicated by cerebral microhaemorrhage on MRI.The distribution and number of microhaemorrhages were recorded using the microbleed anatomical scale,and patients’baseline characteristics and salient test results were also recorded.Results Cerebral microhaemorrhages were noted to have a predilection for the corpus callosum,the juxtacortical white matter and brainstem.All patients had a preceding period of critical illness with respiratory failure and severe hypoxia necessitating intubation and mechanical ventilation.Discussion This study demonstrates a pattern of cerebral microhaemorrhage that is similar to the pattern reported in patients with non-COVID-19 related critical illness and other causes of severe hypoxia.This raises questions regarding whether microhaemorrhage occurs from endothelial dysfunction due the direct effect of SARS-CoV-2 infection or from the secondary effects of critical illness and hypoxia. 展开更多
