Skeletal stem/progenitor cell(SSPC)senescence is a major cause of decreased bone regenerative potential with aging,but the causes of SSPC senescence remain unclear.In this study,we revealed that macrophages in calluses secrete prosenescent factors,including grancalcin(GCA),during aging,which triggers SSPC senescence and impairs fracture healing.Local injection of human rGCA in young mice induced SSPC senescence and delayed fracture repair.Genetic deletion of Gca in monocytes/macrophages was sufficient to rejuvenate fracture repair in aged mice and alleviate SSPC senescence.Mechanistically,GCA binds to the plexin-B2 receptor and activates Arg2-mediated mitochondrial dysfunction,resulting in cellular senescence.Depletion of Plxnb2 in SSPCs impaired fracture healing.Administration of GCA-neutralizing antibody enhanced fracture healing in aged mice.Thus,our study revealed that senescent macrophages within calluses secrete GCA to trigger SSPC secondary senescence,and GCA neutralization represents a promising therapy for nonunion or delayed union in elderly individuals. 展开更多

Stabilization of tearing modes and neoclassical tearing modes is of great importance for tokamak operation.Electron cyclotron waves(ECWs)have been extensively used to stabilize the tearing modes with the virtue of highly localized power deposition.Complete suppression of the m/n=2/1 tearing mode(TM)by electron cyclotron resonance heating(ECRH)has been achieved successfully on the J-TEXT tokamak.The effects of ECW deposition location and power amplitude on the 2/1 TM suppression have been investigated.It is found that the suppression is more effective when the ECW power is deposited closer to the rational surface.As the ECW power increases to approximately 230 k W,the 2/1 TM can be completely suppressed.The island rotation frequency is increased when the island width is reduced.The experimental results show that the local heating inside the magnetic island and the resulting temperature perturbation increase at the O-point of the island play dominant roles in TM suppression.As the ECW power increases,the 2/1 island is suppressed to smaller island width,and the flow shear also plays a stabilizing effect on small magnetic islands.With the stabilizing contribution of heating and flow shear,the 2/1 TM can be completely suppressed. 展开更多

The fruits of Amomum tsao-ko(Cao-Guo)were documented in Chinese Pharmacopoeia for the treatment of abdominal pain,vomiting,and plague.In our previous study,a series of diarylheptanes and flavonoids withα-glucosidase and protein tyrosine phosphatase 1B(PTP1B)inhibitory activity have been reported from the middle-polarity part of A.tsao-ko,whereas the antidiabetic potency of the low-polarity constituents is still unclear.In this study,three new hydroxytetradecenals,(2E,4E,8Z,11Z)-6R-hydroxytetradeca-2,4,8,11-tetraenal(1),(2E,4E,8Z)-6R-hydroxytetradeca-2,4,8-trienal(2)and(2E,4E)-6R-hydroxytetradeca-2,4-dienal(3)were obtained from the volatile oils of A.tsao-ko.The structures of compounds 1–3 were determined using spectroscopic data involving 1D and 2D nuclear magnetic resonance(NMR),high-resolution mass spectra(HRMS),and specific rotation([α]D).Their hypoglycemic activity was evaluated against glycogen phosphorylase(GPa)and PTP1B.Compounds 1 and 2 displayed moderate activity against PTP1B with inhibition rates of 33.8%−50.3%at 100 and 200μM.Moreover,compound 1 exhibited an obvious inhibitory effect on GPa(IC50=31.7μM),whereas compound 2 was inactive.This study demonstrates hydroxytetradecenals as the characteristic components of A.tsao-ko with therapeutic potential in diabetes. 展开更多

Background The co-occurrence of depression and anxiety among adolescents is typically associated with suicide ideation.Aims The study aimed to investigate the symptom-level relationship between suicide ideation and the comorbidity of depression and anxiety.Methods 1501 adolescents aged 12-19 years were assessed using the Patient Health Questionnaire(PHQ-9)and the Generalized Anxiety Disorder Scale,and 716 adolescents who scored≥5 on both scales were selected as participants.Network analysis was used to identify the network structure of depressive symptoms and anxiety symptoms.Participants were categorised into either the suicide ideation or non-suicide ideation groups based on their scoring on the suicide-related item in PHQ-9.A comparison was made between the depression-anxiety symptom networks of the two groups.Results‘Restlessness’,‘sad mood’and‘trouble relaxing’were the most prominent central symptoms in the depression-anxiety symptom network,and‘restlessness’,‘nervousness’and‘reduced movement’were the bridge symptoms in this network.‘Sad mood’was found to be directly related to‘suicide ideation’with the highest variance.The network structure was significantly different in properties between the suicide ideation group and the non-suicide ideation group,with‘restlessness’and‘sad mood’exhibiting significantly higher influence in the network of the suicide ideation group than that in the non-suicide ideation group.Conclusion Restlessness and sad mood could be targeted for the intervention of depression-anxiety symptoms among adolescents with suicide ideation. 展开更多

目的:探讨腹膜后肿瘤切除术后出现功能性胃排空障碍(functional delayed gastric emptying,FDGE)的危险因素。方法:回顾性分析2017年9月至2022年12月于北京大学国际医院行腹膜后肿瘤切除术后出现FDGE的患者67例(观察组),选择同期836例... 目的:探讨腹膜后肿瘤切除术后出现功能性胃排空障碍(functional delayed gastric emptying,FDGE)的危险因素。方法:回顾性分析2017年9月至2022年12月于北京大学国际医院行腹膜后肿瘤切除术后出现FDGE的患者67例(观察组),选择同期836例未出现FDGE的腹膜后肿瘤切除术患者作为对照组,调查和对比两组患者的病史及临床诊治资料,观察腹膜后肿瘤切除术后影响FDGE发生的危险因素。结果:全部患者总体术后FDGE发生率为(7.42%)。观察组存在不良精神因素(37.31%)、术前消化道梗阻(41.79%)、术后腹腔并发症(79.10%)、糖尿病(29.86%)的比例高于对照组,同时在年龄(61.85±6.11)岁、出血量(1011.94±507.30)mL方面高于对照组,差异具有统计学意义(P<0.05)。观察组术前白蛋白水平(38.22±3.75)g/L、术后白蛋白水平(30.22±3.36)g/L低于对照组,差异具有统计学意义(P<0.05)。危险因素分析显示,高龄、糖尿病、术前消化道梗阻、术中出血量、围手术期白蛋白水平、术后腹腔并发症、不良精神因素是术后发生FDGE的影响因素。结论:导致腹膜后肿瘤切除术后胃排空障碍的影响因素种类众多,应予以准确评估,制定合理、详细的预防和诊疗计划。 展开更多

As a promising method for fast ion diagnostics,collective Thomson scattering(CTS)can measure the one-dimensional velocity distribution of fast ions with high spatial and temporal resolution.The feasibility of diagnosing fast ions in a compact high-field tokamak by CTS was studied in this work,and the results showed that a wide range of probing frequencies could be applied.A high-frequency case and a low-frequency case were mainly considered for fast ion diagnostics in a compact high-field tokamak.The use of a high probing frequency could effectively avoid the refraction effect of the beams,while the application of a low probing frequency allows greater flexibility in the selection of scattering angle which may help to improve the spatial resolution.Based on typical plasma conditions(B_(0)=12.2 T,n_(e0)=4.3×10^(20)m^(-3),T_(e0)=22.2 keV,T_(i0)=19.8 keV)for a compact high-field tokamak,a220 GHz CTS diagnostic that utilizes a small scattering angle ofθ=30°and a 160 GHz CTS diagnostic that utilizes an orthogonal geometry were proposed.Further study showed that the high-frequency case could operate in a wider range of plasma conditions and provide more information on fast ions while the low-frequency case could achieve higher spatial resolution of the poloidal direction. 展开更多

背景食管癌发病率及死亡率在全世界都极高,且治疗效果不佳.由于目前对食管癌发病机制尚未完全清楚,这不利于食管癌特异性治疗药物的研究.通过阅读大量文献发现,热休克转录因子1(heat shock transcription fact 1,HSF1)与多种恶性肿瘤的... 背景食管癌发病率及死亡率在全世界都极高,且治疗效果不佳.由于目前对食管癌发病机制尚未完全清楚,这不利于食管癌特异性治疗药物的研究.通过阅读大量文献发现,热休克转录因子1(heat shock transcription fact 1,HSF1)与多种恶性肿瘤的发生发展密切相关,那么HSF1是否也与食管鳞状细胞癌的发生发展密切相关呢?HSF1是否会成为治疗食管鳞状细胞癌的生物靶标呢?在不同中晚期食管鳞状细胞癌患者中,不同患者对放化疗敏感度不一样,已有研究证明,在肝细胞癌中,HSF1会削弱放化疗对肿瘤的毒性作用从而降低疗效,那么HSF1是否会影响食管鳞状细胞癌对放化疗的敏感性呢?目的探究HSF1在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)中表达情况及其表达程度对ESCC放化疗敏感性的影响.方法(1)将92例患者分成4组:ESCCⅠ、Ⅱ期手术切除组20例,ESCCⅢ、Ⅳ期手术切除组18例,ESCCⅢ、Ⅳ期放化疗组44例,其中ESCCⅢ、Ⅳ期放化疗组分为HSF1低表达组16例和高表达组28例食管不典型增生组10例;对照组:食管炎症组10例;(2)取各组病理蜡块通过免疫组化及染色技术检测HSF1在各组表达情况;(3)通过ESCCⅢ、Ⅳ期患者HSF1低表达组及高表达组的非肿瘤病灶大小、肿瘤直径及癌胚抗原(carcinoembryonic antigen,CEA)值在放化疗前后的变化,评估患者对放化疗治疗敏感性;(4)分析影响ESCC手术患者3年生存期的相关因素及ESCC患者3年总体生存率.结果(1)HSF1在各ESCC组中高表达,在食管炎症组及食管不典型增生组中低表达,且各ESCC组与食管炎症组及食管不典型增生组的HSF1表达情况存在差异,P=0.001,有统计学意义;HSF1表达与年龄、性别、肿瘤部位、肿瘤大小、分化程度、T分期、N分期、M分期无关,P>0.05,均无统计学意义;(2)放化疗后,ESCCⅢ、Ⅳ期放化疗组中HSF1低表达组与HSF1高表达组相比,非肿瘤病灶减少更多,肿瘤直径减小更大,CEA值下降更明显,P<0.005,均有统计学意义;(3)在ESCC手术切除组中,3年生存期与年龄(P=0.019)、HSF1表达程度(P=0.028)、T分期(P=0.007)、N分期(P=0.016)有关,均有统计学意义,与性别、肿瘤部位、肿瘤大小、分化程度无关,P>0.05,均无统计学意义;HSF1低表达组3年总体生存率比HSF1高表达组高,P=0.016,有统计学意义;(4)在ESCCⅢ、Ⅳ期放化疗组中,HSF1低表达组1年、2年及3年生存率均比HSF1高表达组高,P<0.05,有统计学意义;HSF1低表达组3年总体生存率比HSF1高表达组高,P=0.03,有统计学意义.结论(1)HSF1在ESCC中高表达且HSF1表达越高,患者预后越差,且HSF1表达与患者临床特征无关;(2)在ESCCⅢ、Ⅳ期放化疗组中,HSF1表达越高,患者对放化疗敏感性越差;(3)在ESCC手术切除组及ESCCⅢ、Ⅳ期放化疗组中,HSF1低表达组3年总体生存率比HSF1高表达组高. 展开更多

AIM To determine whether circular RNAs(circ RNAs) are involved in pathological processes of gastric cancer(GC).METHODS Three circ RNAs with differential expression in GC and colorectal cancer were randomly selected for validation by quantitative reverse transcription-polymerase chain reaction(q RT-PCR), using 20 pairs of gastric tissues and normal tissues. Based on the predicted circ RNAmi RNA network, we then focused on hsa_circ_0000745, which was found to be down-regulated in 20 GC tissues compared with normal tissues. The hsa_circ_0000745 levels were further analyzed by q RT-PCR in 60 GC tissues and paired adjacent non-tumor tissues, as well as 60 plasma samples from GC patients and 60 plasma samples from healthy controls. The associations between the levels of hsa_circ_0000745 and the clinicopathological features of GC patients were statistically assessed. A receiver operating characteristic(ROC) curve was used to evaluate the diagnostic value of hsa_circ_0000745 in GC.RESULTS Hsa_circ_0000745 was down-regulated in GC tissues vs non-tumorous tissues(P < 0.001) and in plasma samples from patients with GC vs healthy controls(P < 0.001). The expression level of hsa_circ_0000745 in GC tissues correlated with tumor differentiation, while the expression level in plasma correlated with tumor-nodemetastasis stage. The area under the ROC curve(AUC) of hsa_circ_0000745 in plasma was 0.683, suggesting good diagnostic value. Plasma hsa_circ_0000745 level combined with carcinoembryogenic antigen(CEA) level increased the AUC to 0.775.CONCLUSION Hsa_circ_0000745 plays an important role in GC and its expression level in plasma in combination with CEA level is a promising diagnostic marker for this malignancy. 展开更多

Background: Pancreatic ductal adenocarcinoma(PDAC) is the most deadly type of tumor, and its pathogenesis remains unknown. Circular RNAs(circRNAs) may be functional and bind to micro RNAs and consequently, influence the activity of targeted mRNAs. Recent researches indicate that one circRNA, ciRS-7, acts as a sponge of miR-7 and thus, inhibits its activity. It is well known that miR-7 is a cancer suppressor in many cancers. However, the relationship between ciRS-7 and miR-7, and the role of ciRS-7 in PDAC, remains to be elucidated. Methods: miR-7 and ciRS-7 expression in 41 pairs of PDAC tumors and their paracancerous tissues were detected by quantitative reverse transcription polymerase chain reaction(qRT-PCR). The relationships between their expression levels and clinicopathological features in PDAC tissues were assessed. The relationship between miR-7 and ciRS-7 was also assessed by Spearman’s correlation. We also used cell lines to evaluate the role of ciRS-7 in cell line behavior. The ciRS-7 interfere RNA(si RNA) and its empty vector were transfected into PDAC cells. PDAC cells proliferation and invasion abilities were detected by MTT assay and invasion analysis. The expression of proteins was assessed by Western blotting. Results: ciRS-7 expression was significantly higher in PDAC tissues than paracancerous tissues( P = 0.002). However, miR-7 expression showed the opposite trend( P = 0.048). Moreover, ciRS-7 expression was inversely correlated with miR-7 in PDAC( r s =-0.353, P = 0.023). ciRS-7 expression was also significantly elevated in venous invasion(3.72 ± 2.93 vs. 2.14 ± 1.26;P = 0.028) and lymph node metastasis(4.19 ± 2.75 vs. 2.32 ± 1.90;P = 0.016) in PDAC patients. Furthermore, ciRS-7 knockdown suppressed cell proliferation and invasion of PDAC cells( P < 0.05), and the downregulation of ciRS-7 resulted in miR-7 overexpression and subsequent inhibition of epidermal growth factor receptor(EGFR) and signal transducer and activator of transcription 3(STAT3). Conclusions: Circular RNA ciRS-7 plays an oncogene role in PDAC, partly by targeting miR-7 and regulating the EGFR/STAT3 signaling pathway. 展开更多

Drought stress is a limiting factor for wheat production and food security.Drought priming has been shown to increase drought tolerance in wheat.However,the underlying mechanisms are unknown.In the present study,the genes encoding the biosynthesis and metabolism of abscisic acid(ABA)and jasmonic acid(JA),as well as genes involved in the ABA and JA signaling pathways were up-regulated by drought priming.Endogenous concentrations of JA and ABA increased following drought priming.The interplay between JA and ABA in plant responses to drought priming was further investigated using inhibitors of ABA and JA biosynthesis.Application of fluridone(FLU)or nordihydroguaiaretic acid(NDGA)to primed plants resulted in lower chlorophyll-fluorescence parameters and activities of superoxide dismutase and glutathione reductase,and higher cell membrane damage,compared to primed plants(PD)under drought stress.NDGA+ABA,but not FLU+JA,restored priming-induced tolerance,as indicated by a finding of no significant difference from PD under drought stress.Under drought priming,NDGA induced the suppression of ABA accumulation,while FLU did not affect JA accumulation.These results were consistent with the expression of genes involved in the biosynthesis of ABA and JA.They suggest that ABA and JA are required for priming-induced drought tolerance in wheat,with JA acting upstream of ABA. 展开更多

The use of supercritical-fluid chromatography for determining partial molar volumes of ethyl esters of cis-5,8,11,14,17-eicosapentaenoic acid (EPA) and cis -4,7,10,13,16,19- docosa-hexaenoic acid (DHA) in supercritical carbon dioxide is presented and discussed. Partial molar volumes of EPA and DHA esters are obtained from the variation of the retention properties with the density of mobile phase at 313.15 K, 323.15 K, 333.15 K and in the pressure range from 9 MPa to 21 MPa. 展开更多

Drought priming is a promising approach to improve tolerance to further drought in wheat.The root apex plays important roles in drought however,its contribution to drought priming remains unknown.To provide mechanistic insights into this process,the transcriptomes and proteomes at three different zones along the root axis under drought stress were analyzed.Physiological assessment of root growth indicated that priming augmented roots growth in response to drought and also the levels of protective proline and glycine betaine.Scanning across the proximal to the distal zones of the root apex indicated increases the transcription of genes involved in primary and secondary metabolism.Conversely,genes related to translation,transcription,folding,sorting and degradation,replication and repair were increased in the apex compared to the proximal zone.A single drought episode suppressed their expression but prior drought priming served to maintain expression with recurrent drought stress.The differentially primed responses genes were mainly involved in the pathways related to plant hormone signaling,stress defense and cell wall modification.The prediction of regulatory hubs using Cytoscape implicated signaling components such as the ABA receptor PYL4 as influencing antioxidant status and the cell cycle.Based our integrative transcriptomic-proteomic assessments we present a model for drought priming protected plant hormone signaling transduction pathways to drive the cell cycle and cell wall loosening to confer beneficial effects on roots to counter the effects of drought.This model provides a theoretical basis for improvement of drought tolerance in wheat,via an increased understanding of drought priming induced drought tolerance. 展开更多

To maximize the catalytic performance of MoS_(2) in the hydrogen evolution reaction,we investigate the electrocatalytic and photocatalytic performance of monolayer MoS_(2) doped with noble metal(Ag,Au,Cu,Pd,and Pt)using first principles calculation combined with the climbing image nudged elastic band method.We find the band gap of the monolayer MoS_(2) is reduced significantly by the noble metal doping,which is unfavorable to improving its photocatalytic performance.The optical absorption coefficient shows that the doping does not increase the ability of the monolayer MoS_(2) to absorb visible light.The monolayer MoS_(2) doped with the noble metal is not a potential photocatalyst for the hydrogen evolution reaction because the band edge position of the conduction band minimum is lower than-4.44 eV,the reduction potential of H^(+)/H_(2).Fortunately,the band gap reduction increases the electron transport performance of the monolayer MoS_(2),and the activation energy of water splitting is greatly reduced by the noble metal doping,especially the Pt doping.On the whole,noble metal doping can enhance the electrocatalytic performance of the monolayer MoS_(2). 展开更多

背景中晚期食管癌的主要症状是吞咽困难,由于无手术指征,为改善患者症状及提高生活质量,食管支架治疗成为主要姑息治疗方法.结合许多研究结果可推测:内放射支架结合普通支架及近距离放疗的优势,治疗效果更佳.目的评价内放射支架(碘125... 背景中晚期食管癌的主要症状是吞咽困难,由于无手术指征,为改善患者症状及提高生活质量,食管支架治疗成为主要姑息治疗方法.结合许多研究结果可推测:内放射支架结合普通支架及近距离放疗的优势,治疗效果更佳.目的评价内放射支架(碘125粒子支架)与普通覆膜支架对中晚期食管癌患者姑息治疗的疗效及并发症的差异.方法应用计算机检索电子数据库:PubMed(1989/2020-03)、Web of Science(2000/2020-03)、Wiley Online Library(1992/2020-03)、中国期刊全文数据库(1978/2020-03)、万方数据库(1997/2020-03)、维普数据库(2000/2020-03)及手动检索Cochrance图书馆.采用Cochrance系统评价员手册(4.2.2版)推荐的方法纳入文章,使用牛津2011年证据水平等级评估对纳入文章进行质量评估并用Revman 5.3软件进行Meta分析.结果最终纳入10篇文献,共有943例患者,其中置入内放射支架有449例,置入普通覆膜支架有494例.对10篇文献进行Meta分析后结果显示:内放射支架组比普通覆膜支架组的平均生存期长3.91 mo(95%CI:1.68-6.13,Z=3.44,P=0.0006),中位生存期长3.12 mo(95%CI:1.78-4.47,Z=4.57,P=0.0001).两组患者的吞咽困难分数在置入支架后1、3 mo内均明显降低(P<0.05),有统计学差异,但两组间无统计学差异(P>0.05).置入支架后,内放射支架组与普通覆膜支架组疼痛发生率差异无统计学意义(OR=0.89,95%CI:0.65-1.21,Z=0.77,P=0.44);出血发生率差异无统计学意义(OR=0.80,95%CI:0.52-1.22,Z=1.03,P=0.30);穿孔发生率差异无统计学意义(OR=1.16,95%CI:0.55-2.43,Z=0.39,P=0.70);支架移位发生率差异无统计学意义(OR=0.66,95%CI:0.31-1.38,Z=1.10,P=0.27);再狭窄发生率差异有统计学意义(OR=0.61,95%CI:0.42-0.87,Z=3.73,P=0.006).结论内放射支架用于治疗中晚期食管癌,可延长患者平均生存期及中位生存期,且术后再狭窄的发生率明显降低,但是在1-3 mo内缓解吞咽困难症状及术后疼痛、出血、穿孔及支架移位的发生率与普通覆膜支架无明显差异. 展开更多

To maximize the potential of monolayer molybdenum disulfide(MoS2)sheet in the disposal of heavy metal ions in wastewater,we compared the adsorption of several common heavy metal ions(including Cr^(3+),Ni^(2+),Cu^(2+),Zn^(2+),Cd^(2+),Hg^(2+),and Pb^(2+))in wastewater on the monolayer MoS2 sheet through first-principles calculation.Our simulation results show that the monolayer MoS2 sheet is a potential heavy metal adsorption material because of the attractive interaction between them.The most negative adsorption energy determines that the TMo site is the most stable adsorption site for the heavy metal ions.The attractive interaction is considered as chemical adsorption,and it is closely related to charge transfer.The orbital hybridization between S p and heavy metal ions p and d states electrons contributes to the adsorption,except the orbital hybridization between S p and Pb p states electrons contributes to the Pb^(2+) adsorption.All the results show that the monolayer MoS2 sheet is most suitable for removing Ni^(2+) and Cr^(3+) ions from wastewater,followed by Cu^(2+) and Pb^(2+).For the ions Cd^(2+),Zn^(2+),and Hg^(2+),its adsorption strength remains to be improved. 展开更多

Objective Cerebral ischemia/reperfusion(I/R)is a potential factor for lethal injury,and currently lacks effective remedies.Bauhinia championii extracts(BCEs)have been reported to exhibit anti-oxidative and anti-hypoxia properties.The current work aimed to study whether BCE could alleviate neuronal injury caused by I/R.Methods To investigate the protective effects of BCE,oxygen-glucose deprivation/reperfusion(OGD/R)was applied to the HT22 cell line in vitro and to a cerebral I/R mouse model in vivo.Results Under OGD/R,the survival of HT22 cells was significantly prolonged after treatment with BCE.In vivo,BCE significantly reduced the infarct area and decreased neuronal apoptosis caused by I/R.It was further found that OGD/R could trigger endoplasmic reticulum(ER)stress and induce ER stress-mediated neuronal apoptosis in vivo and in vitro,while BCE could effectively alleviate ER stress and neuronal apoptosis.Conclusion These results suggested that BCE exhibits neuroprotective effects by reducing ER stress-mediated apoptosis after cerebral I/R injury.BCE may therefore be an effective therapeutic regimen against cerebral I/R damage. 展开更多

Although the maximal COD removal efficiency would reach 35.4% when 5 mL NaClO was added in 100 mL industrial dyes effluent, it is found that by using nano-Fe2O3/goldmine system, the COD removal efficiency of 13,000 mg/L dyes wastewater could reach up to 75.5% in the presence of 30 g/L nano-Fe2O3/goldmine complex and 50 mL/L NaClO at 50。C. 展开更多
