There is an increasing interest in discovering new antibacterial agents derived from nature to enhance the treatment of various bacterial infections.Defensins and their derived peptide fragments exhibit significant antibacterial activity without any cytotoxic effects,making them attractive features for potential novel antibacterial therapeutics.Crassostrea gigas,a traditional seafood that has been used worldwide for centuries,has its shells applied in Chinese medicine as Ostreae concha.In this study,bioinformatics analysis was used to obtain a novel antibacterial peptide,CGD-1,derived from marine Chinese medicine Ostreae concha.The physicochemical characterization and circular dichroism analysis results demonstrated that CGD-1 assembled into anα-helical structure in a simulated membrane environment,and it displayed antibacterial action against Gram-negative bacteria.The minimal inhibitory concentrations against both Pseudomonas aeruginosa ATCC27853 and Escherichia coli ATCC25922 were 25μM.CGD-1 was able to efficiently permeate the cell membrane.Changes in bacterial cell morphology were evaluated using a field emission scanning electron microscope.The results suggested that CGD-1 exerted its antibacterial activity through permeabilizing and disrupting the bacterial cell membrane.Therefore,CGD-1 may have potential applications in fighting against pathogenic bacteria such as P.aeruginosa and E.coli. 展开更多

BACKGROUND Stress-related gastric mucosal damage or ulcer remains an unsolved issue for critically ill patients.Stress ulcer prophylaxis has been part of routine intensive care,but uncertainty and controversy still exist.Co-secreted with mucins,intestinal trefoil factor(ITF)is reported to promote restitution and regeneration of intestinal mucosal epithelium,although the mechanism remains unknown.AIM To elucidate the protective effects of ITF on gastric mucosa and explore the possible mechanisms.METHODS We used a rat model of gastric mucosal damage induced by water immersion restraint stress and lipopolysaccharide-treated human gastric epithelial cell line to investigate the potential effects of ITF on damaged gastric mucosa both in vivo and in vitro.RESULTS ITF promoted the proliferation and migration and inhibited necrosis of gastric mucosal epithelia in vitro.It also preserved the integrity of gastric mucosa by upregulating expressions of occludin and zonula occludens-1.In the rat model,pretreatment with ITF ameliorated the gastric mucosal epithelial damage and facilitated mucosal repair.The protective effects of ITF were confirmed to be exerted via activation of Akt signaling,and the specific inhibitor of Akt signaling LY249002 reversed the protective effects.CONCLUSION ITF might be a promising candidate for prevention and treatment of stressinduced gastric mucosal damage,and further studies should be undertaken to verify its clinical feasibility. 展开更多

BACKGROUND:Paraquat(PQ)-induced acute lung injury(ALI)and pulmonary fi brosis are common diseases with high mortality but without eff ective antidotes in emergency medicine.Our previous study has proved that arctigenin suppressed pulmonary fibrosis induced by PQ.We wondered whether arctigenin could also have a protective eff ect on PQ-induced ALI.METHODS:A PQ-induced A549 cell injury model was used,and the effect of arctigenin was determined by a cell counting kit-8(CCK-8)cell viability assay.In addition,terminal deoxynucleotidyl transferase(TdT)-mediated dUTP nick-end labelling(TUNEL)staining assays and mitochondrial membrane potential assays were performed to evaluate the level of cell apoptosis.The generation of reactive oxygen species(ROS)was refl ected by dihydroethidium(DHE)staining and a 2’,7’-dichlorodihy drofluorescein diacetate(DCFH-DA)assay.Moreover,immunoblotting studies were used to assess the expression of mitogen-activated protein kinases(MAPKs)and p38 MAPK.RESULTS:Arctigenin attenuated PQ-induced inhibition of A549 cell viability in a dose-dependent manner.Arctigenin also significantly reduced PQ-induced A549 cell apoptosis,as refl ected by the TUNEL assay and mitochondrial membrane potential assay,which may result from suppressed ROS/p38 MAPK signaling because we found that arctigenin dramatically suppressed ROS generation and p38 MAPK phosphorylation.CONCLUSION:Arctigenin could attenuate PQ-induced lung epithelial A549 cell injury in vitro by suppressing ROS/p38 MAPK-mediated cell apoptosis,and arctigenin might be considered a potential candidate drug for PQ-induced ALI. 展开更多

目的:评估FS-LASIK手术在制作角膜瓣过程中不透明气泡层(OBL)的发生对准分子激光机眼球旋转追踪补偿的影响。方法:前瞻性临床研究。随机抽取2019-07/09在我院行FS-LASIK手术200例400眼。统计FS-LASIK术中OBL的总体发生率、角膜瓣外OBL... 目的:评估FS-LASIK手术在制作角膜瓣过程中不透明气泡层(OBL)的发生对准分子激光机眼球旋转追踪补偿的影响。方法:前瞻性临床研究。随机抽取2019-07/09在我院行FS-LASIK手术200例400眼。统计FS-LASIK术中OBL的总体发生率、角膜瓣外OBL发生率、角膜瓣内OBL发生率、动态自旋跟踪矫正(DCC)和静态自旋追踪矫正(SCC)的运行成功率,并分析OBL对DCC和SCC运行的影响。结果:本研究共发生OBL 183眼,总体OBL发生率为45.8%,DCC成功运行397眼,总体DCC运行成功率为99.2%;SCC成功运行的手术共计为293眼,总体SCC运行成功率为73.2%;总体OBL、角膜瓣外OBL、角膜瓣内OBL对DCC的运行成败均无影响(P>0.05),但对SCC的运行成败均有影响(P<0.05),OBL发生会导致SCC运行失败。结论:在制作角膜瓣过程中OBL的发生可能会导致FS-LASIK准分子激光机SCC运行的失败。 展开更多

Comparison of age-related changes between corneal and ocular aberration in young and mid-age myopic patients 被引量:4

AIM: To study the dynamic character of aberration between the cornea and the ocular with aging, and to evaluate the symmetry of the aberrations between right and left eye in order to supply the data for clinic to do the refractive surgery reasonably. METHODS: This is a comparative case series study. 82 normal cases (164 eyes) including 37 females (74 eyes) and 45 males (90 eyes) were recruited through the routine examinations,Topolyzer and wavefront analysis. The average age was 25.9 +/- 5.0 years old (range 18 to 49 years old), and the mean spherical equivalent (SE) is -3.82 +/- 2.21D (range -1.00 to -6.00D). The changes of aberrations regarding age, the relationship between anterior corneal and total aberrations were analyzed,as well as the symmetry between right and left eyes by using Zernike terms. RESULTS: The Z (3) (1), RMS3 of corneal aberrations, Z (3) (1), Z (4) (0) RMS3 and RMS4 of ocular aberrations had a positive correlation with age. The zernike terms both in corneal and whole eye were significantly correlated between right and left eyes. CONCLUSION: The corneal horizontal coma, ocular horizontal coma and ocular spherical aberrations become to increase at the age of more than 40 years old. The dynamic change of aberration with aging, balance between corneal and ocular, and the symmetric character between left eye and right eye should be designed carefully in the treatment nomogram before the refractive surgery. 展开更多

角膜缘保护技术在经上皮角膜交联术中的应用效果

目的:评估经上皮角膜交联(Epi-on CXL)术中采用角膜缘保护技术治疗圆锥角膜的临床应用效果。方法:前瞻性临床研究。选取2019-01/12我院收治拟行Epi-on CXL手术的双眼进展期圆锥角膜患者15例30眼,将右眼15眼纳入试验组,术中采用角膜缘保... 目的:评估经上皮角膜交联(Epi-on CXL)术中采用角膜缘保护技术治疗圆锥角膜的临床应用效果。方法:前瞻性临床研究。选取2019-01/12我院收治拟行Epi-on CXL手术的双眼进展期圆锥角膜患者15例30眼,将右眼15眼纳入试验组,术中采用角膜缘保护技术;左眼15眼纳入对照组,术中不采用角膜缘保护技术。比较两组患眼术前和术后最佳矫正远视力、散光度、角膜曲率Sim-K平均值(Km)、角膜最薄点厚度、泪膜破裂时间及术后不适感、角膜上皮愈合时间的差异。结果:与术前相比,术后3mo时两组最佳矫正远视力、散光度均改善(P<0.05),术后角膜最薄点厚度逐渐减小(P<0.05),术后3mo内泪膜破裂时间变短(P<0.05),但两组之间最佳矫正远视力、散光度、Km、角膜最薄点厚度、泪膜破裂时间、术后不适感方面均无差异(P>0.05),且试验组术后角膜上皮愈合时间比对照组短(3.20±0.56d vs 3.73±0.96d,P=0.041)。结论:角膜缘保护技术能够缩短Epi-on CXL术后角膜上皮愈合时间,初步证实了该技术在Epi-on CXL手术中应用的可行性。 展开更多

Considering the intrinsic advantages of natural copiousness and cost-effectiveness of potassium resource,potassium-ion batteries(KIBs) are booming as prospective alternatives to lithium-ion batteries(LIBs) in large-scale energy storage scenarios. Nevertheless, lacking desirable electrodes for reversibly hosting the bulky K+hinders the widespread application of KIBs, and it needs to be urgently solved. Hereon, the porous S-doped Sb_(2)O_(3)-graphene-carbon(SAGC) nanofibers are manufactured through an adjustable and facile approach, which involves electrospinning, in situ etching and sulfuration. The synthesized SAGC is featured by the ultra-small amorphous Sb_(2)O_(3) homogeneously wrapped inside the carbon matrix, as well as the co-incorporation of graphene and sulfur. Tentatively,the SAGC nanofiber sheets are applied as binder-free anodes for KIBs, exhibiting a prominent cycling life(256.72 m Ah·g^(-1) over 150 cycles at 100 m A·g^(-1)) and rate·g^(-1) over 100 cycles at 1 A·g^(-1)). The positive synergy among all the active components accounts for the distinguished performances of the SAGC. By reinforcing the tolerability to the swelling stress, producing the valid electrochemical active sites, and promoting the charge transferring for reversible K+uptake, the SAGC finally renders the excellent cyclability, capacity, and rate capability. Moreover, the extrinsic electrochemical pseudocapacitance characteristics induced by the porous carbon substrate elevate the K-storage capacity of the SAGC as well. It is hoped that the conclusions drawn may offer new insights into a direction for the high-performance binderfree KIB anodes. 展开更多

The objective of this study was to systematically evaluate the efficacy and safety of Huachansu capsules in the treatment of esophageal cancer.We searched all relevant Chine se and English documents in the Cochrane Libraty Clinical Controlled Trials,PubMed,EMB ASE,CNKI,and VIP databases,from the time the databases were established to December 2020.The studies were selected according to the inclusion and exclusion criteria,and their quality was evaluated.We used RevMan 5.4 to conduct the meta-analysis of measurement indicators and intervention measures.Six randomized controlled trials comprising 526 patients with esophageal cancer were included in the study.Compared with radiotherapy and chemotherapy alone,oral Huachansu capsules combined with radiotherapy and chemotherapy improved the clinical efficacy(risk ratio[RR]=1.35,95%confidence interval[CI],1.17-1.55,P<0.0001),quality of life(weighted mean difference[MD]=10.01,95%CI[3.39-16.64],P=0.003),and immune level(CD3+:MD=4.99,95%CI[3.70-6.29],P<0.0001;CD4+:MD=6.49,95%CI[5.55-7.42],P<0.0001;CD5+:MD=0.20,95%CI[0.11-0.28],P<0.0001),and reduced toxic and side effects(reductions in white blood cell count:RR=0.63,95%CI[0.44-0.90],P=0.01;Reductions in blood platelet:RR=0.70,95%CI[0.52-0.94],P=0.02;Gastrointestinal reaction:RR=0.73,95%CI[0.59-0.89],P=0.002).However,the number of included studies was small,and conclusions still need to be further verified using larger samples and high-quality randomized,double-blind controlled trials. 展开更多

Metallodrugs with fine-tuned coordination between metals and bioactive ligands can achieve cytotoxic effects in cancer therapy and have been considered as a new approach for drug design.However,it has yet to be elucidated whether these metallodrugs target epitranscriptomic proteins for gene expression regulation.This report describes a rhein-based Rh(l)-arene complex,Rh1,that exhibited promising antiproliferative ffects in several tumor cellines.Rh1 induced cell death through the autophagy,cell cycle arrest,and accumulation of intracllular reactive oxygen species(ROs),In addition,Rh1 upregulated the global N^(6)-methyladenosine(m^(6)A)levels in A549 cells in the fat mass-and obesity-associated protein(FTO)-dependent manner.Collectively,the metal-based FTO inhibitor Rh1 effectively suppressed tumor cell proliferation and modulated the abundance of cellular m^(6)A,highlighting the potential of metal-based agents to target and regulate epitranscriptomics for tumor suppression. 展开更多
