Objective:The aim of this study was to investigate the growth inhibition effects and the mechanisms of trkA kinase inhibitor on human pterygium fibroblasts(HPF).Methods:Three normal conjunctivas and eleven pterygiums were surgically collected,trkA and p75 was examined in different tissues by immunohistochemistry.trkA and p75 expression was detected in HPF by immumofluorescence method.After being treated with 10-80 nmol/L trkA kinase inhibitor for 24-96 h,the growth activities of HPF were studied by MTT colorimetry.Caspase-3 was inspected in HPF by spectrophotometric method.The apoptosis of HPF was detected by flow cytometry.Results:Expression of trkA and p75 was significantly observed in epithelium and fibroblast of pterygium tissues.Expression of trkA was observed in epithelium and fibroblast of normal conjunctiva.Expression of p75 was only observed in epithelium of normal conjunctiva.trkA kinase inhibitor could effectively inhibit the growth of human pterygium fibroblasts in vitro in time and dose dependent manners.Caspase-3 expression was increased.The rates of apoptosis were 8.26%-29.62%(P < 0.01).Conclusion:TrkA and p75 possibly participate in genesis and development of pterygium.trkA kinase inhibitor could induce apoptosis of human pterygium fibroblasts.Inducing apoptosis through changing the ratio of trkA and p75 in fibroblasts and up-regulate Caspase-3 was probably one of its molecular mechanisms. 展开更多

To the Editor:Anterior chamber(AC)paracentesis is a commonly performed procedure to lower intraocular pressure(IOP),and to sample aqueous humor(AH)for clinical and research purposes.As suggested by an international collaboration team,an immediately performed AC paracentesis before an intravitreal injection can also prevent post-injection IOP spike.[1]While IOP could immediately be reduced by AC paracentesis,the procedure of obtaining an AH sample(liquid biopsy)is more complicated.Several methods have been developed to facilitate AH collection and transfer,[2-7]among which a vacuum-based AH collector(SnovoDAHC I-50,Sightnovo Medical Technology Co.,Beijing,China)has better performance than the commonly used insulin syringe in accurate sampling of about 50μL AH in rabbits by one hand.[7]The current clinical trial aimed to evaluate the performance and safety of this product in human subjects. 展开更多

Sudden unexplained death(SUD)remains a puzzle in forensic medicine.Desmoglein‑2(DSG2)has been linked to arrhythmogenic right ventricular cardiomyopathy which may cause life‑threatening ventricular arrhythmias and sudden death.Fatal arrhythmias resulting in sudden death also occur in the absence of morphologic cardiac abnormalities at autopsy.We hypothesized that DSG2 mutations may be responsible for certain Chinese SUD cases.We sequenced all 15 exons of DSG2 in DNA extracted from postmortem heart tissues of 25 Chinese patients dying from SUD.The primers were designed using the Primer Express 3.0 software.Direct sequencing for both sense and antisense strands was performed with a BigDye Terminator DNA sequencing kit on a 3130 Xl Genetic Analyzer.Mutation damage prediction was made using Mutation Taster,PolyPhen,and SIFT software.In 2 of 25 cases of Chinese SUD samples,two DSG2 heterozygous mutations(p.P927 L and p.T1070M)were identified,and one is probably damaging.We concluded that DSG2 mutations may be related to the occurrence of part of SUD cases in the Chinese Han population. 展开更多
