BACKGROUND Perivascular epithelioid cell tumor(PEComa)is a rare mesenchymal neoplasm that predominantly affects the kidney and uterus.The occurrence of this tumor in the liver,particularly with simultaneous involvemen...BACKGROUND Perivascular epithelioid cell tumor(PEComa)is a rare mesenchymal neoplasm that predominantly affects the kidney and uterus.The occurrence of this tumor in the liver,particularly with simultaneous involvement of the liver and kidney,is exceedingly uncommon.Pathological diagnosis is the gold standard.PEComas usually show positive immunohistochemical staining for melanocytic(HMB-45,Melan-A)and myoid(SMA,muscle-specific actin)markers.CASE SUMMARY We presented a noteworthy case of malignant PEComa affecting both the liver and kidney in a 53-year-old man with tuberous sclerosis complex(TSC).FAT2 and TP73 mutations in the kidney were identified and positive expression of diagnostic markers including HMB-45,Melan A,and TFE3 were detected.In addition,we demonstrated that hepatic artery perfusion chemotherapy was ineffective for hepatic PEComa,while surgery remained the most effective approach.Everolimus showed an excellent efficacy in the postoperative treatment of the tumor.CONCLUSION Surgical treatment is preferred for malignant PEComa affecting liver and kidney,especially with TSC;everolimus is effective postoperatively.展开更多

The major risk gro...The first case of AIDS was reported in 1985 in China, but by the early 21st century, the government estimated that there were 840,000 citizens living with HIV/AIDS. The number is increasing rapidly. The major risk groups are injection drug users (IDUSs; 43%) and former plasma donors (27%), but rates among heterosexual groups are rising rapidly. Sentinel surveillance was initiated in 1986, and now includes IDUs, men-who-have-sex-with-men, sexually transmitted disease clinic attendees, antenatal women, long-distance truck drivers, and sex workers. Although the government was slow to respond to the epidemic in the late 20th century, it has made a vigorous response in the early 21st century. Components of that response include implementation and evaluation of harm reduction programs for IDUs, education to increase knowledge and reduce stigma, treatment and social support for rural and poor HIV/AIDS patients, widespread testing, and increased funding for HIV/AIDS programs. International agencies have been generous in their support of the government initiatives. To successfully combat the epidemic, China needs to develop and train the necessary infra- structure to implement its intervention programs, particularly in the rural areas, to vigorously combat stigma and discrimination, support research especially in the universities and research institutions other than the China Centers for Disease Control, develop a system for efficient exchange of research and program information, and update legislation to reflect the current situation.展开更多
The relationship between parental genetic differences and the quality and yield of japonica hybrid rice strongly influences japonica hybrid rice breeding.In this study,137 parental lines of japonica hybrid rice were g...The relationship between parental genetic differences and the quality and yield of japonica hybrid rice strongly influences japonica hybrid rice breeding.In this study,137 parental lines of japonica hybrid rice were genotyped using 8 K rice SNP-Chips to characterize their genetic diversity,population structure,and indica-genotype proportion.The genetic diversity of total parental lines averaged 0.264,with values of0.287 for restorer lines and 0.148 for the sterile lines.The introduction of indica lineage increased the genetic diversity of restorer lines relative to that of sterile lines.By model-based population structure analysis,the 137 lines were divided into 14 groups.According to the grouping results,eight restorer lines and five sterile lines were selected from different groups for cross breeding,yielding 40 japonica hybrid rice combinations(F1).Investigation of the yield and quality of these combinations showed that highyield combinations could be obtained by increasing parental genetic distance to 0.8–0.9,a result accomplished largely by the introduction of indica genomic components of restorer lines.To further improve grain quality,the genetic distance between parents should be reduced to 0.4–0.5,suggesting an indicagenotype proportion of 30%–40%for restorer lines.This study may provide a reference for breeding of japonica hybrid rice.展开更多

With the help of communication interaction system to supervise the implementation of infection prevention and control, and observe the incorrect situation of pee use and personal behavior before and after the implementation. The disinfection robot and meal delivery robot were used in ward disinfection and life nursing combined with nursing practice. <strong>Result:</strong> Through the supervision of communication interaction system, the frequency of pee use and personal behavior was reduced. The frequency of bad articles before and after improvement was wearing protective clothing (2.80%/0.84%), taking off protective clothing (5.87%/0.84%), personal behavior observation (8.38%/1.90%), P < 0.01. The robot disinfected and delivered medicine for 912.5 h, saving 225 shifts of nursing staff. <strong>Conclusion:</strong> Intelligent equipment is a good option for infection control in isolation ward of COVID-2019. It can not only reduce the workload of health workers, but also the cross-infection.展开更多

Although the overall prevalence rate is still low, with a population of 1.3 billion, high-risk factors which have contributed to the HIV/AIDS epidemics worldwide continue to prevail in China, including a high rate of injecting drug use and needle sharing, commercial sex with low rates of condom use, and concurrent sex with both commercial sex workers and noncommercial casual or steady sex partners. In addition, there are increasing “double risk” populations overlapping drug users and sex workers, as well as increasing rates of STDs and HIV among high-risk populations. Sexual transmission, therefore, may serve as a bridge connecting high-risk populations with general populations. Panicle traits directly associated with yield are a target of selection in rice breeding.Although abundant QTL for panicle traits have been identified,there is little information about the genetic basis of panicle tra...Panicle traits directly associated with yield are a target of selection in rice breeding.Although abundant QTL for panicle traits have been identified,there is little information about the genetic basis of panicle traits in japonica super rice(JSR)cultivars.In this study,we identified QTL for panicle traits in three environments using a population of recombinant inbred lines(RILs)derived from the JSR cultivar Liaoxing 1.A total of 197 RILs were genotyped with 285 polymorphic SNP markers.Phenotypic data and best linear unbiased prediction(BLUP)value of primary branch number(BNP),secondary branch number(BNS),grain number on primary branch(GNP),grain number on secondary branch(GNS),grain number per panicle(GN),panicle length(PL)and grain density(GD)were used for QTL mapping.A total of 105 QTL for seven panicle traits were detected in single environments using their BLUP values.Individual QTL explained 0.51%–52.22%of the phenotypic variation.Of the 105,49 were also detected by joint multi-environment analyses.Five stable QTL:qGD9,qPL9,qGNP9,qGN6,and qBNS6.2 were identified in multiple environments.qGD9,qGNP9,and qPL9,co-localizing on chromosome 9,likely correspond to the known gene DEP1.Importantly,qGN6 and qBNS6.2 in a co-localization region were identified as novel QTL,and their Liaoxing 1 alleles had a positive effect.Several RILs with the QTL allele combinations qGD9/qPL9/qGNP9 and qGN6/qBNS6.2 showed greater GN.Further investigation of the putative gene underlying qGN6/qBNS6.2 would shed light on the molecular mechanism of JSR.展开更多

Rare earth elements(REEs)are used for the developme nt of new energy materials owing to their intrinsic physicochemical property.However,excess REEs in water threaten the safety of animals,plants and humans.An efficie...Rare earth elements(REEs)are used for the developme nt of new energy materials owing to their intrinsic physicochemical property.However,excess REEs in water threaten the safety of animals,plants and humans.An efficient way to separate REEs from the water is therefore needed.In this study,a biosorbent consisting of iron oxide(Fe3 O4),persimmon tannin(PT),and graphene oxide(GO)as Fe3 O4/PT/GO was prepared,and the adsorption of trivalent erbium(Er3+)ions from aqueous solution was investigated.The adsorption process for Er3+ions conforms to pseudo-second order kinetic and the Langmuir isotherm model behavior.Thermodynamic studies indicate that the adsorption process is spontaneous and endothermic.Scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD),thermogravimetric analysis(TGA).Fourier-transform infrared(FT-IR)spectroscopy;Brunauer-Emmett-Teller(BET)analysis,and vibrating sample magnetometer(VSM)were used to assess the adsorption mechanism of Er3+ions onto the Fe3 O4/PT/GO biosorbent.A combination of electrostatic interactions,redox reactivity and chelation are responsible for adsorption of Er3+ions on the Fe3 O4/PT/GO biosorbent,The magnetic Fe3 O4/PT/GO biosorbent can be easily separated under the magnetic field for effective recycle of Er3+ions from aqueous solution.Therefore,this new biomass composite holds great promise for wastewater treatment.展开更多
·AIM:To compare the ciliary body thickness between eyes with primary angle closure(PAC)and primary angle-closure glaucoma(PACG)with the normal eyes,and to investigate the association between ciliary body thicknes...·AIM:To compare the ciliary body thickness between eyes with primary angle closure(PAC)and primary angle-closure glaucoma(PACG)with the normal eyes,and to investigate the association between ciliary body thickness and ciliary processes situation.·METHODS:In this cross-sectional study,57 patients with PAC/PACG were matched to 57 normal subjects after propensity score matching(PSM)adjusting for age and gender.All subjects underwent conventional ocular examinations and ultrasound biomicroscopy(UBM)examination,among which the patients with PAC/PACG performed the examinations one month after laser peripheral iridotomy(LPI).Quantitative parameters were measured,which included ciliary body thickness at the position of 1 mm posterior to the scleral spur(CBT1),trabecular-ciliary process distance(TCPD)and trabecularciliary process angle(TCA).·RESULTS:Eyes with PAC/PACG presented significantly thinner CBT1,shorter TCPD and smaller TCA(P<0.001)than the normal eyes,both in comparison of the means of four quadrants and in comparisons of each quadrant.After removing images with peripheral anterior synechia(PAS),the same results were also found in comparisons between the two groups.Significant correlations were found between TCPD(R~2=0.537,P<0.001)and TCA(R~2=0.517,P<0.001)with CBT1.·CONCLUSION:Eyes with PAC/PACG have thinner ciliary body thickness and more anteriorly situated ciliary processes.Thinner ciliary body thickness is associated with anterior situation of the ciliary processes.展开更多
AIM:To evaluate the morphological changes in anterior segment in Chinese patients with uveal effusion(UE)after the attack of acute primary angle-closure(APAC)using ultrasound biomicroscopy(UBM),and to assess th...AIM:To evaluate the morphological changes in anterior segment in Chinese patients with uveal effusion(UE)after the attack of acute primary angle-closure(APAC)using ultrasound biomicroscopy(UBM),and to assess the clinical course and prognosis of the disease.METHODS:In a retrospective case series,26 eyes in 26 consecutive patients diagnosed with UE after the treatment of intraocular pressure(IOP)-lowering medication for the attack of APAC were enrolled. The unaffected fellow eyes served as controls. The morphological changes were observed by ultrasonography,slit lamp microscopy and gonioscopy. UBM was used to assess the degree and extent of effusion based on the analysis of parameters associated with UE.RESULTS:The mean IOP was 9.2(SD 2.1)mm Hg at the diagnosis of UE after IOP-lowering medication,while 14.1(SD,2.6)mm Hg in the fellow eyes(P=0.000). The anterior chamber depth(ACD)(P=0.000),angle opening distance at 500 μm(AOD500)(P〈0.01)and anterior chamber angle(ACA)(P〈0.05)were decreased significantly,while ciliary body thickness(CBT)(P〈0.05)increased significantly in UE eyes. UE grade analysis showed 7 eyes in grade 1,9 eyes in grade 2,and 10 eyes in grade 3. Quadrant scores were performed of 4 eyes in 1 quadrant,3 eyes in 3 quadrants,and 19 eyes in 4 quadrants. There was the positive correlation between grade and quadrant score(R=0.644,P=0.000). The effusion on all eyes were recovered after medication,which mean IOP was 13.9(SD,2.8)mm Hg.CONCLUSION:UE is a frequent complication in Chinese patients after the attack of APAC,partially associated with hypotony. The severity of UE is correlation with height of effusion,extent of detachment,and shallower ACD.展开更多
Background:Despite the clear link between nonalcoholic fatty liver disease(NAFLD)and type 2 diabetes mellitus,little is understood about how glycemic control impacts histological severity.We aimed to investigate the d...Background:Despite the clear link between nonalcoholic fatty liver disease(NAFLD)and type 2 diabetes mellitus,little is understood about how glycemic control impacts histological severity.We aimed to investigate the deeper association between hemoglobin A1c(HbA1c)and the histologic severity of liver fibrosis.Methods:A total of 568 adults with biopsy-proven NAFLD from the PERSONS cohort in Wenzhou were enrolled.The association between mean HbA1c and hepatic histological features was investigated with ordinal logistic regression.Generalized additive models were used to identify the non-linear relationship between HbA1c and increased fibrosis stage(stage F3).Causal mediation analysis was performed to calculate the indirect effect of the relationship between HbA1c and hepatic fibrosis mediated by cytokines.Results:Every 1%increase in mean HbA1c was associated with 16%higher odds of increased fibrosis stage(odds ratio 1.16,95%CI 1.04–1.30),even after adjustment for confounding factors.There was a non-linear association between HbA1c and increased fibrosis stage(stage F3),and the HbA1c inflection point was 9.2%.In particular,the ORs on the left and right sides of this inflection point were 2.1(95%CI 1.4–3.3)and 0.1(95%CI 0–4.8),respectively.7%of the association(OR 1.07,95%CI 1.01–1.12)between HbA1c and liver fibrosis was mediated by leukemia inhibitory factor.Conclusions:Glycemic control predicts severity of hepatic fibrosis and leukemia inhibitory factor plays an intermediary role between them,and thus optimizing glycemic control may be a means of modifying the risk of fibrosis progression in NAFLD.展开更多
Objective:To investigate the inhibitory effect on Burkholderia pseudomallei(B.pseudomallei)strain HNBP001 of a bacillomycin D-like cyclic lipopeptide compound named bacillomycin DC isolated from Bacillus amyloliquefac...Objective:To investigate the inhibitory effect on Burkholderia pseudomallei(B.pseudomallei)strain HNBP001 of a bacillomycin D-like cyclic lipopeptide compound named bacillomycin DC isolated from Bacillus amyloliquefaciens HAB-2.Methods:The antibacterial effect of bacillomycin DC on B.pseudomallei was determined using the disk diffusion method.The minimum inhibitory concentrations were evaluated by microdilution assay.In addition,transmission electron microscopy was performed and quantitative real-time polymerase chain reaction assay was carried out to determine the expression of Mex B,Opr D2,and qnr S genes.Results:Bacillomycin DC produced an inhibition zone against B.pseudomallei with minimum inhibitory concentration values of 12.5μg/mL 24 h after treatment and 50μg/mL at 48 and 72 h.Transmission electron microscopy showed that bacillomycin DC resulted in roughening cell surface and cell membrane damage.Quantitative real-time polymerase chain reaction analysis showed low expression of Mex B,Opr D2 and qnr S genes.Conclusions:Bacillomycin DC inhibits the growth of B.pseudomallei and can be a new candidate for antimicrobial agents of B.pseudomallei.展开更多

Forty-one epilepsyassociated ion channel genes and their mutations are systematically reviewed. In this paper, we analyzed the genotypes, functional alterations(funotypes), and phenotypes of these mutations. Eleven genes featured loss-offunction mutations and six had gain-of-function mutations.Nine genes displayed diversified funotypes, among which a distinct funotype-phenotype correlation was found in SCN1A. These data suggest that the funotype is an essential consideration in evaluating the pathogenicity of mutations and a distinct funotype or funotype-phenotype correlation helps to define the pathogenic potential of a gene.展开更多
Background The prevalence of zinc deficiency is high in patients with chronic liver disease,but few studies have hitherto explored the relationship between the serum zinc level and hepatitis B virus(HBV)-related acute...Background The prevalence of zinc deficiency is high in patients with chronic liver disease,but few studies have hitherto explored the relationship between the serum zinc level and hepatitis B virus(HBV)-related acute-on-chronic liver failure(ACLF).This study aimed to assess the association between zinc deficiency and infectious complications,and model for end-stage liver disease(MELD)score in patients with HBV-related ACLF.Methods Patients with HBV-related ACLF from the Department of Infectious Diseases of the Third Affiliated Hospital of Sun Yat-sen University(Guangzhou,China)between January 2019 and December 2019 were retrospectively analysed in this study.Their demographic,clinical,and laboratory data were retrieved from the hospital information system and analysed.The Student’s t-test was used for normally distributed continuous variables between two groups and the Chi-square test was used for categorical data.Univariate and multivariate logistic regression analyses were applied to identify independent parameters.Results A total of 284 patients were included in this study,including 205 liver cirrhosis and 79 non-cirrhosis patients.The proportion of patients with zinc deficiency was the highest(84.5%),followed by subclinical zinc deficiency(14.1%)and normal zinc level(1.4%).Patients in the zinc deficiency group had a higher MELD score than the subclinical zinc deficiency or normal zinc group(P=0.021).Age,total bilirubin,and serum zinc level were independent factors for infection(P_(s)<0.05).The serum zinc level in patients without complications at admission was significantly higher than that in patients with complications(P=0.004).Moreover,the serum zinc level in patients with prothrombin time activity(PTA)of<20%was significantly lower than that in patients with 20%≤PTA<30%(P=0.007)and that in patients with 30%≤PTA<40%(P<0.001).Conclusions Zinc deficiency is common in patients with HBV-related ACLF.Zinc deficiency is closely associated with infectious complications and MELD score in patients with HBV-related ACLF.展开更多

Self-transcribing active regulatory region sequencing (STARR-seq) measures the enhancer activity of millions of DNA fragments in parallel. Here we used STARR-seq to generate a quantitative global map of rice enhancers. Most enhancers were mapped within genes,especially at the 5' untranslated regions (5'UTR) and in coding sequences. Enhancers were also frequently mapped proximal to silent and lowly-expressed genes in transposable element (TE)-rich regions. Analysis of the epigenetic features of enhancers at their endogenous loci revealed that most enhancers do not co-localize with DNase I hypersensitive sites (DHSs) and lack the enhancer mark of histone modification H3K4me1. Clustering analysis of enhancers according to their epigenetic marks revealed that about 40%of identified enhancers car-ried one or more epigenetic marks. Repressive H3K27me3 was frequently enriched with positive marks,H3K4me3 and/or H3K27ac,which together label enhancers. Intergenic enhancers were also predicted based on the location of DHS regions relative to genes,which overlap poorly with STARR-seq enhancers. In summary,we quantitatively identified enhancers by functional analysis in the genome of rice,an important model plant. This work provides a valuable resource for further mechanistic studies in different biological contexts.展开更多

文摘The relationship between parental genetic differences and the quality and yield of japonica hybrid rice strongly influences japonica hybrid rice breeding.In this study,137 parental lines of japonica hybrid rice were genotyped using 8 K rice SNP-Chips to characterize their genetic diversity,population structure,and indica-genotype proportion.The genetic diversity of total parental lines averaged 0.264,with values of0.287 for restorer lines and 0.148 for the sterile lines.The introduction of indica lineage increased the genetic diversity of restorer lines relative to that of sterile lines.By model-based population structure analysis,the 137 lines were divided into 14 groups.According to the grouping results,eight restorer lines and five sterile lines were selected from different groups for cross breeding,yielding 40 japonica hybrid rice combinations(F1).Investigation of the yield and quality of these combinations showed that highyield combinations could be obtained by increasing parental genetic distance to 0.8–0.9,a result accomplished largely by the introduction of indica genomic components of restorer lines.To further improve grain quality,the genetic distance between parents should be reduced to 0.4–0.5,suggesting an indicagenotype proportion of 30%–40%for restorer lines.This study may provide a reference for breeding of japonica hybrid rice.
文摘<strong>Objective:</strong> To explore the application of intelligent equipment in non-negative pressure isolation ward for COVID-2019 patients. <strong>Method:</strong> From February 1 to March 17, 2020, intelligent equipment, such as communication interaction system, intelligent disinfection robot, delivering robot, were used in non- negative pressure isolation ward of COVID-2019. With the help of communication interaction system to supervise the implementation of infection prevention and control, and observe the incorrect situation of pee use and personal behavior before and after the implementation. The disinfection robot and meal delivery robot were used in ward disinfection and life nursing combined with nursing practice. <strong>Result:</strong> Through the supervision of communication interaction system, the frequency of pee use and personal behavior was reduced. The frequency of bad articles before and after improvement was wearing protective clothing (2.80%/0.84%), taking off protective clothing (5.87%/0.84%), personal behavior observation (8.38%/1.90%), P < 0.01. The robot disinfected and delivered medicine for 912.5 h, saving 225 shifts of nursing staff. <strong>Conclusion:</strong> Intelligent equipment is a good option for infection control in isolation ward of COVID-2019. It can not only reduce the workload of health workers, but also the cross-infection.

文摘China is currently experiencing one of the most rapidly expanding HIV epidemics in the world. Although the overall prevalence rate is still low, with a population of 1.3 billion, high-risk factors which have contributed to the HIV/AIDS epidemics worldwide continue to prevail in China, including a high rate of injecting drug use and needle sharing, commercial sex with low rates of condom use, and concurrent sex with both commercial sex workers and noncommercial casual or steady sex partners. In addition, there are increasing “double risk” populations overlapping drug users and sex workers, as well as increasing rates of STDs and HIV among high-risk populations. Sexual transmission, therefore, may serve as a bridge connecting high-risk populations with general populations. There is an urgent need to prevent the spread of HIV from these high-risk oooulations into the general oooulation of China.

文摘Panicle traits directly associated with yield are a target of selection in rice breeding.Although abundant QTL for panicle traits have been identified,there is little information about the genetic basis of panicle traits in japonica super rice(JSR)cultivars.In this study,we identified QTL for panicle traits in three environments using a population of recombinant inbred lines(RILs)derived from the JSR cultivar Liaoxing 1.A total of 197 RILs were genotyped with 285 polymorphic SNP markers.Phenotypic data and best linear unbiased prediction(BLUP)value of primary branch number(BNP),secondary branch number(BNS),grain number on primary branch(GNP),grain number on secondary branch(GNS),grain number per panicle(GN),panicle length(PL)and grain density(GD)were used for QTL mapping.A total of 105 QTL for seven panicle traits were detected in single environments using their BLUP values.Individual QTL explained 0.51%–52.22%of the phenotypic variation.Of the 105,49 were also detected by joint multi-environment analyses.Five stable QTL:qGD9,qPL9,qGNP9,qGN6,and qBNS6.2 were identified in multiple environments.qGD9,qGNP9,and qPL9,co-localizing on chromosome 9,likely correspond to the known gene DEP1.Importantly,qGN6 and qBNS6.2 in a co-localization region were identified as novel QTL,and their Liaoxing 1 alleles had a positive effect.Several RILs with the QTL allele combinations qGD9/qPL9/qGNP9 and qGN6/qBNS6.2 showed greater GN.Further investigation of the putative gene underlying qGN6/qBNS6.2 would shed light on the molecular mechanism of JSR.

文摘Rare earth elements(REEs)are used for the developme nt of new energy materials owing to their intrinsic physicochemical property.However,excess REEs in water threaten the safety of animals,plants and humans.An efficient way to separate REEs from the water is therefore needed.In this study,a biosorbent consisting of iron oxide(Fe3 O4),persimmon tannin(PT),and graphene oxide(GO)as Fe3 O4/PT/GO was prepared,and the adsorption of trivalent erbium(Er3+)ions from aqueous solution was investigated.The adsorption process for Er3+ions conforms to pseudo-second order kinetic and the Langmuir isotherm model behavior.Thermodynamic studies indicate that the adsorption process is spontaneous and endothermic.Scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD),thermogravimetric analysis(TGA).Fourier-transform infrared(FT-IR)spectroscopy;Brunauer-Emmett-Teller(BET)analysis,and vibrating sample magnetometer(VSM)were used to assess the adsorption mechanism of Er3+ions onto the Fe3 O4/PT/GO biosorbent.A combination of electrostatic interactions,redox reactivity and chelation are responsible for adsorption of Er3+ions on the Fe3 O4/PT/GO biosorbent,The magnetic Fe3 O4/PT/GO biosorbent can be easily separated under the magnetic field for effective recycle of Er3+ions from aqueous solution.Therefore,this new biomass composite holds great promise for wastewater treatment.
文摘·AIM:To compare the ciliary body thickness between eyes with primary angle closure(PAC)and primary angle-closure glaucoma(PACG)with the normal eyes,and to investigate the association between ciliary body thickness and ciliary processes situation.·METHODS:In this cross-sectional study,57 patients with PAC/PACG were matched to 57 normal subjects after propensity score matching(PSM)adjusting for age and gender.All subjects underwent conventional ocular examinations and ultrasound biomicroscopy(UBM)examination,among which the patients with PAC/PACG performed the examinations one month after laser peripheral iridotomy(LPI).Quantitative parameters were measured,which included ciliary body thickness at the position of 1 mm posterior to the scleral spur(CBT1),trabecular-ciliary process distance(TCPD)and trabecularciliary process angle(TCA).·RESULTS:Eyes with PAC/PACG presented significantly thinner CBT1,shorter TCPD and smaller TCA(P<0.001)than the normal eyes,both in comparison of the means of four quadrants and in comparisons of each quadrant.After removing images with peripheral anterior synechia(PAS),the same results were also found in comparisons between the two groups.Significant correlations were found between TCPD(R~2=0.537,P<0.001)and TCA(R~2=0.517,P<0.001)with CBT1.·CONCLUSION:Eyes with PAC/PACG have thinner ciliary body thickness and more anteriorly situated ciliary processes.Thinner ciliary body thickness is associated with anterior situation of the ciliary processes.

文摘AIM:To evaluate the morphological changes in anterior segment in Chinese patients with uveal effusion(UE)after the attack of acute primary angle-closure(APAC)using ultrasound biomicroscopy(UBM),and to assess the clinical course and prognosis of the disease.METHODS:In a retrospective case series,26 eyes in 26 consecutive patients diagnosed with UE after the treatment of intraocular pressure(IOP)-lowering medication for the attack of APAC were enrolled. The unaffected fellow eyes served as controls. The morphological changes were observed by ultrasonography,slit lamp microscopy and gonioscopy. UBM was used to assess the degree and extent of effusion based on the analysis of parameters associated with UE.RESULTS:The mean IOP was 9.2(SD 2.1)mm Hg at the diagnosis of UE after IOP-lowering medication,while 14.1(SD,2.6)mm Hg in the fellow eyes(P=0.000). The anterior chamber depth(ACD)(P=0.000),angle opening distance at 500 μm(AOD500)(P〈0.01)and anterior chamber angle(ACA)(P〈0.05)were decreased significantly,while ciliary body thickness(CBT)(P〈0.05)increased significantly in UE eyes. UE grade analysis showed 7 eyes in grade 1,9 eyes in grade 2,and 10 eyes in grade 3. Quadrant scores were performed of 4 eyes in 1 quadrant,3 eyes in 3 quadrants,and 19 eyes in 4 quadrants. There was the positive correlation between grade and quadrant score(R=0.644,P=0.000). The effusion on all eyes were recovered after medication,which mean IOP was 13.9(SD,2.8)mm Hg.CONCLUSION:UE is a frequent complication in Chinese patients after the attack of APAC,partially associated with hypotony. The severity of UE is correlation with height of effusion,extent of detachment,and shallower ACD.

文摘Background:Despite the clear link between nonalcoholic fatty liver disease(NAFLD)and type 2 diabetes mellitus,little is understood about how glycemic control impacts histological severity.We aimed to investigate the deeper association between hemoglobin A1c(HbA1c)and the histologic severity of liver fibrosis.Methods:A total of 568 adults with biopsy-proven NAFLD from the PERSONS cohort in Wenzhou were enrolled.The association between mean HbA1c and hepatic histological features was investigated with ordinal logistic regression.Generalized additive models were used to identify the non-linear relationship between HbA1c and increased fibrosis stage(stage F3).Causal mediation analysis was performed to calculate the indirect effect of the relationship between HbA1c and hepatic fibrosis mediated by cytokines.Results:Every 1%increase in mean HbA1c was associated with 16%higher odds of increased fibrosis stage(odds ratio 1.16,95%CI 1.04–1.30),even after adjustment for confounding factors.There was a non-linear association between HbA1c and increased fibrosis stage(stage F3),and the HbA1c inflection point was 9.2%.In particular,the ORs on the left and right sides of this inflection point were 2.1(95%CI 1.4–3.3)and 0.1(95%CI 0–4.8),respectively.7%of the association(OR 1.07,95%CI 1.01–1.12)between HbA1c and liver fibrosis was mediated by leukemia inhibitory factor.Conclusions:Glycemic control predicts severity of hepatic fibrosis and leukemia inhibitory factor plays an intermediary role between them,and thus optimizing glycemic control may be a means of modifying the risk of fibrosis progression in NAFLD.

文摘Objective:To investigate the inhibitory effect on Burkholderia pseudomallei(B.pseudomallei)strain HNBP001 of a bacillomycin D-like cyclic lipopeptide compound named bacillomycin DC isolated from Bacillus amyloliquefaciens HAB-2.Methods:The antibacterial effect of bacillomycin DC on B.pseudomallei was determined using the disk diffusion method.The minimum inhibitory concentrations were evaluated by microdilution assay.In addition,transmission electron microscopy was performed and quantitative real-time polymerase chain reaction assay was carried out to determine the expression of Mex B,Opr D2,and qnr S genes.Results:Bacillomycin DC produced an inhibition zone against B.pseudomallei with minimum inhibitory concentration values of 12.5μg/mL 24 h after treatment and 50μg/mL at 48 and 72 h.Transmission electron microscopy showed that bacillomycin DC resulted in roughening cell surface and cell membrane damage.Quantitative real-time polymerase chain reaction analysis showed low expression of Mex B,Opr D2 and qnr S genes.Conclusions:Bacillomycin DC inhibits the growth of B.pseudomallei and can be a new candidate for antimicrobial agents of B.pseudomallei.

文摘Ion channels are crucial in the generation and modulation of excitability in the nervous system and have been implicated in human epilepsy. Forty-one epilepsyassociated ion channel genes and their mutations are systematically reviewed. In this paper, we analyzed the genotypes, functional alterations(funotypes), and phenotypes of these mutations. Eleven genes featured loss-offunction mutations and six had gain-of-function mutations.Nine genes displayed diversified funotypes, among which a distinct funotype-phenotype correlation was found in SCN1A. These data suggest that the funotype is an essential consideration in evaluating the pathogenicity of mutations and a distinct funotype or funotype-phenotype correlation helps to define the pathogenic potential of a gene.

文摘Background The prevalence of zinc deficiency is high in patients with chronic liver disease,but few studies have hitherto explored the relationship between the serum zinc level and hepatitis B virus(HBV)-related acute-on-chronic liver failure(ACLF).This study aimed to assess the association between zinc deficiency and infectious complications,and model for end-stage liver disease(MELD)score in patients with HBV-related ACLF.Methods Patients with HBV-related ACLF from the Department of Infectious Diseases of the Third Affiliated Hospital of Sun Yat-sen University(Guangzhou,China)between January 2019 and December 2019 were retrospectively analysed in this study.Their demographic,clinical,and laboratory data were retrieved from the hospital information system and analysed.The Student’s t-test was used for normally distributed continuous variables between two groups and the Chi-square test was used for categorical data.Univariate and multivariate logistic regression analyses were applied to identify independent parameters.Results A total of 284 patients were included in this study,including 205 liver cirrhosis and 79 non-cirrhosis patients.The proportion of patients with zinc deficiency was the highest(84.5%),followed by subclinical zinc deficiency(14.1%)and normal zinc level(1.4%).Patients in the zinc deficiency group had a higher MELD score than the subclinical zinc deficiency or normal zinc group(P=0.021).Age,total bilirubin,and serum zinc level were independent factors for infection(P_(s)<0.05).The serum zinc level in patients without complications at admission was significantly higher than that in patients with complications(P=0.004).Moreover,the serum zinc level in patients with prothrombin time activity(PTA)of<20%was significantly lower than that in patients with 20%≤PTA<30%(P=0.007)and that in patients with 30%≤PTA<40%(P<0.001).Conclusions Zinc deficiency is common in patients with HBV-related ACLF.Zinc deficiency is closely associated with infectious complications and MELD score in patients with HBV-related ACLF.

文摘Enhancers activate transcription in a distance-,orientation-,and position-independent manner,which makes them difficult to be identified. Self-transcribing active regulatory region sequencing (STARR-seq) measures the enhancer activity of millions of DNA fragments in parallel. Here we used STARR-seq to generate a quantitative global map of rice enhancers. Most enhancers were mapped within genes,especially at the 5' untranslated regions (5'UTR) and in coding sequences. Enhancers were also frequently mapped proximal to silent and lowly-expressed genes in transposable element (TE)-rich regions. Analysis of the epigenetic features of enhancers at their endogenous loci revealed that most enhancers do not co-localize with DNase I hypersensitive sites (DHSs) and lack the enhancer mark of histone modification H3K4me1. Clustering analysis of enhancers according to their epigenetic marks revealed that about 40%of identified enhancers car-ried one or more epigenetic marks. Repressive H3K27me3 was frequently enriched with positive marks,H3K4me3 and/or H3K27ac,which together label enhancers. Intergenic enhancers were also predicted based on the location of DHS regions relative to genes,which overlap poorly with STARR-seq enhancers. In summary,we quantitatively identified enhancers by functional analysis in the genome of rice,an important model plant. This work provides a valuable resource for further mechanistic studies in different biological contexts.
