MetS affects 20% of adults in the Western world and 33% of adults in China[2] and has become a serious public health problem worldwide. However, the mechanism underlying the occurrence and progression of MetS is still largely unclear. It is now well established that excess fat deposition leads to abdominal obesity, which plays a vital role in the underlying mechanism. Adipose tissue can function as an endocrine organ that secretes various adipokines. The dysregulated expression of adipokines caused by excess adiposity and adipocyte dysfunction, has been linked to the pathogenesis of MetS[3]. AIM: To investigate the apoptotic effects of melittin on SGC-7901 cells via activation of the mitochondrial signaling pathway in vitro.METHODS: SGC-7901 cells were stimulated by melittin, and its effect on proliferati...AIM: To investigate the apoptotic effects of melittin on SGC-7901 cells via activation of the mitochondrial signaling pathway in vitro.METHODS: SGC-7901 cells were stimulated by melittin, and its effect on proliferation and apoptosis of was investigated by methyl thiazolyl tetrazolium assay, morphologic structure with transmission electron microscopy, annexin-V/propidium iodide double-staining assay, measuring mitochondrial membrane potential(MMP) levels, and analyzing reactive oxygen species(ROS) concentrations were analyzed by flow cytometry. Cytochrome C(Cyt C), apoptosis-inducing factor(AIF), endonuclease G(Endo G), second mitochondria-derived activator of caspases(Smac)/direct IAP binding protein with low isoelectric point(Diablo), and FAS were analyzed by western blot. The expression of caspase-3 and caspase-8 was measured using activity assay kits.RESULTS: Melittin was incubated at 1.0, 2.0, 4.0, or 6.0 μg/m L for 1, 2, 4, 6, or 8 h and showed a timeand concentration-dependent inhibition of SGC-7901 cell growth. Melittin induced SGC-7901 cell apoptosis, which was confirmed by typical morphological changes. Treatment with 4 μg/m L melittin induced early apoptosis of SGC-7901 cells, and the early apoptosis rates were 39.97% ± 3.19%, 59.27% ± 3.94%, and 71.50% ± 2.87% vs 32.63% ± 2.75% for 1, 2, and 4 h vs 0 h(n = 3, P < 0.05); the ROS levels were 616.53% ± 79.78%, 974.81% ± 102.40%, and 1330.94% ± 93.09% vs 603.74% ± 71.99%(n = 3, P < 0.05); the MMP values were 2.07 ± 0.05, 1.78 ± 0.29, and 1.16 ± 0.25 vs 2.55 ± 0.42(n = 3, P < 0.05); caspase-3 activity was significantly higher compared to the control(5492.3 ± 321.1, 6562.0 ± 381.3, and 8695.7 ± 449.1 vs 2330.0 ± 121.9), but the caspase activity of the non-tumor cell line L-O2 was not different from that of the control. With the addition of the caspase-3 inhibitor(Ac-DEVD-CHO), caspase-3 activity was significantly decreased compared to the control group(1067.0 ± 132.5 U/g vs 8695.7 ± 449.1 U/g). The expression of the Cyt C, Endo G, and AIF proteins in SGC-7901 cells was significantly higher than those in the control(P < 0.05), while the expression of the Smac/Diablo protein was significantly lower than the control group after melittin exposure(P < 0.01). Ac-DEVD-CHO did not, however, have any effect on the expression of caspase-8 and FAS in the SGC-7901 cells.CONCLUSION: Melittin can induce apoptosis of human gastric cancer(GC) cells through the mitochondria pathways, and it may be a potent agent in the treatment of human GC.展开更多
A model (Bayesian oceanic front detection, BOFD) of sea surface temperature (SST) front detection in satel- lite-derived SST images based on a threshold interval is presented, to be used in different applications ...A model (Bayesian oceanic front detection, BOFD) of sea surface temperature (SST) front detection in satel- lite-derived SST images based on a threshold interval is presented, to be used in different applications such as climatic and environmental studies or fisheries. The model first computes the SST gradient by using a Sobel algorithm template. On the basis of the gradient value, the threshold interval is determined by a gradi- ent cumulative histogram. According to this threshold interval, front candidates can be acquired and prior probability and likelihood can be calculated. Whether or not the candidates are front points can be deter- mined by using the Bayesian decision theory. The model is evaluated on the Advanced Very High-Resolution Radiometer images of part of the Kuroshio front region. Results are compared with those obtained by using several SST front detection methods proposed in the literature. This comparison shows that the BOFD not only suppresses noise and small-scale fronts, but also retains continuous fronts.展开更多

METHODS: RAW 264.7 macrophages were used as a target cell line. Cell viability after treatment with quercetin at different time points was detected by Carboxyfluorescein diacetate, succinimidyl ester. Phagocytic function of macrophages was deter- mined by a fluorometric assay. Cytokine tumor ne- crosis factor a (TNF-a) production was measured by enzyme-linked immunosorbent assay. F-actin cy- toskeleton of L929 cells was stained by Alexa Fluor 488-phalloidin. RESULTS: Pretreatment with quercetin decreasedcell viability only at the highest concentration of 37 μg/mL 2, 24, and 48 h after the treatment. The phagocytic efficiency of macrophages pretreated with quercetin was significantly decreased in a time- and dose-dependent manner. F-actin label- ing showed that the actin cytoskeleton of the cells started to break down 2 h after treatment. More- over, it notably inhibited cytokine TNF-a produc- tion after Candida albicans infection. CONCLUSION: Pretreatment with quercetin in- duced an anti-inflammatory effect against Candida albicans infection in macrophages.展开更多

This study aimed to investigate the neuroprotective effect of Paeoniae Radix Rubra(PRR) on ischemic stroke of mice.Methods: The focal ischemic stroke model was produced via middle cerebral artery occlusion. The experimental mice were divided into four groups: vehicle-sham group, PRR-sham group, vehicle-ischemia group, and PRR-treated ischemia group. The cerebral infarction volume was detected with TTC staining.The number of neurons in the hippocampal CA1 of the ischemic side, and the activation of astrocytes and microglia were observed via immunohistochemical staining. Western blotting was used to determine the expression changes of SOD1, SOD2, and Catalase protein levels in the hippocampus.Results: PRR significantly reduced the cerebral infarct volume induced by ischemic injury and inhibited the astrocytes and microglia activation in the hippocampal CA1 region. The decreased levels of SOD1,SOD2, and Catalase that was induced by ischemic reperfusion were simultaneously improved after PRR treatment.Conclusion: PRR improved neuronal injuries that were induced by transient cerebral ischemia via inhibiting gliosis and elevating anti-oxidants.展开更多
