目的运用液相色谱-质谱法测定茜草药材中芦西丁的含量。方法采用Agilent1290HPLC-G6460三重四级杆质谱仪,在ESI-离子模式下选择离子监测,色谱分离采用AgilentEclipsePlusC18RRHD色谱柱(2.1mm×50mm,1.8μm),以0.1%甲酸-乙腈... 目的运用液相色谱-质谱法测定茜草药材中芦西丁的含量。方法采用Agilent1290HPLC-G6460三重四级杆质谱仪,在ESI-离子模式下选择离子监测,色谱分离采用AgilentEclipsePlusC18RRHD色谱柱(2.1mm×50mm,1.8μm),以0.1%甲酸-乙腈(70∶30)为流动相,柱温30℃,流速0.4mL/min,进样量2μL。结果芦西丁在0.0052~2.64ng范围内呈良好线性关系(r=0.9999),80%、100%、120%浓度水平的加样回收率为94.80%~100.94%(平均97.17%,RSD=6.16%),方法重复性及精密度均良好。结论本研究建立的方法灵敏、准确,重复性好,适用于茜草中芦西丁的含量测定,可用于茜草药材中该成分的限度检查。 展开更多

微孔滤膜对3种常用中药指标成分HPLC测定结果的影响 被引量:5

目的研究采用不同尺寸和材质针式微孔滤膜处理中药饮片,对其中指标性成分野黄芩苷、黄芩苷和甘草酸铵HPLC定量检测结果的影响,以确定最适用的微孔滤膜类型。方法以中药饮片半枝莲、黄芩、甘草为研究对象,采用安谱、津腾、泰坦、岛津等... 目的研究采用不同尺寸和材质针式微孔滤膜处理中药饮片,对其中指标性成分野黄芩苷、黄芩苷和甘草酸铵HPLC定量检测结果的影响,以确定最适用的微孔滤膜类型。方法以中药饮片半枝莲、黄芩、甘草为研究对象,采用安谱、津腾、泰坦、岛津等不同品牌尼龙、聚四氟乙烯(PTFE)等不同材质及0.45、0.22μm等不同尺寸的有机相微孔滤膜滤过以及高速离心进行样品前处理,对3种中药成分(野黄芩苷、黄芩苷、甘草酸铵)分别按《中国药典》2020年版方法进行HPLC定量测定;同时还测定了经上述不同方法处理的不同质量浓度水平(低、中、高)对照品溶液,得到的相应色谱峰的响应值。采用主成分分析(principal component analysis,PCA)进行统计分析,比较采用不同微孔滤膜的处理方式对各定量测定结果的影响。结果与经高速离心处理后的样品测定结果相比,不同样品前处理的对照品溶液峰面积均呈现不同程度的降低。结论微孔滤膜对3种指标成分均具有不同程度的吸附作用;采用PTFE滤膜处理后的含量结果、峰面积结果与高速离心法更接近,对定量测定结果影响更小,更适用于进样前样品(对照品、供试品)溶液的滤过处理。 展开更多

Ulcerative colitis(UC), a chronic inflammatory disease affecting the colon, has a rising incidence worldwide. The known pathogenesis is multifactorial and involves genetic predisposition, epithelial barrier defects, d... Ulcerative colitis(UC), a chronic inflammatory disease affecting the colon, has a rising incidence worldwide. The known pathogenesis is multifactorial and involves genetic predisposition, epithelial barrier defects, dysregulated immune responses, and environmental factors. Nowadays, the drugs for UC include 5-aminosalicylic acid, steroids, and immunosuppressants. Long-term use of these drugs, however, may cause several side effects, such as hepatic and renal toxicity, drug resistance and allergic reactions. Moreover, the use of traditional Chinese medicine(TCM) in the treatment of UC shows significantly positive effects, low recurrence rate, few side effects and other obvious advantages. This paper summarizes several kinds of active compounds used in the experimental research of anti-UC effects extracted from TCM, mainly including flavonoids, acids, terpenoids, phenols, alkaloids, quinones, and bile acids from some animal medicines. It is found that the anti-UC activities are mainly focused on targeting inflammation or oxidative stress, which is associated with increasing the levels of anti-inflammatory cytokine(IL-4, IL-10, SOD), suppressing the levels of pro-inflammatory cytokines(TNF-α, IL-1β, IL-6, IL-8, IL-23, NF-κB, NO), reducing the activity of MPO, MDA, IFN-γ, and iNOS. This review may offer valuable reference for UC-related studies on the compounds from natural medicines. 展开更多

Phenolic compounds,the most abundant secondary metabolites in plants,have received more and more attention in recent years because of their distinct bioactivities.This review summarizes different types of phenolic compounds and their extraction and analytical methods used in the recent reports,involving 59 phenolic compounds from 52 kinds of plants.The extraction methods include solid–liquid extraction,ultrasound-assisted extractions,microwave-assisted extractions,supercritical fluid extraction,and other methods.The analysis methods include spectrophotometry,gas chromatography,liquid chromatography,thin-layer chromatography,capillary electrophoresis,and near-infrared spectroscopy.After illustrating the specific conditions of the analytical methods,the advantages and disadvantages of each method are also summarized,pointing out their respective suitability.This review provides valuable reference for identification and/or quantification of phenolic compounds from natural products. 展开更多
