目的探讨药物联合植物性护肤品治疗多形性日光疹的疗效及安全性。方法多形性日光疹患者96例,随机分为实验组(48例)和对照组(48例),所有患者均给予相同口服药(羟氯喹400 mg/d,分2次服用,氯雷他定5 mg/d,口服4周)及外用药物(糠酸莫米松乳... 目的探讨药物联合植物性护肤品治疗多形性日光疹的疗效及安全性。方法多形性日光疹患者96例,随机分为实验组(48例)和对照组(48例),所有患者均给予相同口服药(羟氯喹400 mg/d,分2次服用,氯雷他定5 mg/d,口服4周)及外用药物(糠酸莫米松乳膏外搽患处,使用2周后,改为外搽氟芬那酸丁酯软膏2周,均每天涂搽2次)。实验组患者在药物治疗的同时均使用植物性护肤品,对照组仅给予药物治疗。于治疗2周后、4周后评价2组疗效。治疗后对所有患者进行随访4周观察其复发率。根据患者临床症状和体征(包括皮损面积大小、红斑丘疹、浸润程度、瘙痒程度)进行疗效评判。结果治疗4周后,实验组有效率为47.92%,对照组有效率为33.33%,两组疗效比较差异有统计学意义(P<0.05)。随访后发现实验组复发率(37.50%)明显较对照组(75.00%)低,差异有统计学意义(P<0.05)。所有患者用药和护肤品期间均未出现严重的不良反应。结论药物联合植物性护肤品治疗多形性日光疹疗效较好,安全性高。 展开更多

These dysfunctions could be decelerated by the preventive effects o... <b>Introduction:</b> Skin aging is the result of many cellular dysfunctions over time particularly the fibroblasts and the keratinocytes. These dysfunctions could be decelerated by the preventive effects of some skin treatments such as Intradermal injections. NCTF 135HA has a polycomponent formulation designed to improve the efficacy of non-cross linked hyaluronic acid (as a micro-filler) on fibroblasts function. Although NCTF 135HA has been used by aesthetic practitioners since 20 years, we have analyzed all <i>in </i><i>vitro</i>, <i>ex vivo</i> and <i>in vivo</i> studies during past 10 years in order to summarized its anti-aging effect. <b>Methods</b><b>:</b> In these series of studies, the known effects of HA have been evaluated with this boosted formula. Collagen I synthesis, antiglycation effect and contractile forces developed by fibroblasts were studied. Fibroblasts and keratinocytes proliferations were evaluated in monolayer cells culture. The filling effect thanks to NCTF 135HA injection was <i>ex vivo </i>performed by fringe projection and the protective effect of NCTF 135HA against solar irradiation was performed <i>ex vivo</i> to study proliferation rate, elastin and collagen expression. Two clinical trials were performed on women to evaluate the anti-aging effect of NCTF 135HA injection. Hydration, firmness, radiance, wrinkles size, pore size, thickness and density of the dermis were analyzed. <b>Results</b><b>:</b> Collagen I, anti-AGE products, cell proliferation and contractile forces were significantly increased with NCTF 135HA in culture medium. <i>Ex vivo</i> studies showed that NCTF 135HA increased, cells proliferation rate, elastin and collagen production. In addition, the filling effect of the NCTF 135HA is significantly superior to placebo. The first clinical trial shows that NCTF 135HA injection significantly increased tonicity, hydration and radiance of the skin and significantly decreased wrinkle score. The second clinical trial demonstrated that NCTF 135HA injection significantly increased dermis density and thickness, skin homogeneity and skin radiance and significantly decreased pore size and wrinkle depth and volume. <b>Conclusion</b>: <i>In vitro</i>, <i>ex vivo</i> and <i>in vivo</i> studies showed that this minimally invasive technique could be used in order to improve the skin quality which could have a visual effect on preventing the aging signs. 展开更多

The purpose of the study was to evaluate the effects of corticosteroids on histamine release and to compare their potency with the MacKenzie classification based on their vasoconstrictor effects. Thanks to ex Vivo cutaneous microdialysis, we studied histamine-induced release over a period of time on excised abdominal skin from women. Eight corticosteroids were topically applied with occlusive dressing onto the skin, above probes, before anti-IgE injection. Histamine levels were assessed by an EIA method. In order to compare the different corticosteroids, AUC was calculated allowing an estimation of the amount of released histamine for 60 min of ex vivo cutaneous microdialysis. Diflucortolone 0.1% and micronized betamethasone dipropionate 0.05% are considered as corticosteroids with high potency in MacKenzie classification. Betamethasone dipropionate associated with propylene glycol 0.05%, belongs to a stronger class in Mackenzie classification. Our results showed that the decrease in histamine release was more important with difluocortolone than with both of these corticosteroids. Therefore there was no correlation between the vasoconstrictor potency of topical corticosteroids and their ability to inhibit histamine release. 展开更多

Inter-Toe Cracks: A Cosmetic Response versus Excipient. Efficacy and Tolerance Evaluation

A randomized, monocentric, double-blind, intra-individual excipient-controlled comparative study was performed to evaluate the efficacy and tolerance of an active peptide (laminin 5 fragment [LN-5]) formulated in cosmetic balm, versus excipient in inter-toe cracks. Two products were tested on 10 healthy volunteers. Each of them suffered from this particular superficial wound characterized by a peeling off a skin tab between at least two inter-toe spaces. The duration of this study belonged of 28 days with an intermediate visit to D14. Initial and outcome evaluation was performed using a 4-grade-scale depending on the severity of the inter-toe cracks. Tolerance and healing were assessed using macrophotographs. At D14 and D 28, the difference of cure between the 2 treatments was in favor of the peptide active product. The clinical score showed a best efficiency of the active compared with the excipient;indeed a significant difference between D28 and D0 was observed for the active ingredient. Active product repairs more quickly inter-toes cracks than the excipient and we noted the complete disappearance of the “severe” and “moderate” stages from the 14th day. This study showed clearly a fast (in 14 days) and beneficial effect of the application of LN-5 compared with the excipient, on the cutaneous repair of inter-toes cracks. Since LN-5 fragment is capable of producing immediate biological activity and reinforcing the dermal-epidermal junction, it can accelerate tissue repair. 展开更多

Efficacy of Oral Micronutrient Supplementation on Linear Nail Growth in Healthy Individuals

Only placebo controlled double blind studies could provide the reliable data considering the physiologic nail growth. <strong><em>Objective:</em></strong> The objective of this study was to evaluate the efficacy of consumption of a micronutrient supplementation on linear nail growth and thickness. <strong><em>Subjects and Method:</em></strong> 60 healthy female volunteers aged 35 to 65 years old were enrolled, randomized blindly in treatment and placebo groups, taking one tablet per day for 3 months. The evaluation was performed on D0 and D90 ± 3 days by measuring the linear nail growth, nail thickness by high frequency ultrasound imaging and also subjects’ self-assessment. <strong><em>Results:</em></strong> All 60 subjects finished the study without any serious adverse event. At D90 both groups revealed a significant linear nail growth (5.20 ± 0.35 for treatment group (p = 0.001) and 5.15 ± 0.30 for placebo group (p = 0.001)). However, the difference between the treatment and placebo group was statistically significant (p = 0.01) demonstrating the efficacy of oral supplementation on linear nail growth. No significant difference was observed at D90 for nail thickness measured with HFUS between 2 groups. The self-assessment score regarding ?brittle nails? and ?split nails? was diminished non-significantly in both groups. <strong><em>Conclusion:</em></strong> The results observed in this trial revealed that the oral micronutrient supplementation can provide beneficial effect on nail growth. This result may be due to the whole formula as the single vitamin treatment like biotin was shown to be non-effective. A longer study would be needed to confirm the efficacy on nail thickness. 展开更多

Thus, a treatment that would prevent or hasten the resolution of bruising should be very useful. Generally, the regression of bruising was conducted with patients or animals models. So we decided firstly to develop an ex vivo model in order to test antibruising properties of topical formulations and secondly to evaluate a curative effect of a cream (mixture of arnica extract and apigenin) in comparison with a positive control (vulnerary cream) and also to estimate the preventive interest of this cream. The results showed that the injection of 25 μl of blood into the dermis of skin fragments was sufficient to create a model of induced-bruise. The duration of 24 hours was chosen to compare the effects of actives on the decrease in the size of the bruise. Joint effects of a pretreatment and a treatment of a mixture of arnica extract and apigenin decreased significantly the area of bruising compared to the treatment group, the control group and the positive control group. Many topical products claim to improve bruising on their package label. Our model can demonstrate their efficacy and determinate the best topical antibruising formulation. The mechanism involved in anti-inflammatory activity of active compounds of topical formulations is often not fully understood. Our blood-induced model may bring some responses through the study of mediators of the inflammation. 展开更多

Mitochondrial function is impaired with an increase in lactate production during aging. Sculptra? (poly-L-lactic acid: PLLA), a biodegradable synthetic polymer, is used for subcutaneous volume restoration. Thus we decided to investigate different fibroblast functions when placed in contact with PLLA. Objectives: The potential of PLLA to compensate for the reduction of metabolic activity, to restore the migration capacity of WF and to inhibit the lactate production, was investigated and compared to NF. Methods: Two different skin samples were used from each of the three women’s facelift (one inside a face wrinkle and one from normal aged skin). Collagen I, lactate productions and proliferation capacities were investigated on monolayer cultures. Migration properties were evaluated using three-dimensional collagen lattices. Results: PLLA increased collagen I synthesis, restored migration capacities and tended to decrease lactate production in WF, whereas PPLA stimulated proliferation in NF and tended to improve the migration of NF. Conclusion: These results suggested that PLLA from Sculptra? acted as a stimulus for collagen production in WF and that it is suitable for correcting skin depressions, such as wrinkles. 展开更多

Efficacy and Safety of a Botanical Topical Serum on Male and Female Androgenetic Alopecia

Androgenetic alopecia (AGA) is a common form of hair loss in both men and women, it affects up to 80% of men and 50% of women during their life. The current treatments approved for AGA are limited, inconstantly effective and might be associated with adverse events. The aim of this study was to evaluate the efficacy and safety of a botanical topical serum (SERUM THICKER) applied for 6 months in 14 males and 10 females with mild to moderate androgenetic alopecia. The hair parameters were assessed using trichoscopy images obtained with high resolution camera FotoFinder Leviacam<sup>&#174;</sup> and analyzed with optimized software from TrichoLAB<sup>TM</sup> and Vitual tattoo<sup>TM</sup> technology. Hair loss was assessed using a wash-test procedure, and self-assessments were collected via questionnaires. Both at 28 days and 168 days, a significant increase of the hair density was observed, respectively +4.8% (p = 0.038);+11.7% (p < 0.001) for 1 application per day and +5.2% (p = 0.032);+9% (p < 0.001) for 2 applications/week. Reported to the whole scalp, this represents an estimation of hair gain of 7000 hair in 6 months for subjects applying SERUM THICKER once a day and 5000 hair for subjects applying SERUM THICKER twice then once a week. Although, the analysis of hair shedding during washing did not permit to show a time effect or any effect of the application frequency, the analysis showed a marginal time effect in the group treated once a day (p = 0.0674). Regarding efficacy on hair loss and regrowth, although no statistical analysis were performed, it seems that for all questions and for both groups the positive answers were higher after 6 months of treatment than after 1 month. SERUM THICKER represents an interesting, effective, well-tolerated and safe multitarget treatment to address mild to moderate AGA in male and female. Efficacy was observed from one application per week and seems greater once applied once daily. 展开更多

Stretch marks are cutaneous lesions occurring frequently in weight gain during puberty and early adult life. Previous studies showed that highest contractile forces were developed by fibroblasts from early stretch marks (SMF) compared to those developed by fibroblasts from normal healthy skin (HF) of the same patient. The aim of the present study was to evaluate a possible preventive effect of a topical preparation (containing vitamins C and E and botanical extracts including natural extract of Calendula, Aloe Vera and Evening Primrose Oil) on the formation of stretch marks using GlasboxPlus? device. In the presence of the tested mixture, a significant overall decrease in contractile forces of SMF was noted for 24 hours of measurements compared to control SMF and made it possible to restore their capacity to the same level as HF indicating a possible preventive effect of the tested mixture. The synergistic effects of these components are certainly responsible for the decrease in contractile forces of early stretch marks fibroblasts. 展开更多
