Citrus yellow vein clearing virus(CYVCV)is a new citrus virus that has become an important factor restricting the development of China’s citrus industry,and the CYVCV coat protein(CP)is associated with viral pathogenicity.In this study,the Eureka lemon zinc finger protein(ZFP)ClDOF3.4 was shown to interact with CYVCV CP in vivo and in vitro.Transient expression of ClDOF3.4 in Eureka lemon induced the expression of salicylic acid(SA)-related and hypersensitive response marker genes,and triggered a reactive oxygen species burst,ion leakage necrosis,and the accumulation of free SA.Furthermore,the CYVCV titer in ClDOF3.4 transgenic Eureka lemon plants was approximately 69.4%that in control plants 6 mon after inoculation,with only mild leaf chlorotic spots observed in those transgenic plants.Taken together,the results indicate that ClDOF3.4 not only interacts with CP but also induces an immune response in Eureka lemon by inducing the SA pathways.This is the first report that ZFP is involved in the immune response of a citrus viral disease,which provides a basis for further study of the molecular mechanism of CYVCV infection. 展开更多

The fatigue performance of optimized welded detail has been investigated by fatigue experiments of three welded specimens under different loadings.In addition,local finite element models of this welded detail were established using finite element software ANSYS.The influences of different factors such as plate thickness,plate gap and initial geometric imperfections on the stress concentration coefficient(SCC) were discussed.The experimental results indicate that the fatigue life of three specimens for this welded detail is 736,000,1,044,200 and 1,920,300 times,respectively.The web thickness,the filler plate thickness and the initial geometric imperfection have relatively less effect on the SCCs of this welded detail.However,cope-hole radius is influential on the SCCs of the web and the weld.The SCC of weld is significantly affected by the weld size and plate gap,but the SCCs of other parts of the welded detail are hardly affected by the plate gap. 展开更多

Outlier Detection and Forecasting for Bridge Health Monitoring Based on Time Series Intervention Analysis

The method of time series analysis,applied by establishing appropriate mathematical models for bridge health monitoring data and making forecasts of structural future behavior,stands out as a novel and viable research direction for bridge state assessment.However,outliers inevitably exist in the monitoring data due to various interventions,which reduce the precision of model fitting and affect the forecasting results.Therefore,the identification of outliers is crucial for the accurate interpretation of the monitoring data.In this study,a time series model combined with outlier information for bridge health monitoring is established using intervention analysis theory,and the forecasting of the structural responses is carried out.There are three techniques that we focus on:(1)the modeling of seasonal autoregressive integrated moving average(SARIMA)model;(2)the methodology for outlier identification and amendment under the circumstances that the occurrence time and type of outliers are known and unknown;(3)forecasting of the model with outlier effects.The method was tested with a case study using monitoring data on a real bridge.The establishment of the original SARIMA model without considering outliers is first discussed,including the stationarity,order determination,parameter estimation and diagnostic checking of the model.Then the time-by-time iterative procedure for outlier detection,which is implemented by appropriate test statistics of the residuals,is performed.The SARIMA-outlier model is subsequently built.Finally,a comparative analysis of the forecasting performance between the original model and SARIMA-outlier model is carried out.The results demonstrate that proper time series models are effective in mining the characteristic law of bridge monitoring data.When the influence of outliers is taken into account,the fitted precision of the model is significantly improved and the accuracy and the reliability of the forecast are strengthened. 展开更多

Calcium channel blockers(CCBs)are widely used to treat cardiovascular diseases such as hypertension,angina pectoris,hypertrophic cardiomyopathy,and supraventricular tachycardia.CCBs selectively inhibit the inward flow of calcium ions through voltage-gated calcium channels,particularly Ca v 1.2,that are expressed in the cardiovascular system.Changes to the molecular structure of Ca v 1.2 channels could affect sensitivity of the channels to blockade by CCBs.Recently,extensive alternative splicing was found in Ca v 1.2 channels that generated wide phenotypic variations.Cardiac and smooth muscles express slightly different,but functionally important Ca v 1.2 splice variants.Alternative splicing could also modulate the gating properties of the channels and giving rise to different responses to inhibition by CCBs.Importantly,alternative splicing of Ca v 1.2 channels may play an important role to influence the outcome of many cardiovascular disorders.Therefore,the understanding of how alternative splicing impacts Ca v 1.2 channels pharmacology in various diseases and different organs may provide the possibility for individualized therapy with minimal side effects. 展开更多

Hv1 is the only voltage-gated proton-selective channel in mammalian cells.It contains a conserved voltage-sensor domain,shared by a large class of voltage-gated ion channels,but lacks a pore domain.Its primary role is to extrude protons from the cytoplasm upon pH reduction and membrane depolarization.The best-known function of Hv1 is the regulation of cytosolic pH and the nicotinamide adenine dinucleotide phosphate oxidase-dependent production of reactive oxygen species.Accumulating evidence indicates that Hv1 is expressed in nervous systems,in addition to immune cells and others.Here,we summarize the molecular properties,distribution,and physiological functions of Hv1 in the peripheral and central nervous systems.We describe the recently discovered functions of Hv1 in various neurological diseases,including brain or spinal cord injury,ischemic stroke,demyelinating diseases,and pain.We also summarize the current advances in the discovery and application of Hv1-targeted small molecules in neurological diseases.Finally,we discuss the current limitations of our understanding of Hv1 and suggest future research directions. 展开更多

Background:The epidemic of overweight and obesity has become a worldwide public health problem.Cardiometabolic diseases may originate in childhood.We investigated the association between percent body fat(PBF)measured by the bioelectrical impedance assay and cardiometabolic risk(CMR)in pediatrics.Methods:This cross-sectional study involved 3819 subjects(6-17 years old)in Shanghai.We assessed the association between PBF and body mass index(BMI)with multiple CMR factors.We examined the risk for cardiometabolic abnormalities attributable to overweight and obesity based on age-and sexspecific PBF Z-scores and BMI Z-scores,respectively.Results:PBF,but not BMI,was positively associated with multiple CMR factors in males and females except for total cholesterol in females(all p<0.05).Compared with the non-overweight group based on PBF,overweight and obese subjects had increasingly higher odds ratio of dyslipidemia(2.90(1.99-4.23),4.59(2.88-7.32)for males and 1.82(1.20-2.75),2.46(1.47-4.11)for females)and elevated blood pressure(BP)(3.26(2.35-4.51),4.55(2.92-7.09)for males and 1.59(1.07-2.34),3.98(2.27-6.17)for females).Obesity females showed a higher likelihood for hyperglycemia(2.19(1.24-3.84))than non-overweight females.In both sexes,the predictive effect of PBF on dyslipidemia and elevated BP in adolescents was better than that in children.For hyperglycemia,the predictive effect of PBF was better in male adolescents and female children.There was no risk difference for cardiometabolic abnormalities attributable to BMI-based obesity categories.Conclusions:PBF but not BMI was associated with CMR.Overweight and obesity categories based on PBF had an increased risk for cardiometabolic abnormalities in children and adolescents. 展开更多
