Role of Cyclin D1b in Inducing Macrophages Toward a Tumor-associated Macrophage-like Phenotype in Murine Breast Cancer

Objective:Tumor-associated macrophages(TAMs)of the M2 phenotype are frequently associated with cancer progression.Invasive cancer cells undergoing epithelial-mesenchymal transition(EMT)have a selective advantage as TAM activators.Cyclin D1b is a highly oncogenic splice variant of cyclin D1.We previously reported that cyclin D1b enhances the invasiveness of breast cancer cells by inducing EMT.However,the role of cyclin D1b in inducing macrophage differentiation toward tumor-associated macrophage-like cells remains unknown.This study aimed to explore the relationship between breast cancer cells overexpressing cyclin Dlb and TAMs.Methods:Mouse breast cancer 4T1 cells were transfected with cyclin D1b variant and co-cultured with macrophage cells in a Transwell coculture system.The expression of characteristic cytokines in differentiated macrophages was detected using qRT-PCR,ELISA and zymography assay.Tumor-associated macrophage distribution in a transplanted tumor was detected by immunofluorescence staining.The proliferation and migration ability of breast cancer cells was detected using the cell counting kit-8(CCK-8)assay,wound healing assay,Transwell invasion assay,and lung metastasis assay.Expression levels of mRNAs were detected by qRT-PCR.Protein expression levels were detected by Western blotting.The integrated analyses of The Cancer Genome Atlas(TCGA)datasets and bioinformatics methods were adopted to discover gene expression,gene coexpression,and overall survival in patients with breast cancer.Results:After co-culture with breast cancer cells overexpressing cyclin D1b,RAW264.7 macrophages were differentiated into an M2 phenotype.Moreover,differentiated M2-like macrophages promoted the proliferation and migration of breast cancer cells in turn.Notably,these macrophages facilitated the migration of breast cancer cells in vivo.Further investigations indicated that differentiated M2-like macrophages induced EMT of breast cancer cells accompanied with upregulation of TGF-β1 and integrinβ3 expression.Conclusion:Breast cancer cells transfected with cyclin D1b can induce the differentiation of macrophages into a tumor-associated macrophage-like phenotype,which promotes tumor metastasis in vitro and in vivo. 展开更多

作为一种意合型语言,汉语由字组词的特性明显,字面义词的词义大体可由其构词结构和语素概念来表达,但对非字面义词的处理存在偏差,这也是语言深度理解中的一个棘手问题.本文从语言认知的角度出发,提出了适用于汉语词的非字面义的知识表... 作为一种意合型语言,汉语由字组词的特性明显,字面义词的词义大体可由其构词结构和语素概念来表达,但对非字面义词的处理存在偏差,这也是语言深度理解中的一个棘手问题.本文从语言认知的角度出发,提出了适用于汉语词的非字面义的知识表示方式:发掘《现代汉语词典》中的3524个非字面义二字词,判定它们作为隐喻或转喻现象的非字面义类型,标注其在《同义词词林》中的源域、目标域,并选取面向计算的适合的字面义词承担者.该工作首次在词汇级别上,系统地揭示了汉语隐喻和转喻现象的数量、类型及语义域映射分布状况,并且在算法框架不变的情况下,显著改善了词义相似度计算效果.这些思路、做法及语言资源建设,有望推动人文领域和计算应用等相关工作的深入开展. 展开更多

The Prandtl-Tomlinson(PT)model is the most widely used and successful minimalistic model to describe atomistic scale friction.It can describe the thermally activated,stress assistant process which shows stick-slip frictional behavior.The relationship between the energy barrier and lateral force is critical to determine how the frictional force depends on velocity and temperature.There is some confusion in the literature to derive such relationship.The underlying assumption and approximations are not stated in a clear way and the rigorous derivations are missing.This study discusses the asymptotic behavior of the energy barrier as the support-spring coupling lowers it to zero and gives a detailed derivation of the asymptotic expression of the energy barrier within the framework of PT model. 展开更多

Monitoring the operating status of a High-Speed Train(HST) at any moment is necessary to ensure its security. Multi-channel vibration signals are collected by sensors installed on bogies and beneficial information are extracted to determine the running condition. Based on multi-view clustering and considering different views of complementary information, this study proposes a Multi-view Kernel Fuzzy C-Means(MvKFCM) model for condition recognition of the HST bogie. First, fast Fourier transform coefficients of HST vibration signals of all channels are extracted. Then, the fuzzy classification coefficient of every channel is calculated after clustering to select the appropriate channels. Finally, the selected channels are used to cluster by MvKFCM and the conditions of HST are determined. Experimental results show that the selection is effective to maintain rich feature information and remove redundancy. Furthermore, the condition recognition rate of MvKFCM is higher than that of single-view and four other multiple-view clustering algorithms. 展开更多
