Establishment and evaluation of Type 2 Diabetes Mellitus andits complications induced by low dose of multiple streptozotocin

Objective: To explore a simple, economical and feasible method for establishing type 2 diabetes mellitus and its complication model. Method: SD rats were randomly divided into two groups: normal group (15 rats) normal diet, model group (20 rats) with high glucose and high fat diet. After 2 weeks of feeding, model group was injected low dose(30mg/Kg) streptozotocin in the abdominal cavity, once a day for 6 days, normal group was injected with the same dose of citric acid buffer. Observe the changes of rat diet, body, hair, spirit, urine volume, and monitor weight, blood glucose. After 6 weeks, fast insulin, blood lipid, serum calcium, phosphorus and other biochemical indexes of rats, after HE staining observe morphologic variations changes of pancreas and kidney using microscope, and the changes of femoral bone in SD rats. Results: Compared with the normal group, the model group showed obvious symptoms, drink more, eat more, polyuria, emaciation, hyperglycemia, low calcium, low phosphorus, hyperlipidemia and other metabolic abnormalities, pancreatic atrophy, tissue fibrosis, glomerular area increased, basement membrane thickening, osteoporosis. Conclusion:High glucose and high fat diet combined with repeated intraperitoneal injecti on of low dose STZ can be successfully established the model of type 2 diabetes, and its high successful rate, low mortality rate, relatively simple, economical and feasible, has a certain significance of research on diabetes and its complications. 展开更多

Study on medication laws of traditional Chinese medicine of 340 cases of COVID-19 based on data mining

Objective:To analyze the prescription rules of traditional Chinese medicine for patients with COVID-19 in Wuhan Third Hospital, and summarize the disease characteristics and clinical treatment ideas, in order to learn from the treatment of this disease by traditional Chinese medicine.Methods: This study established a cloud database based on the ancient and modern medical cases to mine the rules of medication with analyses of frequency of drug used, association analysis, cluster analysis, complex network and other methods.Results:Of 340 COVID-19 patients discharged from our hospital, 143 males and 197 females with male: female = 0.73: 1, and there were 167 patients over 60 years old, and the majority of clinical illnesses were mainly middle-aged and elderly people. Most of them had dry cough, anorexia, fatigue, etc. The traditional Chinese medicine for the treatment of COVID-19 involves a total of 204 flavors. The most frequent use is Fuling of 225 times. The treatment of traditional Chinese medicines is mainly involved with flavors such as warm, spicy and sweet. Mainly detoxification, dampness and phlegm, Qingfei Paidu Decoction is a common clinical prescription, of which the core drugs are Huoxiang, Fabanxia, Chenpi, Baizhu, Cangzhu, Fuling, Chaihu, Huangqin, Xingren, and Gancao.Conclusion: Through this study, the patients with COVID-19 in our hospital are mainly middle-aged and elderly people, and the etiology is characterized by "cold, wet, poisonous, and epidemic".
