Histone H3 lysine 27 trimethylation(H3K27me3) is a histone modification associated with transcriptional repression. However, insights into the genome-wide pattern of H3K27me3 in grapevines are limited. Here, anti-H3K27 chromatin immunoprecipitation(ChIP), high-throughput sequencing, and transcriptome analysis were performed using leaves of Vitis amurensis. The leaves were treated at 4°C for 2 h and 24 h and used to investigate changes in H3K27me3 under chilling treatment. The results show that H3K27me3 is well-distributed both in gene regions(-50%) and in the intergenic region(-50%) in the grapevine genome(Vitis vinifera ‘Pinot Noir PN40024'). H3K27me3 was found to be localized in8 368 annotated gene regions in all detected samples(leaves at normal temperature and under chilling treatments) and mainly enriched in gene bodies with the adjacent promoter and downstream areas. The short-term chilling treatments(4°C for 2 h) induced 2 793 gains and 305losses in H3K27me3 modification. Subsequently, 97.3% of the alterations were restored to original levels after 24 h treatment. The ChIP-qPCR for five differential peaks showed similar results to the data for ChIP-seq, indicating that the chilling-induced H3K27me3 modification is reliable.Integrative analysis of transcriptome and ChIP-seq results showed that the expression of H3K27me3 target genes was significantly lower than those of non-target genes, indicating transcriptional repression of H3K27me3 in grapevine leaves. Furthermore, histone methylation alterations were detected in 82 genes and were related to either repression or activation of their expression during chilling stress. The findings provide the genome-wide H3K27me3 patterns in grapevines and shed light on uncovering its regulation in chilling stress responses. 展开更多

The results showed that the selection on breeding goal traits of each line was effective since 14 generations, especially the selection on growth traits did not cause adverse influence on reproductive performance. After the 6^ th generation, main performances of each line maintained in higher level, but the breeding progress obviously declined. It illustrated that current breeding means was effective for maintaining and consolidating breeding goal traits of Dahen broiler, and it should timely adjust breeding scheme to obtain more genetic progress. 展开更多

Anthraquinone dyes are a class of typical carcinogenic and hard-biodegradable organic pollutants.This study aimed to enhance the decolorization of anthraquinone dye by rationally designing an expected immobilized system.Reactive blue 4(RB4) was used as a substrate model and a previous isolated dyedegrading strain Aspergillus flavus A5pl was purposefully immobilized.Considering the effects of cell attachment and mass transfer,the polyurethane foam(PUF) with open pore structure was selected as the immobilization carrier.Results showed that the RB4 decolorization efficiency was significant enhanced after immobilization.Compared to the free mycelium system,the decolorization time of200 mg·L^(-1)RB4 was shortened from 48 h to 28 h by the PUF-immobilized cell system.Moreover,the PUF-immobilized system could tolerate RB4 up to 2000 mg-L^(-1).In the packed bed bioreactor(PBBR),an average decolorization efficiency of 93.3% could be maintained by the PUF-immobilized system for26 days.The decolorization process of RB4 was well described by the logistic equation and the degradation pathway was discussed.It was found that the higher specific growth rate of the PUF-immobilized cells was one of reasons for the enhanced decolorization.The good performance of the PUFimmobilized cell system would make it have potential application value for RB4 bioremediation. 展开更多

The anode-free design is a promising strategy to increase the energy density of aqueous Zn metal batteries(AZMBs).However,the scarcity of Zn-rich cathodes and the rapid loss of limited Zn greatly hinder their commercial applications.To address these issues,a novel anode-free Zniodine battery(AFZIB)was designed via a simple,low-cost and scalable approach.Iodine plays bifunctional roles in improving the AFZIB overall performance:enabling high-performance Zn-rich cathode and modulating Zn deposition behavior.On the cathode side,the ZnI_(2) serves as Zn-rich cathode material.The graphene/polyvinyl pyrrolidone heterostructure was employed as an efficient host for ZnI_(2) to enhance electron conductivity and suppress the shuttle effect of iodine species.On the anode side,trace I_(3)^(−) additive in the electrolyte creates surface reconstruction on the commercial Cu foil.The in situ formed zincophilic Cu nanocluster allows ultralow-overpotential and uniform Zn deposition and superior reversibility(average coulombic efficiency>99.91% over 7,000 cycles).Based on such a configuration,AFZIB exhibits significantly increased energy density(162 Wh kg^(−1)) and durable cycle stability(63.8% capacity retention after 200 cycles)under practical application conditions.Considering the low cost and simple preparation methods of the electrode materials,this work paves the way for the practical application of AZMBs. 展开更多

The strong push/pull electron capability of dithienopyrrolobenzothiadiazole/ phenothiazine and large π-conjugated dithienopyrrolobenzothiadiazole facilitate hole mobility and high conductivity. The devices using the dithienopyrrolobenzothiadiazole/phenothiazine-based hole trans-porting material achieved a power conversion efficiency of 14.2% under 1 sun illumination and improved stability under 20% relative humidity at room temperature without encapsulation. The present finding highlights the potential of dithienopyrrolobenzothiadiazole-based donor-acceptor-donor small molecular hole transporting materials for perovskite solar cells. 展开更多

Stocking density is an important factor for animal production performance.It appears particularly urgent and important to research systematically on stocking density for the dual restriction of environmental protection and animal welfare.This review dealt with the influence of stocking density on ducks'productivity and health through production performance,animal behavior and animal welfare,and analyzed the possible mechanism of high stocking density reducing production performance,resulting in abnormal behavior and stress,causing welfare and health problems.We consider that it might be associated with heat stress,more spatial competition(to gather the food,drinking water,sports area,etc.),as well as the quality deterioration of litter and house air caused by high stocking density.Finally,we put forward some suggestions on the study of stocking density's effects on ducks,and proposed the future work that need to be studied and problems that need to be solved imminently. 展开更多

[Objectives]This study was conducted to improve the production efficiency of dark-shank partridge-like plumage high-quality broilers and ensure animal welfare,so as to meet the development needs of China’s yellow-feather broiler industry.[Methods]Sichuan Animal Science Academy and Sichuan Dahen Poultry Breeding Co.,Ltd.formed a joint breeding research team.The team bred Dahen 799 broiler complete set line with local chicken breeds as the main material and improving production efficiency,stabilizing meat quality and flavor and achieving autosexing of chicks as the main breeding goals by modern poultry breeding technologies,after 10 years and 8 generations.[Results]The Dahen 799 broiler complete set line was approved by National Commission of Livestock and Poultry Genetic Resources in December 2020.The 66-week-old egg production of the breed reached 189 eggs.For the commercial generation,cocks and hens showed 10-week-old weights of 2668 and 2269 g,respectively;the feed conversion ratio was 2.26-2.35;the survivability was 96.7%-97.2%;and the average intramuscular fat contents of cocks and hens were in the range of 1.8%-2.1%,and the inosinic acid contents were in the range of 2.3-3.2 mg/g.The production performance and meat flavor of Dahen 799 broilers were better than Dahen 699 broilers,Sichuan mountainous bone-black chickens and two other social breeds.Compared with Sichuan mountainous bone-black chickens,the egg production at the age of 66 weeks increased by 47 eggs;the average weight of cocks and hens of the commercial generation at 10 weeks increased by 45.21%;and the autosexing rate of chicks of the commercial generation reached 99.3%.[Conclusions]Dahen 799 broilers have the advantages of good production performance,high feed conversion ratio and excellent meat flavor and the characteristic of autosexing which other breeds do not have,and can fully meet the industrial development needs of dark shanks and partridge-like plumage broilers. 展开更多

The relevance of genetic mechanism to the phenotype of hypoxic adaptation remains elusive.Tibetan chickens typically used to investigate the mechanism for the adaptation of hypoxia and the recognition of hypoxia-related piRNA remains an open issue.The purpose of this study was to illustrate whether the piRNAs were related to hypoxic adaptation.First of all,the differentially expressed piRNAs(DEpiRNAs)were identified through RNA sequencing between the Tibetan chickens and Daheng broilers.Subsequently,the target genes of DEpiRNAs were predicted and annotated by software.The network was constructed by Cytoscape.In our study,a total of 277 DEpiRNAs(33 down-regulated,244 up-regulated)were identified in the Tibetan chickens compared with the Daheng broilers.All of the 277 DEpiRNAs predicted 36658 targeted genes.Gene Ontology(GO)analysis showed that the target genes were significantly enriched in the biological process correlated with proliferation and apoptosis of cells,including cell cycle,mitochondrial outer membrane permeabilization,and positive regulation of stress-activated mitogen-activated protein kinase cascade.Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis implicated that the DEpiRNAs were mainly involved in immune and metabolism,including natural killer cell-mediated cytotoxicity,toll-like receptor signaling pathway and fatty acid metabolism.Furthermore,a predicted network with four piRNAs acted on 11 pathways via interacting with 22 target genes,in which piR-gga-1368839 regulated metabolic pathways by acting on DHCR24.In conclusion,we determined the DEpiRNAs in the Tibetan chickens and found that these piRNAs were associated with metabolism,which may be favorable for researching the biological adaptation to hypoxic stress. 展开更多

Sterol regulatory element-binding factor-1 and-2(SREBF1 and SREBF2)are important transcription factors involved in the regulating lipid homeostasis.Based on the essential role of SREBF1 and SREBF2,we measured the mRNA expression levels of the two genes in six various tissues at different growth points.Our results showed that the SREBF1 and SREBF2 were expressed in all six tissues examined in Erlang mountainous chicken(SD02)at 42 d,and were expressed abundantly in the uropygial gland and liver,with relatively lowest levels of expression in the abdominal fat,sebum cutaneum and leg muscle.The expression ratio of SREBF1 and SREBF2 in breast muscle,leg muscle,sebum cutaneum and uropygial gland exhibited a"decline-rise"trend.However,in liver,the expression ratio of these two genes exhibited a"decline-rise-decline"trend.Meanwhile,the expression level of SREBF1 gene of all tissues was lower than that of SREBF2 except for uropygial gland.The findings will provide important references for further function investigation of the two genes involved in fat deposition in chickens. 展开更多

Risk of extended viral shedding of Omicron BA.2 in Shanghai:Implications for vaccination strategy optimization

Background:In late March 2022,an outbreak of coronavirus disease 2019(COVID-19)caused by the Omicron BA.2 strain occurred in Shanghai,China.This retrospective study aimed to investigate the clinical characteristics,laboratory parameters,and vaccine protectiveness related to this disease in China.Methods:We conducted a single-center retrospective study on 735 patients with COVID-19 hospitalized from March 17 to May 14,2022.Clinical characteristics were analyzed based on vaccination status and viral shedding time(VST).The least absolute shrinkage and selection operator(LASSO)regression and 5-fold cross-validation were applied to screen factors linked to the rate of the VST.Generalized linear models were further applied to estimate the odds ratios for factors influencing the VST.Results:The median VST of unvaccinated patients was 13(11-16)days,which was longer than that of patients vaccinated with one or two doses(11[9-13]days)and with completed booster doses(11[8-12]days).A LASSO regression model and 5-fold cross-validation showed that age of≥60 years(β=0.01),pneumonia(β=0.53),and higher number of comorbidities(β=0.69)were positively associated with the VST,whereas the platelet count(β=-8.0×10-5)was inversely associated with the VST.Subgroup analysis revealed that the number of vaccinations was significantly associated with a decreased VST among patients with renal dysfunction(odds ratio[OR],0.65;95%confidence interval[CI],0.44-0.97;P=0.034)and patients with two or more comorbidities(OR,0.09;95%CI,0.03-0.28;P<0.001).The lymphocyte count was significantly associated with a decreased VST among patients aged<60 years(OR,0.51;95%CI,0.30-0.85;P=0.011),patients with normal renal function(OR,0.41;95%CI,0.21-0.80;P=0.009),and patients with fewer than two comorbidities(OR,0.49;95%CI,0.30-0.80;P=0.005).Conclusion:Our preliminary results suggest that the complete and booster vaccination contributes to the viral clearance of Omicron BA.2 variants,while the protectiveness of vaccination is most imperative in patients with impaired renal function and more comorbidities. 展开更多

High monomer concentration is a requisite for engendering the aggregation-induced emssion(AIE)phenomenon as well as the formation of supramolecular polymers.Therefore,this is supposed to ensure the generation of AIE supramolecular polymers,wherein the monomer soluability takes effect.Nevertheless,parts of supramolecular monomers are considered as poessessing different soluability towards the same sovlent,through which the polymerzation process is thus hard to proceed.Interfacial polymerzation gets over the soluabilty restriction,providing a facile method for propelling the reaction of thesemonomers.Herein,we had prepared M1 containing tetraphenylethene(TPE)functionalized with two terpyridine derivatives,then making M1 dissolving in CHCl3 to give solutions.Cu^(2+)solutions were fabricated through dissolving CuCl_(2)into H2O.Towards mixing those solutions,AIE interfacial supramolecular polymers(AIEISPs)displaying green fluorescence were generated at the interface of two phases on the basis of metal-coordination between terpyridine and Cu^(2+).These AIEISPs were certificated to possess the stimuli-responsiveness,for which the excessive addition of tetrabutylammonium hydroxide would cause the structure destruction owing to the stronger bonding ability with Cu^(2+)than that of terpyridine.These fabricated AIEISPs had provided a new avenue to prepare AIE supramolecular polymers. 展开更多

After the impoundment of the Three Gorges Reservoir(TGR), the hydrological situation of the reservoir has changed greatly. The concentration and distribution of typical persistent organic pollutants in water and sediment have also changed accordingly. In this study, the concentration, distribution and potential sources of 16 polycyclic aromatic hydrocarbons(PAHs) and 6 phthalic acid esters(PAEs) during the water drawdown and impoundment periods were investigated in water and sediment from the TGR. According to our results, PAHs and PAEs showed temporal and spatial variations. The mean ΣPAH and ΣPAE concentrations in water and sediment were both higher during the water impoundment period than during the water drawdown period. The water samples from the main stream showed larger ΣPAH concentration fluctuations than those from tributaries. Both the PAH and PAE concentrations meet the Chinese national water environmental quality standard(GB 3838-2002). PAH monomers with 2–3 rings and 4 rings were dominant in water, and 4-ring and 5–6-ring PAHs were dominant in sediment. Di-n-butyl phthalate(DBP) and di-2-ethylhexyl phthalate(DEHP)were the dominant PAE pollutants in the TGR. DBP and DEHP had the highest concentrations in water and sediment, respectively. The main source of PAHs in water from the TGR was petroleum and emissions from coal and biomass combustion, whereas the main sources of PAHs in sediments included coal and biomass combustion, petroleum, and petroleum combustion. The main source of PAEs in water was domestic waste, and the plastics and heavy chemical industries were the main sources of PAEs in sediment. 展开更多

The heterogeneous integration of silicon thin film and lithium niobate(LN) thin film combines both the advantages of the excellent electronics properties and mature micro-processing technology of Si and the excellent optical properties of LN,comprising a potentially promising material platform for photonic integrated circuits. Based on ion-implantation and wafer-bonding technologies, a 3 inch wafer-scale hybrid mono-crystalline Si/LN thin film was fabricated. A high-resolution transmission electron microscope was used to investigate the crystal-lattice arrangement of each layer and the interfaces. Only the H-atom-concentration distribution was investigated using secondary-ion mass spectroscopy. Highresolution X-ray-diffraction ω–2θ scanning was used to study the lattice properties of the Si/LN thin films. Raman measurements were performed to investigate the bulk Si and the Si thin films. Si strip-loaded straight waveguides were fabricated, and the optical propagation loss of a 5-μm-width waveguide was 6 d B/cm for the quasi-TE mode at1550 nm. The characterization results provide useful information regarding this hybrid material. 展开更多

以木质素为原料,在水相中合成一种适用于木材胶黏剂的木质素环氧树脂。为了考察木质素环氧化反应条件对木质素环氧树脂的羟基和环氧基的影响,以及对胶合板的胶合强度的影响,采用FT-IR和31PNMR对木质素环氧树脂结构进行表征,并用TG和DTG... 以木质素为原料,在水相中合成一种适用于木材胶黏剂的木质素环氧树脂。为了考察木质素环氧化反应条件对木质素环氧树脂的羟基和环氧基的影响,以及对胶合板的胶合强度的影响,采用FT-IR和31PNMR对木质素环氧树脂结构进行表征,并用TG和DTG对木质素环氧树脂的热稳定性进行分析。结果表明,环氧化反应主要发生在酚羟基上,在反应过程中,相比环氧氯丙烷,NaOH的加入量对木质素环氧树脂结构和胶合板的胶合强度影响更大。随NaOH加入量增加,木质素环氧树脂中环氧基团逐渐增多,胶合板的胶合强度呈现先升高、后降低的趋势。当木质素的羟基与NaOH摩尔比为1:1时,由木质素环氧树脂制得的胶合板胶合强度达到最大,湿强度达1.61 MPa,超过国家标准II类板的要求(≥0.7 MPa)。采用扫描电镜研究了黏接机理,发现环氧化程度提高时,固化后的木质素环氧树脂的结构更加稳定且致密,导致胶合板的胶合强度也提高。但过高的环氧化程度会增大胶黏剂的粒径,导致胶黏剂与木板不能形成更好的机械互锁结构,从而降低胶合板的胶合强度。还进一步简化了木质素环氧树脂木材胶黏剂的合成工艺,使环氧化反应后的体系无需处理即可直接应用于木材胶黏剂,减少了胶合板生产工艺流程。此外,经过30天的储存期,胶黏剂黏合强度没有明显下降。通过与商业脲醛树脂木材胶黏剂对比,其黏接强度可以达到商业脲醛树脂的水平。 展开更多

Fresh water microplastic pollution is of pressing concern globally,but its distribution and sources in reservoirs are poorly documented.Danjiangkou Reservoir is the second largest reservoir in China and is divided into the Han Reservoir and Dan Reservoir.In this work,microplastic abundances and morphological characteristics of the reservoir were investigated.The microplastic abundance of 15 main tributaries of the reservoir was also measured.The vertical distribution(in water column and sediment),horizontal distribution(in Han Reservoir and Dan Reservoir)and source of microplastics were analyzed.Microplastics accumulated in the middle layer of the reservoir,and the size and color of the microplastic particles changed from the surface to the bottom,which implies that surveys of surface water are not enough to determine the microplastic contamination for deep water reservoirs.In the surface water,the microplastic abundance in the Han Reservoir was lower than that in the Dan Reservoir(p<0.05),but microplastic abundance did not differ significantly in the intermediate and bottom water.Tributaries were one of the main sources of microplastics for Han Reservoir but not for Dan Reservoir.Agricultural cultivation in the hydro-fluctuation belt might be an important source of microplastics in the Dan Reservoir,which should be given additional attention.The results of this study can provide valuable information for developing microplastic sampling strategies in deep water reservoirs.Further studies are recommended to investigate the process through which microplastics in the hydro-fluctuation belt enter the reservoir and the sinking behavior of microplastics in the reservoir. 展开更多

Air route network optimization,one of the essential parts of the airspace planning,is an effective way to optimize airspace resources,increase airspace capacity,and alleviate air traffic congestion.However,little has been done on the optimization of air route network in the fragmented airspace caused by prohibited,restricted,and dangerous areas(PRDs).In this paper,an air route network optimization model is developed with the total operational cost as the objective function while airspace restriction,air route network capacity,and non-straight-line factors(NSLF) are taken as major constraints.A square grid cellular space,Moore neighbors,a fixed boundary,together with a set of rules for solving the route network optimization model are designed based on cellular automata.The empirical traffic of airports with the largest traffic volume in each of the 9 flight information regions in China's Mainland is collected as the origin-destination(OD) airport pair demands.Based on traffic patterns,the model generates 35 air routes which successfully avoids 144 PRDs.Compared with the current air route network structure,the number of nodes decreases by 41.67%,while the total length of flight segments and air routes drop by 32.03% and 5.82% respectively.The NSLF decreases by 5.82% with changes in the total length of the air route network.More importantly,the total operational cost of the whole network decreases by 6.22%.The computational results show the potential benefits of the model and the advantage of the algorithm.Optimization of air route network can significantly reduce operational cost while ensuring operation safety. 展开更多

目的:探讨飞秒激光小切口角膜基质透镜取出术(SMILE)前后全眼高阶像差和角膜生物力学参数的变化,并分析角膜生物力学对全眼高阶像差的影响。方法:前瞻性临床研究。选择2019年3月至2020年3月在西安爱尔眼科医院行SMILE的屈光不正患者160... 目的:探讨飞秒激光小切口角膜基质透镜取出术(SMILE)前后全眼高阶像差和角膜生物力学参数的变化,并分析角膜生物力学对全眼高阶像差的影响。方法:前瞻性临床研究。选择2019年3月至2020年3月在西安爱尔眼科医院行SMILE的屈光不正患者160例,术后随访6个月。手术前、后使用iTrace视觉功能分析仪测量角膜及全眼球差、彗差、三叶草、总高阶像差(HOA);使用Corvis-ST生物力学分析仪测量角膜硬度参数(SP-A1)和最大形变幅度(DA)。采用配对t检验、单因素方差分析、Pearson线性相关分析等对高阶像差变化量(角膜ΔHOA、全眼ΔHOA)及生物力学改变量(ΔSP-A1、ΔDA)进行分析。结果:SMILE术后6个月,150例(295眼)(随访率为94%)角膜HOA为(0.37±0.01)μm,全眼HOA为(0.24±0.01)μm,角膜HOA较术前增加15.63%,全眼HOA较术前减少35.14%,角膜HOA、全眼HOA的手术前后差异均有统计学意义(t=3.76,P=0.002;t=10.04,P=0.001);SP-A1为(77.29±11.86)mmHg/mm,较术前下降29.6%,DA为(1.19±0.68)mm,较术前增加10.4%,手术前、后SP-A1、DA差异均有统计学意义(t=32.98、20.62,P<0.001)。ΔSP-A1、ΔDA与全眼ΔHOA均呈正相关(r=0.19,P<0.001;r=0.17,P=0.002)。结论:SMILE术后6个月的全眼HOA减小,角膜生物力学改变与全眼HOA的变化具有相关性。 展开更多
