METHODS: The human fetal hepatocytes were cultured in serum-free medium, HBV-positive serum was added into the medium to study the susceptibility of hepatocytes to HBV infection. The supernatant was collected for ELISA assay of HBsAg and HBeAg, and quantitative fluorescence PCR for HBV-DNA assay daily. Albumin and HBcAg, CK8 and CK18 expressions were detected by immunohistochemistry in cultured hepatocytes. Content of lactate dehydrogenate (LDH) was measured to find out the integrity of the cell membrane. RESULTS: A stable hepatocyte culture system was established. HBV could infect the hepatocytes and replicate, and HBcAg expression could be detected by immunohistochemistry in hepatocyte-like cells. HBV- DNA in the supernatant could be detected from d 2 to d 18 and HBsAg and HBeAg were positive on d 3-d 18 after HBV infection. HBV in medium increased from d 0 to d 6 and subsequently decreased as the cells were progressively loosing their hepatocyte phenotypes. CONCLUSION: HBV could infect human fetal hepato- cytes and replicate. This in vitro model allowed a detailed study on early events associated with human HBV entry into cells and subsequent replication.展开更多

The intrinsic viscosity of polysaccharide was found to be 78.95 mL/g in DMSO solution at 25℃. The isolated polysaccharide was reacted with chlorosulfonic acid to obtain pachyman sulfate using the improved Wolfrom method. The results of the orthogonality experiment on the sulfation reaction identified that the effectiveness of the reaction conditions on the degree of sulfation and the value of intrinsic viscosity is in the following order: molar ratio of chlorosulfonic acid to glucoside (3-5) 〉 reaction temperature (60-80℃) 〉 reaction time (1 2 h), The kinetic studies of the pachyman sulfation indicated that the hydrolysis is accompanied with the sulfation process. The decrease in intrinsic viscosity of the sulfated pachyman is proportional to the increase in the degree of sulfation under the mild reaction conditions of 〈 80℃, chlorosulfonic acid/glucoside mole ratio 〈 5, and reaction time 〈 2 h. Beyond the above reaction conditions, excessive loss of --OH group occurs during hydrolysis. The NMR results indicated a complete sulfation on C-6 and a partial sulfation on the C-2 and C-4 of glucoside.展开更多
BACKGROUND Trocar site hernia(TSH)is a rare but potentially dangerous complication of laparoscopic surgery,and the drain-site TSH is an even rarer type.Due to the difficulty to diagnose at early stages,TSH often leads...BACKGROUND Trocar site hernia(TSH)is a rare but potentially dangerous complication of laparoscopic surgery,and the drain-site TSH is an even rarer type.Due to the difficulty to diagnose at early stages,TSH often leads to a delay in surgical intervention and eventually results in life-threatening consequences.Herein,we report an unusual case of drain-site TSH,followed by a brief literature review.Finally,we provide a novel,simple,and practical method of prevention.CASE SUMMARY A 54-year-old female patient underwent laparoscopic subtotal hysterectomy and bilateral adnexectomy for uterine fibroids 8 d ago in another hospital.She was admitted to our hospital with a 2-d history of intermittent abdominal pain,nausea,vomiting,and abdominal enlargement with an inability to pass stool and flatus.The emergency computed tomography scan revealed the small bowel herniated through a 10 mm trocar incision,which was used as a drainage port,with diffuse bowel distension and multiple air-fluid levels with gas in the small intestines.She was diagnosed with drain-site strangulated TSH.The emergency exploratory laparotomy confirmed the diagnosis.A herniorrhaphy followed by standard intestinal resection and anastomosis were performed.The patient recovered well after the operation and was discharged on postoperative day 8 and had no postoperative complications at her 2-wk follow-up visit.CONCLUSION TSH must be kept in mind during the differential diagnosis of post-laparoscopic obstruction,especially after the removal of the drainage tube,to avoid the serious consequences caused by delayed diagnosis.Furthermore,all abdomen layers should be carefully closed under direct vision at the trocar port site,especially where the drainage tube was placed.Our simple and practical method of prevention may be a novel strategy worthy of clinical promotion.展开更多

The experimental uptakes of 1-naphthol and 1-naphthylamine in all binary-component systems of different molar ratios were obviously higher than the corresponding uptakes predicted by the extended Langmuir model, assuming no interaction between the adsorbed molecules of the two components. This phenomenon was attributed to the cooperative adsorption effect arising from the hydrogen bonding interaction between 1-naphthol and 1-naphthylamine molecules. A modified extended Langmuir model was proposed to describe the binary adsorption behavior by means of introducing a fitting parameter related with the cooperative adsorption effect of the adsorbates.展开更多

MMPs and TIMPs expressiond were explored in this rat model of F.solani keratitis using real-time polymerase chain reaction(PCR) and DIF.GM6001(400 μmol/m L) was used to treat infected corneas. The keratitis duration, amount and area of corneal neovascularization(CNV) were evaluated.·RESULTS: MMP-3 expression was 66.3 times higher in infected corneas compared to normal corneas. MMP-8,-9,and-13 expressions were significantly upregulated in the mid-period of the infection, with infected- to-normal ratios of 4.03, 39.86, and 5.94, respectively. MMP-2 and-7expressions increased in the late period, with the infected-to-normal ratios of 5.94 and 16.22, respectively.TIMP-1 expression was upregulated in the early period,and it was 43.17 times higher in infected compared to normal corneas, but TIMP-2,-3, and-4 expressions were mildly downregulated or unchanged. The results of DIF were consistent with the result of real-time PCR.GM6001, a MMPs inhibitor, decreased the duration of F.solani infection and the amount and area of CNV.·CONCLUSION: MMPs and TIMPs contributed into the progress of F.solani keratitis.展开更多

All adsorption isotherms can be well fitted by Langmuir and Freundlich equations. Compared with D301 driven by hydrogen bonding interaction only and CHA111 driven by Van der Waals interaction only, phenol adsorption on aminated adsorbents driven by both hydrogen bonding and Van der Waals interactions were apparently different, i.e., negative effect for NDA105, positive effect for NDA101 and synergistic effect for NDA103. In this synergistic action, some weak interactions would contribute more or less to the adsorption than they work individually.展开更多

BACKGROUND In most elderly patients with intertrochanteric fractures,satisfactory fracture reduction can be achieved by closed reduction using a traction table.However,intertrochanteric fractures cannot achieve satisf...BACKGROUND In most elderly patients with intertrochanteric fractures,satisfactory fracture reduction can be achieved by closed reduction using a traction table.However,intertrochanteric fractures cannot achieve satisfactory reduction in a few patients,which is called irreducible intertrochanteric fractures.Especially for type 31A3 irreducible intertrochanteric fractures,limited open reduction of the broken end with different intraoperative reduction methods is required to achieve satisfactory reduction and fixation.AIM To discuss clinical efficacy of intracortical screw insertion plus limited open reduction in type 31A3 irreducible intertrochanteric fractures in the elderly.METHODS A retrospective analysis was performed on 23 elderly patients with type 31A3 irreducible intertrochanteric fractures(12 males and 11 females,aged 65-89-yearsold)who received treatment at the orthopedics department.After type 31A3 irreducible intertrochanteric fractures were confirmed by intraoperative C-arm,all of these cases received intracortical screw insertion plus limited open reduction in the broken end with intramedullary screw internal fixation.The basic information of surgery,reduction effects,and functional recovery scores of the hip joint were assessed.RESULTS All patients were followed up for 13.8 mo on average.The operation time was 53.8±13.6 min(40-95 min).The intraoperative blood loss was 218.5±28.6 mL(170-320 mL).The average number of intraoperative X-rays was 22.8±4.6(18-33).The average time to fracture union was 4.8±0.7 mo.The reduction effect was assessed using Kim’s fracture reduction evaluation.Twenty cases achieved grade I fracture reduction and three cases grade II fracture reduction.All of them achieved excellent or good fracture reduction.Upon the last follow-up,the functional recovery scores score was 83.6±9.8,which was not significantly different from the functional recovery scores score(84.8±10.7)before the fracture(t=0.397,P=0.694).CONCLUSION With careful preoperative preparation,intracortical screw insertion plus limited open reduction contributed to high-quality fracture reduction and fixation.Good clinical outcomes were achieved without increasing operation time and intraoperative blood loss.展开更多

The ratio of the oil in inclusion complex was used to evaluate the technology based on the orthogonal design. [Results] The best volatile oil extraction condition was to add 400 mL of water into the mixture of crude drugs, and to extract the herbal medicine for 2 h with advanced soaking for 6 h with water. The swollen degree of the mice s toes in the different dose groups of volatile oil from O. basilicum L. was significantly lower ( P <0.05), and the high-dose and middle-dose groups had better inhibition.[Conclusions] The volatile oil from O. basilicum L. in Bozhou had a significant inhibitory effect on xylene-induced mouse ear swelling and carrageenan-induced mouse paw swelling. It proved and clarified the anti-inflammatory effects of volatile oil from O. basilicum L. in Bozhou.展开更多

In this way, NaNO 2-Al (NO 3) 3-NaOH was used as color developing agent, and the theoretical extraction of flavone in ginkgo leaf can reach 11.74 mg/g.[Conclusions] This method can be used to extract total flavonoids from G. biloba leaves with simple operation, low cost and good repeatability.展开更多

文摘Serum pharmacochemistry of traditional Chinese medicine has become a more accurate and rapidly developing reasonable way to analyze the effective material basis of traditional Chinese medicine in recent years. Through this method, we can select very complex Chinese medicine components and clarify the effective substances consistent with the main functions, and then separate and purify effective ingredients in traditional Chinese medicine, so as to explain the material basis of the drug effect and prove the rationality and principle of action of Chinese medicine compounds. It can show the multi-component and multi-target characteristic of traditional Chinese medicine, and after analyzing the migrating components of serum, we can explore the direct effective substances that act on the patient’s body, so as to accelerate and accurately complete the in-depth study of the effective substances of Chinese medicine.

文摘AIM: To investigate matrix metalloproteinases(MMPs)and tissue inhibitor of metalloproteinases(TIMPs)expression during the progress of fusarium solani(F.solani) keratitis in a rat model.· METHODS: A rat model of F.solani keratitis was produced using corneal scarification and a hand-made contact lens. MMPs and TIMPs expressiond were explored in this rat model of F.solani keratitis using real-time polymerase chain reaction(PCR) and DIF.GM6001(400 μmol/m L) was used to treat infected corneas. The keratitis duration, amount and area of corneal neovascularization(CNV) were evaluated.·RESULTS: MMP-3 expression was 66.3 times higher in infected corneas compared to normal corneas. MMP-8,-9,and-13 expressions were significantly upregulated in the mid-period of the infection, with infected- to-normal ratios of 4.03, 39.86, and 5.94, respectively. MMP-2 and-7expressions increased in the late period, with the infected-to-normal ratios of 5.94 and 16.22, respectively.TIMP-1 expression was upregulated in the early period,and it was 43.17 times higher in infected compared to normal corneas, but TIMP-2,-3, and-4 expressions were mildly downregulated or unchanged. The results of DIF were consistent with the result of real-time PCR.GM6001, a MMPs inhibitor, decreased the duration of F.solani infection and the amount and area of CNV.·CONCLUSION: MMPs and TIMPs contributed into the progress of F.solani keratitis.

文摘Pressure is an effective and clean way to modify the electronic structures of materials,cause structural phase transitions and even induce the emergence of superconductivity.Here,we predicted several new phases of the Zr XY family at high pressures using the crystal structures search method together with first-principle calculations.In particular,the Zr Ge S compound undergoes an isosymmetric phase transition from P4/nmm-I to P4/nmm-II at approximately 82 GPa.Electronic band structures show that all the high-pressure phases are metallic.Among these new structures,P4/nmm-II Zr Ge S and P4/mmm Zr Ge Se can be quenched to ambient pressure with superconducting critical temperatures of approximately 8.1 K and 8.0 K,respectively.Our study provides a way to tune the structure,electronic properties,and superconducting behavior of topological materials through pressure.

文摘Adsorption behaviors of phenol from aqueous solutions have been investigated in batch systems at 303 K and 318 K respectively, using hypercrosslinked polymeric adsorbent (CHA111), aminated hypercrosslinked polymeric adsorbents (NDA101, NDA103, NDA105) and weakly basic polymeric adsorbent (D301) with a view to studying the effect of hydrogen bonding and Van der Waals interactions between adsorbate and the adsorbent. All adsorption isotherms can be well fitted by Langmuir and Freundlich equations. Compared with D301 driven by hydrogen bonding interaction only and CHA111 driven by Van der Waals interaction only, phenol adsorption on aminated adsorbents driven by both hydrogen bonding and Van der Waals interactions were apparently different, i.e., negative effect for NDA105, positive effect for NDA101 and synergistic effect for NDA103. In this synergistic action, some weak interactions would contribute more or less to the adsorption than they work individually.

文摘A novel ZIF-8-CMC hybrid material was fabricated from the hybridization of ZIF-8 and carboxymethylcellulose(CMC) by impregnation method for n-hexane/3-methylpentane separation.The surface properties of ZIF-8 were tailored by introducing CMC into ZIF-8 nanoparticles.In this work,adsorption separation of n-hexane(nHEX) and 3-methylpentane(3 MP) on ZIF-8-CMC were investigated by batch vapor-phase adsorption and liquid-phase breakthrough adsorption.The adsorption selectivity of nHEX/3 MP reversed from preferable adsorption of nHEX to preferable adsorption of 3 MP upon the increasing of CMC containing in the hybrid materials.As the temperature increases,the adsorption amounts of nHEX and 3 MP decrease.With the increasing of CMC contents,the nHEX uptake decreased,the uptake capacity of 3 MP increased gradually.For liquid-phase breakthrough adsorption,the dynamic adsorption capacity of nHEX also decreased with the increasing of temperature.

文摘BACKGROUND In most elderly patients with intertrochanteric fractures,satisfactory fracture reduction can be achieved by closed reduction using a traction table.However,intertrochanteric fractures cannot achieve satisfactory reduction in a few patients,which is called irreducible intertrochanteric fractures.Especially for type 31A3 irreducible intertrochanteric fractures,limited open reduction of the broken end with different intraoperative reduction methods is required to achieve satisfactory reduction and fixation.AIM To discuss clinical efficacy of intracortical screw insertion plus limited open reduction in type 31A3 irreducible intertrochanteric fractures in the elderly.METHODS A retrospective analysis was performed on 23 elderly patients with type 31A3 irreducible intertrochanteric fractures(12 males and 11 females,aged 65-89-yearsold)who received treatment at the orthopedics department.After type 31A3 irreducible intertrochanteric fractures were confirmed by intraoperative C-arm,all of these cases received intracortical screw insertion plus limited open reduction in the broken end with intramedullary screw internal fixation.The basic information of surgery,reduction effects,and functional recovery scores of the hip joint were assessed.RESULTS All patients were followed up for 13.8 mo on average.The operation time was 53.8±13.6 min(40-95 min).The intraoperative blood loss was 218.5±28.6 mL(170-320 mL).The average number of intraoperative X-rays was 22.8±4.6(18-33).The average time to fracture union was 4.8±0.7 mo.The reduction effect was assessed using Kim’s fracture reduction evaluation.Twenty cases achieved grade I fracture reduction and three cases grade II fracture reduction.All of them achieved excellent or good fracture reduction.Upon the last follow-up,the functional recovery scores score was 83.6±9.8,which was not significantly different from the functional recovery scores score(84.8±10.7)before the fracture(t=0.397,P=0.694).CONCLUSION With careful preoperative preparation,intracortical screw insertion plus limited open reduction contributed to high-quality fracture reduction and fixation.Good clinical outcomes were achieved without increasing operation time and intraoperative blood loss.

文摘[Objectives] To optimize volatile oil extraction from Ocimum basilicum with the water vapor distillation. [Methods] An orthogonal design was carried out to determine the volume of volatile oil by 3 factors: the immersion time, distillation time, amount of water. The ratio of the oil in inclusion complex was used to evaluate the technology based on the orthogonal design. [Results] The best volatile oil extraction condition was to add 400 mL of water into the mixture of crude drugs, and to extract the herbal medicine for 2 h with advanced soaking for 6 h with water. [Conclusions] The process is stable, reasonable, and feasible.

文摘[Objectives] The research aimed to study the anti-inflammatory effects of volatile oil from Ocimum basilicum L. in Bozhou through the experimental animal model of inflammation.[Methods] Xylene-induced mouse ear swelling model and carrageenan-induced mouse paw swelling model were used to compare the anti-inflammatory effects of volatile oil from O. basilicum L.[Results] The extremely high, high, medium, and low doses of volatile oil from O. basilicum L. had a certain inhibitory effect on the ear swelling of mice induced by xylene ( P <0.05), and the inhibitory effect of high-dose group on the ear swelling of mice was better. The swollen degree of the mice s toes in the different dose groups of volatile oil from O. basilicum L. was significantly lower ( P <0.05), and the high-dose and middle-dose groups had better inhibition.[Conclusions] The volatile oil from O. basilicum L. in Bozhou had a significant inhibitory effect on xylene-induced mouse ear swelling and carrageenan-induced mouse paw swelling. It proved and clarified the anti-inflammatory effects of volatile oil from O. basilicum L. in Bozhou.

文摘[Objectives] To optimize the extraction of total flavone in folium Ginkgo biloba leaves.[Methods] Using the ultrasonic extraction method, the three levels of maximum impact were selected by each factor selection, and the optimal extraction technology was obtained by box-benhnken test design analysis.[Results] After analyzing the results, the optimal process was as follows: a time of 28.89 min and liquid/material ratio of 31.38∶ 1, the temperature was 40.74℃. In this way, NaNO 2-Al (NO 3) 3-NaOH was used as color developing agent, and the theoretical extraction of flavone in ginkgo leaf can reach 11.74 mg/g.[Conclusions] This method can be used to extract total flavonoids from G. biloba leaves with simple operation, low cost and good repeatability.

This paper aims to review the latest progress in phytochemical research and pharmacological research of O. basilicum,which will assist various practitioners and clinicians in understanding the traditional and modern uses of O. basilicum.
