目的 观察1种新型细胞递送工具(MSC-exo)转运靶向基因调控胰腺癌增殖效应。方法 透射电子显微镜(transmission electron microscope,TEM)和纳米颗粒跟踪分析技术(nanoparticle tracking analysis,NTA)鉴定人间充质干细胞外泌体(human me... 目的 观察1种新型细胞递送工具(MSC-exo)转运靶向基因调控胰腺癌增殖效应。方法 透射电子显微镜(transmission electron microscope,TEM)和纳米颗粒跟踪分析技术(nanoparticle tracking analysis,NTA)鉴定人间充质干细胞外泌体(human mesenchymal stem cell exosomes,MSC-exo)并转运miR-450a-5p进入CFPAC-1,探讨miR-450a-5p靶向BZW2抑制胰腺癌细胞增殖效应。基因技术处理Pc-BZW2,CCK-8、EdU、细胞划痕、Transwell验证MSC-exo与MSC-exo-miR-450a-5p对细胞的抑制作用。结果 与胰腺正常组织相比miR-450a-5p在胰腺癌组织中低表达(P<0.05),CFPAC-1细胞MSC-exo-miR-450a-5p外泌体标记蛋白CD63、TSG101表达高于MSC-exo(P<0.05)。CCK-8、EdU、细胞划痕、Transwell实验显示MSC-exo-miR-450a-5p较MSC-exo可显著抑制CFPAC-1细胞增殖、侵袭和迁移(P<0.05)。通过双荧光素酶实验证实,miR-450a-5p靶向BZW2,并且RT-qPCR和免疫印迹检测miR-450a-5p和BZW2表达成负性相关(P<0.05)。过表达BZW2,CCK-8、EdU、细胞划痕、Transwell实验均证实,pc-BZW2逆转MSC-exo-miR-450a-5p对CFPAC-1的抑癌功能,免疫印迹检测PCNA、Ki-67、MMP2、MMP9,结果与上述实验一致(P<0.05)。结论 hMSC-exo是1种新的递送系统,靶向BZW2转运miR-450a-5p抑制胰腺癌细胞的生物学恶性,为胰腺癌靶向治疗研究提供了重要线索。 展开更多

以微晶纤维素、壳聚糖为双网络单元,以氢氧化钠/尿素/水为溶剂体系、甲基三甲氧基硅烷(MTMS)为改性剂,采用“冻融-浸渍”两步法制得超疏水纤维素/壳聚糖复合气凝胶。研究结果表明,该复合气凝胶呈现三维互穿多孔结构,其接触角随着MTMS的... 以微晶纤维素、壳聚糖为双网络单元,以氢氧化钠/尿素/水为溶剂体系、甲基三甲氧基硅烷(MTMS)为改性剂,采用“冻融-浸渍”两步法制得超疏水纤维素/壳聚糖复合气凝胶。研究结果表明,该复合气凝胶呈现三维互穿多孔结构,其接触角随着MTMS的加入,先逐渐增大后急剧减小,当MTMS的浓度为1.96 g/μL时,疏水效果最好,接触角可达到151°。此外,该气凝胶具有良好的油/水选择性吸收能力,吸油量最高达到9.3 g/g,且经过5次再生循环后,对于异丙醇的吸附量仍能保持初始值的92.8%,是一种良性可再生的生物质基吸油材料。 展开更多

HMA2(heavy metal ATPase 2)plays a crucial role in extracellular and intracellular Zn^(2+)transport across biomembranes,maintaining ion homeostasis,and playing an important role in the normal physiological metabolism,growth,and development of plants.In our study,a novel HMA2 gene,named MaHMA2,was isolated and cloned from white mulberry(Morus alba L.).The gene sequence obtained was 1,342 bp long,with an open reading frame of 1,194 bp,encoding a protein of 397 amino acids,with a predicted molecular mass of 42.852 kD and an isoelectric point of 7.53.This protein belonged to the PIB-type ATPase transport protein family.We analyzed the expression of the MaHMA2 gene by quantitative real-time PCR.The results showed that the level of MaHMA2 gene expression decreased to a Zn concentration of 800 mg/kg.Malondialdehyde and proline levels increased and responded to increasing Zn when the MaHMA2 gene was silenced,whereas the activities of peroxidase and superoxide dismutase tended to increase in response to increasing Zn^(2+)ion stress concentrations but were lower in the gene-silenced plants.These findings suggested that the MaHMA2 gene played an active role in the tolerance response of mulberry to Zn stress. 展开更多

Background:The reproductive health addresses the reproductive processes,functions and system at all stages of life.Enhancing the level of global reproductive health is the goal of sustained attention and struggle by the international community.The social and economic development in Southeast Asia is lagging behind,and its female reproductive health is worrying,while the differences of female reproductive health among different regions are significant.Objective:To obtains the necessity and urgency of strengthening the reproductive health level of Southeast Asian countries,so as to provide the basis for the priorities and target to policy-makers and health administrators to improve reproductive health.Methods:Literature review were searched in PubMed,Web of Science databases,Google scholar database,and WHO's webpages.Maternal mortality ratio,contraceptive rates,unmet need for family planning,antenatal and postnatal care coverage,and sexually transmitted disease were the five key indicators and the influence factors for female reproductive health status in Southeast Asian countries.Results:The reproductive health of Southeast Asian women were still at a lower level overall and varied in different regions and conntries.Women's education and attitude,accessibility of service,socioeconomic and cultural factors,etc.were the potential influencing factors.Conclusion:There is left quite large space for improvement to the reproductive health in Southeast Asian countries and efficient interventions can be achieved for the key and easier-improved risk factors such as education and in high-risk areas. 展开更多

Recrystallization effect on surface passivation of Hastelloy X alloy fabricated by laser powder bed fusion

Post-heat treatment is generally adopted in the additive manufacturing field due to its alleviation of high residual stress and modification of rapid-solidified multilevel heterogeneous microstructure,and the related performance of the heat-treated counterparts calls for a systemic investigation to build a criterion of the heat treatment procedure.In this work,we focus on the heat treatment effects on the recrystallization of the Hastelloy X alloy produced by the laser powder bed fusion(LPBF)method,and the related surface passivation of the heat-treated counterparts is meticulously assessed as well.Results show that the multilevel heterostructure for LPBF Hastelloy X alloy consists of sub-micro dislocation cell substructures with Cr/Mo elemental segregation,fine columnar grains,and periodically-distributed molten pools.After heat treatment,partially and fully recrystallized structures for LPBF Hastelloy X alloys were achieved at 1100 and 1200℃for 1 h,respectively.Furthermore,the as-built LPBF Hastelloy X alloy shows superior corrosion resistance while the heat-treated one(1100℃)exhibits the worst in the borate buffer solution.The growth of passive film exhibited a highly linear correlation with the nucleation process controlled by diffusion,and high dislocation density and low angle grain boundary decreased the diffusion coefficient of cation vacancies,augmenting the nucleation sites of the passive film and enhancing its growth rate.Moreover,the micro-galvanic effect resulting from the partially recrystallized microstructure actively facilitated the formation of inhomogeneous porous passive films,leading to the worst corrosion resistance. 展开更多

Laser powder bed fusion (LPBF) is a commonly used additive manufacturing (AM) method for efficiently producing intricate geometric components. This investigation examines factors such as pores, cellular structure, grain size, and inclusions from the manufacturing process that contribute to the corrosion resistance of LPBF DSS. Furthermore, the as-built LPBF duplex stainless steel (DSS) is primarily ferrite due to the rapid cooling process. Therefore, the transformation of ferrite to austenite after various heat treatments in LPBF DSS and its corresponding corrosion resistance are presented. Additionally, a new mixed powder method is proposed to increase the austenite content in the as-built LPBF DSS. This review also focuses on the passivation capability and pitting corrosion performance in LPBF and conventional DSS. This article summarizes the variations in microstructure between as-built and heat-treated LPBF DSS, with their impacts on corrosion resistance, offering insights for manufacturing highly corrosion-resistant LPBF DSS. 展开更多

Objective:To investigate the risk factors of severe postpartum hemorrhage(PPH)in cesarean section of twin pregnancy,and to provide clinical basis for pregnancy management and perioperative obstetric management of twin pregnancy.Methods:The clinical data of 631 twin pregnancies with gestational age28 weeks delivered by cesarean section at Peking University People's Hospital(PKUPH)from January 2004 to January 2017 were retrospectively analyzed.Methods of conception,the combined weight of twins,serum albumin level before cesarean section,operation time and other factors on the amount of blood loss during cesarean section were analyzed.Results:The proportion of severe PPH was significantly higher in in vitro fertilization-embryo transfer(IVF-ET)group,the combined weight of twins>6000g group,serum albumin before cesarean section<30 g/dl group than in the natural pregnancy group,4000–6000g group,<4000g group and serum albumin30 g/dl group respectively(P<0.05).The proportion of severe PPH in the elective surgery group of twin pregnancy was higher than that in the emergency surgery group,but the difference was not statistically significant(P>0.05).Moreover,according to the surgical indications,the emergency surgery group was divided into premature rupture of membranes(PROM),labor,fetal distress and others groups,no significant difference were detected among these groups(P>0.05).Conclusion:IVF-ET,the combined weight of twins,serum albumin before operation were significantly correlated with severe PPH of twin pregnancies delivered by cesarean section,revealing that it is necessary to strengthen pregnancy management of twin pregnancy. 展开更多

以微晶纤维素(Microcrystalline cellulose,MCC)和海藻酸钠(Sodium alginate,SA)为网络框架,海泡石(Sepiolite,SEP)为功能单元,采用悬浮液滴法构建纤维素-海藻酸钠-海泡石(MCC-SA-SEP)双网络多孔复合微球。通过SEM和TG对复合微球结构和... 以微晶纤维素(Microcrystalline cellulose,MCC)和海藻酸钠(Sodium alginate,SA)为网络框架,海泡石(Sepiolite,SEP)为功能单元,采用悬浮液滴法构建纤维素-海藻酸钠-海泡石(MCC-SA-SEP)双网络多孔复合微球。通过SEM和TG对复合微球结构和热稳定性能进行表征,并研究该微球对亚甲基蓝(Methylene blue,MB)水溶液的吸附性能。结果表明,MCC-SA-SEP复合微球呈现三维网络多孔结构,且随着SEP含量的增加热稳定性逐渐提高。吸附结果显示MCC-SA-SEP符合准二级动力学模型和Langmuir等温线,对MB的饱和吸附容量高达333.3 mg/g。经过五次再生循环后,对MB吸附能力仍能维持85.4%,表明该多孔复合微球可以作为一种高效可再生的有机-无机复合吸附剂用于染料废水处理。 展开更多
