The importance of mistuning analysis lies on understanding the vibration characteristics of the bladed disk.Previous works generally ignored the nonlinear effect of prestress,which increase the gap between the actual results and research results.In this paper,a unique way to address the reduced-order model is presented,where disk and each blade component is attached as individual substructures with free-interface component mode synthesis considering prestress.The influence of nonlinear inertial effect on mistuning characteristics of rotors is compared with linear inertial effect.Results show that ignoring nonlinearity of the rotational speed,the influence of speed on mode localization may be different from the true situation. 展开更多

Integrated Skin Microbiome-Metabolome Analysis Reveals Altered Bacterial Community Composition and Metabolites in Psoriasis

Objective:Current theories highlight the role of the microbiome in the pathogenesis of psoriasis.Additionally,abnormal metabolism can alter disease processes in terms of occurrence,progression,and prognosis.Therefore,an integrative microbiome and metabolome analysis of the skin may aid in understanding the disease pathogenesis and identify therapeutic targets for psoriasis.Methods:We recruited 22 patients with psoriasis and 22 age-and sex-matched healthy controls.Skin swabs were collected from the participants’scalps.All samples underwent amplicon sequencing of the internal transcribed spacer 1 and V3V416S rRNA regions and metabolome analysis.For amplicon data,analysis of the alpha diversities,microbial community structures and principal coordinate analysis were performed.Differential metabolites and pathway enrichment were analyzed for metabolome data.Studentt test and Wilcoxon rank sum test were used for statistical analysis.Results:The psoriatic lesions were characterized by higher bacterial diversity,significantly higher abundances ofCorynebacterium(P<0.001)andStaphylococcus(P=0.012),and a lower abundance ofCutibacterium(P<0.001)compared with healthy controls.However,no significant alterations in the fungal diversity or fungal taxonomies were detected.Metabolome analysis revealed that prostaglandin-related metabolites,nucleotides,and cysteine-and methionine-related metabolites were significantly enriched in patients with psoriasis,and these metabolites were positively correlated with the disease-associated bacteriaStaphylococcus andCorynebacterium.Conclusions:We demonstrated significant alterations in the skin microbiome and metabolome in patients with psoriasis compared with healthy controls. 展开更多

Background:Benvitimod cream,a novel synthetic small molecule,was effective in treating mild-to-moderate plaque psoriasis.We conducted a phase III clinical trial to assess the efficacy and safety of benvitimod cream in patients with mild-to-moderate plaque psoriasis.Methods:We randomly assigned 686 patients(2:1:1)to receive 1%benvitimod cream,0.005%calcipotriol ointment or placebo twice a day for 12 weeks.The primary efficacy end points were the percentage of patients with a 75%or greater reduction from baseline in the psoriasis area and severity index(PASI 75)score and with a score of 0 or 1 in static physician’s global assessment(sPGA)at week 12.Results:The results showed that 50.4%of patients in the benvitimod group achieved PASI 75,which was significantly higher than that in the calcipotriol(38.5%,P<0.05)and placebo(13.9%,P<0.05)groups.The proportion of patients achieving an sPGA score 0 or 1 was 66.3%in the benvitimod group and 63.9%in the calcipotriol group,which were both significantly higher than that in the placebo group(34%,P<0.05).In the long-term follow-up study,50.8%of patients experienced recurrence.After retreatment with 1%benvitimod,73.3%of patients achieved an sPGA score of 0 or 1 again at week 52.Adverse events included application site irritation,follicular papules,and contact dermatitis.No systemic adverse reactions were reported.Conclusion:During this 12-week study,benvitimod cream was demonstrated with high effectiveness and safety in patients with mild-to-moderate plaque psoriasis. 展开更多

White-opaque Switching in Different Mating Type-like Locus Gene Types of Clinical Candida albicans Isolates

Background:Candida albicans (C.albicans) can become a pathogen causing superficial as well as life-threatening systemic infections,especially in immunocompromised patients.Many phenotypic attributes contribute to its capacity to colonize human organs.In our study,93 C.albicans isolates from patients of various candidiasis in a hospital of China were surveyed.We aimed to investigate the white-opaque (WO) switching competence,drug sensitivity,and virulence of mating type-like (MTL) a/α isolates.Methods:Internal transcribed spacer (ITS) gene and the MTL configuration were detected in all the isolates by reverse transcription-polymerase chain reaction.White/opaque phenotype and doubling time of cell growth were determined.The minimum inhibitory concentrations of antifungal agent were measured using broth microdilution method.Results:Sixty-four isolates (69.6%) were classified to serotype A,19 (20.6%) to serotype B,and 9 (9.8%) to serotype C.Moreover,phylogenetic analysis showed that these isolates were divided into four different subgroups of ITS genotypes.Most of our clinical isolates were MTLa/α type,while 6.8% remained MTLa or MTLα type.The frequency of opaque phenotype was 71.0% (66 isolates).Following the guidelines of Clinical and Laboratory Standards Institute M27-A3,all isolates were susceptible to caspofungin and a few (0.6-3.2%) of them showed resistance against amphotericin B,flucytosine,fluconazole,itraconazole,and voriconazole.Conclusions:From these analyses,there were comparatively more C.albicans strains classified into serotype B,and the frequency of opaque phase strains was significant in the clinical isolates from China.Genetic,phenotypic,or drug susceptibility patterns were not significantly different from previous studies.MTLa/α isolates could also undergo WO switching which facilitates their survival. 展开更多

Background:Psoriasis is a chronic inflammatory skin disease,affecting about 0.6%of the Chinese population.Many patients are not well controlled by conventional treatments,thus there is need for new treatment regimens.In this study,we assessed the efficacy and safety of secukinumab in Chinese patients with moderate to severe plaque psoriasis.Methods:This study was a 52-week,multicentre,randomized,double-blind,placebo-controlled,parallel-group,Phase 3 trial.A sub-population of study participants(≥18 years)of Chinese ethnicity were randomized to receive subcutaneous injections of 300 or 150 mg secukinumab,or placebo.The co-primary endpoints were psoriasis area severity index(PASI)75 and Investigator’s Global Assessment(IGA)0/1 at Week 12.Results:A total of 441 Chinese patients were enrolled in this study.Co-primary outcomes were achieved;300 and 150 mg secukinumab were superior to placebo as shown in the proportion of patients that achieved PASI 75(97.7%and 87.2%vs.3.7%,respectively;P<0.001),and IGA 0/1(82.3%and 69.7%vs.2.7%;P<0.001)at Week 12.Treatment efficacy was maintained until Week 52.There was no increase in overall adverse events with secukinumab relative to placebo throughout the 52-week period.Conclusion:Secukinumab is highly effective and well tolerated in Chinese patients with moderate to severe plaque psoriasis. 展开更多

Photoenzymatic reversible addition-fragmenatation chain transfer(RAFT)emulsion polymerization,surfactant-free or ab initio,of various monomers is reported with oxygen toleranee.In surfactant-free emulsion polymerizatoin,poly(N,N-dimethylacrylamide)s were used as stabilizer blocks for emulsion polymerization of methyl acrylate,n-butyl acrylate and styrene,producing well-defined amphiphilic block copolymers,including those with an ultrahigh molecular weight,at quantitative conversions.The controlled character of surfactant-free emulsion polymerization was confirmed by kinetic studies,chain extension studies and GPC analyses.Temporal control was demonstrated by light ON/OFF experiments.In ab initio emulsion polymerization of methyl acrylate and methyl methacrylate,low-dispersity hydrophobic polymers were synthesized with predictable molecular weights.This study extends the monomer scope suitable for photoenzymatic RAFT polymerization from hydrophilic to hydrophobic monomers and demonstrates that oxygen-toleranee can be equally achieved for emulsion polymerization with excellent RAFT control. 展开更多

As a result of fundamental changes in the International Code of Nomenclature on the use of separate names for sexual and asexual stages of fungi,generic names of many groups should be reconsidered.Members of the ECMM/ISHAM working group on Pseudallescheria/Scedosporium infections herein advocate a novel nomenclature for genera and species in Pseudallescheria,Scedosporium and allied taxa.The generic names Parascedosporium,Lomentospora,Petriella,Petriellopsis,and Scedosporium are proposed for a lineage within Microascaceae with mostly Scedosporium anamorphs producing slimy,annellidic conidia.Considering that Scedosporium has priority over Pseudallescheria and that Scedosporium prolificans is phylogenetically distinct from the other Scedosporium species,some name changes are proposed.Pseudallescheria minutispora and Petriellidium desertorum are renamed as Scedosporium minutisporum and S.desertorum,respectively.Scedosporium prolificans is renamed as Lomentospora prolificans. 展开更多

To the Editor:A 13-year-old girl with Crohn disease(CD)from Shandong Province transferred to our inflammatory bowel disease(IBD)center during April 2013.Three months previously,the patient presented with hematochezia and fever,but antibiotic treatment was unsuccessful.After admission,a colonoscopy indicated manifestations of segmental lesions,cobble stoning,and stricture[Figure 1 A].Cytomegalovirus,Epstein-Barr virus,Clostridium difficile^and amoebae were excluded as possible causes.There were also no indications of Behcet disease,systemic vasculitis,or other autoimmune diseases.Our initial diagnosis was active severe CD(Vienna typing,A1L3B2).This study was approved by the Ethical Committee of First Hospital of Peking University(No.2019153).An appropriate signed patient consent form was obtained. 展开更多
