针对矩阵式测试盘上芯片偏移检测问题,搭建一套基于机器视觉的芯片偏移检测系统,主要包括上下料流道、三维移动平台和视觉检测模块。检测系统以一标准样板为基准,构建模板匹配与仿射变换相结合的图像矫正算法,实现不同待测样本间检测的... 针对矩阵式测试盘上芯片偏移检测问题,搭建一套基于机器视觉的芯片偏移检测系统,主要包括上下料流道、三维移动平台和视觉检测模块。检测系统以一标准样板为基准,构建模板匹配与仿射变换相结合的图像矫正算法,实现不同待测样本间检测的通用性;以灰度梯度及梯度方向为依据,设计针对芯片区域的矩形测量算法;系统以芯片左上角和右下角点连线的中心为基准点计算芯片偏移量。实验结果表明,本系统针对芯片偏移检测的误差范围-2.145~4.257μm,单片芯片偏移计算算法执行的平均时间为72.56 ms,检测轴运行速度为20 mm/s,对行列数为5×12的矩阵式测试盘上芯片的平均检测时间为64.5 s/盘,可满足实际加工过程的需求;使用测量系统分析(MSA)中的偏倚与线性度分析方法对本系统的准确性进行评估,结果表明系统的偏倚和线性均满足生产需求。 展开更多

Bovine cumulus-oocyte complexes were cultured in the maturation medium containing 4 different concentrations of verapamil and trifluoperazine to testify the necessity of extracellular Ca2+ and Ca2+-calmodulin complex for the resumption and completion of meiosis as well as cumulus expansion.Ultrastructure of the treated oocytes was also observed to investigate the cytoplasm maturation. The results showed that verapamil didn't influence the cumulus expansion,meiosis resumption and completion and cytoplasm maturation significantly.TFP inhibited cumulus expansion in a dose-dependent manner.25 μm trifluoperazine significantly inhibited the GVBD and maturation(p<0.01),wherease 1 μm TFP had no effect.Both oocytes and cumulus cells treated with 25 μm TFP severely degenerated.Our observations suggest that the resumption and completion of meiosis and cumulus expansion are Ca2+ -CaM dependent and blocking membrane Ca2+ channel does not influence oocyte germinal vesicle breakdown,nuclear and cytoplasm maturation significantly in cattle. 展开更多

The in vitro maturation, fertilization and development of bovine ovary oocytes in two differentcultural systems A and B were studied under the conditions of 38.5℃,5%CO2,95%air and 100%humidity.The maturation rates were 94.5%and 91.3%,respectively,and the difference wasextremely significant.Frozen semen were thawed and sperm were capacitated with three kinds ofcapacitation agents for fertilization.The pronucleus rates were 76%,65%-68%and 62%respectively.The rates of embryos developed to morula and blastocyst were 19%,16% and 17%respectively.The developmental rates of embryos cocultured with bovine oviductal epithelium cellsand bovine granulosa cells were 25% and 23.4% respectively,with no significant difference. Freshembryos were transplanted into 15 recipicns,and three of them were pregnant and calves were bornin 1990 and 1991.The pregnant rate was 20%.The emryos developed faster before 8-cell stage andslower after 8-cell stage,in vitro than in vivo. 展开更多

目的:比较两种精蛋白锌重组赖脯胰岛素混合注射液(50R)(万邦生化)和优泌乐50(礼来制药)治疗2型糖尿病(T2DM)的有效性和安全性。方法:本研究为多中心、随机、平行、阳性对照、Ⅲ期临床研究。自2018年8月30日至2020年5月27日从全国36个中... 目的:比较两种精蛋白锌重组赖脯胰岛素混合注射液(50R)(万邦生化)和优泌乐50(礼来制药)治疗2型糖尿病(T2DM)的有效性和安全性。方法:本研究为多中心、随机、平行、阳性对照、Ⅲ期临床研究。自2018年8月30日至2020年5月27日从全国36个中心入选612例口服降糖药血糖控制不佳的T2DM患者,采用随机数字表法按1∶1分配进入试验组(万邦生化)和对照组(优泌乐50,礼来制药),治疗16周。比较两组患者治疗前后糖化血红蛋白(HbA 1c)、空腹血糖(FPG)、标准餐餐后2 h血糖(2hPG)、体重、血脂、HbA 1c达标率以及低血糖事件、不良事件的发生率和胰岛素抗体阳性率。组间比较采用t检验、Wilcoxon秩和检验、χ2检验或Fisher精确概率法。对HbA 1c、FPG和2hPG治疗前后变化的组间比较采用协方差分析法进行检验。结果:612例患者中,进入安全性数据集609例,其中试验组305例,对照组304例;进入符合方案集534例,其中试验组267例,对照组267例。符合方案集中,试验组和对照组HbA 1c分别下降了1.85%±1.23%和1.90%±1.27%,FPG分别下降了(3.56±2.99)和(3.72±3.26)mmol/L,2hPG分别下降了(6.94±5.41)和(7.07±5.10)mmol/L,上述指标两组间差异均无统计学意义(P>0.05)。试验组和对照组总体低血糖发生率[分别为51.15%(156/305)和45.72%(139/304)]、不良事件发生率[分别为76.39%(233/305)和77.63%(236/304)]以及胰岛素抗体转阳率[分别为22.85%(61/267)和19.10%(51/267)]差异均无统计学意义(P>0.05)。结论:精蛋白锌重组赖脯胰岛素混合注射液50R(万邦生化)治疗T2DM的有效性和安全性与优泌乐50(礼来制药)相当,具有良好的耐受性。 展开更多

A method for DNA isolation from early development of blastocyst and further analysis of nuclear and mitochondrial DNA was developed in present study. Total DNA was prepared from interspecies reconstructed blastocyst and a giant panda specific microsatellite locus g010 was successfully amplified. DNA sequencing of the PCR product showed that two sequences of reconstructed blastocysts are the same as that of positive control giant panda. Our results prove that the nucleus of interspecies reconstructed blastocyst comes from somatic nucleus of donor giant panda. 展开更多

Interspecies somatic nuclear transfer (NT) may provide a new approach for preservation of the endangered rare species. Previous interspecies cloning studies have shown that a nucleus from a quiescent somatic cell supports early development of reconstructed embryos in the ooplasm from another species. In this study, we transferred nonqui-escent somatic cells from a giant panda into the perivitelline space of the enucleated rabbit oocytes. After electrofusion (at the rate of 71.6%) and electrical activation, 4.2% of the panda-rabbit reconstructed embryos developed to blastocyst in vitro. For improving the development rate of reconstructed embryos, we used serial NT in this study, i.e. blas-tomeres from reconstructed morulae were transferred into the perivitelline space of the enucleated rabbit oocytes. The fusion rates in the groups of serial I, serial Ⅱ and serial Ⅲ were 79.5%, 84.1% and 78.0%, respectively, having no difference with that of somatic group. Oocytes collected from prepubertal gilt ovaries were matured in vitro (IVM), and fertilized in vitro (IVF) or electrically activated. Phosphorylation of mitogen-activated protein kinase (MAPK) was detected with SDS-PAGE and Western blotting, and translocation of ERK2 was observed with immunofluorescent cytochemistry. We found that the quantity of MAPK kept unchanged during oocyte maturation. There was no phosphorylated MAPK in porcine oocytes at the germinal vesicle (GV) stage; a little MAPK was phosphorylated at 20 h of IVM; a high level phosphory-lation was detected at 30 h, while MAPK phosphorylation decreased at 36 h; and then MAPK phosphorylation increased again to the peak level from 40 to 60 h. ERK2 translocated from the peripheral cytoplasm to inner cytoplasm and nuclear area during oocyte maturation. There was nearly no phosphorylated MAPK at 18 and 20 h of electrically activated oocytes, but phosphorylation increased at 22 h. There was no phosphorylated MAPK at 12 h of IVF, while 展开更多
