The process of entrainment-mixing between cumulus clouds and the ambient air is important for the development of cumulus clouds.Accurately obtaining the entrainment rate(λ)is particularly important for its parameterization within the overall cumulus parameterization scheme.In this study,an improved bulk-plume method is proposed by solving the equations of two conserved variables simultaneously to calculateλof cumulus clouds in a large-eddy simulation.The results demonstrate that the improved bulk-plume method is more reliable than the traditional bulk-plume method,becauseλ,as calculated from the improved method,falls within the range ofλvalues obtained from the traditional method using different conserved variables.The probability density functions ofλfor all data,different times,and different heights can be well-fitted by a log-normal distribution,which supports the assumed stochastic entrainment process in previous studies.Further analysis demonstrate that the relationship betweenλand the vertical velocity is better than other thermodynamic/dynamical properties;thus,the vertical velocity is recommended as the primary influencing factor for the parameterization ofλin the future.The results of this study enhance the theoretical understanding ofλand its influencing factors and shed new light on the development ofλparameterization. 展开更多

Cloud microphysical properties are significantly affected by entrainment and mixing processes.However,it is unclear how the entrainment rate affects the relative dispersion of cloud droplet size distribution.Previously,the relationship between relative dispersion and entrainment rate was found to be positive or negative.To reconcile the contrasting relationships,the Explicit Mixing Parcel Model is used to determine the underlying mechanisms.When evaporation is dominated by small droplets,and the entrained environmental air is further saturated during mixing,the relationship is negative.However,when the evaporation of big droplets is dominant,the relationship is positive.Whether or not the cloud condensation nuclei are considered in the entrained environmental air is a key factor as condensation on the entrained condensation nuclei is the main source of small droplets.However,if cloud condensation nuclei are not entrained,the relationship is positive.If cloud condensation nuclei are entrained,the relationship is dependent on many other factors.High values of vertical velocity,relative humidity of environmental air,and liquid water content,and low values of droplet number concentration,are more likely to cause the negative relationship since new saturation is easier to achieve by evaporation of small droplets.Further,the signs of the relationship are not strongly affected by the turbulence dissipation rate,but the higher dissipation rate causes the positive relationship to be more significant for a larger entrainment rate.A conceptual model is proposed to reconcile the contrasting relationships.This work enhances the understanding of relative dispersion and lays a foundation for the quantification of entrainment-mixing mechanisms. 展开更多

TNF-α促进上皮间质转化在肥胖相关性结肠炎-癌进展中的作用

目的研究上皮间质转化(EMT)在肥胖相关性结肠炎-癌演变进程中的作用及其可能的机制。方法昆明小鼠30只,以高脂饲料喂养成功诱导小鼠肥胖后,将24只高脂肥胖小鼠随机分为空白对照组(n=6)、普通饲喂模型组(n=8)和高脂模型组(n=10),随后采... 目的研究上皮间质转化(EMT)在肥胖相关性结肠炎-癌演变进程中的作用及其可能的机制。方法昆明小鼠30只,以高脂饲料喂养成功诱导小鼠肥胖后,将24只高脂肥胖小鼠随机分为空白对照组(n=6)、普通饲喂模型组(n=8)和高脂模型组(n=10),随后采用促炎剂葡聚糖硫酸钠(DSS)及基因毒性造模剂氧化偶氮甲烷(AMO)模拟小鼠结肠炎-癌演进模型,HE染色观察结肠炎-癌进展的组织病理学改变,揭示高脂诱导的肥胖与结肠癌之间的关系;IHC检测TNF-α、E-cadherin以及N-cadherin的表达,计算阳性染色细胞数进行统计学分析,进一步阐明EMT与肥胖相关性结肠癌之间的关系。结果高脂模型组小鼠造模后肠上皮细胞失去原有的结构,并有大量炎症细胞浸润,出现大量类似上皮内瘤变的病理改变,而普通饲喂组小鼠在造模后结肠组织腺体消失,炎症细胞浸润明显,但上皮内瘤变的形成明显少于高脂模型组;空白对照组、普饲模型组及高脂模型组小鼠的结肠组织中TNF-α阳性染色细胞计数分别为(1.333±0.492)个/视野、(15.60±2.261)个/视野、(25.82±6.353)个/视野,与空白对照组比较,普饲及高脂模型组小鼠的结肠组织中TNF-α阳性染色细胞数明显增加,而且高脂模型组小鼠TNF-α阳性染色细胞数增加较普饲模型组更多,差异均具有显著统计学意义(P<0.01);空白对照组小鼠E-cadherin和N-cadherin阳性染色细胞数分别为(38.77±4.969)个/视野、(2.667±0.866)个/视野,饲模型组小鼠分别为(22.38±2.785)个/视野、(5.273±1.794)个/视野,而高脂模型组小鼠E-cadherin阳性染色细胞数为(9.583±2.392)个/视野,明显减少,N-cadherin阳性染色细胞数为(16.36±3.641)个/视野,显著增加,与空白对照组和普饲模型组相比较,差异均具有显著统计学意义(P<0.01)。结论高脂诱导的肥胖与结肠癌的发生存在一定的关系,其具体机制可能与肥胖引起的慢性炎症导致EMT的发生有关。 展开更多

Due to the critical correlation between inflammation and carcinogenesis, a therapeutic candidate with anti-inflammatory activity may find application in cancer therapy. Here, we report the therapeutic efficacy of celastrol as a promising candidate compound for treatment of pancreatic carcinoma via na?ve neutrophil membrane-coated poly(ethylene glycol) methyl ether-block-poly(lactic-co-glycolic acid)(PEG-PLGA) nanoparticles. Neutrophil membranecoated nanoparticles(NNPs) are well demonstrated to overcome the blood pancreas barrier to achieve pancreas-specific drug delivery in vivo. Using tumor-bearing mice xenograft model, NNPs showed selective accumulations at the tumor site following systemic administration as compared to nanoparticles without neutrophil membrane coating. In both orthotopic and ectopic tumor models,celastrol-loaded NNPs demonstrated greatly enhanced tumor inhibition which significantly prolonged the survival of tumor bearing mice and minimizing liver metastases. Overall, these results suggest that celastrol-loaded NNPs represent a viable and effective treatment option for pancreatic carcinoma. 展开更多
