Acidic digestion is an important digestive process of marine fish.In fish stomach, two enzymes are involved in the secretion of hydrochloric acid(HCl)and proteomic digestion:H^(+)/K^(+)-ATPase and pepsinogen. However, the starting of digestive function in fish is still unclear. To reveal the details of acidic digestion of turbot Scophthalmus maximus in early development,a 40 day of turbot larvae culture was conducted. The H^(+)/K^(+)-ATPase gene from the turbot S.maximus(smH^(+)/K^(+)-ATPase)was identified and characterized.Based on our previous discription on pepsinogen of turbot S.maximus,we combined pepsinogen and H^(+)/K^(+)-ATPase and analyzed the mechanism of acidic digestion in turbot. Results show that the spatial and temporal expression profiles of H^(+)/K^(+)-ATPase agreed with pepsinogen A and C in turbot, indicating a synergetic action between H^(+)/K^(+)-ATPase and pepsinogen during the acidic digestion process. In addition, the turbot juveniles showed a faster growth after the expressions of H^(+)/K^(+)-ATPase gene and pepsinogen gene, demonstrating that pepsin had a higher digestive efficiency, for which a compound diet should be provided to the fish from Day 22 onward. This study provided a reference for biology research and aquaculture of turbot and other marine fishes. 展开更多

Dissolved oxygen(DO)is one of most important factors which affect wide range physiologic features of including immune responses and intestinal bacterial community. However, the underlying mechanisms remain enigmatic. To address this question, the intestinal bacterial community compositions and the immune features of Atlantic salmon(Salmo salar)grown in recirculating aquaculture systems(RAS)were characterized.Fish were reared under different DO saturation levels,e.g.,200% saturation named high group(H),100%saturation named control group(CK),and 60%saturation named lower group(L).Large variations in the operational taxonomic units(OTUs)frequency distribution for the intestinal bacterial community of Atlantic salmon were observed. The intestinal bacterial community of all groups was dominated mainly by three phyla,e.g.,Proteobacteria.Firmicutes,and Bacteroidetes.Interestingly,Acinetobacter baumannii,an opportunistic pathogen of salmon was increased significantly in L group. We further monitored the immunity features of fish under different DO levels. The results show that leucocyte number, cortisol level, the expressions of interleukin-1β(IL-1β),Toll-like receptor 4(TLR4),and nucleotide-binding oligomerization domain like protein 2(NOD2)were higher at significant levels in the L group than those in the other two groups.TLR4 and NOD2 are usually related with the bacterial infections;therefore,it is reasonable to believe that the stronger immune responses observed in the L group might be related with the higher abundance of A.baumannii in the inte stine of Atlantic salmon.Overall, these findings demonstrated that low DO level may induce stronger immunity responses in Atlantic salmon. 展开更多

In most fish, reproduction is seasonal or periodic under the suitable conditions. In turbot(Scophthalmus maximus)farms, one of the most economically important marine flatfish species, changes in daylength could cause changes in the spawning time.In this study,to characterize the regulation of reproductive physiology following light signals, three melatonin receptors(Mtnr)investigated in turbot were named sm Mtnr1,sm Mtnr2,and sm Mtnr1 c.Distinct expression profiles demonstrated that Mtnr m RNAs were concentrated in the brain(as detected in the hypothalamus(Hy)and mesencephalon(Me)),gonad and eye.The most abundant Mtnr1 and Mtnr2 m RNA expression levels were detected in the central nervous system at the beginning of the breeding season, suggesting that Mtnr1 and Mtnr2 may play vital roles in the regulation of turbot gonadal development. In addition, the melatonin profiles gradually increased and reached to the highest level at the spawning stage, indicating that melatonin is a potent hormone in the regulation of fish oocyte growth and maturation. The results of this study suggested that melatonin is the primary factor that transduces the light signal and regulates the physiological functions of turbot seasonal reproduction. Moreover, the results of this study may establish a foundation for further research seeking to identify fish melatonin receptors involved in the gonadal development and gamete maturation. 展开更多

A bidirectional in vitro brain–computer interface(BCI)directly connects isolated brain cells with the surrounding environment, reads neural signals and inputs modulatory instructions. As a noninvasive BCI,it has clear advantages in understanding and exploiting advanced brain function due to the simplified structure and high controllability of ex vivo neural networks. However, the core of ex vivo BCIs,microelectrode arrays(MEAs),urgently need improvements in the strength of signal detection, precision of neural modulation and biocompatibility.Notably,nanomaterial-based MEAs cater to all the requirements by converging the multilevel neural signals and simultaneously applying stimuli at an excellent spatiotemporal resolution,as well as supporting long-term cultivation of neurons. This is enabled by the advantageous electrochemical characteristics of nanomaterials, such as their active atomic reactivity and outstanding charge conduction efficiency, improving the performance of MEAs.Here,we review the fabrication of nanomaterial-based MEAs applied to bidirectional in vitro BCIs from an interdisciplinary perspective. We also consider the decoding and coding of neural activity through the interface and highlight the various usages of MEAs coupled with the dissociated neural cultures to benefit future developments of BCIs. 展开更多

Black rockfish(Sebastes schlegelii)is one of the most important marine economic viviparous fishes.Recently, germplasm degradation and genetic diversity reduction have occurred due to overfishing and long-term artificial breeding.Germ cell transplantation combined with cryopreservation may be an alternative way to protect genetic resources. However,in viviparous fish that undertake fertilization and embryo development in vivo, transplantation is more difficult than in oviparous fish, including selection of transplantation stage, isolation of germ stem cells, and preparation of sterile recipients. This seriously restricts the development of viviparous transplantation. Therefore,in this study,we aimed to explore a transplantation method suitable for these species.Donor cells were isolated from cryopreserved whole testes of 300–400g male Sebastes schlegelii in May,labeled by PKH26,and intra-peritoneally transplanted into allogeneic larvae at 5–10 days post-birth.Subsequently,the development of donor-derived cells in recipients were continuously detected by fluorescence labeling, histology, microsatellite markers, and fecundity tests. The results showed that donors were rich in spermatogonia(75%)and recipients maintained a high survival rate after transplantation, with a rate of>20%at sexual maturity.Further,donor-derived cells successfully migrated(100%),colonized,and incorporated into the developing recipient gonad(93.33%).Finally,transplanted recipients could normally develop and differentiate into male and female individuals, with donor-derived gametes found in 65.38%of mature recipients. In the present study,we first establish a simple and suitable transplantation method for Sebastes schlegelii using immature males and specific larvae, which will serve as a promising tool in the protection of germplasm resources for this transplantation-restricted marine viviparous species. 展开更多

This paper presents structures and principles of a fast stockpile simulation engine that enhances automatic material process by providing accurate digital information of material quality distribution within stockpiles.Compared with traditional measurementbased approaches, our simulation technology reduces operation cost by eliminating frequent physical measuring;improves operation accuracy through all-time whole-area contour simulation(instead of measuring a few positions of material layers now and then);and speeds the process through fast simulation and instant response to quality information query.Based on simplified grain dynamics and cellular automata models, the technology can achieve real-time/super-real-time simulation, which is critical to be adopted by the industry.Application of the technology in analyzing mineral ores handling is also discussed. 展开更多

Grid cells with stable hexagonal firing patterns in the medial entorhinal cortex(MEC)carry the vital function of serving as a metric for the surrounding environment.Whether this mechanism processes only spatial information or involves nonspatial information remains elusive. Here,we fabricated an MEC-shaped microelectrode array(MEA)to detect the variation in neural spikes and local field potentials of the MEC when rats forage in a square enclosure with a planar,three-dimensional object and social landmarks in sequence. The results showed that grid cells exhibited rate remapping under social conditions in which spike firing fields closer to the social landmark had a higher firing rate.Furthermore, global remapping showed that hexagonal firing patterns were rotated and scaled when the planar landmark was replaced with object and social landmarks. In addition, when grid cells were activated, the local field potentials were dominated by the theta band(5–8 Hz),and spike phase locking was observed at troughs of theta oscillations. Our results suggest the pattern separation mechanism of grid cells in which the spatial firing structure and firing rate respond to spatial and social information, respectively, which may provide new insights into how the brain creates a cognitive map. 展开更多
