Firstly, a sparse measurement matrix construction method based on a logistic sequence is proposed, which conducts nonlinear transformation for the determined logistic sequence, making it obey uniform distribution, then conducts sign function mapping, and generates the pseudorandom sequence with Bernoulli distribution, thus leading to good signal recovery under down-sampling and easy availability for engineering realization. Secondly, in combination with the RMA imaging approach, the dictionary with all scene information and phase error correction is constructed for CS signal recovery and error correction. Finally, the non-quadratic solution model jointing imaging and phase error correction based on regularization is built, and it is solved by two steps - the separable surrogate functionals (SSF) iterative shrinkage algorithm is adopted to realize target scattering estimate; the iteration mode is adopted for the correction of the dictionary model, so as to achieve the goal of error correction and highly-focused imaging. The proposed approach proves to be effective through numerical simulation and real measurement in anechoic chamber. The results show that, the proposed approach can realize high-resolution imaging in the case of less data; the designed measurement matrix has better non-coherence and easy availability for engineering realization. A R. simsii variety "Purple Crane" was selected as the experimental material, and its flower organs were sampled at nine floral developmental stages to measure the contents of gibberellic acid-3 (GA3 ), abscisic acid (ABA), indole-3-acetic acid (IAA), and zeatin riboside (ZR) through enzyme-linked immunosorbent assay (ELISA). The results showed that the content of ZR was highest at full flowering stage; IAA and GA3 contents were higher than other stages in the process of flower bud differentiation and flowering. ABA played an important role in the regulation of flower bud formation. The results will provide theoretical references for exploring the regulation of plant hormones towards Rhododendron flower traits and breeding new varieties.展开更多

Zn supplements may reduce injury to cell membranes of the thyroid gland due to iodine deficiency. OBJECTIVE: To establish an iodine deficiency rat model using low-iodine food, which was supplemented with compound Zn and Zn gluconate, to observe the effects of Zn on brain development, as well as pituitary gland and thyroid gland function in iodine-deficient rats. DESIGN, TIME AND SETTING: Randomized grouping study of neural development was performed in the central laboratory of Shandong Institute for Prevention and Treatment of Endemic Disease from 1998 to 1999. MATERIALS: A total of 270 Wistar, female rats, one month after weaning, were used in this study, including 150 pregnant and 120 neonatal rats. Rats were randomly divided into six groups: normal control, model, iodine, compound Zn, iodine and compound Zn, and zinc gluconate. Each group contained 25 pregnant rats and 20 neonatal rats. METHODS: The pregnant rats and 20 neonatal rats, and well as the normal group, were fed standard chow and allowed free access to tap water (containing 5 u g/L iodine and 1 mg/L Zn). The remaining five groups were fed low-iodine chow. However, the model group received distilled water, the iodine group received potassium-iodide distilled water (containing 300 u g/L iodine), the compound Zn group received distilled water and intragastrically administrated 10 mL/kg compound Zn solution, once per day, the iodine and compound Zn group received distilled water with 300 u g/L iodine and intragastrically administrated 10 mL/kg compound Zn solution, once per day. All treatments lasted 90 days. MAIN OUTCOME MEASURES: All pregnant rats were sacrificed on the day 21 of pregnancy. Body mass number and rate of fetal absorption, as well as fetal death and malformation, were determined. Thyroid and pituitary gland weights were measured, as well as serum levels of thyroid hormone, gonadotropin, and sex hormones. In the experimental study of neonatal rats, the animals normally gave birth at day 21. At day 45 after birth of the neonatal rats, thyroid and pituitary gland weights were measured, and protein, DNA, and RNA concentrations were measured. RESULTS: Pregnant rats in the iodine group exhibited decreased urine iodine and body mass (F = 7.37, P 〈 0.01 ), increased thyroid absolute and relative weight (F= 7.01, 50.27, P 〈 0.01), as well as decreased T4 and FF4 (F = 7.01, 29.32, P 〈 0.01 ) and increased T3 and VI'3 (F = 41.20, 5.94, P 〈 0.01). Gonadotropic and sexual hormones were abnormal. The pregnant rats displayed decreased weight gain, and the rates of malformation, dead, and absorbed fetuses were increased. Compared with the control group, the neonatal rats with iodine deficiency exhibited lower brain weights (P 〈 0.01 ). Brain protein, DNA, and RNA, concentrations were decreased, with a rate of RNA/DNA (F = 5.70, 55.86, 25.65, 5.44, P 〈 0.01). Body mass was gradually increased (F= 6.74, P 〈 0.01), and the thyroid glands were enlarged (F= 50.01, 76.13, P 〈 0.01). Following Zn administration, thyroid gland weight was decreased in pregnant rats (P 〈 0.01 ). Thyroid hormone, gonadotropic hormones, and sexual hormones were restored to some degree. Fetal weight was increased, and the rates of malformation, dead, and absorbed fetuses were decreased. At the same time, neonatal rats gained body weight, displayed decreased thyroid gland weight, as well as increased protein, DNA, and RNA concentrations in the brain. The ratio of RNA/DNA and protein/DNA increased following Zn administration (P 〈 0.01 ). CONCLUSION: Zn supplementation may decrease the degree of goiter, ameliorate thyroid hormone disorder, as well as gonadotropic and sexual hormone disorders, and increase protein, DNA, and RNA content. Zn supplementation antagonized reproductive abnormalities in pregnant rats, decreased fetal growth,and disturbed brain development in neonatal rats as a result of iodine deficiency.展开更多
(–)-Epicatechin-3-gallate(ECG),a bioactive polyphenolic compound,has contributed a lot to the health benefits of green tea.Great attention has been focused on(–)-epigallocatechin-3-gallate(EGCG),but limited research...(–)-Epicatechin-3-gallate(ECG),a bioactive polyphenolic compound,has contributed a lot to the health benefits of green tea.Great attention has been focused on(–)-epigallocatechin-3-gallate(EGCG),but limited research has been performed towards ECG.Like EGCG,ECG also possesses various pharmacological and physiological properties,such as mediation of antioxidant activities,anti-inflammation response,regulation of cell proliferation and apoptosis,as well as anticancer properties during angiogenesis,invasion and metastasis stages.Nontoxic ECG has various molecular targets within the cells,including CYP enzymes,phaseⅡdetoxification and antioxidant enzymes,as well as pro-inflammatory mediators.The antineoplastic mechanism contains inhibition of phase 1 CYP enzymes,induction of phaseⅡdetoxification and antioxidant enzymes,high anti-inflammatory efficacy,arrest of cell cycle progression,regulation of apoptosis,as well as mediation of metastasis processes.In particular,the gallate moiety of ECG is critical for mediating inhibitory effects towards cancer cells.Besides regulation of intracellular signaling pathways,ECG also inhibits RNase A and matrix metalloproteinase enzymatic activity via chelating metals(copper and zinc)in cancer cells.This review has summarized recent studies on pharmacological properties of ECG,and discussed corresponding mechanism on modulation of cellular signaling events by ECG,hoping to broaden its multiple usage.展开更多

In total, 5 differential fragments were cloned and sequenced. Among 356 positive colonies verifi...In this study, an (AG) n microsatellite-enriched library of Rhododendron simsii Planch. was constructed with FIASCO ( fast Isolation by AFLP sequences containing repeats) method. Among 356 positive colonies verified by colony PCR of the (AG)n microsatellite-enriched library, 233 colonies were selected for further sequencing. According to the results, 202 colonies containing microsatellites were obtained, accounting for 86.7%. Discarding sequences with miscella- neous peaks, overlapping peaks and weak signals, 39 sequences were screened finally for subsequent primer exploitation, including 22 perfect type SSR loci, 5 im- perfect type SSR loci and 12 compound type SSR loci. The FIASCO method is efficient and feasible for constructing microsatellite-enriched libraries of Rhododendron spp. , which laid a solid foundation for isolation of microsatellite loci and investigation of genetic diversity and genetic structure of Rhododendron spp.展开更多

Azalea is one of the ten famous flowers in China,as well a famous flower in the world,and the whole plant has medicinal value. This study investigated the change profile of gene methylation during the four flowering stages of Rhododendron pulchrum by methylation sensitive amplification polymorphism( MSAP),and 412 differential bands were obtained after selective amplification with screened 18 pairs of effective primers. Among the differential bands,23 differential bands were selected and subjected to recovery,sequencing and data alignment,and it was found that chalcone synthase,cowpox mitochondria,birch liana mitochondria,HDR reductase in camellia,Vaccinium macrocarpon mitochondria,Cyperus rotundus mitochondria protein,Shigella flexneri plasmid and Scophthalmus maximus cD NA-AFLP fragment have a regulating effect on azalea epigenetic inheritance,and next,genes related to the flowering period of R. pulchrum would be further studied,thereby improving the economic value of R. pulchrum.展开更多
The coronavirus disease 2019(COVID-19),caused by SARS-CoV-2,has spread around the world with high mortality.To diagnose promptly and accurately is the vital step to effectively control its pandemic.Dynamic characteris...The coronavirus disease 2019(COVID-19),caused by SARS-CoV-2,has spread around the world with high mortality.To diagnose promptly and accurately is the vital step to effectively control its pandemic.Dynamic characteristics of SARSCoV-2-specific antibodies which are important for diagnosis of infection have not been fully demonstrated.In this retrospective,single-center,observational study,we enrolled the initial 131 confirmed cases of COVID-19 at Jin-Yin-Tan Hospital who had at least one-time antibody tested during their hospitalization.The dynamic changes of IgM and IgG antibodies to SARS-CoV-2 nucleocapsid protein in 226 serum samples were detected by ELISA.The sensitivities of IgM and IgG ELISA detection were analyzed.Result showed that the sensitivity of the IgG ELISA detection(92.5%)was significantly higher than that of the IgM(70.8%)(P\0.001).The meantimes of seroconversion for IgM and IgG were6 days and 3 days,respectively.The IgM and IgG antibody levels peaked at around 18 days and 23 days,and then IgM fell to below the baseline level at about day 36,whereas IgG maintained at a relatively high level.In conclusion,antibodies should be detected to aid in diagnosis of COVID-19 infection.IgG could be a sensitive indicator for retrospective diagnosis and contact tracing,while IgM could be an indicator of early infection.展开更多

文摘(–)-Epicatechin-3-gallate(ECG),a bioactive polyphenolic compound,has contributed a lot to the health benefits of green tea.Great attention has been focused on(–)-epigallocatechin-3-gallate(EGCG),but limited research has been performed towards ECG.Like EGCG,ECG also possesses various pharmacological and physiological properties,such as mediation of antioxidant activities,anti-inflammation response,regulation of cell proliferation and apoptosis,as well as anticancer properties during angiogenesis,invasion and metastasis stages.Nontoxic ECG has various molecular targets within the cells,including CYP enzymes,phaseⅡdetoxification and antioxidant enzymes,as well as pro-inflammatory mediators.The antineoplastic mechanism contains inhibition of phase 1 CYP enzymes,induction of phaseⅡdetoxification and antioxidant enzymes,high anti-inflammatory efficacy,arrest of cell cycle progression,regulation of apoptosis,as well as mediation of metastasis processes.In particular,the gallate moiety of ECG is critical for mediating inhibitory effects towards cancer cells.Besides regulation of intracellular signaling pathways,ECG also inhibits RNase A and matrix metalloproteinase enzymatic activity via chelating metals(copper and zinc)in cancer cells.This review has summarized recent studies on pharmacological properties of ECG,and discussed corresponding mechanism on modulation of cellular signaling events by ECG,hoping to broaden its multiple usage.

文摘Persimmon ( Diospyros kaki Thunb. ) , a member of Ebenaceae family, is known as "woody grain" and "iron-stem crop". However, the epigenetie research has been rarely reported. In this study, tannin content in late fruit development periods was determined. Moreover, the MSAP (methylation-sensitive amplified polymorphism) method was used to analyze methylation profile, and 13 primer combinations were selected. In total, 5 differential fragments were cloned and sequenced. This research would be helpful for further gene cloning, research on deastringency mechanism, as well as molecular breeding of D. kaki.

文摘In this study, an (AG) n microsatellite-enriched library of Rhododendron simsii Planch. was constructed with FIASCO ( fast Isolation by AFLP sequences containing repeats) method. Among 356 positive colonies verified by colony PCR of the (AG)n microsatellite-enriched library, 233 colonies were selected for further sequencing. According to the results, 202 colonies containing microsatellites were obtained, accounting for 86.7%. Discarding sequences with miscella- neous peaks, overlapping peaks and weak signals, 39 sequences were screened finally for subsequent primer exploitation, including 22 perfect type SSR loci, 5 im- perfect type SSR loci and 12 compound type SSR loci. The FIASCO method is efficient and feasible for constructing microsatellite-enriched libraries of Rhododendron spp. , which laid a solid foundation for isolation of microsatellite loci and investigation of genetic diversity and genetic structure of Rhododendron spp.

At the school level,the guiding principles of student orientation and respecting teachers and teaching must be adhered to,and corresponding reforms in the school's system,policies and environment needs to be carried out. At the university teacher level,the concept of student orientation must be adhered to in teaching,and it is regarded as the guiding concept of teaching and education and is implemented in classroom teaching and practical teaching. At the level of college administrator,the concept of student orientation must be adhered in the course of daily management and service to do better management,service and education work. In order to better implement the student-oriented concept and promote the growth and development of college students,we needs to build a mentor team for college students' growth based on political workers,establish a team of academic tutors for college students based on professional teachers and build a platform for cooperation between schools and enterprises. The innovative training mode can promote students to grow into talents.

文摘DNA methylation is a kind of important epigenetic phenomenon,which could regulate gene expression in cells by affecting the transcription and duplication of DNA without changing cell DNA base sequence. Azalea is one of the ten famous flowers in China,as well a famous flower in the world,and the whole plant has medicinal value. This study investigated the change profile of gene methylation during the four flowering stages of Rhododendron pulchrum by methylation sensitive amplification polymorphism( MSAP),and 412 differential bands were obtained after selective amplification with screened 18 pairs of effective primers. Among the differential bands,23 differential bands were selected and subjected to recovery,sequencing and data alignment,and it was found that chalcone synthase,cowpox mitochondria,birch liana mitochondria,HDR reductase in camellia,Vaccinium macrocarpon mitochondria,Cyperus rotundus mitochondria protein,Shigella flexneri plasmid and Scophthalmus maximus cD NA-AFLP fragment have a regulating effect on azalea epigenetic inheritance,and next,genes related to the flowering period of R. pulchrum would be further studied,thereby improving the economic value of R. pulchrum.
文摘The coronavirus disease 2019(COVID-19),caused by SARS-CoV-2,has spread around the world with high mortality.To diagnose promptly and accurately is the vital step to effectively control its pandemic.Dynamic characteristics of SARSCoV-2-specific antibodies which are important for diagnosis of infection have not been fully demonstrated.In this retrospective,single-center,observational study,we enrolled the initial 131 confirmed cases of COVID-19 at Jin-Yin-Tan Hospital who had at least one-time antibody tested during their hospitalization.The dynamic changes of IgM and IgG antibodies to SARS-CoV-2 nucleocapsid protein in 226 serum samples were detected by ELISA.The sensitivities of IgM and IgG ELISA detection were analyzed.Result showed that the sensitivity of the IgG ELISA detection(92.5%)was significantly higher than that of the IgM(70.8%)(P\0.001).The meantimes of seroconversion for IgM and IgG were6 days and 3 days,respectively.The IgM and IgG antibody levels peaked at around 18 days and 23 days,and then IgM fell to below the baseline level at about day 36,whereas IgG maintained at a relatively high level.In conclusion,antibodies should be detected to aid in diagnosis of COVID-19 infection.IgG could be a sensitive indicator for retrospective diagnosis and contact tracing,while IgM could be an indicator of early infection.
