[Objectives]This study was conducted to explore the quality control method and establish the quality standard of Zhuang medicine Buddlejae Flos.[Methods]The microscopic identification method was adopted to identify the characters and microscopic characteristics of dried Buddlejae Flos in combination with traditional experience.The moisture,ash,acid-insoluble components and extracts of Buddlejae Flos were determined with reference to general rules of Chinese Pharmacopoeia(2015 edition).TLC identification was performed.Liquid chromatography-mass spectrometry(LC-MS)was used to determine the contents of linarin and verbascoside in Buddlejae Flos,and gas chromatography-mass spectrometry(GC-MS)was adopted to detect pesticide residues in Buddlejae Flos.Heavy metal elements As,Cd and Pb were detected by inductively coupled plasma mass spectrometry(ICP-MS).[Results]The established method is simple and accurate.Clear spots were observed on the thin layer chromatograms,and the resolution was good.The average value of moisture content was 12.24%;the average value of ash content was 5.1%;the average value of acid-insoluble content was 7.5%;and the average value of extract content was 27.3%.The regression equation of rutin in Buddlejae Flos was y=11.896x-0.0049,R^(2)=0.9996,and the contents of linarin and verbascoside were 5680 and 2080 mg/kg,respectively.No pesticide residues and heavy metals were detected in the medicinal materials of Buddlejae Flos.[Conclusions]This study can provide reference for the quality standard control of Guangxi Zhuang medicine Buddlejae Flos. 展开更多

Effects of Radix Puerariae, Radix Rehmanniae and Their Compatibility on Blood Glucose and Blood Lipids in Mice

[Objectives]To explore the effects of the compatibility of Radix Puerariae and Radix Rehmanniae on blood glucose and blood lipids in diabetic mouses.[Methods]Diabetic mouse model was established.The body weight and fasting blood glucose of mice were measured after 7 and 14 d of administration,and the biochemical indicators of blood lipids(TC,HDL-C,and LDL-C)were detected after 14 d of administration.[Results]Compared with the Radix Puerariae group and Radix Rehmanniae group,the compatibility group(1:2)had the best hypoglycemic effect(P<0.05),and TC and LDL-C in the compatibility group(2:1)significantly decreased(P<0.05),while HDL-C in the compatibility group(1:1)significantly increased(P<0.05).[Conclusions]Radix Puerariae,Radix Rehmanniae and their combination can reduce the blood glucose of diabetic mice.The compatibility group(1:2)had a significant hypoglycemic effect(P<0.05),and LDL-C in the compatibility group(2:1)significantly declined,while HDL-C in the compatibility group(1:1)rose significantly. 展开更多

[Objectives]The anti-tumor activity of fractions from Buddleja officinalis Maxim.by petroleum ether,ethyl acetate,n-butanol and water solvent was studied.[Methods]The ethanol extract from B.officinalis Maxim.was extracted and then concentrated with petroleum ether,ethyl acetate,n-butanol and water,respectively,and the extracts were obtained.The inhibitory effects of the four different fractions on the growth of three tumor cell lines in vitro were detected by CCK-8 method,and the median inhibitory concentration(IC 50 value)was calculated.[Results]The four fractions inhibited the growth of the three tumor cell lines in vitro,among which the n-butanol fraction had the best anti-tumor activity.The IC 50 values of the n-butanol fraction on human gastric cancer(SGC-7901),human breast cancer(MCF-7)and human liver cancer(BEL-7404)cell lines were 0.08,1.58 and 0.12 mg/mL,respectively.[Conclusions]Petroleum ether,ethyl acetate,n-butanol and water fractions from the ethanol extract of B.officinalis Maxim.had certain anti-tumor effects,and the n-butanol fraction had the best anti-tumor activity. 展开更多

Protective Effect of Ethanol Extract from Sweet Potato Leaves on CCL_(4)-Induced Liver Injury in Mice

[Objectives]To investigate the protective effect of ethanol extract from sweet potato leaves on liver injury induced by CCl_(4)in mice.[Methods]25 ICR mice were randomly divided into blank group,model group,high-dose extract group(200 mg/kg),low-dose extract group(100 mg/kg)and positive control group(2 mg/kg colchicine),with 5 mice in each group.All groups except the blank group were given intraperitoneal injection of 20%CCl 4 olive oil solution(2 mL/kg),and the blank group was given the same dose of olive oil solution three times a week.After 4 weeks,each administration group was given the corresponding dose of drugs(10 mL/kg),and the blank group and model group were given the corresponding amount of normal saline for 2 weeks.After the last intragastric administration,fasting was required,but water was allowed,blood was taken from eyeballs,and upper serum was taken by static centrifugation.Serum AST,ALT,CRP,IL-6 and SOD levels were detected by the kit.[Results]Compared with the blank group,the serum AST and ALT levels in the model group were significantly increased;compared with the model group,the ethanol extract of sweet potato leaves could decrease the levels of ALT,AST,CRP,IL-6 and increase the level of SOD in serum.[Conclusions]The ethanol extract of sweet potato leaves had protective effect on the mice with liver injury induced by CCl_(4),and its mechanism may be to protect the liver by lowering enzymes,inhibiting inflammation and antioxidant stress. 展开更多

[Objectives]To study the anti-fatigue effect of water extract from Centella asiatica(L.)Urban on mice.[Methods]After intragastric administration of low,medium and high concentration of water extract from Centella asiatica(L.)Urban for 14 d,the rotarod time and the content of serum lactic acid in mice were determined,respectively.[Results]Compared with the control group,the rotarod time of mice in the low and medium concentration groups was significantly prolonged(P<0.05),but there was no significant difference between the control group and the high concentration group;the content of serum lactic acid in the medium concentration group was significantly lower than that in the control group(P<0.01),but there was no significant difference between low concentration group and high concentration group and the control group.[Conclusions]A certain concentration of water extract from Centella asiatica(L.)Urban had a good anti-fatigue effect. 展开更多

[Objectives]To explore the anti-tumor activity of the extracts of petroleum ether,ethyl acetate,n-butanol and aqueous solution from Mahoniae caulis.[Methods]The extracts were extracted with petroleum ether,ethyl acetate,n-butanol and aqueous solution respectively,and then concentrated.The inhibitory effects of these extracts on the growth of three tumor cell lines in vitro were detected by CCK-8 method,and the IC 50 value was calculated.[Results]The four extracts inhibited the growth of the three tumor cell lines in vitro,among which the n-butanol extract had the best anti-tumor activity.The IC 50 values of the n-butanol extract on human gastric cancer(SGC-7901),human breast cancer(MCF-7)and human liver cancer(BEL-7404)cell lines were 0.23,0.25 and 0.58 mg/mL,respectively.[Conclusions]The ethanol extract of Mahoniae caulis under petroleum ether,ethyl acetate,n-butanol and aqueous solution had certain anti-tumor effect,and n-butanol extract had the best anti-tumor activity. 展开更多

[Objectives]The analgesic and hemostatic effects of Paris polyphylla,Scutellaria baicalensis,and their compatibility were studied.[Methods](i)In the experiment on the writhing of mice induced by acetic acid,60 Kunming SPF mice were randomly selected,and were randomly divided into 6 groups according to the male-to-female ratio of 1∶1,including blank control group,P.polyphylla group,S.baicalensis group,and three P.polyphylla and S.baicalensis compatibility groups(with the ratio of 1∶2,1∶1 and 1∶2).There were 10 mice in each group.In the experimental groups,the mice were given 1 g/mL Chinese medicine extract according to a dose of 20 mL/kg.In the control group,the mice were given 0.9%normal saline in equal volumes.Gavage was performed one time every 24 h and lasted for 14 d.1 h after the gavage on the 14th day,they were given 0.5%glacial acetic acid solution via intraperitoneal injection.(ii)In the hot plate experiment,60 Kunming SPF mice were selected,and the ratio of male to female,grouping,administration and gavage were the same as those of the glacial acetic acid-induced writhing experiment.Gavage was lasted for 14 d.1 h after the gavage on the 14th day,the mice were placed on a hot plate apparatus at(55±0.5)℃,and the time of licking hindfoot was measured.(iii)The hemostatic effect was explored through three experimental methods of tail hemostasis,femoral artery hemostasis and liver hemostasis.60 Kunming SPF mice were taken in each of the three experiments,and the male-female ratio and grouping were the same as above.The powder was covered on the surface of the wound sites,and the bleeding was observed.The hemostatic time was recorded,and the hemostatic time was recorded as 3 min if it exceeded 3 min.[Results](i)In the acetic acid-induced writhing experiment,compared with the blank control group,the pain of mice in the experimental groups was inhibited.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the inhibition rate of writhing was 20.43%,28.32%,52.30%,32.79%,and 39.02%,respectively.The analgesic effect of the compatibility 1∶2 group was the most obvious.(ii)In the hot plate experiment,compared with the blank control group,the experimental groups had analgesic effect.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the increase rate of pain threshold was 29.17%,47.83%,61.54%,and 50.61%,and 53.83%,respectively.The analgesic effect of the compatibility 2∶1 group was the most significant.(iii)In the hemostatic experiment,there was a significant difference in hemostatic time between the compatibility 1∶2 group and the blank control group,P.polyphylla group,and S.baicalensis group(P<0.05).The hemostatic effect of the compatibility 1∶2 group was the best.[Conclusions]P.polyphylla,S.baicalensis,and its compatibility had good analgesic and hemostatic effects,and the best compatibility ratio was 1∶2. 展开更多

[Methods] Phenol-sulfuric acid method was used to determine polysaccharide's content. Based on single-factor experiment and orthogonal test,the influences of solid-liquid ratio( A),ultrasonic extraction temperature( B),ultrasonic power( C) and ultrasonic time( D) on extraction rate of O. benghalensis polysaccharide were studied,and the optimal process of ultrasonic extraction was selected. [Results] The influence order of solid-liquid ratio( A),ultrasonic extraction temperature( B),ultrasonic power( C) and ultrasonic time( D) on extraction rate of O. benghalensis polysaccharide was A > D > B > C. Via the orthogonal test,it was determined that the optimal extraction condition of O. benghalensis polysaccharide was as below: solid-liquid ratio 1∶ 25( g/mL),extraction temperature 60℃,extraction power 80 W,extraction time 40 min. Under the condition,extraction rate of O. benghalensis polysaccharide was9. 80%. And the test was done about its scavenging effect on DPPH radicals and superoxide anions,and the antioxidant activity was evaluated. [Results] The optimum extraction process by cellulase method was as follows: p H value of 7; enzyme dosage of 4 ml; enzymolysis temperature of 40 ℃; extraction time of 2. 5 h. The average extraction rate of Herba Lycopi polysaccharide was 10. 70%,and RSD was 0. 17%. The antioxidant activity test showed that Herba Lycopi polysaccharide had a significant scavenging effect on radicals. [Conclusions] Herba Lycopi polysaccharides had strong antioxidant activity,and could provide a scientific basis for the development and utilization of Herba Lycopi medicinal resources. 展开更多

[Methods]Pigment of Myrica ruba leaves was extracted by ethanol extraction method. Absorbance was measured at wavelength of 300 nm. Effects of ethanol volume fraction,solid-to-liquid ratio,temperature,and extraction time were studied. L_9( 3~3) orthogonal experiment was carried out to optimize the extraction process,and DPPH scavenging ability was measured.[Results]The optimum extraction process of Myrica ruba leaf pigment was as follows: the ethanol volume fraction was 50%; the solid-to-liquid ratio was 1: 60( w/v),extraction temperature 70℃; extraction time was 60 min; after the concentration of purified Myrica ruba leaf pigment reached 0. 4455 mg/mL,the DPPH scavenging ability was basically the same as the same concentration Vc.[Conclusions] The experiment indicates that the optimized extraction process has a high yield rate,and Myrica ruba leaf pigment has high anti-oxidant activity. 展开更多

Objective:The study aims to explore the interventional effect of Abrus cantonensis and Armeniaca mume Sieb and their compatibility on blood glucose and blood lipid in diabetic mice and to explore the optimal ratio of efficacy of their compatibility.Methods:SPF Kunming male mice were fed with high sugar and high fat diet for 6 weeks and then intraperitoneally injected with STZ solution to establish a diabetes model.The successfully modeled mice were randomly divided into 6 groups according to their body weight:model group,Abrus Cantoniensis group,Armeniaca mume Sieb group,as well as Abrus Cantoniensis and Armeniaca mume Sieb group divided into 1:2,1:1 and 2:1 groups according to volume ratio with 10 mice in each group.Another 10 healthy mice were taken as blank group and intraperitoneally injected with sodium citrate buffer.The corresponding solution was given intragastric administration by body mass once a day for 20 days.After the last administration,fasting was performed for 12 hours.Serum was collected by the method of eyeball blood collection to determine the contents of fasting blood glucose,total cholesterol Triacylglycerol,triglyceride,low density lipoprotein and high-density lipoprotein.Results:Compared with Abrus Cantoniensis and Armeniaca mume Sieb groups,Glu,TG,TC and LDL-C of the three compatibility groups significantly decreased,with the most significant decrease in the compatibility group 1:2,while there was no significant difference in HDL-C among all groups.Conclusion:Abrus Cantoniensis and Armeniaca mume Sieb and their combination have synergistic effects on the improvement of blood glucose and lipid levels in mice with high glucose and high fat combined with STZ-induced diabetes mellitus model.Among all the experimental groups,the compatibility 1:2 group had the best effect. 展开更多

[Objectives]To study the anti-inflammatory and analgesic effects of Millettia speciosa and Tinpspora sinensis and their compatibility.[Methods](i)In the glacial acetic acid writhing experiment,60 SPF-grade Kunming mice were adopted,and the mice were randomly divided into 6 groups at male-female ratio of 1∶1,namely,the blank control group,M.speciosa group,T.sinensis group,M.speciosa compatible with T.sinensis group at the ratio of 1∶2(expressed as 1∶2 compatibility group),M.speciosa compatible with T.sinensis group at the ratio of 1∶1(expressed as 1∶1 compatibility group),and M.speciosa compatible with T.sinensis group at the ratio of 2∶1(expressed as 2∶1 compatibility group),12 mice for each group.Mice of the experimental groups were administered at a dose of 20 mL/kg,and the corresponding concentration of the Chinese medicine extract was given at 1 g/mL.The control group was administered with an equal volume of 0.9%physiological saline,and was intragastrically administered once every 24 h for 14 d.After intragastric administration for one hour on day 14,intraperitoneal injection of 0.5%glacial acetic acid solution was performed to induce pain.(ii)In the hot plate experiment,60 Kunming female SPF mice were adopted,grouped,intragastrically administered with the same glacial acetic acid writhing experiment for 14 d.After intragastric administration for one hour on day 14,the mice were placed on a hot plate apparatus at(55±0.5)℃.to measure the time of licking their hind feet.(iii)In the anti-inflammatory experiment,60 Kunming SPF mice were adopted,grouped,intragastrically administered with the same glacial acetic acid writhing experiment for 14 d.After intragastric administration for one hour on day 14,xylylene was administered to the left ears of mice at a dose of 50μL/piece to induce inflammation.[Results](i)In the glacial acetic acid writhing experiment,compared with the blank control group,the experimental group showed analgesic effects.Specifically,M.speciosa group,T.sinensis group,1∶2 compatibility group,1∶1 compatibility group,2∶1 compatibility group showed significant effect(P<0.05),the writhing inhibition rate was 17.65%,20.59%,29.41%,26.47%,and 44.12%,respectively,and 2∶1 compatibility group showed the most significant analgesic effects.(ii)In the hot plate experiment,compared with the control group,all experimental groups showed analgesic effect.Specifically,M.speciosa group,T.sinensis group,1∶2 compatibility group,1∶1 compatibility group,2∶1 compatibility group showed significant effect(P<0.05),the pain threshold improvement rates were 16.13%,14.55%,14.96%,29.95%,and 58.68%,respectively,and 2∶1 compatibility group showed the most significant analgesic effect.(iii)In the anti-inflammatory experiment,the swelling degree of the 1∶2 compatibility group was significantly different from that of the blank control group,M.speciosa group,T.sinensis group(P<0.05).and 1∶2 compatibility group showed the most significant anti-inflammatory effect.[Conclusions]M.speciosa,T.sinensis,and their compatibility had anti-inflammatory and analgesic effects.The 2∶1 compatibility group had the best analgesic effects,and 1∶2 compatibility group had the best anti-inflammatory effects. 展开更多

[Objectives]To investigate the acute toxicity of petroleum ether extract,n-butanol extract,ethyl acetate extract and water extract to mice.[Methods]The medium lethal dose(LD50)in mice could not be determined,so the maximum dose method was used for experiment.Mice were not fed but allowed to drink water for 16 h before the experiment.There were 74 SPF Kunming Mice,half male and half female,and it was divided into extract group and blank control group.The toxicity and death were recorded after intragastric administration and regular close observation.If any mice died,the dead mice were dissected.The changes of heart,liver,spleen,lung,kidney,brain,stomach,small intestine and other organs were observed with the naked eye.At the end of the 14 d observation period,the surviving mice in each group were killed and dissected.The gross pathological changes of the main organs were observed with the naked eye according to the same method mentioned above.[Results]There were no obvious toxic symptoms and no death in the mice.[Conclusions]The maximum doses of petroleum ether extract,n-butanol extract,ethyl acetate extract and water extract in mice were 1.61,1.71,1.65 and 1.79 g/L,respectively.Thus it was found that the quarterly tests of petroleum ether extract,n-butanol extract,ethyl acetate extract and water extract suggested that the medicine was safe. 展开更多

[Objectives] To study the in vitro antioxidant activity of resveratrol in peanut shell.[Methods] The crude extracts and purified substances were obtained by ultrasonic extraction and extraction,respectively.The scavenging ability and reducing ability of DPPH free radicals,hydroxyl radicals and superoxide radicals were studied and compared with BHT and Vc.[Results]Regardless of crude extract or purified substances,resveratrol in peanut shells had reducing ability,but the crude extract had greater reducing ability than purified substances,Vc and BHT,and the purified substances had greater reducing ability than BHT.At the same concentration,the effects of purified substances on scavenging DPPH free radicals were better than BHT,and the scavenging rate of crude extracts for superoxide radicals was higher than BHT,purified substances,and Vc;the purified substances had certain scavenging effects on the DPPH free radicals,hydroxyl radicals and superoxide radicals;the scavenging effects of crude extract on superoxide radicals increased with the increase of the concentration,but the scavenging effects on DPPH free radicals and hydroxyl radicals dropped with the increase of the concentration.[Conclusions]Crude extracts and purified substances of resveratrol in peanut shell have antioxidant activity. 展开更多

When the extraction temperature was 70℃,the extraction time was 1. 5 h and the solid-liquid ratio was 1: 10,the ethanol percentage was 60%,the extraction effect was the best,extraction of flavonoids was 0. 107 5 mg/m L.[Conclusions] This study is expected to provide a theoretical basis for further development and utilization of C. lacryma-jobi L. 展开更多

[Results] The extraction rate of ethanol extraction method was 14. 76 mg/100 g; the extraction rate of ultrasonic extraction method was 56. 85 mg/100 g; the extraction rate of acid water extraction method was 56. 63 mg/100 g; the extraction rate of Soxhlet extraction method was 58. 19 mg/100 g.[Conclusions]Results indicate that Soxhlet extraction method can obtain the highest extraction rate. 展开更多

[Objectives]To explore the hypoglycemic effect of Radix Et Rhizoma Salviae Miltiorrhizae,Guixi green tea and their compatibility on mice,so as to provide more options for the adjuvant treatment of diabetes and improve the comprehensive utilization value of Guixi green tea.[Methods]After being fed adaptively for 7 d,60 SPF-grade male mice were intraperitoneally injected with alloxouracil(180 mg/kg)to induce hyperglycemia model,from which 35 mice with a successful model were selected.They were randomly divided into 5 groups(Danshen group,green tea group,compatible group,positive control group and blank control group),with 7 animals in each group.The mice in all groups were administered according to the dose of 200 mg/kg once a day for 15 d.After the last administration,their body weight was measured on the 15 th day after fasting for 12 h,and their fasting blood glucose was measured by tail clipping.[Results]After modeling,the mice in each group showed typical hyperglycemia symptoms,namely obvious polydipsia,polyphagia,polyuria and weight loss.Compared with the blank control group,Danshen group and green tea group had obvious hypoglycemic effect(P<0.05),and compatible group had significant hypoglycemic effect(P<0.01).Compared with the blank control group,the weight loss of diabetic mice could also be inhibited in green tea group and compatible group(P<0.05).[Conclusions]Danshen,Guixi green tea and their compatibility had the effect of lowering blood sugar,among which the compatible group had the most obvious effect. 展开更多
