目的:探讨非SMC凝缩蛋白I复合物亚基D2(non-structural maintenance of chromosomes condensin I complex subunit D2,NCAPD2)在乳腺癌中的表达及其临床病理学意义。方法:利用UALCAN(The University of ALabama at Birmingham Cancer da...目的:探讨非SMC凝缩蛋白I复合物亚基D2(non-structural maintenance of chromosomes condensin I complex subunit D2,NCAPD2)在乳腺癌中的表达及其临床病理学意义。方法:利用UALCAN(The University of ALabama at Birmingham Cancer data analysis Portal)和UCSC Xena(University of California Santa Cruz Xena)数据库分析癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库中乳腺癌患者的测序数据,得到NCAPD2 mRNA在乳腺癌组织和正常乳腺组织中的表达差异以及其表达水平与患者临床病理特征间的关系。采用免疫组织化学法检测NCAPD2蛋白在135例乳腺癌组织和36例癌旁正常组织中的表达水平,分析其表达水平与乳腺癌临床病理特征之间的关系。利用Kaplan-Meier Plotter和ROC Plotter数据库分析NCAPD2mRNA表达水平与乳腺癌患者生存期的关系以及乳腺癌患者对化疗的响应能力。利用LinkedOmics、Metascape和STRING(Search Tool for the Retrieval of Interaction Gene/Proteins)数据库分析NCAPD2在乳腺癌中可能参与的生物学功能。应用蛋白质印迹法检测乳腺癌细胞株中NCAPD2蛋白的表达水平。采用MTT法和平板克隆实验检测敲减NCAPD2对乳腺癌细胞增殖能力的影响,应用蛋白质印迹法和FCM法检测敲减NCAPD2对乳腺癌细胞周期的影响。结果:UALCAN和UCSC Xena数据库数据均显示NCAPD2 mRNA的表达水平在乳腺癌组织中上调,并与临床分期、临床分型和淋巴结转移有关(P均<0.05)。免疫组织化学染色结果显示NCAPD2蛋白在乳腺癌中的阳性表达率(76.30%)和强阳性率(51.85%)均高于癌旁正常组织,其高表达与乳腺癌淋巴结转移呈正相关(P=0.004)。生存分析结果显示,NCAPD2高表达患者的总生存期短于低表达的患者(P均<0.05)。NCAPD2高表达患者对化疗的响应能力低于低表达者(P均<0.05),NCAPD2对化疗药物(紫杉烷和蒽环类)的敏感性具有较好的预测能力。生物信息学分析结果显示,与NCAPD2相关的差异表达基因共有9733个(上调基因有4873个,下调基因有4860个),主要参与细胞周期、有丝分裂等生物学进程,且NCAPD2与非SMC凝缩蛋白I复合物亚基H(non-structural maintenance of chromosomes condensin I complex subunit H,NCAPH)和染色质结构维持蛋白4(structural maintenance of chromosomes 4,SMC4)等蛋白存在相互作用。乳腺癌细胞MCF-7和Hs578T中NCAPD2蛋白的表达水平较高,敲减NCAPD2表达可抑制这2种乳腺癌细胞的增殖。细胞周期实验结果显示,与对照组相比,敲减Hs578T细胞中NCAPD2的表达后,G1期细胞所占百分比明显提高,而G2期细胞所占百分比明显减少(P均<0.05),同时G0/G1期检查点标志物细胞周期蛋白激酶1(cyclin-dependent kinase 1,CDK1)和细胞周期蛋白激酶6(cyclin-dependent kinase 6,CDK6)的蛋白表达水平明显降低,细胞周期蛋白依赖性激酶抑制剂1A(cyclin-dependent kinase inhibitor 1A,p21)蛋白的表达水平明显上调(P均<0.05)。结论:NCAPD2在乳腺癌中高表达,与患者的不良预后呈正相关,敲减其表达可抑制乳腺癌细胞的增殖和迁移,因此NCAPD2可能是潜在的乳腺癌预后因子及治疗靶点。展开更多

The effects of annealing on the parameters in the BAC model for InGaNP are also discussed in this work. In addition, the origins for improving luminescence efficiency by annealing are explained.
