Matrix vesicles(MVs)have shown strong effects in diseases such as vascular ectopic calcification and pathological calcified osteoarthritis and in wound repair of the skeletal system due to their membranous vesicle cha...Matrix vesicles(MVs)have shown strong effects in diseases such as vascular ectopic calcification and pathological calcified osteoarthritis and in wound repair of the skeletal system due to their membranous vesicle characteristics and abundant calcium and phosphorus content.However,the role of MVs in the progression of osteoporosis is poorly understood.Here,we report that annexin A5,an important component of the matrix vesicle membrane,plays a vital role in bone matrix homeostasis in the deterioration of osteoporosis.We first identified annexin A5 from adherent MVs but not dissociative MVs of osteoblasts and found that it could be sharply decreased in the bone matrix during the occurrence of osteoporosis based on ovariectomized mice.We then confirmed its potential in mediating the mineralization of the precursor osteoblast lineage via its initial binding with collagen type I to achieve MV adhesion and the subsequent activation of cellular autophagy.Finally,we proved its protective role in resisting bone loss by applying it to osteoporotic mice.Taken together,these data revealed the importance of annexin A5,originating from adherent MVs of osteoblasts,in bone matrix remodeling of osteoporosis and provided a new strategy for the treatment and intervention of bone loss.展开更多
目的通过比较危重症患者膀胱温与直肠温、耳温的差异性及相关性,探讨膀胱温应用于危重症患者体温监测的可行性。方法2018年1月至2019年5月,便利抽样法选取广州市某三级甲等医院重症监护室(intensive care unit,ICU)收治的47例重症患者...目的通过比较危重症患者膀胱温与直肠温、耳温的差异性及相关性,探讨膀胱温应用于危重症患者体温监测的可行性。方法2018年1月至2019年5月,便利抽样法选取广州市某三级甲等医院重症监护室(intensive care unit,ICU)收治的47例重症患者为研究对象,分别测量其入科后24h内6个时间点(2:00、6:00、10:00、14:00、18:00、22:00)的膀胱温与直肠温、耳温。采用重复测量方差分析及组内相关系数,比较三种体温测量结果间的差异性及相关性。结果膀胱温与直肠温、耳温的差异均有统计学意义(均P<0.001),与测量时间无交互作用。直肠温(r=0.91,P<0.05)、耳温(r=0.70,P<0.05)与膀胱温呈正相关。结论膀胱温与直肠温、耳温相关性较好,是ICU重症患者监测体温的可靠方法之一;且膀胱温能够持续监测,有助于减轻护士工作量,降低交叉感染发生率,值得在ICU中推广。展开更多

Tomato leaf mold is a common disease in tomato production and severely impacts the growth,fruit quality and yield of tomato plants.Research on tomato leaf mold has a long history and has focused mainly on the differen...Tomato leaf mold is a common disease in tomato production and severely impacts the growth,fruit quality and yield of tomato plants.Research on tomato leaf mold has a long history and has focused mainly on the differentiation of pathogen races,the structure and function of pathogen Avr gene products,the cloning of Cf resistance genes and the analysis of disease resistance mechanisms.Interactions between Cf and Avr are in accordance with the"gene-for-gene"hypothesis and typical Cf/Avr interactions are part of effector-triggered immunity(ETI).However,an increasing number of studies have proven that pathogen-associated molecular pattern(PAMP)-triggered immunity(PTI)is involved in the disease resistance response system mediated by Cf genes.In addition,different Cf genes have both similar and different roles in the disease resistance response,indicating that the disease resistance mechanism of Cf genes is complex.In this paper,progress in tomato leaf mold research was reviewed,and the regulatory mechanism underlying the Cf-mediated disease resistance response was thoroughly explored.We hope this summary will lay a foundation for research on tomato leaf mold disease resistance mechanisms and is applicable to breeding for disease resistance.展开更多

文摘本研究旨在探讨S100钙结合蛋白A16 (S100 calcium binding protein A16, S100A16)在肝细胞脂质代谢中的作用及可能的生物学机制。用脂肪酸培养HepG2细胞(人肝癌细胞系)以建立脂肪酸培养模型,对照模型不加脂肪酸培养,每一模型分成3组细胞,分别用S100a16过表达质粒、shRNA质粒、Vector质粒转染。用试剂盒检测细胞甘油三酯浓度,用油红O染色观察脂滴聚集情况,用免疫沉淀和质谱分析寻找和S100A16相互作用的兴趣蛋白,并用免疫共沉淀验证,用Western blot和qRTPCR进行相关机制研究。结果显示,和对照模型相比,脂肪酸培养模型细胞内脂肪和甘油三酯浓度显著增加。S100a16过表达组细胞内脂肪积累显著高于Vector质粒转染组。热休克蛋白A5 (heat shock protein A5, HSPA5)与S100A16之间存在相互作用。S100a16过表达可上调内质网应激的HSPA5/肌醇依赖酶1α-X结合蛋白1 (inositol-requiring enzyme 1α-X binding protein 1,IRE1α-XBP1)通路相关蛋白(HSPA5、IRE1α和pIREα1)表达水平,并上调脂肪合成相关基因Srebp1c、Acc和Fas mRNA表达水平,而转染S100A16 shRNA质粒可使上述蛋白和mRNA水平低于Vector质粒转染组。以上结果提示,S100A16可能通过内质网应激HSPA5/IRE1α-XBP1通路促进HepG2细胞脂质合成。
