Integrated ribosome and proteome analyses reveal insights into sevoflurane-induced long-term social behavior and cognitive dysfunctions through ADNP inhibition in neonatal mice

A growing number of studies have demonstrated that repeated exposure to sevoflurane during development results in persistent social abnormalities and cognitive impairment.Davunetide,an active fragment of the activity-dependent neuroprotective protein(ADNP),has been implicated in social and cognitive protection.However,the potential of davunetide to attenuate social deficits following sevoflurane exposure and the underlying developmental mechanisms remain poorly understood.In this study,ribosome and proteome profiles were analyzed to investigate the molecular basis of sevoflurane-induced social deficits in neonatal mice.The neuropathological basis was also explored using Golgi staining,morphological analysis,western blotting,electrophysiological analysis,and behavioral analysis.Results indicated that ADNP was significantly down-regulated following developmental exposure to sevoflurane.In adulthood,anterior cingulate cortex(ACC)neurons exposed to sevoflurane exhibited a decrease in dendrite number,total dendrite length,and spine density.Furthermore,the expression levels of Homer,PSD95,synaptophysin,and vglut2 were significantly reduced in the sevoflurane group.Patch-clamp recordings indicated reductions in both the frequency and amplitude of miniature excitatory postsynaptic currents(mEPSCs).Notably,davunetide significantly ameliorated the synaptic defects,social behavior deficits,and cognitive impairments induced by sevoflurane.Mechanistic analysis revealed that loss of ADNP led to dysregulation of Ca^(2+)activity via the Wnt/β-catenin signaling,resulting in decreased expression of synaptic proteins.Suppression of Wnt signaling was restored in the davunetide-treated group.Thus,ADNP was identified as a promising therapeutic target for the prevention and treatment of neurodevelopmental toxicity caused by general anesthetics.This study provides important insights into the mechanisms underlying social and cognitive disturbances caused by sevoflurane exposure in neonatal mice and elucidates the regulatory pathways involved. 展开更多

冷拔态GH4738合金热变形本构方程及热加工图

目的研究紧固件用冷拔态GH4738合金棒材在不同工艺参数下的热变形行为,为紧固件热加工工艺参数优化提供理论指导。方法采用Gleeble-3500热模拟实验机对冷拔态GH4738合金棒材在变形温度1000~1080℃、应变速率1~10 s^(−1)条件下进行了热... 目的研究紧固件用冷拔态GH4738合金棒材在不同工艺参数下的热变形行为,为紧固件热加工工艺参数优化提供理论指导。方法采用Gleeble-3500热模拟实验机对冷拔态GH4738合金棒材在变形温度1000~1080℃、应变速率1~10 s^(−1)条件下进行了热压缩实验,变形量为50%。计算了该合金的材料常数和变形激活能Q,建立了基于峰值应力的冷拔态GH4738合金的本构方程,根据动态材料模型理论绘制了冷拔态GH4738合金的能量耗散图和失稳图,获得了合金在不同应变下的热加工图,并讨论了显微组织演变情况。结果冷拔态GH4738合金的流变应力随着变形温度的增加或应变速率的减小而降低。线性回归的相关系数证实了描述该材料热变形行为的本构方程的准确性。基于冷拔态GH4738合金的热加工图及显微组织验证结果可得,冷拔态GH4738合金的主要失稳区工艺参数区间为1000~1035℃/0.12~3s^(−1),1030~1072℃/0.25~10 s^(−1)和1075~1080℃/2.72~10 s^(−1)。热加工较佳工艺条件为1000~1028℃/0.02~0.14 s^(−1)和1040~1080℃/0.06~0.74 s^(−1)。结论通过对冷拔态GH4738合金热变形本构方程和热加工图进行研究,获得了冷拔态GH4738合金优化的热变形工艺参数,可用于指导冷拔态GH4738合金的紧固件热加工成形。 展开更多

目的提高航空发动机叶片抗外物损伤的性能。方法采用薄壁件激光冲击强化工艺,对某型发动机TC4钛合金叶片包含一阶弯曲振动节线区域的表面进行处理,随后在叶片前缘一阶弯曲振动节线位置设计不同应力集中系数的缺口。参考有限元仿真软件... 目的提高航空发动机叶片抗外物损伤的性能。方法采用薄壁件激光冲击强化工艺,对某型发动机TC4钛合金叶片包含一阶弯曲振动节线区域的表面进行处理,随后在叶片前缘一阶弯曲振动节线位置设计不同应力集中系数的缺口。参考有限元仿真软件分析结果和相关标准要求,预制应力集中系数Kt为3.2的缺口。通过力值校核和有限元仿真之间的多次迭代,明确应力测试位置与缺口危险点应力之间的关系。通过振动疲劳试验对激光冲击强化效果进行评价。通过扫描电子显微镜观察疲劳断口的形貌,采用残余应力仪对梯度残余应力进行测试,并提取相应位置的半峰全宽值,对激光冲击强化提升缺口叶片疲劳强度的原因进行分析。结果经激光冲击强化处理后的钛合金缺口叶片在107次循环下的疲劳强度提升了63.2%;残余压应力层深度可达1.5 mm,且表层位错密度提升了67.5%;经激光冲击强化处理后钛合金缺口叶片裂纹萌生于近表面。结论激光冲击强化引入的表层梯度残余压应力和位错增殖是缺口叶片疲劳强度提升的主要原因。 展开更多

目的钛合金关键承力接耳孔边疲劳断裂是影响飞机飞行安全的重难点问题,采用激光冲击强化技术对TC4钛合金小孔件进行强化,提高其疲劳寿命。方法开展TC4钛合金小孔件单点有无填充、多点搭接激光冲击强化有限元数值模拟研究,确定最优强化工... 目的钛合金关键承力接耳孔边疲劳断裂是影响飞机飞行安全的重难点问题,采用激光冲击强化技术对TC4钛合金小孔件进行强化,提高其疲劳寿命。方法开展TC4钛合金小孔件单点有无填充、多点搭接激光冲击强化有限元数值模拟研究,确定最优强化工艺,并设计带双孔疲劳试样,进行疲劳试验验证。结果直径3mm光斑单点激光冲击强化的有效范围仅为1.9mm。孔内有填充,最内圈光斑圆心距孔边0.75mm时,单光斑激光冲击强化孔边残余应力场分布均匀,且不会引入残余拉应力。双面依次强化会使先强化面残余压应力值略高于后强化面。46.5%径向搭接率下,孔边多点搭接激光冲击强化应力场均匀性优于36.5%和56.5%径向搭接率。强化后,试样的疲劳寿命得到提升,提升效果随最大加载力的减小而显著增大。断口分析表明,强化后,孔边裂纹源位置向深度方向移动,疲劳裂纹扩展区的疲劳条带间距明显减小。结论最优强化工艺为:周向搭接率56.5%,径向搭接率46.5%,最内圈光斑圆心距孔边0.75mm,孔填充双面同时强化。激光冲击强化在孔边表面引入600~800MPa的残余压应力,模拟件疲劳寿命提升了6.98%~60.96%。 展开更多

To further improve the performance of UKF(Unscented Kalman Filter) algorithm used in BDS/SINS(BeiDou Navigation Satellite System/Strap down Inertial Navigation System), an improved GM-UKF(Gaussian Mixture Unscented Kalman Filter) considering non-Gaussian distribution is discussed in this paper. This new algorithm using SVD(Singular Value Decomposition) is proposed to alternative covariance square root calculation in UKF sigma point production. And to end the rapidly increasing number of Gaussian distributions, PDF(Probability Density Function) re-approximation is conducted. In principle this efficiency algorithm proposed here can achieve higher computational speed compared with traditional GM-UKF. And simulation experiment result show that, compared with UKF and GM-UKF algorithm, new algorithm implemented in BDS/SINS tightly integrated navigation system is suitable for handling nonlinear/non-Gaussian integrated navigation position calculation, for its lower computational complexity with high accuracy. 展开更多

Results and Analyses of BDS Precise Orbit Determination with the Enhancement of Fengyun-3C 被引量:9

Global navigation satellite system occultation sounder (GNOS) Fengyun-3C was launched successfully on September 23, 2013, which carried GPS/BDS receiver for the first time. This provides the convenience to study the enhancement results of low earth orbiter satellite (LEO) to BDS precise orbit determination (POD). First the data characteristic and code observation noise of GNOS are analyzed. Then the enhancement experiments in the case of global and regional ground observation stations layout are processed with four POD schemes: BDS single system, GPS/BDS double system, BDS single system with GNOS observations, GPS/BDS double system with GNOS observations. The precision of BDS orbits and clocks are compared via overlapping arcs. Results show that in the case of global station layout the along directional precision of GEO satellite has the biggest improvement, with the improvement percentage 60%. Then the precision of cross direction and the along direction of remaining satellites shows the second biggest improvement. The orbit precision of BDS-only POD in part of observation arcs some satellite even suffers a slight decline. The root mean square (RMS) of overlapping clock difference of visible arcs in GPS/BDS POD experiments improves by 0.1 ns level. As to the experiments of regional station layout with 7 ground stations, the orbit and clock overlapping precision and orbit predicting precision are analyzed. Results show that the predicting precision of BDS GEO satellites in the along direction improves by 85%. The remaining also has a substantial improvement, with the average percentage 21.7%. RMS of overlapping clock difference of visible arcs improves by 0.5 ns level. 展开更多

There is increasing concern about the uncombined(UC)observation model in the field of global navigation satellite system(GNSS).Based on the global positioning system(GPS)and the third-generation BeiDou navigation satellite system(BDS-3),this study processed the UC precision orbit determination(POD)for single and dual systems.First,a UC observation model suitable for multi-GNSS POD was derived,and the ionospheric-free(IF)combination observation model was presented.Although the ambiguity parameters of UC and IF strategies were different after reparameterization,the difference could be removed when processing ambiguity resolution,and the equivalence was proved theoretically.To demonstrate the accuracy of BDS-3 orbits fully,the observation data of approximately 1 month were selected for determining the precise orbit for global positioning system(GPS)only,BDS-3 only,and GPS/BDS-3 systems based on the UC and IF models.The orbit precision of BDS-3 satellites was validated by using metrics,including comparison with precision products released by Wuhan University,orbit boundary discontinuity,and satellite laser ranging(SLR)residuals.The results show that the orbit accuracies of the IF and UC models are almost the same,the difference in orbits is approximately several millimeters,and the clock difference is within 0.01 ns.The GPS/BDS-3 combined solution shows better accuracy compared to other solutions.The average accuracies in the R and 3D directions are approximately 4 and 15 cm,and the clock standard deviation is approximately 0.2 ns compared to external orbit product.The root mean square of SLR residuals is approximately 4 cm. 展开更多
