Objective The prevalence of carbapenem-resistant Klebsiella pneumoniae(CR-KP)is a global public health problem.It is mainly caused by the plasmid-carried carbapenemase gene.Outer membrane vesicles(OMVs)contain toxins ...Objective The prevalence of carbapenem-resistant Klebsiella pneumoniae(CR-KP)is a global public health problem.It is mainly caused by the plasmid-carried carbapenemase gene.Outer membrane vesicles(OMVs)contain toxins and other factors involved in various biological processes,includingβ-lactamase and antibiotic-resistance genes.This study aimed to reveal the transmission mechanism of OMV-mediated drug resistance of Klebsiella(K.)pneumoniae.Methods We selected CR-KP producing K.pneumoniae carbapenemase-2(KPC-2)to study whether they can transfer resistance genes through OMVs.The OMVs of CR-KP were obtained by ultracentrifugation,and incubated with carbapenem-sensitive K.pneumoniae for 4 h.Finally,the carbapenem-sensitive K.pneumoniae was tested for the presence of bla_(KPC-2)resistance gene and its sensitivity to carbapenem antibiotics.Results The existence of OMVs was observed by the electron microscopy.The extracted OMVs had bla_(KPC-2)resistance gene.After incubation with OMVs,bla_(KPC-2)resistance gene was detected in sensitive K.pneumoniae,and it became resistant to imipenem and meropenem.Conclusion This study demonstrated that OMVs isolated from KPC-2-producing CR-KP could deliver bla_(KPC-2)to sensitive K.pneumoniae,allowing the bacteria to produce carbapenemase,which may provide a novel target for innovative therapies in combination with conventional antibiotics for treating carbapenem-resistant Enterobacteriaceae.展开更多
To study the influence of thickness on the magnetic properties of ReBCO(Re = Y, Gd, Sm, Nd, etc.) bulk superconductors, a single domain gadolinium barium copper oxide(GdBCO) bulk superconductor fabricated by the Re + ...To study the influence of thickness on the magnetic properties of ReBCO(Re = Y, Gd, Sm, Nd, etc.) bulk superconductors, a single domain gadolinium barium copper oxide(GdBCO) bulk superconductor fabricated by the Re + 011 top seeded infiltration growth(Re + 011 TSIG) method was continuously sliced along the bottom to obtain samples of different thickness. The levitation force and attractive force of these samples were tested at 77 K in the zero-field-cooled(ZFC)state. It is found that as the sample thickness decreases, the levitation force decreases gradually whereas the attractive force increases. This is related to the varied ability to resist the penetration of magnetic field occasioned by varying sample thickness, which are deeply revealed by combining with the characteristics of the non-ideal type-II superconductor. Further,the levitation force exhibits a trend of slow initial change followed by rapid change, which may be attributed to the growth of the sample. Measurement of the trapped field shows that a similar distribution of trapped field at the top and bottom surfaces can be achieved by removing some materials from the bottom of the bulk. These results provide a reference for meeting the actual requirements of ReBCO bulks of different thicknesses and greatly contribute to practical designs and applications.展开更多
Lightweight,ultra-flexible,and robust crosslinked transition metal carbide(Ti3C2 MXene)coated polyimide(PI)(C-MXene@PI)porous composites are manufactured via a scalable dip-coating followed by chemical crosslinking ap...Lightweight,ultra-flexible,and robust crosslinked transition metal carbide(Ti3C2 MXene)coated polyimide(PI)(C-MXene@PI)porous composites are manufactured via a scalable dip-coating followed by chemical crosslinking approach.In addition to the hydrophobicity,anti-oxidation and extreme-temperature stability,efficient utilization of the intrinsic conductivity of MXene,the interfacial polarization between MXene and PI,and the micrometer-sized pores of the composite foams are achieved.Consequently,the composites show a satisfactory X-band electromagnetic interference(EMI)shielding effectiveness of 22.5 to 62.5 dB at a density of 28.7 to 48.7 mg cm−3,leading to an excellent surface-specific SE of 21,317 dB cm^(2)g^(−1).Moreover,the composite foams exhibit excellent electrothermal performance as flexible heaters in terms of a prominent,rapid reproducible,and stable electrothermal effect at low voltages and superior heat performance and more uniform heat distribution compared with the commercial heaters composed of alloy plates.Furthermore,the composite foams are well attached on a human body to check their electromechanical sensing performance,demonstrating the sensitive and reliable detection of human motions as wearable sensors.The excellent EMI shielding performance and multifunctionalities,along with the facile and easy-to-scalable manufacturing techniques,imply promising perspectives of the porous C-MXene@PI composites in next-generation flexible electronics,aerospace,and smart devices.展开更多
The formation of nerve bundles,which is partially regulated by neural cell adhesion molecule 1(NCAM1),is important for neural network organization during peripheral nerve regeneration.However,little is known about how...The formation of nerve bundles,which is partially regulated by neural cell adhesion molecule 1(NCAM1),is important for neural network organization during peripheral nerve regeneration.However,little is known about how the extracellular matrix(ECM)microenvironment affects this process.Here,we seeded dorsal root ganglion tissue blocks on different ECM substrates of peripheral nerve ECM-derived matrixgel,Matrigel,laminin 521,collagen I,and collagen IV,and observed well-aligned axon bundles growing in the peripheral nerve ECM-derived environment.We confirmed that NCAM1 is necessary but not sufficient to trigger this phenomenon.A protein interaction assay identified collagen VI as an extracellular partner of NCAM1 in the regulation of axonal fasciculation.Collagen VI interacted with NCAM1 by directly binding to the FNIII domain,thereby increasing the stability of NCAM1 at the axolemma.Our in vivo experiments on a rat sciatic nerve defect model also demonstrated orderly nerve bundle regeneration with improved projection accuracy and functional recovery after treatment with 10 mg/m L Matrigel and 20μg/m L collagen VI.These findings suggest that the collagen VI-NCAM1 pathway plays a regulatory role in nerve bundle formation.This study was approved by the Animal Ethics Committee of Guangzhou Medical University(approval No.GY2019048)on April 30,2019.展开更多
Objective The influential factors of chemotherapy-induced myelosuppression in esophageal cancer in central China are unclear.This study aimed to investigate the effect of commonly used chemotherapy regimens on the inc...Objective The influential factors of chemotherapy-induced myelosuppression in esophageal cancer in central China are unclear.This study aimed to investigate the effect of commonly used chemotherapy regimens on the incidence of myelosuppression in clinical treatment of esophageal cancer.Methods In this retrospective study,624 patients with esophageal cancer who received six different chemotherapy regimens between 2013 and 2020 at our institute were included.Chemotherapy consisted of lobaplatin,5-fluorouracil(5-F),lobaplatin and 5-F,nedaplatin,nedaplatin and paclitaxel(PTX),cisplatin and PTX.Multivariable logistic regression analysis was used to explore the risk of myelosuppression among the six different chemotherapy regimens.Results Compared with lobaplatin group,the incidence of myelosuppression in patients treated with chemotherapy regimens of lobaplatin and 5-F,nedaplatin,nedaplatin and PTX and cisplatin and PTX were significantly ameliorated.The dose of lobaplatin was significantly reduced(P=0.007)when lobaplatin was combined with 5-F,and the combination could significantly reduce the risk of myelosuppression(P=0.022).Furthermore,chemotherapeutic regimens,the dose of platinum,hemoglobin and uric acid levels,age,sex,total bilirubin and immune-enhancing drugs were found to be strong predictors of developing myelosuppression.Conclusion Targeted preventive interventions that enhance immune function,reduce uric acid levels and choose combined medication during chemotherapy should be implemented for high-risk patients to reduce the occurrence of myelosuppression.In addition,the dose of lobaplatin should be adjusted when combined with other chemotherapy drugs to reduce the incidence of myelosuppression.展开更多
During the pipeline plugging process,both the pipeline and the pipe isolation tool(PIT)will be greatly damaged,due to the violent vibration of the flow field.In this study,it was proposed for the first time to reduce ...During the pipeline plugging process,both the pipeline and the pipe isolation tool(PIT)will be greatly damaged,due to the violent vibration of the flow field.In this study,it was proposed for the first time to reduce the vibration of the flow field during the plugging process by optimizing the surface structure of the PIT.Firstly,the central composite design(CCD)was used to obtain the optimization schemes,and the drag coefficient and pressure coefficient were proposed to evaluate the degree of flow field changes.Secondly,a series of computational fluid dynamics(CFD)simulations were performed to obtain the drag coefficient and pressure coefficient during dynamic plugging.And the mathematical model of drag coefficient and pressure coefficient with the surface structure of the PIT were established respectively.Then,a modified particle swarm optimization(PSO)was applied to predict the optimal value of the surface structure of the PIT.Finally,an experimental rig was built to verify the effectiveness of the optimization.The results showed that the improved method could reduce the flow field vibration by 49.56%.This study provides a reference for the design of the PIT surface structure for flow field vibration technology.展开更多
All-solid-state batteries(ASSBs) hold great promise for next-generation energy storage technologies owing to their advantage in different aspects such as energy density,safety,and wide temperature tolerance.However,th...All-solid-state batteries(ASSBs) hold great promise for next-generation energy storage technologies owing to their advantage in different aspects such as energy density,safety,and wide temperature tolerance.However,the use of solid-state electrolytes(SSEs) instead of liquid ones meanwhile brings serious concerns related to the point-to-point contact between SSEs and electrodes,which is known to result in high interface resistance and inhomogeneous distribution of charges during the Li^(+)plating/stripping process,eventually leading to a premature failure of ASSBs.This review focuses on the garnet-type SSEs in the formula of Li_(7)La_(3)Zr_(2)O_(12)(LLZO),and discusses the structure-performance relationship of this ceramic electrolyte in detail to achieve a clear understanding of its Li^(+)transmission mechanism.Meanwhile,the challenges of cubic phase LLZO(c-LLZO) for their application in solidstate batteries(SSBs) are demonstrated by the Li/LLZO interface,which features the importance of Li metal wettability and dendrite suppression for sustainable performance.Furthermore,this review summarizes the recent research strategies to combat these contact issues at the Li/LLZO interface,highlighting the essential role played by surface modification of LLZO electrolytes.Following the obtained insights,perspectives for future research on LLZO to accelerate its potential development of SSBs in commercialized applications are also provided.展开更多

BACKGROUND Globally,the World Health Organization ranks major depressive disorder(MDD)as the leading cause of disability.However,MDD molecular etiology is still poorly understood.AIM To explore the possible associatio...BACKGROUND Globally,the World Health Organization ranks major depressive disorder(MDD)as the leading cause of disability.However,MDD molecular etiology is still poorly understood.AIM To explore the possible association between mitochondrial ND6 T14502C mutation and MDD.METHODS Clinical data were collected from two pedigrees,and detailed mitochondrial genomes were obtained for the two proband members.The assessment of the resulting variants included an evaluation of their evolutionary conservation,allelic frequencies,as well as their structural and functional consequences.Detailed mitochondrial whole genome analysis,phylogenetic,and haplotype analysis were performed on the probands.RESULTS Herein,we reported the clinical,genetic,and molecular profiling of two Chinese families afflicted with MDD.These Chinese families exhibited not only a range of onset and severity ages in their depression but also extremely low penetrances to MDD.Sequence analyses of mitochondrial genomes from these pedigrees have resulted in the identification of a homoplasmic T14502C(I58V)mutation.The polymorphism is located at a highly conserved isoleucine at position 58 of ND6 and distinct mitochondrial DNA(mtDNA)polymorphisms originating from haplogroups M10 and H2.CONCLUSION Identifying the T14502C mutation in two individuals with no genetic relation who exhibit symptoms of depression provides compelling evidence that this mutation may be implicated in MDD development.Nonetheless,the two Chinese pedigrees that carried the T14502C mutation did not exhibit any functionally significant mutations in their mtDNA.Therefore,the phenotypic expression of the T14502C mutation related to MDD may be influenced by the nuclear modifier gene(s)or environmental factors.展开更多

Fifteen batches were identified to have been processed with sulfur fumigation, while others were not.Conclusions: The HPLC method was found to be most appropriate for the determination of sulfur fumigation for moutan cortex.展开更多

文摘All-solid-state batteries(ASSBs) hold great promise for next-generation energy storage technologies owing to their advantage in different aspects such as energy density,safety,and wide temperature tolerance.However,the use of solid-state electrolytes(SSEs) instead of liquid ones meanwhile brings serious concerns related to the point-to-point contact between SSEs and electrodes,which is known to result in high interface resistance and inhomogeneous distribution of charges during the Li^(+)plating/stripping process,eventually leading to a premature failure of ASSBs.This review focuses on the garnet-type SSEs in the formula of Li_(7)La_(3)Zr_(2)O_(12)(LLZO),and discusses the structure-performance relationship of this ceramic electrolyte in detail to achieve a clear understanding of its Li^(+)transmission mechanism.Meanwhile,the challenges of cubic phase LLZO(c-LLZO) for their application in solidstate batteries(SSBs) are demonstrated by the Li/LLZO interface,which features the importance of Li metal wettability and dendrite suppression for sustainable performance.Furthermore,this review summarizes the recent research strategies to combat these contact issues at the Li/LLZO interface,highlighting the essential role played by surface modification of LLZO electrolytes.Following the obtained insights,perspectives for future research on LLZO to accelerate its potential development of SSBs in commercialized applications are also provided.

文摘Redox-active organic materials are emerging as the new playground for the design of new exciting battery materials for rechargeable batteries because of the merits including structural diversity and tunable electrochemical properties that are not easily accessible for the inorganic counterparts.More importantly,the sustainability developed by using naturally abundant chemical elements(C,H,N,O and S) makes them as an ideal alternative material for Li-ion batteries(LIBs).However,the identification and screening of proper organic materials is still challenging in the past decades.Assisted by the artificial intelligence,this review will give a summary of the theoretical design aspects of redox-active organic materials from densityfunctional theory(DFT) to machine learning(ML) methods in the past two decades,with a particular emphasis on the calculation method to predict the chemical/electrochemical stability and reversibility.This review will also analyze and discuss the challenges and perspectives for the development of organic battery materials.

文摘BACKGROUND Globally,the World Health Organization ranks major depressive disorder(MDD)as the leading cause of disability.However,MDD molecular etiology is still poorly understood.AIM To explore the possible association between mitochondrial ND6 T14502C mutation and MDD.METHODS Clinical data were collected from two pedigrees,and detailed mitochondrial genomes were obtained for the two proband members.The assessment of the resulting variants included an evaluation of their evolutionary conservation,allelic frequencies,as well as their structural and functional consequences.Detailed mitochondrial whole genome analysis,phylogenetic,and haplotype analysis were performed on the probands.RESULTS Herein,we reported the clinical,genetic,and molecular profiling of two Chinese families afflicted with MDD.These Chinese families exhibited not only a range of onset and severity ages in their depression but also extremely low penetrances to MDD.Sequence analyses of mitochondrial genomes from these pedigrees have resulted in the identification of a homoplasmic T14502C(I58V)mutation.The polymorphism is located at a highly conserved isoleucine at position 58 of ND6 and distinct mitochondrial DNA(mtDNA)polymorphisms originating from haplogroups M10 and H2.CONCLUSION Identifying the T14502C mutation in two individuals with no genetic relation who exhibit symptoms of depression provides compelling evidence that this mutation may be implicated in MDD development.Nonetheless,the two Chinese pedigrees that carried the T14502C mutation did not exhibit any functionally significant mutations in their mtDNA.Therefore,the phenotypic expression of the T14502C mutation related to MDD may be influenced by the nuclear modifier gene(s)or environmental factors.

文摘Objective: Establish a quick, precise and specific method to determine whether the moutan cortex obtained from market is processed with sulfur; provide a reliable method for the scientific evaluation.Methods: Three methods, including acid-base titration method, high-performance liquid-chromatography(HPLC) method, and ultraperformance liquid chromatography coupled with time-of-flight mass spectrometry(UHPLC-TOF-MS) method were used to detect whether30 batches of samples were fumigated with sulfur.Results: The results of three methods were substantially the same. Fifteen batches were identified to have been processed with sulfur fumigation, while others were not.Conclusions: The HPLC method was found to be most appropriate for the determination of sulfur fumigation for moutan cortex.
文摘Objective To review and explore the advantages and disadvantages of the teaching of the reproductive system in Shanghai Jiao Tong University School of Medicine,in order to provide a feasible scheme for the subsequent reform of the teaching system,including the syllabus design and the optimization of the teaching method,etc.Methods With the one-year practice in the teaching of the reproductive system,this paper has explored the achievements in the implementation of this teaching system from three aspects,including the teaching experience and feelings of teachers,student questionnaires and test scores.Meanwhile,this paper has also deeply analyzed some problems that may exist or be derived from the teaching practice in that teaching model.Results The new teaching method has huge differences from the traditional"discipline-oriented"teaching method in the curricular design,teachers’lecture form,students’learning skills and examination types,etc.What is more important,the new teaching method will significantly renew the concepts of both teaching and learning.Conclusion The method of the systemic teaching is one of the critical ways to realize the reform of the medical education,and can also provide the necessary basis for the cultivation of professional high-quality teachers required in the medical education.
