为研究香椿籽多糖(Polysaccharides of seeds of Toona sinensis,STSP)不同组分的体外抗凝血活性,以水提醇沉法得到的香椿籽粗多糖为实验材料,通过聚酰胺法脱蛋白、活性炭脱色,经离子交换柱DEAE Sepharose CL-6进行纯化。采用高效凝胶...为研究香椿籽多糖(Polysaccharides of seeds of Toona sinensis,STSP)不同组分的体外抗凝血活性,以水提醇沉法得到的香椿籽粗多糖为实验材料,通过聚酰胺法脱蛋白、活性炭脱色,经离子交换柱DEAE Sepharose CL-6进行纯化。采用高效凝胶过滤色谱(HPGFC)和离子色谱分别对多糖的分子质量和其单糖组成进行分析,初步研究了纯化后的香椿籽多糖组分的体外抗凝血活性。结果表明:共得到4种纯化多糖组分即STSP-1、STSP-2、STSP-3、STSP-4;HPGFC法测定STSP-2、STSP-3皆为单峰,为均一多糖,其重均分子质量分别为13218 u和36080 u;单糖测定表明,香椿籽多糖主要由阿拉伯糖(Ara)、鼠李糖(Rha)、氨基葡萄糖(GluN)、半乳糖(Gal)、葡萄糖(Glu)、木糖(Xyl)、甘露糖(Man)、葡萄糖醛酸(GluA)、半乳糖醛酸(GalA)组成。体外抗凝血实验表明,STSP-3能显著延长凝血酶原时间(Prothrombin time,PT)和凝血酶时间(Thrombin time,TT),表明其抗凝血活性是通过影响外源性途径、共同途径实现。实验结果为香椿籽多糖的纯化及资源利用奠定了基础。展开更多
