The business features and the operation pattern ofassembly line based on mass customization are analyzed. And the research emphatically studiesvarious technologic factors to improve customer satisfaction and their corresponding implementmethods in operating assembly line. In addition, the models are proposed for operating assembly lineunder dynamic process environment in mass customization. A genetic approach is developed to providethe optimal solution to the models. The products were characterized by G...A two-step process was used to synthesize the high molecular weight poly (lactic acid) using thionyl chloride as an activator for the polycondensation of prepolymer of lactic acid. The products were characterized by GPC, DSC, and 1H-NMR. Poly (L-lactic acid) (PLLA) with molecular weight Mw: 32875 was obtained when thionyl chloride was used, while low molecular weight PLLA Mw: 7350 was yielded without thionyl chloride. Effects of the concentration of thionyl chloride, polymerization time and bases on the molecular weight of poly (lactic acid) were investigated. In DSC scans the glass transition temperatures (Tg) of the resulting polymers varied from 25.3 to 57.6℃, and the Tg of poly(L-lactic acid) was higher than that of poly (D,L-lactic acid) (PDLLA). The melting points of poly (L-lactic acid) and poly (D, L-lactic acid) could not be found unless the polymers were obtained by thionyl chloride activator.展开更多

E1A have the ability to integrate into the host genome, resulting in long-time ex...Adenovirus 5 type E1A as a tumor suppressor gene can inhibit tumor growth and enhance the sensitivity of chemotherapy and radiotherapy. E1A have the ability to integrate into the host genome, resulting in long-time expres-sion that induces Rb gene inactivation and animal cells im-mortalization. This prompted us to select the E1A protein for treatment of cancer in order to overcome the limitations of E1A gene therapy. Thus, we firstly constructed E1A eu-caryotic expression vector (pPIC9/E1A), transformated the pichia pastoris yeast cells (GS115) and screened the high-expressing recombinant strains. The positive yeast strains were cultured in the shake flask, and induced for 3 d. The crude E1A protein was purified using two steps of col-umn chromatography on HiTrap Q and HiTrap SP. The pu-rified E1A protein was identified by SDS-PAGE and Western blot. E1A protein was mostly located at cellular nuclear when Chariot delivered E1A protein into cells. The analysis in vitro indicated that the E1A protein arrested LN686 cell cycle at G2/M phase, and significantly inhibited the growth of LN686 tumor cells. The current studies firstly provided an experimental basis to further develop E1A protein for tumor treatment.展开更多
