目的探讨微信群平台联合Seminar教学法在ERCP初学者后续培训中的作用效果。方法选择ERCP初学者微信群,共62人,应用微信群平台联合Seminar教学,观察1年,采用理论考核及问卷调查的方式对教学效果进行主客观评价。结果通过1年教学,教学结... 目的探讨微信群平台联合Seminar教学法在ERCP初学者后续培训中的作用效果。方法选择ERCP初学者微信群,共62人,应用微信群平台联合Seminar教学,观察1年,采用理论考核及问卷调查的方式对教学效果进行主客观评价。结果通过1年教学,教学结束时学员的理论考试成绩(84.96±3.63)分高于教学开始时(74.81±4.94)分,P<0.05)。学员主观感受,包括提高学习效率,临床应变能力,ERCP技能提高,自学及科研能力,团队沟通合作等方面均有明显提高。结论微信群平台联合Seminar教学法能提高ERCP初学者的客观理论成绩,改善主观感受,效果良好。 展开更多

In this article,the FCC diesel fuel is chemically treated with GZ reagent.The existent gum,insolubles after oxidation,and iodine munber are greatly reduced after chermical treatment.This method has a good effect and is simple.This method is characterized by high recovery rate of fuel(>99.7?and low cost.The property of the mixture of chemically treated diesel fuel with straight-run diesel fuel can meet the requirements of national standard GB252-2000.The color and oxidation stability of the mixed fuel after having been stored for three months were still lower than the values specified in the national standard. 展开更多

The business features and the operation pattern ofassembly line based on mass customization are analyzed. And the research emphatically studiesvarious technologic factors to improve customer satisfaction and their corresponding implementmethods in operating assembly line. In addition, the models are proposed for operating assembly lineunder dynamic process environment in mass customization. A genetic approach is developed to providethe optimal solution to the models. A two-step process was used to synthesize the high molecular weight poly (lactic acid) using thionyl chloride as an activator for the polycondensation of prepolymer of lactic acid. The products were characterized by GPC, DSC, and 1H-NMR. Poly (L-lactic acid) (PLLA) with molecular weight Mw: 32875 was obtained when thionyl chloride was used, while low molecular weight PLLA Mw: 7350 was yielded without thionyl chloride. Effects of the concentration of thionyl chloride, polymerization time and bases on the molecular weight of poly (lactic acid) were investigated. In DSC scans the glass transition temperatures (Tg) of the resulting polymers varied from 25.3 to 57.6℃, and the Tg of poly(L-lactic acid) was higher than that of poly (D,L-lactic acid) (PDLLA). The melting points of poly (L-lactic acid) and poly (D, L-lactic acid) could not be found unless the polymers were obtained by thionyl chloride activator. 展开更多

‘月月粉’月季PP2C家族基因鉴定及非生物胁迫响应分析

利用生物信息方法从月季‘月月粉’(Rosa chinensis‘Old Blush’)基因组中共鉴定出69个PP2C家族成员,其编码氨基酸的长度在122~1082 aa之间,等电点介于4.03~9.34,大部分蛋白呈亲水性。系统进化树分析将其分为12个亚族,同一亚族内基因... 利用生物信息方法从月季‘月月粉’(Rosa chinensis‘Old Blush’)基因组中共鉴定出69个PP2C家族成员,其编码氨基酸的长度在122~1082 aa之间,等电点介于4.03~9.34,大部分蛋白呈亲水性。系统进化树分析将其分为12个亚族,同一亚族内基因结构和保守基序具有较高的相似性。基因片段复制事件是PP2C家族基因扩张的主要原因。启动子顺式作用元件分析表明约85%的月季PP2C含有逆境应答元件、激素应答元件等,推测其在响应逆境胁迫的过程中发挥重要作用。月季PP2C基因的表达具有组织和发育时期特异性,且部分基因参与干旱和高温胁迫响应过程,其中3个A亚族基因在干旱胁迫中响应明显,1个A亚族基因和2个G亚族基因在高温胁迫中响应明显。通过qRT-PCR分析11个不同亚族PP2C基因在月季失水胁迫和ABA处理下的表达模式,发现6个A亚族基因(RC0G0099200、RC1G0458700、RC1G0569200、RC2G0066800、RC2G0089100和RC5G0571100)和1个G亚族基因(RC3G0083700)的表达量显著升高,表明这些基因可能受ABA诱导并参与植物失水胁迫响应。 展开更多

Adenovirus 5 type E1A as a tumor suppressor gene can inhibit tumor growth and enhance the sensitivity of chemotherapy and radiotherapy. E1A have the ability to integrate into the host genome, resulting in long-time expres-sion that induces Rb gene inactivation and animal cells im-mortalization. This prompted us to select the E1A protein for treatment of cancer in order to overcome the limitations of E1A gene therapy. Thus, we firstly constructed E1A eu-caryotic expression vector (pPIC9/E1A), transformated the pichia pastoris yeast cells (GS115) and screened the high-expressing recombinant strains. The positive yeast strains were cultured in the shake flask, and induced for 3 d. The crude E1A protein was purified using two steps of col-umn chromatography on HiTrap Q and HiTrap SP. The pu-rified E1A protein was identified by SDS-PAGE and Western blot. E1A protein was mostly located at cellular nuclear when Chariot delivered E1A protein into cells. The analysis in vitro indicated that the E1A protein arrested LN686 cell cycle at G2/M phase, and significantly inhibited the growth of LN686 tumor cells. The current studies firstly provided an experimental basis to further develop E1A protein for tumor treatment. 展开更多
