Epidermal growth factor (EGF) induced intracellular free calcium ([Ca2+]i) response was studied in fura-2- or fluo-3-loaded human hepatoma cells of BEL-7404 cell line. Single cell [Ca2+]i analysis and [Ca2+], measurement in cell populations revealed that EGF triggered a rapid [Ca2+]iincrease in the dose-dependent and time- dependent manner. Pretreatment of cells with an endoplasmic reticulum (ER) Ca2+-ATPase inhibitor, thapsigargin (TG) at 100 nM concentration for 20 min, completely abolished EGF-induced [Ca2+]i increase, and chelating extracellular calcium by excess EGTA partially inhibited the increase.Furthermore, the expression of antisense EGF receptor sequence in BEL-7404 cells suppressed the [Ca2+]i response to EGF. The results suggest that EGF receptor-mediated [Ca2+]i increase in the human hepatoma cells is essentially dependent on the Ca2+ storage in ER. 展开更多

Objective: To test the expression of HER4 in non-small cell lung cancer (NSCLC) and elucidate the relationship between its over-expression and the clinical pathology of NSCLC. Methods: 70 cases of paraffin-embedded tissues from informative NSCLC were tested for the expression of HER4 by means of immunohistochemical assay. Results: HER4 were overexpressed in NSCLC in 91.4%. The overexpression of HER4 correlated only with the lymph node metastasis, TNM staging and survival after operation. Conclusion: ErbB4 is one of the genes to regulate the growth of NSCLC in advanced stages and artificial interference of the overexpression of HER4 in NSCLC might be a good way for the treatment of NSCLC in advanced stages. 展开更多

The coronavirus disease 2019(COVID-19)caused by SARS-CoV-2 was identified in December 2019.The symptoms include fever,cough,dyspnea,early symptom of sputum,and acute respiratory distress syndrome(ARDS).Mesenchymal stem cell(MSC)therapy is the immediate treatment used for patients with severe cases of COVID-19.Herein,we describe two confirmed cases of COVID-19 in Wuhan to explore the role of MSC in the treatment of COVID-19.MSC transplantation increases the immune indicators(including CD4 and lymphocytes)and decreases the inflammation indicators(interleukin-6 and C-reactive protein).High-flow nasal cannula can be used as an initial support strategy for patients with ARDS.With MSC transplantation,the fracrion of inspired O2(Fi02)of the two patients gradually decreased while the oxygen saturation(Sa02)and partial pressure of oxygen(P02)improved.Additionally,the patients9 chest computed tomography showed that bilateral lung exudate lesions were adsorbed after MSC infusion.Results indicated that MSC transplantation provides clinical data on the treatment of COVID-19 and may serve as an alternative method for treating COVID-19,particularly in patients with ARDS. 展开更多
