To balance the relationship between high yield and low nitrogen supply,the nitrogen utilization efficiency of watermelon needs to be improved urgently.Nodule inception-like Protein(NLP)transcription factors play a key...To balance the relationship between high yield and low nitrogen supply,the nitrogen utilization efficiency of watermelon needs to be improved urgently.Nodule inception-like Protein(NLP)transcription factors play a key node role in nitrate response and growth and development of plant,however,comprehensive analysis of the NLP gene family in watermelon is unclear.This study explored the functional classification,evolutionary characteristics,and expression profile of the ClNLP gene family.Three ClNLPs were categorized into three groups according to their gene structure and phylogeny.All of them contained the conserved RWP-RK and PB1 domains.Evolutionary analysis of ClNLPs revealed that ClNLP1 and ClNLP3 underwent strong purified selection.In addition,cis-acting elements related to plant hormones and abiotic stresses were present in the ClNLP promoter.According to tissue-specific analysis ClNLP was widely expressed in roots,stems,leaves,flowers and fruits,and ClNLP1 was significantly induced in the roots of different nitrogen utilization varieties under different nitrate nitrogen supply.The SRTING functional protein association network suggested that ClNLP1 is associated with most genes,such as NRT1.1,NRT2.1,NIA1,and NIR1,and the dual-luciferase reporter assay found that ClNLP1 positively regulates the expression of ClNRT2.1.We speculated that ClNLP1 might play a central role in regulating the response of watermelon to nitrate nitrogen.展开更多

Fourteen SD rats were randomly and evenly allocated t...Purpose: To establish an animal model of autologous oral mucosa grafting for limbal stem cell deficiency.Methods: The study was carried from August to October2012. Fourteen SD rats were randomly and evenly allocated to study group A and control group B. Limbal stem cell deficiency was established by alkali burn in the right eye of each rat in both groups. Rats in group A received autologous oral mucosa strip transplantation following the chemical burn. Rats in group B did not receive surgery after the chemical burn.Topical antibiotics and dexamethasone were used in all rats.Corneal clarity, corneal fluorescein staining, oral mucosal graft survival, and complications at postoperative days 1,3,7,14 were observed.Results: The oral mucosa strip graft was detached in one rat in group A. Reepithelialization was observed starting from the graft position and was completed within 14 days in the remaining 6 eyes in group A. However, persistent corneal epithelium defect was observed in all eyes in group B, among which corneal melting and perforation was observed in 2 eyes and corneal opacification with neovascularization was observed in the remaining 5 eyes.Conclusion: Autologous oral mucosa strip grafting for limbal stem cell deficiency can be achieved by a rat model following chemical burn. The fate of the transplanted oral mucosal epithelial cells warrants further study.展开更多

We have isolated a population of mesenchymal stem cell-like pluripotent cells from human epidermis, ...Skin is the largest organ of the human body and a possible source of stem cells for research and cell-based therapy. We have isolated a population of mesenchymal stem cell-like pluripotent cells from human epidermis, termed human (h) EMSCPCs. This preliminary study tested if these hEMSCPCs can be induced to differentiate into neural-like cells. Human EMSCPCs were first cultured for four to seven days in a serum-free neural stem cell (NSC) medium for pre-induction. During pre-induction, hEMSCPCs coalesced into dense spheres that resembled neural rosettes. In the presence of a conditioned differentiation medium, pre-induced cells took on the morphological characteristics of neural cells, including slender projections with inflated or claw-like ends that contacted the soma or projections of other cells as revealed by confocal microscopy. Moreover, these differentiating cells expressed the neural-specific markers β-III tubulin, MAP2, GFAP, and synapsin I as evidenced by immunocytochemistry. Both pre-induced hEMSCPCs and uninduced hEMSCPCs were labeled with CM-DiI and transplanted into the vitreous cavities of nude mice. Transplanted cells were examined four weeks later in frozen eyeball sections by immunofluorescence staining, which demonstrated superior retinal migration and neural differentiation of pre-induced cells. Our study is the first to demonstrate that hEMSCPCs possess the capacity to differentiate into neural-like cells, suggesting potential uses for the treatment of retinal diseases such as age-related macular degeneration.展开更多

Compound fault,as a primary failure leading to unexpected downtime of rotating machinery,dramatically increases the difficulty in fault diagnosis.To deal with the difficulty encountered in implementing compound fault ...Compound fault,as a primary failure leading to unexpected downtime of rotating machinery,dramatically increases the difficulty in fault diagnosis.To deal with the difficulty encountered in implementing compound fault diagnosis(CFD),researchers and engineers from industry and academia have made numerous significant breakthroughs in recent years.Admittedly,many systematic surveys focused on fault diagnosis have been conducted by reputable researchers.Nevertheless,previous review articles paid more attention to fault diagnosis with several single or independent faults,resulting in that there is still lacking a comprehensive survey on CFD.Therefore,to fulfill the above requirements,it is necessary to provide an in-depth overview of fault diagnosis methods or algorithms for compound faults of rotating machinery and uncover potential challenges or opportunities that would guide and inspire readers to devote their efforts to promoting fault diagnosis technology more effective and practical.Specifically,the backgrounds,including the related definitions and a new taxonomy of CFD methods,are detailed according to the way of implementing compound fault recognition.Then,the stateof-the-art applications of CFD are overviewed based on relevant publications in the past decades.Finally,the challenges and opportunities associated with implementing CFD are concluded and followed by a conclusion for ending this survey.We believe that this review article can provide a systematic guideline of CFD from different aspects for potential readers and seasoned researchers.展开更多
Background:A variety of experimental animal models are used in basic ophthalmological research to elucidate physiological mechanisms of vision and disease pathogenesis.The choice of animal model is based on the measur...Background:A variety of experimental animal models are used in basic ophthalmological research to elucidate physiological mechanisms of vision and disease pathogenesis.The choice of animal model is based on the measurability of specific parameters or structures,the applicability of clinical measurement technologies,and the similarity to human eye function.Studies of eye pathology usually compare optical parameters between a healthy and altered state,so accurate baseline assessments are critical,but few reports have comprehensively examined the normal anatomical structures and physiological functions in these models.Methods:Three cynomolgus monkeys,six New Zealand rabbits,ten Sprague Dawley(SD)rats,and BALB/c mice were examined by fundus photography(FP),fundus fluorescein angiography(FFA),and optical coherence tomography(OCT).Results:Most retinal structures of cynomolgus monkey were anatomically similar to the corresponding human structures as revealed by FP,FFA,and OCT.New Zealand rabbits have large eyeballs,but they have large optic disc and myelinated retinal nerve fibers in their retinas,and the growth pattern of retinal vessels were also different to the human retinas.Unlike monkeys and rabbits,the retinal vessels of SD rats and BALB/c mice were widely distributed and clear.The OCT performance of them were similar with human beings except the macular.Conclusions:Monkey is a good model to study changes in retinal structure associated with fundus disease,rabbits are not suitable for studies on retinal vessel diseases and optic nerve diseases,and rats and mice are good models for retinal vascular diseases.These measures will help guide the choice of model and measurement technology and reduce the number of experimental animals required.展开更多

Corneal inflammation was evaluated by inflammation index(IF) grading and NF-κB immunohistochemistry at the same time points. The association between lymphatic vessel counting(LVC) and the IF scores was then examined.Results:.LYVE-1 positive lymphatic vessels occurred in the corneal stroma on day 3,.developed throughout days 7 and10,.and peaked in number at day 14 after keratoplasty.Corneal inflammation was strong on day 3, and then resolved gradually,.but increased again from days 7 to 14 after the transplantation..LVC was strongly and positively correlated with IF after keratoplasty(r=0.41;P<0.05). However, changes in IF scores and LVC were not parallel.Conclusion:.A close,.but not parallel,.relationship was found between corneal lymphangiogenesis and corneal inflammation after corneal transplantation.展开更多
Glaucoma is the leading cause of irreversible blindness worldwide.In the pathogen-esis of glaucoma,activated microglia can lead to retinal ganglion cells(RGCs)apoptosis and death,however,the molecular mechanisms remai...Glaucoma is the leading cause of irreversible blindness worldwide.In the pathogen-esis of glaucoma,activated microglia can lead to retinal ganglion cells(RGCs)apoptosis and death,however,the molecular mechanisms remain largely unknown.We demonstrate that phospholipid scramblase 1(PLSCR1)is a key regulator promoting RGCs apoptosis and their clearance by microglia.As evidenced in retinal progenitor cells and RGCs of the acute ocular hypertension(AOH)mouse model,overexpressed PLSCR1 induced its translocation from the nucleus to the cytoplasm and cytomembrane,as well as elevated phosphatidylserine exposure and reactive oxygen species generation with subsequent RGCs apoptosis and death.These damages were effectively attenuated by PLSCR1 inhibition.In the AOH model,PLSCR1 led to an increase in M1 type microglia activation and retinal neuroinflammation.Upregulation of PLSCR1 resulted in strongly elevated phagocytosis of apoptotic RGCs by activated microglia.Taken together,our study provides important insights linking activated microglia to RGCs death in the glaucoma pathogenesis and other RGC-related neurodegenerative diseases.展开更多

A simplified unconventional Hamilton's variational principle corresponding to initial value problems is proposed, which is convenient for applications. The displacement and mo- mentum are approximated with the same Lagrange interpolation. After the numerical integration and variational operation, the original problems are expressed as algebraic equations with the displacement and momentum at the interpolation points as unknown variables. Some particular variational integrators are derived. An optimal scheme of choosing initial values for the Newton-Raphson method is presented for the nonlinear dynamic system. In addition, specific examples show that the proposed integrators are symplectic when the interpolation point coincides with the numerical integration point, and both are Gaussian quadrature points. Meanwhile, compared with the same order symplectic Runge-Kutta methods, although the accuracy of the two methods is almost the same, the proposed integrators are much simpler and less computationally expensive.展开更多
Background lnterleuldn-18(IL-18)plays a key role in the development,progression and outcome of coronary artery disease and its complications.However,its variability relation to the characterization of atherosclerotic ...Background lnterleuldn-18(IL-18)plays a key role in the development,progression and outcome of coronary artery disease and its complications.However,its variability relation to the characterization of atherosclerotic plaque and percutaneous coronary intervention are still unknown.Methods Fifty four patients with coronary artery disease[22 patients with stable angina(SA)and 32 patients with acute coronary syndrome(ACS)]were enrolled in this study.All patients underwent percutaneous coronary intervention(PCI).The stability of the plaques at the criminal vessels was assessed with analogical IVUS.Serum IL-18 levels were measured at the time points of 5 rain before PCI,and Oh,6h,24h and lmonth after PCI in all patients.Results ACS group consisted mainly of lipidic unstable plaques while SA group of fibrous stable plaques.Moreover,compared with those in SA group,eccentricity index(EI)and remodeling index(RI)were significantly higher in ACS group.Positive remodeling was seen in ACS group while negative or no remodeling in SA group.Further,serum IL-18 levels were significantly elevated in patients with ACS than those in SA group before PCI,increased at Oh,6h,24h after PCI(P<0.05)and were not significant different at 1 month after PCI from those before PCI.Conclusions There is significant difference in the composition and structural characteristics of atherosclerotic plaques between ACS and UA groups.PCI triggersd and enhances the inflammatory response in a short time.Serum levels of IL-18 are the predictors of progression of unstable plaque in atherosclerosis.Post-operative complications of PCI might be reduced by inhibiting IL-18.展开更多

Rats in group B did not receive surgery after the chemical burn.Topical antibiotics and dexamethasone were used in all rats.Corneal clarity, corneal fluorescein staining, oral mucosal graft survival, and complications at postoperative days 1,3,7,14 were observed.Results: The oral mucosa strip graft was detached in one rat in group A. Reepithelialization was observed starting from the graft position and was completed within 14 days in the remaining 6 eyes in group A. However, persistent corneal epithelium defect was observed in all eyes in group B, among which corneal melting and perforation was observed in 2 eyes and corneal opacification with neovascularization was observed in the remaining 5 eyes.Conclusion: Autologous oral mucosa strip grafting for limbal stem cell deficiency can be achieved by a rat model following chemical burn. The fate of the transplanted oral mucosal epithelial cells warrants further study.
文摘Skin is the largest organ of the human body and a possible source of stem cells for research and cell-based therapy. We have isolated a population of mesenchymal stem cell-like pluripotent cells from human epidermis, termed human (h) EMSCPCs. This preliminary study tested if these hEMSCPCs can be induced to differentiate into neural-like cells. Human EMSCPCs were first cultured for four to seven days in a serum-free neural stem cell (NSC) medium for pre-induction. During pre-induction, hEMSCPCs coalesced into dense spheres that resembled neural rosettes. In the presence of a conditioned differentiation medium, pre-induced cells took on the morphological characteristics of neural cells, including slender projections with inflated or claw-like ends that contacted the soma or projections of other cells as revealed by confocal microscopy. Moreover, these differentiating cells expressed the neural-specific markers β-III tubulin, MAP2, GFAP, and synapsin I as evidenced by immunocytochemistry. Both pre-induced hEMSCPCs and uninduced hEMSCPCs were labeled with CM-DiI and transplanted into the vitreous cavities of nude mice. Transplanted cells were examined four weeks later in frozen eyeball sections by immunofluorescence staining, which demonstrated superior retinal migration and neural differentiation of pre-induced cells. Our study is the first to demonstrate that hEMSCPCs possess the capacity to differentiate into neural-like cells, suggesting potential uses for the treatment of retinal diseases such as age-related macular degeneration.

The dispatching management system applied in the warehouse has a wide range of applications in the logistics storage system. Improving the operational efficiency of the dispatching management system can effectively improve the efficiency of the automated warehouse system and reduce the cost of logistics transportation. Therefore, this paper designs a set of dispatching systems with the tracking car as the control target and realizes the positioning and path planning functions of the tracking car in the system. At present, the tracking car can completely receive the commands sent by the system, accurately follow the established path. At the same time, the system software can also obtain the video images taken by the camera on the car and play them smoothly.

文摘Compound fault,as a primary failure leading to unexpected downtime of rotating machinery,dramatically increases the difficulty in fault diagnosis.To deal with the difficulty encountered in implementing compound fault diagnosis(CFD),researchers and engineers from industry and academia have made numerous significant breakthroughs in recent years.Admittedly,many systematic surveys focused on fault diagnosis have been conducted by reputable researchers.Nevertheless,previous review articles paid more attention to fault diagnosis with several single or independent faults,resulting in that there is still lacking a comprehensive survey on CFD.Therefore,to fulfill the above requirements,it is necessary to provide an in-depth overview of fault diagnosis methods or algorithms for compound faults of rotating machinery and uncover potential challenges or opportunities that would guide and inspire readers to devote their efforts to promoting fault diagnosis technology more effective and practical.Specifically,the backgrounds,including the related definitions and a new taxonomy of CFD methods,are detailed according to the way of implementing compound fault recognition.Then,the stateof-the-art applications of CFD are overviewed based on relevant publications in the past decades.Finally,the challenges and opportunities associated with implementing CFD are concluded and followed by a conclusion for ending this survey.We believe that this review article can provide a systematic guideline of CFD from different aspects for potential readers and seasoned researchers.

文摘Background:A variety of experimental animal models are used in basic ophthalmological research to elucidate physiological mechanisms of vision and disease pathogenesis.The choice of animal model is based on the measurability of specific parameters or structures,the applicability of clinical measurement technologies,and the similarity to human eye function.Studies of eye pathology usually compare optical parameters between a healthy and altered state,so accurate baseline assessments are critical,but few reports have comprehensively examined the normal anatomical structures and physiological functions in these models.Methods:Three cynomolgus monkeys,six New Zealand rabbits,ten Sprague Dawley(SD)rats,and BALB/c mice were examined by fundus photography(FP),fundus fluorescein angiography(FFA),and optical coherence tomography(OCT).Results:Most retinal structures of cynomolgus monkey were anatomically similar to the corresponding human structures as revealed by FP,FFA,and OCT.New Zealand rabbits have large eyeballs,but they have large optic disc and myelinated retinal nerve fibers in their retinas,and the growth pattern of retinal vessels were also different to the human retinas.Unlike monkeys and rabbits,the retinal vessels of SD rats and BALB/c mice were widely distributed and clear.The OCT performance of them were similar with human beings except the macular.Conclusions:Monkey is a good model to study changes in retinal structure associated with fundus disease,rabbits are not suitable for studies on retinal vessel diseases and optic nerve diseases,and rats and mice are good models for retinal vascular diseases.These measures will help guide the choice of model and measurement technology and reduce the number of experimental animals required.

文摘背景与目的非小细胞肺癌的免疫治疗药物pembrolizumab需要基于包括免疫组织化学(immunohistochemistry,IHC)在内的检测结果,即通过检测程序性死亡配体1(programmed cell death protein 1,PD-L1)的表达等手段来预测治疗反应。评估肺腺癌细胞学标本免疫细胞化学(immunocytochemistry,ICC)方法检测PD-L1的可行性,并探讨PD-L1表达与临床病理及分子特征的相关性。方法收集60例肺腺癌胸水细胞学标本应用PD-L1 sp263试剂按照生产说明书进行免疫细胞化学染色,同时对胸水细胞学标本做高通量二代测序(next-generation sequencing,NGS),探讨PD-L1与驱动基因突变的相关性。结果60例肺腺癌胸水细胞块标本ICC检测中,有35例PD-L1表达阳性,阳性表达率为58.3%。本院57例组织学标本PD-L1 IHC表达的阳性率为33.3%,细胞学标本与组织学标本差异无统计学意义(P>0.05)。60例细胞学标本中26例接受NGS检测,15例(57.7%)发现表皮生长因子受体(epidermal growth factorreceptor,EGFR)突变,经统计PD-L1表达与EGFR突变未发现相关性。PD-L1的阳性表达率与研究人群的年龄、性别、是否淋巴结或远处转移及是否进行放化疗或靶向治疗均未发现相关性(P>0.05)。结论在无手术标本可取时,胸水细胞学细胞块标本可以对PD-L1进行免疫细胞化学检测,其结果具有可行性。

文摘In machinery fault diagnosis,labeled data are always difficult or even impossible to obtain.Transfer learning can leverage related fault diagnosis knowledge from fully labeled source domain to enhance the fault diagnosis performance in sparsely labeled or unlabeled target domain,which has been widely used for cross domain fault diagnosis.However,existing methods focus on either marginal distribution adaptation(MDA)or conditional distribution adaptation(CDA).In practice,marginal and conditional distributions discrepancies both have significant but different influences on the domain divergence.In this paper,a dynamic distribution adaptation based transfer network(DDATN)is proposed for cross domain bearing fault diagnosis.DDATN utilizes the proposed instance-weighted dynamic maximum mean discrepancy(IDMMD)for dynamic distribution adaptation(DDA),which can dynamically estimate the influences of marginal and conditional distribution and adapt target domain with source domain.The experimental evaluation on cross domain bearing fault diagnosis demonstrates that DDATN can outperformance the state-of-the-art cross domain fault diagnosis methods.

文摘In this study,the bipartite time-varying output formation tracking problem for heterogeneous multi-agent systems(MASs)with multiple leaders and switching commu-nication networks is considered.Note that the switching communication networks may be connected or disconnected.To address this problem,a novel reduced-dimensional observer-based fully distributed asynchronous dynamic edge-event-triggered output feedback control protocol is developed,and the Zeno behavior is ruled out.The theoretical analysis gives the admissible switching frequency and switching width under the proposed control protocol.Different from the existing works,the control protocol reduces the dimension of information to be transmitted between neighboring agents.Moreover,since an additional positive internal dynamic variable is introduced into the triggering functions,the control protocol can guarantee a larger inter-event time interval compared with previous results.Finally,a simulation example is given to verify the effectiveness and performance of the theoretical result.

文摘Purpose:.To examine the relationship between corneal inflammation and corneal lymphangiogenesis after keratoplasty.Methods:.Rat corneal lymphangiogenesis was examined by lymphatic vessel endothelial receptor(LYVE-1) immunohistochemistry and whole mount immunofluorescence at 1, 3, 7,10, and 14 days after corneal transplantation. Corneal inflammation was evaluated by inflammation index(IF) grading and NF-κB immunohistochemistry at the same time points. The association between lymphatic vessel counting(LVC) and the IF scores was then examined.Results:.LYVE-1 positive lymphatic vessels occurred in the corneal stroma on day 3,.developed throughout days 7 and10,.and peaked in number at day 14 after keratoplasty.Corneal inflammation was strong on day 3, and then resolved gradually,.but increased again from days 7 to 14 after the transplantation..LVC was strongly and positively correlated with IF after keratoplasty(r=0.41;P<0.05). However, changes in IF scores and LVC were not parallel.Conclusion:.A close,.but not parallel,.relationship was found between corneal lymphangiogenesis and corneal inflammation after corneal transplantation.

文摘Glaucoma is the leading cause of irreversible blindness worldwide.In the pathogen-esis of glaucoma,activated microglia can lead to retinal ganglion cells(RGCs)apoptosis and death,however,the molecular mechanisms remain largely unknown.We demonstrate that phospholipid scramblase 1(PLSCR1)is a key regulator promoting RGCs apoptosis and their clearance by microglia.As evidenced in retinal progenitor cells and RGCs of the acute ocular hypertension(AOH)mouse model,overexpressed PLSCR1 induced its translocation from the nucleus to the cytoplasm and cytomembrane,as well as elevated phosphatidylserine exposure and reactive oxygen species generation with subsequent RGCs apoptosis and death.These damages were effectively attenuated by PLSCR1 inhibition.In the AOH model,PLSCR1 led to an increase in M1 type microglia activation and retinal neuroinflammation.Upregulation of PLSCR1 resulted in strongly elevated phagocytosis of apoptotic RGCs by activated microglia.Taken together,our study provides important insights linking activated microglia to RGCs death in the glaucoma pathogenesis and other RGC-related neurodegenerative diseases.

文摘This paper develops a new approach to construct variational integrators. A simplified unconventional Hamilton's variational principle corresponding to initial value problems is proposed, which is convenient for applications. The displacement and mo- mentum are approximated with the same Lagrange interpolation. After the numerical integration and variational operation, the original problems are expressed as algebraic equations with the displacement and momentum at the interpolation points as unknown variables. Some particular variational integrators are derived. An optimal scheme of choosing initial values for the Newton-Raphson method is presented for the nonlinear dynamic system. In addition, specific examples show that the proposed integrators are symplectic when the interpolation point coincides with the numerical integration point, and both are Gaussian quadrature points. Meanwhile, compared with the same order symplectic Runge-Kutta methods, although the accuracy of the two methods is almost the same, the proposed integrators are much simpler and less computationally expensive.
文摘Background lnterleuldn-18(IL-18)plays a key role in the development,progression and outcome of coronary artery disease and its complications.However,its variability relation to the characterization of atherosclerotic plaque and percutaneous coronary intervention are still unknown.Methods Fifty four patients with coronary artery disease[22 patients with stable angina(SA)and 32 patients with acute coronary syndrome(ACS)]were enrolled in this study.All patients underwent percutaneous coronary intervention(PCI).The stability of the plaques at the criminal vessels was assessed with analogical IVUS.Serum IL-18 levels were measured at the time points of 5 rain before PCI,and Oh,6h,24h and lmonth after PCI in all patients.Results ACS group consisted mainly of lipidic unstable plaques while SA group of fibrous stable plaques.Moreover,compared with those in SA group,eccentricity index(EI)and remodeling index(RI)were significantly higher in ACS group.Positive remodeling was seen in ACS group while negative or no remodeling in SA group.Further,serum IL-18 levels were significantly elevated in patients with ACS than those in SA group before PCI,increased at Oh,6h,24h after PCI(P<0.05)and were not significant different at 1 month after PCI from those before PCI.Conclusions There is significant difference in the composition and structural characteristics of atherosclerotic plaques between ACS and UA groups.PCI triggersd and enhances the inflammatory response in a short time.Serum levels of IL-18 are the predictors of progression of unstable plaque in atherosclerosis.Post-operative complications of PCI might be reduced by inhibiting IL-18.
