DEAR EDITOR,Pore-forming proteins(PFPs)are widely distributed among all kingdoms of life and can oligomerize to form pores/channels in membrane systems.Extracellular vesicles(EVs)circulate in all biological fluids and can trigger biological responses at a distance,thus emerging as an additional means of intercellular communication through the release of cellular cargo,such as lipids,nucleic acids,metabolites,and proteins.To date,however,the mechanism by which EV contents are released into extracellular space remains unclear.In our previous study on toads(Bombina maxima),we identified a PFP and trefoil factor complexβγ-CAT(βγ-crystallin fused aerolysin-like protein(α-subunit)and trefoil factor(β-subunit)complex,hence namedβγ-CAT),which assembled under strict regulation in response to environmental cues.Here,we observed that the PFPβγ-CAT colocalized with EVs in the skin of B.maxima in vivo.Using small EVs(sEVs)isolated from B.maxima dermal fibroblast cells and murine fibroblast cells,we found thatβγ-CAT could specifically target and oligomerize on purified EVs,rather than disrupt membrane integrity,and promote the release of different metabolites.Our analysis revealed that a secretory PFP drove metabolite release from EVs through channel formation,providing novel clues for the delivery of EV content into extracellular space. 展开更多

Cationic antimicrobial peptides (AMPs) are considered as important candidate therapeutic agents, which exert potent microbicidal properties against bacteria, fungi and some viruses. Based on our previous findings king cobra cathelicidin (OH-CATH) is a 34-amino acid peptide that exerts strong antibacterial and weak hemolytic activity. The aim of this research is to evaluate the efficacy of both OH-CATH30 and its analog D-OH-CATH30 against clinical isolates comparing with routinely utilized antibiotics in vitro. In this study, 584 clinical isolates were tested (spanning 2013-2016) and the efficacy of the candidate peptides and antibiotics were determined by a broth microdilution method according to the CLSI guidelines. Among the 584 clinical isolates, 85% were susceptible to OH-CATH30 and its analogs. Both L- and D-OH-CATH30 showed higher efficacy against (toward) Gram-positive bacteria and stronger antibacterial activity against nearly all Gram-negative bacteria tested compare with antibiotics. The highest bactericidal activity was detected against Acinetobacter spp., including multi-drug-resistant Acinetobacter baumannfi (MRAB) and methicillin-resistant Staphylococcus aureus (MRSA). The overall efficacy of OH-CATH30 and its analogs was higher than that of the 9 routinely used antibiotics. OH-CATH30 is a promising candidate drug for the treatment of a wide variety of bacterial infections which are resistant to many routinely used antimicrobial agents. 展开更多

As the oldest venomous animals,centipedes use their venom as a weapon to attack prey and for protection.Centipede venom,which contains many bioactive and pharmacologically active compounds,has been used for centuries in Chinese medicine,as shown by ancient records.Based on comparative analysis,we revealed the diversity of and differences in centipede toxin-like molecules between Scolopendra mojiangica,a substitute pharmaceutical material used in China,and S.subspinipes mutilans.More than 6000 peptides isolated from the venom were identified by electrospray ionization-tandem mass spectrometry(ESI-MS/MS)and inferred from the transcriptome.As a result,in the proteome of S.mojiangica,246 unique proteins were identified:one in five were toxin-like proteins or putative toxins with unknown function,accounting for a lower percentage of total proteins than that in S.mutilans.Transcriptome mining identified approximately 10 times more toxin-like proteins,which can characterize the precursor structures of mature toxinlike peptides.However,the constitution and quantity of the toxin transcripts in these two centipedes were similar.In toxicity assays,the crude venom showed strong insecticidal and hemolytic activity.These findings highlight the extensive diversity of toxin-like proteins in S.mojiangica and provide a new foundation for the medical-pharmaceutical use of centipede toxin-like proteins. 展开更多

目的了解广州某高等医学院校研究生乙型肝炎病毒(HBV)感染状况,为乙型肝炎防治工作提供依据。方法对广州某高等医学院校2003-2005年入学的3052名研究生进行入学体检现况调查,采集血标本采用ELISA法检测乙型肝炎表面抗原(HBsAg)和表面抗... 目的了解广州某高等医学院校研究生乙型肝炎病毒(HBV)感染状况,为乙型肝炎防治工作提供依据。方法对广州某高等医学院校2003-2005年入学的3052名研究生进行入学体检现况调查,采集血标本采用ELISA法检测乙型肝炎表面抗原(HBsAg)和表面抗体(抗-HBs);用SAS统计软件包对结果进行2χ检验分析。结果广州某高等医学院校研究生HBsAg阳性率为4.75%,其中男性(5.94%)高于女性(3.68%),不同性别HBsAg阳性率差异有统计学意义(χ2=8.5964,P<0.01);不同年级研究生HBsAg阳性率(03、04、05级分别为5.66%、5.40%、3.37%),差异有统计学意义(χ2=7.2629,P<0.05),不同年级研究生抗-HBs阳性率(03、04、05级分别为69.06%、71.36%、78.31%),差异有统计学意义(χ2=24.3465,P<0.01);各年龄组HBsAg阳性率差异无统计学意义(2χ=4.7220,P>0.05);各年龄组抗-HBs阳性率差异有统计学意义(2χ=17.0761,P<0.01)。结论男性群体较女性群体更易成为乙型肝炎病毒的易感人群,是乙型肝炎疫苗接种的重要群体;随着年龄的增加,抗-HBs阳性率逐渐降低,加强学生的乙型肝炎疫苗接种工作对乙型肝炎的防治非常必要。 展开更多

目的了解广州某高等医学院校研究生乙肝疫苗接种情况,为乙肝防治工作提供依据。方法对广州某高等医学院校2006年入学的1139名研究生进行入学体检现况调查,采集血标本用ELISA法检测乙型肝炎表面抗原(HBsAg)和表面抗体(HBsAb)。同... 目的了解广州某高等医学院校研究生乙肝疫苗接种情况,为乙肝防治工作提供依据。方法对广州某高等医学院校2006年入学的1139名研究生进行入学体检现况调查,采集血标本用ELISA法检测乙型肝炎表面抗原(HBsAg)和表面抗体(HBsAb)。同时发放乙肝疫苗接种情况调查表,调查乙肝疫苗接种的年代、次数,是否加强接种及时间。用SAS统计软件包对结果进行X^2检验分析。结果广州某高等医学院校06级研究生HBsAg阳性率为2.90%,曾注射乙肝疫苗组,HBsAg阳性率显著低于从未注射疫苗组(1.15% vs. 21.69%,P〈0.0001),而HBsAb阳性率则显著高于从未注射疫苗组(81.54% vs.44.58%,P〈0.0001)。有17.31%曾接种乙肝疫苗者未能达到预期预防效果。不同年龄研究生乙肝疫苗接种效果有差异(P=0.0462),随着年龄的增加,HBsAb阳性率有下降趋势。女性乙肝疫苗接种效果较男性好(80.0% vs.84.87%,P=0.0468)。接种年限在3年内者,HBsAb阳性率较其他年限高(0—3年 vs.4—6年,P=0.0089;0—3年 vs.7—9年,P=0.0172;0—3年 vs.〉9年,P=0,0474)。注射大于3针(即加强接种)免疫效果较注射3针者好,差异有统计学意义(P=0.0093)。结论随着年龄的增加,乙肝疫苗接种效果(HBsAb阳性率)逐渐降低。男性群体较女性群体更易成为乙型肝炎病毒的易感人群。对接种年限大于3年者,可进行抗-HBs监测,及时进行加强免疫。 展开更多

The arachidonic acid(AA)metabolic pathway participates in various physiological processes as well as in the development of malignancies.We analyzed genomic alterations in AA metabolic enzymes in the Cancer Genome Atlas(TCGA)prostate cancer(PCa)dataset and found that the gene encoding soluble epoxide hydrolase(EPHX2)is frequently deleted in PCa.EPHX2 mRNA and protein expression in PCa was examined in multiple datasets by differential gene expression analysis and in a tissue microarray by immunohistochemistry.The expression data were analyzed in conjunction with clinicopathological variables.Both the mRNA and protein expression levels of EPHX2 were significantly decreased in tumors compared with normal prostate tissues and were inversely correlated with the Gleason grade and disease-free survival time.Furthermore,EPHX2 mRNA expression was significantly decreased in metastatic and recurrent PCa compared with localized and primary PCa,respectively.In addition,EPHX2 protein expression correlated negatively with Ki67 expression.In conclusion,EPHX2 deregulation is significantly correlated with the clinical characteristics of PCa progression and may serve as a prognostic marker for PCa. 展开更多
