AIM:To investigate the adjunct anticancer effect of Astragalus polysaccharides in H22 tumor-bearing mice.METHODS:To establish a solid tumor model,5.0 × 10 6 /mL H22 hepatoma cells were inoculated subcutaneously i...AIM:To investigate the adjunct anticancer effect of Astragalus polysaccharides in H22 tumor-bearing mice.METHODS:To establish a solid tumor model,5.0 × 10 6 /mL H22 hepatoma cells were inoculated subcutaneously into the right armpit region of Kunming mice(6-12 wk old,18-22 g).When the tumors reached a size of 100 mm 3,the animals were treated as indicated,and the mice were randomly assigned to seven groups(n = 10 each).After ten days of treatment,blood samples were collected from mouse eyes,and serum was harvested by centrifugation.Mice were sacrificed,and the whole body,tumor,spleen and thymus were weighed immediately.The rate of tumor inhibition and organ indexes were calculated.The expression levels of serum cytokines,P-glycoprotein(P-GP) and multidrug resistance(MDR) 1 mRNA in tumor tissues were detected using enzyme-linked immunosorbent assay,Western blotting,and quantitative myeloid-derived suppressor cells reverse transcription-polymerase chain reaction,respectively.RESULTS:The tumor inhibition rates in the treatment groups of Adriamycin(ADM) + Astragalus polysaccharides(APS)(50 mg/kg),ADM + APS(100 mg/kg),and ADM + APS(200 mg/kg) were significantly higher than in the ADM group(72.88% vs 60.36%,P = 0.013;73.40% vs 60.36%,P = 0.010;77.57% vs 60.36%,P = 0.001).The spleen indexes of the above groups were also significantly higher than in the ADM group(0.65 ± 0.22 vs 0.39 ± 0.17,P = 0.023;0.62 ± 0.34 vs 0.39 ± 0.17,P = 0.022;0.67 ± 0.20 vs 0.39 ± 0.17,P = 0.012),and the thymus indexes of the ADM + APS(100 mg/kg) and ADM + APS(200 mg/kg) groups were significantly higher than in the ADM group(0.20 ± 0.06 vs 0.13 ± 0.04,P = 0.029;0.47 ± 0.12 vs 0.13 ± 0.04,P = 0.000).APS was found to exert a synergistic antitumor effect with ADM and to alleviate the decrease in the sizes of the spleen and thymus induced by AMD.The expression of interleukin-1α(IL-1α),IL-2,IL-6,and tumor necrosis factor-α(TNF-α) was significantly higher in the ADM + APS(50 mg/kg),ADM + APS(100 mg/kg) and ADM + APS(200 mg/kg) groups than in the ADM group;and IL-10 was significantly lower in the above groups than in the ADM group.APS could increase IL-1α,IL-2,IL-6,and TNF-α expression and decrease IL-10 levels.Compared with the ADM group,APS treatment at a dose of 50-200 mg/kg could downregulate MDR1 mRNA expression in a dose-dependent manner(0.48 ± 0.13 vs 4.26 ± 1.51,P = 0.000;0.36 ± 0.03 vs 4.26 ± 1.51,P = 0.000;0.21 ± 0.04 vs 4.26 ± 1.51,P = 0.000).The expression level of P-GP was significantly lower in the ADM + APS(200 mg/kg) group than in the ADM group(137.35 ± 9.20 mg/kg vs 282.19 ± 20.54 mg/kg,P = 0.023).CONCLUSION:APS exerts a synergistic anti-tumor effect with ADM in H22 tumor-bearing mice.This may be related to its ability to enhance the expression of IL1α,IL-2,IL-6,and TNF-α,decrease IL-10,and downregulate MDR1 mRNA and P-GP expression levels.展开更多
Chronic stress is strongly associated with the occurrence and development of depression and cardiovascular disease.Stress can induce altered mitochondrial function and activation of apoptosis in the cardio-cerebral sy...Chronic stress is strongly associated with the occurrence and development of depression and cardiovascular disease.Stress can induce altered mitochondrial function and activation of apoptosis in the cardio-cerebral system.However,it is unknown whether the protein kinase C ε(PKCε)-aldehyde dehydrogenase 2(ALDH2) pathway is altered under chronic stress,and this study sought to address this question.A rat model of depression was established using a chronic unpredictable mild stress(CUMS) protocol.After experiencing CUMS for 4 weeks,the sucrose preference test and the forced swim test verified depressive-like behaviors.Enzyme linked immunosorbent assays showed that ALDH2 activity was decreased in the rat hippocampus and prefrontal cortex,but was not altered in the myocardium.Western blot assays demonstrated reduced levels of ALDH2 and PKCε,but increased levels of 4-hydroxy-2-nonenal(4 HNE) adducts.Caspase-3 expression did not obviously alter,but active forms of caspase-3 were increased in the hippocampus and prefrontal cortex.In the myocardium,expression of ALDH2,PKCε and 4 HNE adducts did not remarkably alter;while caspase-3 expression was reduced and the active forms of caspase-3 were upregulated.Pearson's correlation test demonstrated that expression of 4 HNE adducts was positively correlated with levels of the active forms of caspase-3 in the hippocampus and prefrontal cortex,but not in the myocardium.In conclusion,chronic stress can damage the PKCε-ALDH2 signaling pathway in the hippocampus and prefrontal cortex,but not in the myocardium.Moreover,4 HNE is associated with active forms of caspase-3 in the hippocampus and prefrontal cortex.展开更多

Additionally, the radial differences in microstructure and mechanical property of joints were investigated by hierarchy slicing method. The results displayed that the width of weld zone and heat-affected zone of joints became wider along radial direction. Meanwhile, the tensile strength of joints decreased gradually along the radial direction. According to the theoretical analysis, the temperature gradient and inhomogeneous forging pressure leaded to the radial differences. Through K-type thermocouples, the actual temperatures at different locations were measured, and the results were consistent with the theoretical analysis. Theoretically, the radial differences of rotary friction welding joint are an inherent phenomenon; thus, the size of weldment should be limited strictly below the corresponding critical size. In order to prevent radial differences from enlarging, the welding surface profile of weldment can be processed into oval shape, and a larger forging pressure can be used within the scope of the joint deformation allowed according to causes for radial differences.展开更多
In this paper,ZnZnFe_(2)O_(4) nanorods were prepared using Zn/Fe metal organic framework(MOF)as precursors,and ZnZnFe_(2)O_(4)/reduced graphene oxide(rGO)was prepared by hydrothermal method.The morphology and composit...In this paper,ZnZnFe_(2)O_(4) nanorods were prepared using Zn/Fe metal organic framework(MOF)as precursors,and ZnZnFe_(2)O_(4)/reduced graphene oxide(rGO)was prepared by hydrothermal method.The morphology and composition of the ZnZnFe_(2)O_(4)/rGO nanocomposite were characterized,and the results showed that the MOF-derived ZnZnFe_(2)O_(4) nanorods are uniformly modified on the surface of rGO.The ZnZnFe_(2)O_(4)/rGO nanocomposite exhibits better SO_(2) gas sensing performance than the single ZnZnFe_(2)O_(4) nanorods at room temperature.The sensing characteristics of single ZnZnFe_(2)O_(4) film sensor,single rGO film sensor and ZnZnFe_(2)O_(4)/rGO composite film sensor at SO_(2)gas concentration(1×10^(-6)-100×10^(-6))were tested.The response of ZnZnFe_(2)O_(4)/rGO composite sensor can reach 18.32%at room temperature.Compared with single ZnZnFe_(2)O_(4) and rGO film sensors,the ZnZnFe_(2)O_(4)/rGO composite sensor has better transient response,good sensitivity and selectivity.In this work,the improvement of the sensor performance is not only due to the p-n heterostructure between ZnZnFe_(2)O_(4) nanorods and rGO nanosheets,but also to the excellent electrical properties of rGO.It provides a new idea for the detection of SO_(2) at room temperature.展开更多
