Klebsiella pneumoniae(K.pneumonia,KpⅠ)is a predominate inducement of bovine mastitis,which is associated with high mortality and milk yield reduction.However,data is lacking on the molecular characteristics of bovine K.pneumoniae,limiting the risk assessment of its transmission through the food chain.Herein,we investigated the prevalence of K.pneumoniae in 6301 clinical mastitis(CM)milk samples from dairy cattle in northern area of China.In total,183 K.pneumoniae isolates were recovered,with detection rates of 3.0% and 2.8% in 2018 and 2019,respectively.Like human clinical K.pneumoniae,all CM K.pneumoniae isolates belonged to one of three phylogroups:KpⅠ(n=143),Klebsiella.quasipneumoniae subsp.similipneumoniae(KpⅡ-B)(n=37),and Klebsiella variicola(KpⅢ)(n=3).We detected the extendedspectrum β-lactamase-encoding genes bla_(SHV-2a),blac_(CTX-M-14),and bla_(CTX-M-15),as well as clpC,lpfA,lacI,lacZ,lacY,and the fecABDEIR operon in the KpⅠ isolates,which may contribute to their pathogenicity and host adaptability in cows.The high prevalence of KpⅠ in dairy farms may be problematic,as it showed relatively higher rates of antibiotic resistance and virulence gene carriage than the KpⅡ-B and KpⅢ isolates.Furthermore,we observed distinct differences in population structure between CM-and human infection-associated KpⅠ isolates,with the genes associated with invasive infection in humans rarely being observed in bovine isolates,indicating that few CM-associated K.pneumoniae isolates pose a threat to human health.Nevertheless,bovine KpⅡ-B isolates shared a high level of nucleotide sequence identity with isolates from human infections and frequently carried the nitrogen-fixation gene nif,suggesting an association between KpⅡ-B isolates from cattle and humans,and plant-derived bacteria. 展开更多

An Ethnographic Study:The Role of“Play”in the Development of English Listening and Speaking Ability of Two Chinese Children in a Multilingual Setting

Much scholarship on“play”activities has focused on enhancing children’s learning processes and developing interest towards learning second language[1].Many of these studies,however,adopt the survey to conduct their study.This study explored how“play with local kids”activities effect the development of English speaking and listening of two Chinese children in the Philippines,employing an ethnographically based approach to observe the two participants playing with local kids,which lasted about 4 months.The findings of the study revealed that“play”activities can not only help young children have a better understanding of their English conversations,and make them have more confidence to speak English,but also inspire them to speak English in such environment.In view of the results,it is obvious that“play with local kids”is a very good way for young children to acquire a second language in a multilingual setting.It is strongly recommended that language researchers and teachers should pay more attention and do more research on the relationship between“play”and second language acquisition. 展开更多

背景与目的耐药的产生是肺癌死亡率居高不下的主要原因,本研究旨在探究叶酸(folic acid,FA)调控双特异性磷酸酶1(dual specificity phosphatase 1,DUSP1)甲基化对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞奥西替尼(Osimertin... 背景与目的耐药的产生是肺癌死亡率居高不下的主要原因,本研究旨在探究叶酸(folic acid,FA)调控双特异性磷酸酶1(dual specificity phosphatase 1,DUSP1)甲基化对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞奥西替尼(Osimertinib,OSM)耐药性的影响。方法浓度梯度递增法建立OSM耐药NSCLC细胞株PC9R。将PC9R细胞随机分为对照组、OSM组(5μmol/L OSM)、FA组(600 nmol/L FA)、甲基化抑制剂地西他滨组(Decitabine,DAC)(10μmol/L DAC)、FA+OSM组(600 nmol/L FA+5μmol/L OSM)和FA+OSM+DAC组(600 nmol/L FA+5μmol/L OSM+10μmol/L DAC)。CCK-8法检测细胞增殖能力。划痕实验检测细胞迁移能力。Transwell实验检测细胞侵袭能力。流式细胞术测定分析各组细胞凋亡率。实时荧光定量聚合酶链式反应(real-time fluorescence quantitative polymerase chain reaction,RT-qPCR)法检测细胞DUSP1 mRNA的表达水平。甲基化特异性PCR(methylation specific PCR,MSP)检测各组细胞中DUSP1启动子区甲基化状态。Western blot分析各组细胞DUSP1蛋白、DUSP1下游丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路关键蛋白的表达水平。结果与对照组相比,OSM组细胞OD_(450)值(48、72 h)、划痕愈合率、细胞侵袭数目、DUSP1表达显著下降(P<0.05),细胞凋亡率、DUSP1甲基化水平、p38 MAPK蛋白表达、细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK)磷酸化水平显著上升(P<0.05);DAC组细胞OD_(450)值(48、72 h)、划痕愈合率、细胞侵袭数目、DUSP1表达显著上升(P<0.05),细胞凋亡率、p38 MAPK蛋白表达、ERK磷酸化水平、DUSP1甲基化水平显著下降(P<0.05)。与OSM组相比,FA+OSM组细胞OD_(450)值(48、72 h)、划痕愈合率、细胞侵袭数目、DUSP1表达显著下降(P<0.05),细胞凋亡率、DUSP1甲基化水平、p38 MAPK蛋白表达、ERK磷酸化水平显著上升(P<0.05)。与FA+OSM组相比,FA+OSM+DAC组细胞OD_(450)值(48、72 h)、划痕愈合率、细胞侵袭数目、DUSP1水平显著升高,细胞凋亡率、DUSP1甲基化水平、p38 MAPK蛋白表达、ERK磷酸化水平显著降低(P<0.05)。结论FA可能通过增强DUSP1甲基化抑制DUSP1表达,调控下游MAPK信号通路,进而促进细胞凋亡,抑制细胞侵袭转移,最终减弱NSCLC细胞OSM耐药性。 展开更多
