目的 探讨妊娠早期七氟醚全麻对仔鼠海马、顶叶皮质中干扰素诱导蛋白(AIM2)表达及炎性反应的影响。方法 将24只胎龄为5~7 d的Sprague-Dawley(SD)大鼠按照随机数字表法随机分为对照组、七氟醚全麻1组(S1组)和七氟醚全麻2组(S2组),每组8... 目的 探讨妊娠早期七氟醚全麻对仔鼠海马、顶叶皮质中干扰素诱导蛋白(AIM2)表达及炎性反应的影响。方法 将24只胎龄为5~7 d的Sprague-Dawley(SD)大鼠按照随机数字表法随机分为对照组、七氟醚全麻1组(S1组)和七氟醚全麻2组(S2组),每组8只。大鼠分娩后每组随机抽取8只仔鼠,维持生存30 d后,通过Morris水迷宫实验评估仔鼠学习记忆能力。收集出生后30 d的仔鼠脑标本,通过苏木精/伊红染色、尼氏染色观察海马和顶叶皮质神经元结构的形态变化和尼氏体。通过免疫组化法和蛋白质印迹法检测仔鼠海马和顶叶皮质中AIM2、CD45、白细胞介素1β(IL-1β)、pro-caspase-1和caspase-1 p10表达。结果 与对照组相比,S1组和S2组仔鼠长期学习和记忆能力较差,对海马和顶叶皮质的损伤程度不同。与对照组相比,S1组和S2组仔鼠中的AIM2、CD45、IL-1β、pro-caspase-1和caspase-1 p10表达升高(P均<0.05),且S2组高于S1组(P均<0.05)。结论 妊娠早期七氟醚全麻可促进仔鼠海马和顶叶皮质中AIM2表达及炎性反应的发生。 展开更多
