PTEN-induced putative kinase 1(PINK1),a mitochondrial kinase that phosphorylates Parkin and other proteins,plays a crucial role in mitophagy and protection against neurodegeneration.Mutations in PINK1 and Parkin can lead to loss of function and early onset Parkinson's disease.However,there is a lack of strong in vivo evidence in rodent models to support the theory that loss of PINK1 affects mitophagy and induces neurodegeneration.Additionally,PINK1 knockout pigs(Sus scrofa)do not appear to exhibit neurodegeneration.In our recent work involving non-human primates,we found that PINK1 is selectively expressed in primate brains,while absent in rodent brains.To extend this to other species,we used multiple antibodies to examine the expression of PINK1 in pig tissues.In contrast to tissues from cynomolgus monkeys(Macaca fascicularis),our data did not convincingly demonstrate detectable PINK1expression in pig tissues.Knockdown of PINK1 in cultured pig cells did not result in altered Parkin and BAD phosphorylation,as observed in cultured monkey cells.A comparison of monkey and pig striatum revealed more PINK1-phosphorylated substrates in the monkey brain.Consistently,PINK1 knockout in pigs did not lead to obvious changes in the phosphorylation of Parkin and BAD.These findings provide new evidence that PINK1expression is specific to primates,underscoring the importance of non-human primates in investigating PINK1function and pathology related to PINK1 deficiency. 展开更多

BACKGROUND This study aimed to identify characteristic gut genera in obese and normal-weight children(8-12 years old)using 16S rDNA sequencing.The research aimed to provide insights for mechanistic studies and prevention strategies for childhood obesity.Thirty normal-weight and thirty age-and sex-matched obese children were included.Questionnaires and body measurements were collected,and fecal samples underwent 16S rDNA sequencing.Significant differences in body mass index(BMI)and body-fat percentage were observed between the groups.Analysis of gut microbiota diversity revealed lowerα-diversity in obese children.Differences in gut microbiota composition were found between the two groups.Prevotella and Firmicutes were more abundant in the obese group,while Bacteroides and Sanguibacteroides were more prevalent in the control group.AIM To identify the characteristic gut genera in obese and normal-weight children(8-12-year-old)using 16S rDNA sequencing,and provide a basis for subsequent mechanistic studies and prevention strategies for childhood obesity.METHODS Thirty each normal-weight,1:1 matched for age and sex,and obese children,with an obese status from 2020 to 2022,were included in the control and obese groups,respectively.Basic information was collected through questionnaires and body measurements were obtained from both obese and normal-weight children.Fecal samples were collected from both groups and subjected to 16S rDNA sequencing using an Illumina MiSeq sequencing platform for gut microbiota diversity analysis.RESULTS Significant differences in BMI and body-fat percentage were observed between the two groups.The Ace and Chao1 indices were significantly lower in the obese group than those in the control group,whereas differences were not significant in the Shannon and Simpson indices.Kruskal-Wallis tests indicated significant differences in unweighted and weighted UniFrac distances between the gut microbiota of normal-weight and obese children(P<0.01),suggesting substantial disparities in both the species and quantity of gut microbiota between the two groups.Prevotella,Firmicutes,Bacteroides,and Sanguibacteroides were more abundant in the obese and control groups,respectively.Heatmap results demonstrated significant differences in the gut microbiota composition between obese and normal-weight children.CONCLUSION Obese children exhibited lowerα-diversity in their gut microbiota than did the normal-weight children.Significant differences were observed in the composition of gut microbiota between obese and normal-weight children. 展开更多

The analysis of microstates in EEG signals is a crucial technique for understanding the spatiotemporal dynamics of brain electrical activity.Traditional methods such as Atomic Agglomerative Hierarchical Clustering(AAHC),K-means clustering,Principal Component Analysis(PCA),and Independent Component Analysis(ICA)are limited by a fixed number of microstate maps and insufficient capability in cross-task feature extraction.Tackling these limitations,this study introduces a Global Map Dissimilarity(GMD)-driven density canopy K-means clustering algorithm.This innovative approach autonomously determines the optimal number of EEG microstate topographies and employs Gaussian kernel density estimation alongside the GMD index for dynamic modeling of EEG data.Utilizing this advanced algorithm,the study analyzes the Motor Imagery(MI)dataset from the GigaScience database,GigaDB.The findings reveal six distinct microstates during actual right-hand movement and five microstates across other task conditions,with microstate C showing superior performance in all task states.During imagined movement,microstate A was significantly enhanced.Comparison with existing algorithms indicates a significant improvement in clustering performance by the refined method,with an average Calinski-Harabasz Index(CHI)of 35517.29 and a Davis-Bouldin Index(DBI)average of 2.57.Furthermore,an information-theoretical analysis of the microstate sequences suggests that imagined movement exhibits higher complexity and disorder than actual movement.By utilizing the extracted microstate sequence parameters as features,the improved algorithm achieved a classification accuracy of 98.41%in EEG signal categorization for motor imagery.A performance of 78.183%accuracy was achieved in a four-class motor imagery task on the BCI-IV-2a dataset.These results demonstrate the potential of the advanced algorithm in microstate analysis,offering a more effective tool for a deeper understanding of the spatiotemporal features of EEG signals. 展开更多

The vast majority of in vitro studies have demonstrated that PINK1 phosphorylates Parkin to work together in mitophagy to protect against neuronal degeneration.However,it remains largely unclear how PINK1 and Parkin are expressed in mammalian brains.This has been difficult to address because of the intrinsically low levels of PINK1 and undetectable levels of phosphorylated Parkin in small animals.Understanding this issue is critical for elucidating the in vivo roles of PINK1 and Parkin.Recently,we showed that the PINK1 kinase is selectively expressed as a truncated form(PINK1–55)in the primate brain.In the present study,we used multiple antibodies,including our recently developed monoclonal anti-PINK1,to validate the selective expression of PINK1 in the primate brain.We found that PINK1 was stably expressed in the monkey brain at postnatal and adulthood stages,which is consistent with the findings that depleting PINK1 can cause neuronal loss in developing and adult monkey brains.PINK1 was enriched in the membrane-bound fractionations,whereas Parkin was soluble with a distinguishable distribution.Immunofluorescent double staining experiments showed that PINK1 and Parkin did not colocalize under physiological conditions in cultured monkey astrocytes,though they did colocalize on mitochondria when the cells were exposed to mitochondrial stress.These findings suggest that PINK1 and Parkin may have distinct roles beyond their well-known function in mitophagy during mitochondrial damage. 展开更多

In high-altitude cold areas,freeze-thaw(F-T)cycles induced by day-night and seasonal temperature changes cause numerous rock mass slope engineering disasters.To investigate the dynamic properties of rock in the natural environment of a high-altitude cold area,standard specimens were drilled from the slope of the Jiama copper mine in Tibet,and dynamic compression tests were performed on watersaturated and frozen sandstone with different numbers of F-T cycles(0,10,20,30,and 40)by the split Hopkinson pressure bar(SHPB)system with a cryogenic control system.The influence of water-saturated and frozen conditions on the dynamic performance of sandstone was investigated.The following conclusions are drawn:(1)With increasing strain rate,the attenuation factor(la)of water-saturated sandstone and the intensifying factor(li)of frozen sandstone linearly increase.As the number of F-T cycles increases,the dependence factor(ld)of water-saturated sandstone linearly decreases,whereas the ld of frozen sandstone linearly increases.(2)The prediction equation of the dynamic compressive strength of water-saturated and frozen sandstone is obtained,which can be used to predict the dynamic compressive strength of sandstone after various F-T cycles based on the strain rate.(3)The mesoscopic mechanism of water-saturated and frozen sandstone’s dynamic compressive strength evolution is investigated.The water softening effect causes the dynamic compressive strength of water-saturated sandstone to decrease,whereas the strengthening effect of pore ice causes it to increase.(4)The decrease in the relative dynamic compressive strength of water-saturated sandstone and the increase in the relative dynamic compressive strength of frozen sandstone can be attributed to the increased porosity. 展开更多

目的研究食管上段胃黏膜异位(HGMUE)的内镜检出率、内镜下表现和合并症。方法回顾性研究2019年1月-2020年6月该院消化内镜中心经电子食管胃镜检查且诊断为HGMUE的患者资料。分析HGMUE的检出率、内镜下表现和合并症。结果 51 326例接受... 目的研究食管上段胃黏膜异位(HGMUE)的内镜检出率、内镜下表现和合并症。方法回顾性研究2019年1月-2020年6月该院消化内镜中心经电子食管胃镜检查且诊断为HGMUE的患者资料。分析HGMUE的检出率、内镜下表现和合并症。结果 51 326例接受胃镜检查的患者中,375例诊断为HGMUE,检出率为0.73%。内镜下主要表现为玫瑰色-橘红色的天鹅绒样斑块,与周围食管黏膜界限清楚,可有多个,且大小不一。内镜下合并症包括:内镜阳性的胃食管反流病75例(20.00%),其中反流性食管炎44例(11.73%)、Barrett食管26例(6.93%)、两者合并存在5例(1.13%);糜烂性胃炎56例(14.93%);息肉54例(14.40%),其中胃息肉47例(12.53%)、十二指肠球部息肉7例(1.87%);消化性溃疡42例(11.20%),其中十二指肠球部溃疡21例(5.60%)、胃溃疡17例(4.53%)、两者合并存在4例(1.07%);恶性肿瘤9例(2.40%),其中胃癌3例(0.80%)、食管癌6例(1.60%),无合并症的HGMUE 139例(37.06%)。结论HGMUE合并酸相关性疾病较为常见;合并上消化道恶性肿瘤尤其是食管恶性肿瘤的占比较高,这可能是上消化道肿瘤的预警因子之一,应予以重视。 展开更多

Background:Old-growth forests are irreplaceable with respect to climate change mitigation and have considerable carbon(C)sink potential in soils.However,the relationship between the soil organic carbon(SOC)turnover rate and forest development is poorly understood,which hinders our ability to assess the C sequestration capacity of soil in old-growth forests.Methods:In this study,we evaluated the SOC turnover rate by calculating the isotopic enrichment factor β(defined as the slope of the regression between ^(13)C natural abundance and log-transformed C concentrations)along 0-30 cm soil profiles in three successional forests in subtropical China.A lower β(steeper slope)is associated with a higher turnover rate.The three forests were a 60-year-old P.massoniana forest(PF),a 100-year-old coniferous and broadleaved mixed forest(MF),and a 400-year-old monsoon evergreen broadleaved forest(BF).We also analyzed the soil physicochemical properties in these forests to examine the dynamics of SOC turnover during forest succession and the main regulators.Results:The β value for the upper 30-cm soils in the BF was significantly(p<0.05)higher than that in the PF,in addition to the SOC stock,although there were nonsignificant differences between the BF and MF.The β value was significantly(p<0.05)positively correlated with the soil recalcitrance index,total nitrogen,and available nitrogen contents but was significantly(p<0.01)negatively correlated with soil pH.Conclusions:Our results demonstrate that SOC has lower turnover rates in old-growth forests,accompanied by higher soil chemical recalcitrance,nitrogen status,and lower soil pH.This finding helps to elucidate the mechanism underlying C sequestration in old-growth forest soils,and emphasizes the important value of old-growth forests among global C sinks. 展开更多

Central nerve signal evoked by thoughts can be directly used to control a robot or prosthetic devices without the involvement of the peripheral nerve and muscles.This is a new strategy of human-computer interaction.A method of electroencephalogram(EEG) phase synchronization combined with band energy was proposed to construct a feature vector for pattern recognition of brain-computer interaction based on EEG induced by motor imagery in this paper,rhythm and beta rhythm were first extracted from EEG by band pass filter and then the frequency band energy was calculated by the sliding time window;the instantaneous phase values were obtained using Hilbert transform and then the phase synchronization feature was calculated by the phase locking value(PLV) and the best time interval for extracting the phase synchronization feature was searched by the distribution of the PLV value in the time domain.Finally,discrimination of motor imagery patterns was performed by the support vector machine(SVM).The results showed that the phase synchronization feature more effective in4s-7s and the correct classification rate was 91.4%.Compared with the results achieved by a single EEG feature related to motor imagery,the correct classification rate was improved by 3.5 and4.3 percentage points by combining phase synchronization with band energy.These indicate that the proposed method is effective and it is expected that the study provides a way to improve the performance of the online real-time brain-computer interaction control system based on EEG related to motor imagery. 展开更多

Objective Gestational diabetes mellitus(GDM)and gestational weight gain(GWG)are important risk factors that are known to affect offspring growth,but these outcomes are inconsistent and it remains unknown if both risk factors have a synergetic effect on early childhood growth.The present study aimed to conduct offspring body mass index-for-age Z-scores(BMIZ)trajectories and to evaluate the independent and interactive effect of the status of GDM and excessive GWG on the risks of overweight/obesity from birth to 24 months of age.Methods A total of 7949 mother-child pairs were enrolled in this study.The weight and length of children were measured at birth,6,12,and 24 months of age to calculate BMIZ.Results The status of GDM was positively associated with offspring BMIZ and risk of macrosomia at birth but was not associated with offspring BMIZ or the risks of overweight/obesity at 6,12,and 24 months of age.In contrast,excessive GWG was positively linked to offspring BMIZ,the stable high BMIZ trajectory pattern,and risks of overweight/obesity in the first 24 months of age.These two risk factors also had a significant synergistic effect on macrosomia at birth,but the interactive effect was only significant in boys during the follow-up years in the sex-stratified analyses.Conclusion The maternal GWG was a more pronounced predictor than GDM with relation to BMIZ and risk of overweight/obesity in early childhood.The interactive effect between these risk factors on offspring overweight/obesity may vary by sex. 展开更多

Food,energy and water(FEW)are indispensable,irreplaceable and safeguarding resources with tangled interlinkages among each other,often framed in terms of‘nexus’.Cities have become the bustling hubs of resource consumption and production,facing the most intense stress.Globally,food security,fresh water shortage and fossil energy exhaustion have become the greatest challenge for the sustainable development of cities in the future.Systematic analysis and simulation of dynamic evolution of urban FEW demand and supply and its nexus would add significant and practical value for macro-management such as ensuring the security of urban resource.Taking Beijing as the case study,we established a dynamic model for the FEW demand and supply from the perspective of cross-ectoral and cross-regional nexus with the Stella modelling platform,then it was run for simulating the dynamic changes and nexus characteristics of FEW during the interval between 2016 and 2035.The results showed that:①The gap between local production and demand of these three resources in Beijing will continue to increase.In 2035,Beijing will need 2.16 million tons of grain,0.83 million tons of meat,4.6 billion m3 of water and 91.65 million tons of standard coal of energy while the resources produced and supplied by locality independently will be 0.21 million tons,0.23 million tons,3.3 billion cubic meters and 16.5 million tons of standard coal respectively.Beijing has to draw on cross-boundary production to meet FEW-use with the increasing gap between local supply and demand.②There is a developing trend of the absolute quantity of interaction among FEW.Structurally,the‘outsourcing’feature is obvious and the indigenous nexus links of three resources are shifting to back-end supply chain such as transportation and treatment.③The change of nexus indexes are mainly affected by the mutual and strong dependence between food and water.Food and energy depend more and more on each other;in contrast,the inter-relationship between water and energy is more stable.In addition,inter-regional FEW nexus is strengthened.④To improve systematic management of urban resources,we need to capture multi-tradeoffs of the FEW system comprehensively and establish a multi-regional and multi-factor integrated and collaborative resource management mechanism. 展开更多

Objective:In various cancers,migration and invasion inhibitory protein(MIIP)is expressed at low level and is involved in cancer pathogenesis.Herein,we sought to explore the function of MIIP in clear cell renal cell carcinoma(ccRCC).Methods:CCK-8,colony formation,cell cycle,and endothelial cell tube formation assays were performed to evaluate the roles of MIIP in ccRCC proliferation and angiogenesis.To explore the underlyi ng mechanism,we con ducted RNA-sequencing,GSEA,qRT-PCR,Western blot,ELISA,cell transfection,coimmunoprecipitation,and ubiquitination assays in ccRCC cell lines.Furthermore,xenograft tumor growth in nude mice,and Ki-67 and CD31 staining in xenograft tissues were examined.Finally,the association of MIIP expression with clinical pathology and the expression status of HIF-2a and cysteine-rich 61(CYR61)were further analyzed in human RCC tissues through Western blot and immunohistochemistry.Results:Both in vitro and in vivo functional experiments indicated that forced expression of MIIP inhibited ccRCC proliferation and angiogenesis,whereas silencing MIIP either in normal HK-2 cells or in ccRCC cells had the opposite effect(P<0.05).Mechanistically,CYR61 was identified as a gene significantly downregulated by MIIP overexpression,and was required for the suppressive role of MIIP in ccRCC.MIIP was found to promote HSP90 acetylation and thus impair its chaperone function toward HIF-2a.Consequently,RACK1 binds HIF-2a and causes its ubiquitination and proteasomal degradation,thus decreasing the transcription of its target,CYR61.Finally,analyses of clinical samples demonstrated that MIIP is significantly downregulated in cancer vs.normal tissues in RCC cases,and its expression is negatively associated with histological grade,metastasis,the prognosis of patients with RCC,and the expression of HIF-2a and CYR61(P<0.05).Conclusions:MIIP is a novel tumor suppressor in ccRCC via negative regulation of HIF-2a-CYR61 axis. 展开更多

The solar energetic particle(SEP)event is a kind of hazardous space weather phenomena,so its quantitative forecast is of great importance from the aspect of space environmental situation awareness.We present here a set of SEP forecast tools,which consists of three components:(1)a simple polytropic solar wind model to estimate the background solar wind conditions at the inner boundary of 0.1 AU(about 20 R⊙);(2)an ice-cream-cone model to estimate the erupted coronal mass ejection(CME)parameters;and(3)the improved Particle Acceleration and Transport in the Heliosphere(i PATH)model to calculate particle fluxes and energy spectra.By utilizing the above models,we have simulated six realistic SEP events from 2010 August 14 to 2014 September 10,and compared the simulated results to the Geostationary Operational Environmental Satellites(GOES)spacecraft observations.The results show that the simulated fluxes of>10 Me V particles agree with the observations while the simulated fluxes of>100 Me V particles are higher than the observed data.One of the possible reasons is that we have adopted a simple method in the model to calculate the injection rate of energetic particles.Furthermore,we have conducted the ensemble numerical simulations over these events and investigated the effects of different background solar wind conditions at the inner boundary on SEP events.The results imply that the initial CME density plays an important role in determining the power spectrum,while the effect of varying background solar wind temperature is not significant.Naturally,we have examined the influence of CME initial density on the numerical prediction results for virtual SEP cases with different CME ejection speeds.The result shows that the effect of initial CME density variation is inversely associated with CME speed. 展开更多

Computed tomography(CT)examination is the major measure for detecting and diagnosis of foreign bodies in human body.Although CT has high sensitivity in diagnosis of foreign body,some interference factors may still lead to missed diagnosis or misdiagnosis.Here we report a rare case that a bamboo stick accidentally pierced into the left chest of a young man who was drunk and unware of this hurt.The patient experienced cough,chest pain,fever,hemoptysis,and was misdiagnosed as primary and secondary tuberculosis based on chest CT examinations at a local hospital,although no tubercular bacillus detected by sputum smear.He subsequently received anti-tuberculous treatments in the following three years,but no improvement of his symptoms was observed.Until one month before his death,the bamboo stick was detected by spiral CT examination as well as three-dimensional image reconstruction at another hospital.Postmortem examination revealed pneumonia,pulmonary infarction,and abscess as the causes of his death.We analyze the potential reasons of misdiagnosis in this case,aiming to provide reference for the diagnosis and treatment of pulmonary inflammation associated with foreign body in the future. 展开更多

Recognition of Bragg Wavelength Disturbed by Time Delay of Fiber Length in Prepositive Tunable Filter

The wavelength shift in fiber Bragg grating does not depend directly on the total light levels, losses in the connecting fibers and couplers, or source power. However, if the tunable Fabry-Perot filter is place on the end of incident fiber, the detected time delay of modulation light is occurred due to the unmatch between the scanning time and light transmission time in the transmission fiber. Consequently, the detected peak wavelength shifts with the length of transmission fiber. Thus, the peak wavelength shift effect of Bragg reflective light transmitted in fiber with different fiber length can be obvious in the demodulator with a prepositive tunable Fabry-Perot filter. The experiment indicates the shift rates of 0.109 - 0.126 nm/km increase approximately linearly with the original peak wavelength of 1532.917 - 1560.300 nm at the fiber length of 0 - 6 km. To certify the consistency of measurement data, the criterion correction is introduced. By using the differential method of two fiber Bragg gratings with an optical path, the differential worth is compensated from the disturbance modulated by the time delay of fiber length. 展开更多
