Construction of the panoptosis-related gene model and characterization of tumor microenvironment infiltration in hepatocellular carcinoma

Hepatocellular carcinoma(HCC)is the most common fatal cancer worldwide,patients with HCC have a high mortality rate and poor prognosis.PANoptosis is a novel discovery of programmed cell death associated with cancer development.However,the role of PANoptosis in HCC remains obscure.In this study,we enrolled 274 PANoptosisrelated genes(PANRGs)and screened 8 genes to set up a prognostic model.A previous scoring system calculated PANscore was utilized to quantify the individual risk level of each HCC patient,and the reliability of the prognostic model has been validated in an external cohort.Nomogram constructed with PANscore and clinical characteristics were used to optimize individualized treatment for each patient.Single-cell analysis revealed a PANoptosis model associated with tumor immune cell infiltration,particularly natural killer(NK)cells.Further exploration of hub genes and assessment of the prognostic role of these 4 hub genes in HCC by quantitative real-time PCR(qRT-PCR)and immunohistochemistry(IHC).In conclusion,we evaluated a PANoptosis-based prognostic model as a potential prognostic biomarker for HCC patients.

一种基于异或运算的(k,n)门限秘密共享算法

现有典型的秘密共享算法多基于SHAMIR算法构建,涉及复杂的多项式计算,数据处理速度较慢。为提高计算效率,提出一种仅基于异或运算的秘密共享算法。根据k、n输入值计算待生成的线索个数,通过与随机二进制序列进行连续异或运算生成算法所需的全部线索,并借助排列组合将所有线索进行分组形成影子秘密。实验结果表明,该算法能够实现任意份额(k,n)门限秘密共享,相较于SHAMIR算法处理速度得到大幅提升,且不存在部分秘密信息泄露的安全隐患。

Contralateral needling at unblocked collaterals for hemiplegia following acute ischemic stroke

Hemiplegia caused by stroke indicates dysfunction of the network between the brain and limbs, namely collateral shock in the brain. Contralateral needling is the insertion of needles into acupoints on the relative healthy side of the body to treat diseases such as apoplexy. However, there is little well-designed and controlled clinical evidence for this practice. This study investigated whether contralateral needling could treat hemiplegia after acute ischemic stroke in 106 randomly selected patients with acute ischemic stroke. These patients were randomly assigned to three groups： 45 in the contralateral needling group, receiving acupuncture on the unaffected limbs; 45 in the conven- tional acupuncture group, receiving acupuncture on the hemiplegic limbs; and 16 in the control group, receiving routine treatments without acupuncture. Acupuncture at acupoints Chize （LU5） in the upper limb and Jianliao （TEl4） in the lower limb was performed for 45 minutes daily for 30 consecutive days. The therapeutic effective rate, Neurological Deficit Score, Modified Barthel Index and FugI-Meyer Assessment were evaluated. The therapeutic effective rate of contralateral nee- dling was higher than that of conventional acupuncture （46.67% vs. 31.11%, P 〈 0.05）. The neuro- logical deficit score of contralateral needling was significantly decreased compared with conven- tional acupuncture （P 〈 0.01）. The Modified Barthel Index and FugI-Meyer Assessment score of contralateral needling increased more significantly than those of conventional acupuncture （both P 〈 0.01）. The present findings suggest that contralateral needling unblocks collaterals and might be more effective than conventional acupuncture in the treatment of hemiplegia following acute ischemic stroke.

Idiopathic chronic ulcerative enteritis with perforation and recurrent bleeding: A case report

Idiopathic chronic ulcerative enteritis (ICUE) is a distinct entity without a defined etiology and is rarely seen in the clinic. Patients with ICUE mainly present with insidious abdominal symptoms such as chronic abdominal pain and intermittent gastrointestinal hemorrhage and symptoms of malnourishment in the early stages of the disease. ICUE is always difficult to diagnose. However, as the disease progresses, patients have a variety of acute abdominal complications such as hemorrhage, perforation, or ileus. Surgical intervention is always needed, and the condition can recur and require repeat laparotomy. When diffuse ulceration of the small bowel is present in the absence of recognizable causes, it is classified as nonspecific or idiopathic. The histological examination always demonstrates an acute, chronic inflammatory infiltration without giant cells, granulomas, or villous atrophy. The etiology of ICUE has not been identified, and its pathogenesis is poorly understood; therefore, radical surgical resection is considered the best available treatment. Here, we report a case of ICUE characterized by nonspecific, multiple, small intestinal ulcers resulting in perforation and recurrent bleeding. The differential diagnosis and the treatment are also discussed.

Heat-related illness in Jinshan District of Shanghai:A retrospective analysis of 70 patients

BACKGROUND: This study aimed to investigate the epidemiological and clinical characteristics of patients with heat-related illness, and guide the prevention, diagnosis and treatment of heat-related illness.METHODS: From June 2013 to August 2013, seventy patients with heat-related illness were treated at Jinshan Hospital of Fudan University, and their epidemiological characteristics, laboratory results, treatment and prognosis were retrospectively analyzed.RESULTS: In the 70 patients, 18 patients suffered from heat stroke and 52 patients from nonheat stroke. When the environmnent temperature was above 35 °C, the body temperature of the patients began to increase markedly. The patients with heat stroke were significantly older than those with non-heat stroke(P<0.05). The body temperature, heart rate, blood glucose, blood lactate dehydrogenase and blood creatine kinase in the patients with heat stroke were higher than those in the patients with non-heat stroke(P<0.05). Blood lactate dehydrogenase and blood creatine kinase were positively correlated with body temperature(r=0.801).CONCLUSION: When the environmental temperature goes above 35 °C, heat-related illness should be prevented, especially in the elderly. The body temperature, heart rate, blood glucose, blood lactate dehydrogenase and blood creatine kinase in patients with heat stroke are higher than those in patients with non-heat stroke. Blood lactate dehydrogenase and blood creatine kinase are positively correlated with body temperature, but their relationship with heat-related illness awaits further study.

The pharmacokinetic study of rutin in rat plasma based on an electrochemically reduced graphene oxide modified sensor

An electrochemical method based on a directly electrochemically reduced graphene oxide （ERGO） film coated on a glassy carbon electrode （GCE） was developed for the rapid and convenient determination of rutin in plasma. ERGO was modified on the surface of GCE by one-step electro-deposition method. Electrochemical behavior of rutin on ERGO/GCE indicated that rutin underwent a surface-controlled quasi-reversible process and the electrochemical parameters such as charge transfer coefficient （α）, electron transfer number （n） and electrode reaction standard rate constant （ks） were 0.53, 2 and 3.4 s -1, respectively. The electrochemical sensor for rutin in plasma provided a wide linear response range of 4.70 × 10 ^-7 1.25 × 10^-5 M with the detection limit （s/n=3） of 1.84 × 10^-8 M. The assay was success- fully used to the pharmacokinetic study of rutin. The pharmacokinetic parameters such as elimination rate half-life （t1/2）, area under curve （AUC）, and plasma clearance （CL） were calculated to be 3.345 ± 0.647 rain, 5750 ±656.0 μg min/mL, and 5.891± 0.458 mL/min/kg, respectively. The proposed method utilized a small sample volume of 10 μL and had no complicated sample pretreatment （without deproteinization）, which was simple, eco-friendly, and time- and cost-efficient for rutin pharmacokinetic studies.

Real-time surface electromyography in Parkinson's disease patients during exercise-induced muscle fatigue

To explore the mechanisms underlying exercise-induced local muscle fatigue in patients with idiopathic Parkinson＇s disease （PD）,we used surface electromyography to record myoelectric signals from the tibialis anterior muscle during isometric contraction-induced fatigue until exhaustion.The results revealed no significant differences between patients with idiopathic PD and healthy controls in maximum voluntary contraction of the tibialis anterior muscle.The basic characteristics of surface electromyography were also similar between the two groups.The duration of isometric contraction at 50% maximum voluntary contraction was shortened in PD patients.In addition,PD patients exhibited a stronger increase in mean square amplitude,but a weaker decrease in median frequency and mean power frequency compared with healthy controls during isometric contraction.The skeletal muscles of PD patients revealed specificity of surface electromyography findings,indicating increased fatigability compared with healthy controls.

Chemical Characteristics Combined with Bioactivity for Comprehensive Evaluation of Tumuxiang Based on HPLC-DAD and Multivariate Statistical Methods

Background: The dried roots of Inula helenium L.(IH) and Inula racemosa Hook f.(IR) are used commonly as folk medicine under the name of "tumuxiang(TMX)". Phenolic acid compounds and their derivatives, as main active constituents in IH and IR, exhibit prominent anti-inflammation effect.Objective: To develop a holistic method based on chemical characteristic and anti-inflammation effect for systematically evaluating the quality of twenty-seven TMX samples(including 18 IH samples and 9 IR samples) from different origins.Methods: HPLC fingerprints data of AL(Aucklandia lappa Decne.) whose dried root was similar with HR was added for classification analysis. The HPLC fingerprints of twenty-seven TMX samples and four AL samples were evaluated using hierarchical clustering analysis(HCA) and principle component analysis(PCA). The spectrum-efficacy model between HPLC fingerprints and anti-inflammatory activities was investigated by principal component regression(PCR) and partial least squares(PLS).Results: All samples were successfully divided into three main clusters and peaks 7, 9, 11, 22, 24 and 26 had a primary contribution to classify these medicinal herbs. The results were in accord with the appraisal results of herbs. The spectrum-efficacy relationship results indicated that citric acid, quinic acid, caffeic acid-β-D-glucopyranoside, chlorogenic acid, caffeic acid, 1,3-O-dicaffeoyl quinic acid, tianshic acid and 3β-Hydroxypterondontic acid had main contribution to anti-inflammatory activities.Conclusion: This comprehensive strategy was successfully used for identification of IH, IR and AL, which provided a reliable and adequate theoretical basis for the bioactivity relevant quality standards and studying the material basis of anti-inflammatory effect of TMX.

Dock4 is required for the maintenance of cochlear hair cells and hearing function

Auditory hair cells(HCs)are the mechanosensory receptors of the cochlea,and HC loss or malfunction can result from genetic defects.Dock4,a member of the Dock180-related protein superfamily,is a guanine nucleotide exchange factor for Rac1,and previous reports have shown that Dock4 mutations are associated with autism spectrum disorder,myelodysplastic syndromes,and tumorigenesis.Here,we found that Dock4 is highly expressed in the cochlear HCs of mice.However,the role of Dock4 in the inner ear has not yet been investigated.Taking advantage of the piggyBac transposon system,Dock4 knockdown(KD)mice were established to explore the role of Dock4 in the cochlea.Compared to wild-type controls,Dock4 KD mice showed significant hearing impairment from postnatal day 60.Dock4 KD mice showed hair bundle deficits and increased oxidative stress,which eventually led to HC apoptosis,late-onset HC loss,and progressive hearing loss.Furthermore,molecular mechanism studies showed that Rac1/β-catenin signaling was significantly downregulated in Dock4 KD cochleae and that this was the cause for the disorganized stereocilia and increased oxidative stress in HCs.Overall,our work demonstrates that the Dock4/Rac1/β-catenin signaling pathway plays a critical role in the maintenance of auditory HCs and hearing function.

Impact of the COVID-19 kindergarten closure on overweight and obesity among 3-to 7-year-old children

Background Knowledge on the impact of the temporary kindergarten closure policy under COVID-19 in 2020 on childhood overweight and obesity is inadequate.We aimed to examine differences in rates of overweight and obesity from 2018 to 2021 among kindergarten children aged 3–7 years.Methods Overweight was defined as body mass index(BMI)>1 standard deviation(SD)for age and sex,and obesity was defined as BMI>2 SD for age and sex.Generalized linear mixed modeling was used for analysis.Results A total of 44,884 children and 71,216 growth data points from all 57 public kindergartens in Jiading District,Shanghai,China were analyzed.The rates of obesity from 2018 to 2021 were 6.9%,6.6%,9.5%,and 7.3%in boys and 2.8%,2.8%,4.5%,and 3.1%in girls,respectively.The rates of overweight from 2018 to 2021 were 14.3%,14.3%,18.2%,and 15.3%in boys and 10.6%,10.9%,13.9%,and 11.6%in girls.The rates of obesity and overweight among kindergarten children in 2020 were significantly higher than those in 2018,2019,and 2021.Compared to 2020,the odds ratios of the obesity rate in 2018,2019,and 2021 were 0.67[95%confidence interval(CI)=0.58–0.77,P<0.001],0.72(95%CI=0.64–0.80,P<0.001)and 0.81(95%CI=0.72–0.92,P=0.001),respectively.The odds ratios of the overweight rate in 2018,2019,and 2021 were 0.75(95%CI=0.69–0.82,P<0.001),0.78(95%CI=0.72–0.84,P<0.001),and 0.89(95%CI=0.81–0.97,P=0.008),respectively,compared to 2020.Conclusions The rates of overweight and obesity significantly increased among kindergarten children in 2020 after the 5-month kindergarten closure.It was critical to provide guidance to caregivers on fostering a healthy lifestyle for children at home under public health emergencies.

Hearing impairment constitutes a global public health problem with severe social,political,and economic consequences,and the World Health Organization estimates that about 466 million individuals globally have some degree of hearing loss.Genetic factors account for about half of all deafness cases.Currently,more than 100 genes are known to be involved in hearing loss,but the underlying molecular mechanisms behind these deafness genes have not been fully characterized.The main clinical treatment for deafness is hearing aids and cochlear implants,but their efficacy depends heavily on the quantity and quality of residual hair cells(HCs)and spiral ganglion neurons and has only limited auditory effects.

创业是否加剧了工作-家庭冲突:来自中国时间利用调查的证据

本文从中国创业情境出发,对我国创业者的工作家庭冲突的异质性进行了深入考察。基于2017年中国时间利用调查(CTUS)数据,从时间维度构造表征工作-家庭冲突的代理指标,比较分析创业者与普通劳动者、创业者群体内部的工作家庭冲突的异质性。研究结果表明,创业者发生工作家庭冲突的概率显著高于普通劳动者。与已婚女性普通劳动者相比,已婚女性创业者发生工作家庭冲突的概率显著增加,而已婚男性创业者发生工作家庭冲突的概率与已婚男性普通劳动者之间不存在显著差异。与普通劳动者相比,生存型创业者发生工作家庭冲突的概率不存在显著差异,而机会型创业者发生工作家庭冲突的概率显著增加。特别是,女性机会型创业者发生工作家庭冲突的概率比女性普通劳动者显著增加约14%。本文不但拓展了家庭嵌入视角下的创业研究,而且为创业者的工作与家庭关系的维护提供了建议。

熔融沉积成型加工的结晶性聚合物结构与性能

熔融沉积成型(fused deposition modeling,FDM)作为一种新型的聚合物加工方式,可制备外形复杂、性能可靠、功能集成的聚合物制品,并随着新打印工艺和新材料体系的不断涌现,显示出巨大的应用和发展潜力.但层层堆积的加工方式为FDM成型件引入了薄弱层间结合界面和较大残余内应力,限制了可FDM加工的聚合物材料种类,并制约着FDM成型件的性能和功能.本文结合FDM加工原理分析了可FDM加工的聚合物材料特性,重点关注FDM成型件的微观结构包括多层次聚集态结构、层间结合界面,总结FDM成型件的宏观力学性能及其各向异性,以此构建FDM成型的聚合物结构-性能关系,并展望高性能FDM成型件的发展前景.

电针围刺法治疗急性乳腺炎后乳房包块临床观察（英文）

目的：观察电针围刺法治疗急性乳腺炎后乳房包块的临床疗效。方法：将符合纳入标准30例急性乳腺炎后乳房包块患者给予电针围刺法治疗,取穴：包块周围4至8针、足三里、三阴交、丰隆、乳根,足三里、三阴交、丰隆三对穴接电针仪。每日治疗1次,7次为一疗程,每个疗程间隔3周。下一疗程开始前采用彩色超声复查乳腺,包块消失则停止治疗,未消失则继续治疗,最多治疗3个疗程。结果：经1至3个疗程后,痊愈16例,占53.3%（16/30）;显效8例,占26.7%（8/30）;有效4例,占13.3%（4/30）;无效2例,占6.7%（2/30）。结论：电针围刺法治疗急性乳腺炎后乳房包块临床有效。

Unmanned aerial vehicle swarm mission reliability modeling and evaluation method oriented to systematic and networked mission

With the development of Unmanned Aerial Vehicle(UAV) system autonomy, network communication technology and group intelligence theory, mission execution in the form of a UAV swarm will be an important realization of future applications. Traditional single-UAV mission reliability modeling methods have been unable to meet the requirements of UAV swarm mission reliability modeling. Therefore, the UAV swarm mission reliability modeling and evaluation method is proposed. First, aimed at the interdependence among the multiple layers, a multi-layer network model of a UAV swarm is established. At the same time, based on the system having the following characteristics—using a mission chain to complete the mission and applying the connectivity of the mission network—the mission network model of a UAV swarm is established. Second, vulnerability and connectivity are selected as two indicators to reflect the reliability of the mission, and aimed at random attack and deliberate attack, vulnerability and connectivity evaluation methods are proposed. Finally, the validity and accuracy of the constructed model are verified through simulations,and the model and selected indicators can meet the reliability requirements of the UAV swarm mission. In this way, this study provides quantitative reference for UAV-swarm-related decisionmaking work and supports the development of UAV-swarm-related work.

Effect of Yiqiyangyin recipe on heparanase and nephrin in rats with adriamycin-induced nephropathy

OBJECTIVE: To discuss the mechanism of Yiqiyangyin recipe in rats with adriamycin (ADR)-induced nephropathy. METHODS: We randomly divided 30 Sprague-Dawley rats into 5 groups: control, model, glucocorticoid, Chinese herb, and Chinese herb plus glucocorticoid groups. The 24-h urine volume was collected on days 7, 14, 21, and 28 after ADR injection, and all rats were killed on day 28. We measured the 24-h levels of urinary protein, serum cholesterol, and serum triglycerides, and renal function of all rats by using routine biochemical methods. Pathological changes in the rat kidneys were observed under light and electron microscopes. Heparanase (HPA) mRNA expression levels were measured using re-al-time fluorescence-quantitative polymerase chain reaction. Urine levels of HPA in all groups were measured using enzyme-linked immunosorbent assay. The expression of nephrin was detected by immunohistochemical staining and quantitatively analyzed using Motic image analysis 3.2 software. RESULTS: The levels of urinary protein and serum triglycerides and cholesterol in rats with ADR-induced nephropathy increased on day 14. The serum albumin levels simultaneously decreased. All the changes reached the peak on day 28. Examination under the light microscope showed inflammatory cells and slight fibroplasia in the renal interstitium in the model group, but fewer inflammatory cells were observed in the intervention groups than those in the model group. Examination under the electron microscope showed extensive fusion of foot processes in ADR rats. HPA mRNA expression was higher in the model group than that in the control group. The HPA mRNA levels in the intervention groups, especially in the Chinese herb group, and Chinese herb plus glucocorticoid group were significantly lower than the level in the model group. The HPA expression levels correlated significantly with the proteinuria level. No significant difference was found in the HPA level in urine between the intervention groups and the model group, whereas the model group had a higher urinary HPA level than the control group. Nephrin mRNA expression levels in the model group were higher than those in the control group. Nephrin mRNA expression levels were significantly lower in the intervention groups than that in the model group, especially the Chinese herb plus glucocorticoid group. Compared with the control group, themodel group showed increased nephrin expression in the kidney. Nephrin levels in the other groups, especially in the Chinese herb plus glucocorticoid group, were significantly lower than that in the model group. The nephrin levels in the kidney were negatively correlated with the proteinuria level. CONCLUSION: Yiqiyangyin recipe could attenuate foot process injury particularly in combination with a steroid reduce the development of proteinuria possibly by inhibition of HPA in the kidney, and regulate the expression of nephrin in rats with ADR-induced nephropathy. Our study showed that treatment with Yiqiyangyin recipe plus glucocorticoid was better than a singular intervention, and we explored the pharmacological mechanism of this combination by biochemical and molecular biological analysis to provide a basis for clinical application.
