Objective: The aim of this trial was to compare both the efficacy and the safety of a weekly nanoparticle albumin-bound paclitaxel(nab-paclitaxel) plus cisplatin vs. gemcitabine plus cisplatin in patients with advanced non-small-cell lung cancer(NSCLC).Methods: A total of 84 participants received either 100 mg/m^2 nab-paclitaxel each week on d 1, 8 and 15 of a 28 day cycle, as well as cisplatin 75 mg/m^2 on d 1 every three weeks(nab-TP arm); or gemcitabine 1,000 mg/m^2 on d 1 and 8, plus cisplatin 75 mg/m^2 on d 1 every three weeks(GP arm). The primary end point was progression-free survival(PFS). The secondary end points were overall response rate(ORR) and overall survival(OS).Results: According to our analysis, the median PFS was 4.8 months for the nab-TP arm vs. 5.2 months for the GP arm(P=0.55). Analysis showed the median OS was 14.6 months for participants who were in the nab-TP arm vs. 15.1 months for those in the GP arm(P=0.94). Besides, nab-TP showed OS advantages over GP in patients harboring epidermal growth factor receptor(EGFR) mutation(26.7 vs. 15.3 months, P=0.046) and patients with a performance status of 0(23.5 vs. 14.7 months, P=0.020). It was found that incidences of drug-related grade 3 or 4 toxicities were comparable between the two treatment arms.Conclusions: Therefore, it can be seen that weekly nab-TP treatment has a similar efficacy and tolerability to GP treatment for patients who are undergoing their first-line treatment for NSCLC. It could be that survival differences among platinum doublets in the context of both EGFR mutation and performance status have the potential to be the basis for our further clinical trials. 展开更多

The MGM(1,m) with optimized background value is used to eliminate the random fluctuations or errors of the observational data of all variables, and the combined prediction model together with the multiple linear regression is established in order to improve the simulation and prediction accuracy of the combined model. Finally, a combined model of the MGM(1,2) with optimized background value and the binary linear regression is constructed by an example. Iron is one of the essential trace elements in the human body.It plays an important role in human biology and pathology.Deregulation of iron levels in cells is associated with disease development.In this work,we synthesized a novel near-infrared intramolecular charge transfer(ICT)based ratiometric fluorescent probe to detect Fe^(2+),by using naphthalimide and indole moieties as building blocks.Our work showed that the radiometric probe has excellent selectivity,sensitivity and rapid response.Moreover,we could successfully perform real-time monitoring of Fe^(2+) in He La cells and C.elegans. 展开更多

Bisulfite(HSO_3) is an important sulfur dioxide(SO_2) derivative, which plays a major role in many physiological processes and is also closely associated with a variety of diseases. Thus the development of highly selective and sensitive fluorescent probes is essential to detect HSO_3 in living cells. In this work,we report the synthesis and analysis of a ratiometric fluorescent probe for selective detection of HSO_3 based on the 1,4-nucleophilic addition reaction with the carbazole as an electron donor(D) and aldehyde group as an electron acceptor(A). The addition of HSO_3 and other ions to our probe can be observed by UV vis and fluorescence spectrometry. Our investigation proved that the probe is highly selective and sensitive for HSO_3 and ratiometric changes. Moreover, the probe has good cell permeability and was successfully applied to the detection of exogenous HSO_3 in Hela cells and C. elegans. 展开更多

Contribution of Winter SSTA in the Tropical Eastern Pacific to Changes of Tropical Cyclone Precipitation over Southeast China 被引量:1

Tropical cyclone precipitation(TCP)accounts for 10%-40%of the boreal summer precipitation that occurs over Southeast China(SEC),causing flood disasters and serious damage.On the decadal scale,TCP increases significantly in SEC while TC frequency decreases in the western North Pacific(WNP)during 1980-2019.Therefore,variations in TCP and the corresponding physical mechanism are investigated in this study.First,an empirical statistical method is introduced to quantify the TCP amount based on accumulated cyclone energy(ACE)and TC frequency with the TCP anomaly decomposed into three items(rainfall frequency,rainfall intensity,and nonlinear item).ACE,as the integration of TC intensity and frequency,is a more effective index than TC frequency for depicting the characteristics of TCP because the contribution of rainfall frequency represented by ACE is higher than that of TC frequency.Then,the physical mechanism affecting the WNP TC activities and TCP in SEC are inspected.Positive sea surface temperature anomaly(SSTA)over the tropical eastern Pacific(TEP)in winter can trigger variations of air-sea interaction over the tropical Pacific,including low-level divergent winds,mid-tropospheric descent flows,high-level convergent winds coupled with negative anomalies of vorticity and humidity over the tropical western Pacific(TWP)in the next summer.These dynamic conditions provide unfavorable environments for TC activities in the WNP and constrain TCP in SEC.Furthermore,more significantly negative SSTA events in the TEP facilitate enhanced ACE along with positive relative vorticity,relative humidity,and upwelling vertical winds anomalies over the coast of SEC after 1998,which is a reasonable explanation for the increasing TCP in SEC. 展开更多

Hydrogen sulfide(H_(2) S) is a signaling molecule that plays important roles in biological systems.The exploration of H_(2) S as a new drug release trigger and its related fluorescent theranostic system is crucial for cancer bio-imaging and therapy.Herein,we designed a new two-photon ratiometric fluorescent theranostic prodrug(compound 1) and studied its spectroscopic properties and application in in vivo imaging.Compound 1 specifically reacted with H_(2) S and released the free active therapeutic component of 7-ethyl-10-hydroxycamptothecin,which was accompanied with a red-shift fluorescence emission signal from 460 nm to 545 nm.The exogenous and endogenous H_(2) S in living cells were imaged by compound 1 under one-photon and two-photon excitation.Furthermore,compound 1 monitored the H_(2) S concentration changes in Caenorhabditis elegans by fluorescence imaging.Additionally,it showed effective drug release activation in situ tumor with exogenous and endogenous H_(2) S as the trigger.The H_(2) S-sensitive activation and drug-release properties highlight the potential of theranostic compound 1 in future cancer treatment and therapy. 展开更多
