TiZrTaNb-based high-entropy alloys(HEAs)are research frontier of biomedical materials due to their high hardness,good yield strength,excellent wear resistance and corrosion resistance.Sn,as an essential trace element in the human body that plays a significant role in physiological process.It has stable chemical properties and a low elastic modulus.In this study,a new material,TiZrTaNbSn HEAs,was proposed as a potential biomedical alloy.The Ti_(35)Zr_(25)Ta_(15)Nb_(15)Sn_(10)biomedical high-entropy alloys(BHEAs)were successfully prepared through an arc melting furnace and then remelted using a German high-temperature and high-pressure apparatus under GPa-level(4 GPa and 7 GPa).The precipitation behavior of the needle-like HCP-Zr_(5)Sn_(3)phase that precipitates discontinuously at the grain boundary was successfully controlled.The phase constitution,microstructure,and corrosion resistance of the alloy were studied.The results show that the needle-like HCP-Zr_(5)Sn_(3)phase is eliminated and the(Zr,Sn)-rich nano-precipitated phase is precipitated in the microstructure under high pressure,which leads to the narrowing of grain boundaries and consequently improves the corrosion resistance of the alloy.In addition,the formation mechanisms of(Zr,Sn)-rich nanoprecipitates in BHEAs were discussed.More Zr and Sn dissolve in the matrix due to the effect of high pressure,during the cooling process,they precipitate to form a(Zr,Sn)-rich nano-precipitated phase. 展开更多

BACKGROUND The importance of age on the development of ocular conditions has been reported by numerous studies.Diabetes may have different associations with different stages of ocular conditions,and the duration of diabetes may affect the development of diabetic eye disease.While there is a dose-response relationship between the age at diagnosis of diabetes and the risk of cardiovascular disease and mortality,whether the age at diagnosis of diabetes is associated with incident ocular conditions remains to be explored.It is unclear which types of diabetes are more predictive of ocular conditions.AIM To examine associations between the age of diabetes diagnosis and the incidence of cataract,glaucoma,age-related macular degeneration(AMD),and vision acuity.METHODS Our analysis was using the UK Biobank.The cohort included 8709 diabetic participants and 17418 controls for ocular condition analysis,and 6689 diabetic participants and 13378 controls for vision analysis.Ocular diseases were identified using inpatient records until January 2021.Vision acuity was assessed using a chart.RESULTS During a median follow-up of 11.0 years,3874,665,and 616 new cases of cataract,glaucoma,and AMD,respectively,were identified.A stronger association between diabetes and incident ocular conditions was observed where diabetes was diagnosed at a younger age.Individuals with type 2 diabetes(T2D)diagnosed at<45 years[HR(95%CI):2.71(1.49-4.93)],45-49 years[2.57(1.17-5.65)],50-54 years[1.85(1.13-3.04)],or 50-59 years of age[1.53(1.00-2.34)]had a higher risk of AMD independent of glycated haemoglobin.T2D diagnosed<45 years[HR(95%CI):2.18(1.71-2.79)],45-49 years[1.54(1.19-2.01)],50-54 years[1.60(1.31-1.96)],or 55-59 years of age[1.21(1.02-1.43)]was associated with an increased cataract risk.T2D diagnosed<45 years of age only was associated with an increased risk of glaucoma[HR(95%CI):1.76(1.00-3.12)].HRs(95%CIs)for AMD,cataract,and glaucoma associated with type 1 diabetes(T1D)were 4.12(1.99-8.53),2.95(2.17-4.02),and 2.40(1.09-5.31),respectively.In multivariable-adjusted analysis,individuals with T2D diagnosed<45 years of age[β95%CI:0.025(0.009,0.040)]had a larger increase in LogMAR.Theβ(95%CI)for LogMAR associated with T1D was 0.044(0.014,0.073).CONCLUSION The younger age at the diagnosis of diabetes is associated with a larger relative risk of incident ocular diseases and greater vision loss. 展开更多

AIM:To investigate whether the apoptotic activities of 8-bromo-7-methoxychrysin(BrMC) involve reactive oxygen species(ROS) generation and c-Jun N-terminal kinase(JNK) activation in human hepatocellular carcinoma cells(HCC).METHODS:HepG2,Bel-7402 and L-02 cell lines were cultured in vitro and the apoptotic effects of BrMC were evaluated by flow cytometry(FCM) after propidium iodide(PI) staining,caspase-3 activity using enzymelinked immunosorbent assay(ELISA),and DNA agarose gel electrophoresis.ROS production was evaluated by FCM after dichlorodihydrofluorescein diacetate(DCHFDA) probe labeling.The phosphorylation level of JNK and c-Jun protein was analyzed by Western blotting.RESULTS:FCM after PI staining showed a dose-dependent increase in the percentage of the sub-G1 cell pop-ulation(P < 0.05),reaching 39.0% ± 2.8% of HepG2 cells after 48 h of treatment with BrMC at 10 μmol/L.The potency of BrMC to HepG2 and Bel-7402(32.1% ± 2.6%) cells was found to be more effective than the lead compound,chrysin(16.2% ± 1.6% for HepG2 cells and 11.0% ± 1.3% for Bel-7402 cell) at 40 μmol/L and similar to 5-flurouracil(33.0% ± 2.1% for HepG2 cells and 29.3% ± 2.3% for Bel-7402 cells) at 10 μmol/L.BrMC had little effect on human embryo liver L-02 cells,with the percentage of sub-G1 cell population 5.4% ± 1.8%.Treatment of HepG2 cells with BrMC for 48 h also increased the levels of active caspase-3,in a concentration-dependent manner.z-DEVD-fmk,a caspase-3specific inhibitor,prevented the activation of caspase-3.Treatment with BrMC at 10 μmol/L for 48 h resulted in the formation of a DNA ladder.Treatment of cells with BrMC(10 μmol/L) increased mean fluorescence intensity of DCHF-DA in HepG2 cells from 7.2 ± 1.12 at 0 h to 79.8 ± 3.9 at 3 h and 89.7 ± 4.7 at 6 h.BrMC did not affect ROS generation in L-02 cells.BrMC treatment failed to induce cell death and caspase-3 activation in HepG2 cells pretreated with N-acetylcysteine(10 mmol/L).In addition,in HepG2 cells treated with BrMC(2.5,5.0,10.0 μmol/L) for 12 h,JNK activation was observed.Peak JNK activation occurred at 12 h post-treatment and this activation persisted for up to 24 h.The expression of phosphorylated JNK and c-Jun protein after 12 h with BrMC-treated cells was inhibited by N-acetylcysteine and SP600125 pre-treatment,but GW9662 had no effect.SP600125 substantially reduced BrMC-induced cell death and caspase-3 activation of HepG2 cells.N-acetylcysteine and GW9662 also attenuated induction of cell death and caspase-3 activation in HepG2 cells treated with BrMC.CONCLUSION:BrMC induces apoptosis of HCC cells by ROS generation and sustained JNK activation. 展开更多

The effects of Ni addition and aging treatments on the microstructure and properties of a Cu–3Ti alloy were investigated. The microstructure and precipitation phases were characterized by X-ray diffraction, scanning electron microscopy, and transmission electron microscopy; the hardness, electrical conductivity, and elastic modulus of the resulting alloys were also tested. The results show that Ni addition increases the electrical conductivity and elastic modulus, but decreases the hardness of the aged Cu–3Ti alloy. Within the range of the experimentally investigated parameters, the optimal two-stage aging treatment for the Cu–3Ti–1Ni and Cu–3Ti–5Ni alloy was 300°C for 2 h and 450°C for 7 h. The hardness, electrical conductivity, and elastic modulus of the Cu–3Ti–1Ni alloy were HV 205, 18.2% IACS, and 146 GPa, respectively, whereas the hardness, electrical conductivity, and elastic modulus of the Cu–3Ti–5Ni alloy were HV 187, 31.32% IACS, and 147 GPa, respectively. Microstructural analyses revealed that β′-Ni3 Ti and β′-Cu4 Ti precipitate from the Cu matrix during aging of the Cu–3Ti–5Ni alloy and that some residual Ni Ti phase remains. The increased electrical conductivity is ascribed to the formation of Ni Ti, β′-Ni3 Ti, and β′-Cu4 Ti phases. 展开更多

Objective:To investigate the role of oxidative stress in human renal tubular epithelial cells(HK-2)induced by high glucose and the underlying signal pathway in vitro.Methods:MYPT1,pro-caspase-3,PGC-1α,and Drpl protein expressions were measured by Western blot.MnSOD2,Drp1 and PGC-1αmRNA expressions were detected by real time PCR.Results:Results showed that high glucose significantly up-regulated the protein expressions of MYPT1,pro-caspase-3 and the mRNA expression of MnSOD2 in HK-2 cells;while Rho kinase inhibitor fasudil and ROCK1 siRNA inhibited protein expressions of pro-caspase-3 and the mRNA expression of MnSOD2 in HK-2 cells induced by high glucose.Importantly,fasudil and ROCK1 siRNA markedly inhibited the expressions of mitochondrial motor proteins Drp1 and mitochondrial gene PGC-la in HK-2 cell=s induced by high glucose.Conclusions:Our findings suggest that Rho kinase signal pathway is involved in mitochondrial oxidative damage and apoptosis in high glucose-induced renal tubular epithelial cells by regulating mitochondrial motor proteins Drp1 and mitochondrial gene PGC-1α.Targeting Rho kinase signal pathway might be a potential strategy for the treatment of diabetic nephropathy. 展开更多

METHODS: Western blotting was used to analyze the expression profile of Axl glycoprotein in mouse hepa-tocellular carcinoma cell line Hca-F treated with tunicamycin and PNGase F 3-(4,5)-dimethylthiazol(-zyl)-3,5- diphenyltetrazolium bromide (MTT) assay, extracellular matrix (ECM) invasion assay (in vitro ) and tumor metastasis assay (in vivo ) were utilized to evaluate the effect of Axl deglycosylation on the Hca-F cell proliferation, invasion and lymphatic metastasis. RESULTS: Tunicamycin and PNGase F treatment markedly inhibited Axl glycoprotein synthesis and expression, proliferation, invasion, and lymphatic metastasis both in vitro and in vivo . In the MTT assay, proliferation was apparent in untreated Hca-F cells compared with treated Hca-F cells. In the ECM invasion assay (in vitro ), treated cells passed through the ECMatrix gel in significantly smaller numbers than untreated cells (tunicamycin 5 μg/mL: 68 ± 8 vs 80 ± 9, P=0.0222; 10 μg/mL: 50 ± 6vs 80 ± 9,P=0.0003; 20 μg/mL: 41 ± 4 vs 80 ± 9, P=0.0001); (PNGase F 8 h: 66 ± 7 vs 82 ± 8, P=0.0098; 16 h: 49 ± 4 vs 82 ± 8, P=0.0001; 24 h: 34 ± 3 vs 82 ± 8, P=0.0001). In the tumor metastasis assay (in vivo ), average lymph node weights of the untreated Hca-F group compared with treated Hca-F groups (tunicamycin 5 μg/mL: 0.84 ± 0.21 g vs 0.72 ± 0.19 g, P=0.3237; 10 μg/mL: 0.84 ± 0.21 g vs 0.54 ± 0.11 g, P=0.0113; 20 μg/mL: 0.84 ± 0.21 g vs 0.42 ± 0.06 g, P=0.0008); (PNGase F 8 h: 0.79 ± 0.15 g vs 0.63 ± 0.13 g, P=0.0766; 16 h: 0.79 ± 0.15 g vs 0.49 ± 0.10 g, P=0.0022; 24 h: 0.79 ± 0.15 g vs 0.39 ± 0.05 g, P=0.0001). AIM: To conduct a comparative study of effectiveness of silicone hydrogel contact lens and hydrogel contact lens, which are used in patients after laser-assisted subepithelial keratomileusis(LASEK).·METHODS: Sixty-three patients(121 eyes) with a spherical equivalent ≤-5.0 D were chosen after undergoing LASEK in 2012 at Guangdong General Hospital. They were randomly divided into 2 groups. The silicone hydrogel group included 32 cases(61 eyes) that wore silicone hydrogel contact lenses for 4-6d after the operation, while the hydrogel group included 31 cases(60 eyes) who wore hydrogel contact lenses for 4-6d after the operation. Patients’ self-reported postoperative symptoms(including pain, photophobia, tears, and foreign body sensation) were evaluated. The healing time of the corneal epithelium, the visual acuity of patients without contact lens after epithelial healing, and the incidence of delayed corneal epithelial shedding were also assessed. The follow-up time was 1mo.·RESULTS: Postoperative symptoms were milder in the silicone hydrogel group than in the hydrogel group.There were significant differences in pain, foreign body sensation, and photophobia between the 2 groups(P <0.05),although there was no significant difference in postoperative tearing(P >0.05). The healing time of the corneal epithelium in the silicone hydrogel lens group was markedly shorter than that in the hydrogel group(4.07±0.25 vs 4.33±0.82 d, t =2.43, P =0.02). Visual acuity without contact lenses after healing of the corneal epithelium was better in the silicone hydrogel group compared with the hydrogel group(χ~2=7.76, P =0.02).There was no significant difference in the occurrence ofdelayed corneal epithelial shedding between the 2 groups(P >0.05).· CONCLUSION: Patients with LASEK using silicon hydrogel contact lenses had less discomfort and shorter corneal epithelial healing time compared with those using hydrogel contact lenses, suggesting that silicon hydrogel contact lenses may be considered to be a better choice of bandage contact lens after LASEK. 展开更多

Efficacy and safety of glucocorticoids in the treatment of community-acquired pneumonia: A meta-analysis of randomized controlled trials 被引量:7

BACKGROUND: Community-acquired pneumonia(CAP) is pneumonia acquired infectiously from normal social contact as opposed to being acquired during hospitalization. CAP is a leading cause of illness and death. This review aims to determine the efficacy and safety of glucocorticoids in the treatment of community-acquired pneumonia(CAP).DATA SOURCES: We searched randomized controlled trials(RCTs) from Pubmed, EMBASE, Cochrane Library, Chinese Journal Full-text Database, and Chinese Biomedical Literature Database to obtain the information by using steroids, glucocorticoids, cortisol, corticosteroids, community-acquired pneumonia and CAP as key words. The quality of RCTs was evaluated. A Meta-analysis was made using Rev Man 5.0 provided by the Cochrance Collaboration.RESULTS: Seven RCTs involving 944 patients were included in the meta-analysis. The mean length of hospital stay in glucocorticoids treatment group was significantly shorter than that in standard treatment group(WMD=–1.70, 95%CI 2.01–1.39, Z=10.81, P<0.00001). No statistically significant differences were found in the mortality rate(RR=0.77,95%CI 0.46–1.27, Z=1.03, P=0.30), the mean length of hospital stay in ICU(WMD=1.17, 95%CI 1.68–4.02, Z=0.81, P=0.42), the incidence of super infection(RR=1.32, 95%CI 0.66–2.63, Z=0.79, P=0.43), the incidence of hyperglycemia(RR=1.84, 95%CI 0.76–4.41, Z=1.36, P=0.17), the incidence of upper gastrointestinal bleeding(RR=1.98, 95%CI 0.37–10.59, Z=0.80, P=0.42) between the standard treatment group and the glucocorticoids treatment group.CONCLUSIONS: The use of glucocorticoids in patients with community-acquired pneumonia can significantly shorten the duration of illness and have a favorable safety profile. We collected almost all Highly Processed Data Products (HPDP) of ISO SWS01 spectra for the Galactic visual carbon stars,infrared carbon stars,extreme carbon stars and carbon-rich proto-planetary nebulae (PPNs).Those infrared spectra are primarily analyzed and discussed.It is shown that either spectral shapes/peaks,or main molecular/dust features are evidenced to change in the sequence of visual carbon stars,infrared carbon stars,extreme carbon stars and carbon-rich PPNs.Statistically,in this sequence,continua are gradually changed from blue to red and locations of spectral peaks of continua are also gradually changed from short wavelengths to long wavelengths.In addition,in this sequence,intensities of main molecular/dust features are also gradually changed from prominent in the short wavelengths to prominent in the long wavelengths.Furthermore,from 2MASS and IRAS photometric data,the sequence is also proved.Results in this paper strongly support the previous suggestion for the evolution sequence of carbon-rich objects in our Galaxy,that is the sequence of visual carbon stars → infrared carbon stars → extreme carbon stars → carbon-rich PPNs. 展开更多

We investigate the spectral variations of seven carbon stars in the infrared using ISO SWS spectral data.Continuum variations of those carbon stars show that during the central star pulsations when carbon stars become fainter in the infrared,their ISO SWS spectra become redder and the near-IR temperature(Tnir) decreases.When carbon stars become brighter in the infrared,their ISO SWS spectra become bluer and T nir increases. Furthermore,it is found that the shorter the wavelength of spectral features,such as the 2.48+2.58(C2H2+HCN+CO+C2) ,3.05(C2H2+HCN) and 3.90(C2H2) μm features,is,the better the correlation of their relative integrated fluxes with the fluxes of the continuum is.The changes of the 5.2(C3) ,11.30(SiC) and 13.70(C2H2) μm features do not obviously correlate with the fluxes of the continuum. 展开更多

Objective To observe the anesthetic effect of 2%lidocaine with Pneumatic injector spray in facial beauty therapy.Methods 240 patients were randomly divided into two groups:the observation group(n=120)was treated with 2%lidocaine before the treatment,and the observation group(n=120)was treated with 2%lidocaine.120 cases of control group were treated with compound lidocaine cream before treatment.The onset time of anesthesia,the effect of anesthesia and adverse reactions were recorded.Results In the pneumatic spray group,the effective time of anesthesia was 5 minutes.Acupuncture had no pain,and the incidence of allergy was 0.8%.The control group:the effective time of anesthesia was 20 minutes.Acupuncture has pain in the skin and can be tolerated.The incidence of allergy was 10.8%.There was significant difference between the two groups(χ2=8.82 P<0.01).Conclusion The injection of 2%lidocaine in facial beauty therapy by air pressure spray instrument has the advantages of quick onset,good anaesthesia effect and less side effects,so it is worth popularizing and applying in clinic. 展开更多

Background:Cervical cancer has the fourth highest incidence and mortality rate of all cancers in women worldwide;让seriously harms their physical and mental health.The aim of this study was to observe the roles and preliminary mechanism of Taurine(Tau)-induced apoptosis in cervical cancer cells.Methods:Cells from the human cervical cancer cell line SiHa were transfected with the recombinant plasmid pEGFP-N1-MST1(mammalian sterile 20-like kinase 1);then,the cell proliferation activity was analyzed by the MTT assay,cell apoptosis by flow cytometry,and the related protein levels by Western blotting.Results:Tau inhibited the proliferation of SiHa cells and induced apoptosis in these cells(the apoptotic rate was 21.95%in the Tau 160 mmol/L group and 30%in the Tau 320 mmol/L group),upregulated the expression of the MST1(control,0.53;Tau 40-320 mmol/L groups,0.84-1.45)and Bax(control,0.45;Tau 40-320 mmol/L groups,0.64-1.51)proteins(P<0.01),and downregulated the expression of Bcl-2(control,1.28,Tau 40-320 mmol/L groups,0.93-0.47)(P<0.01).The overexpression of MST1 promoted the apoptosis of SiHa cells,enhanced the apoptosis-inductive effects of Tau(P<0.01),upregulated the expression of the proapoptotic proteins p73,p53,PUMA(p53 upregulated modulator of apoptosis),and caspase-3,and promoted the phosphorylation of YAP(Yes-associated protein).Conclusions:Tau inhibited the proliferation and induced the apoptosis of cervical cancer SiHa cells.The MST1 protein plays an important role in the Tau-induced apoptosis of cervical cancer cells. 展开更多

Background:As of 2 March,2020,at least 80151 coronavirus disease 2019(COVID-19)cases were reported in China.Most of the patients had a history of visiting Hubei Province or contacting with people who had ever stayed in or passed by Hubei Province or were exposed to symptoms.Some patients got infected through only asymptomatic contact.This study aimed to report the epidemic features and lab identification of a patient confirmed with COVID-19 infection through only asymptomatic contact.Case presentation:A 44-year-old man,who lived in Nanchang,Jiangxi Province,China until 6 March 2020,suffered from cough on 27 January 2020.Fever symptoms appeared on 28 January,with a maximum temperature of 38.8℃,accompanied by cough,sore throat,headache,fatigue,muscle ache,joint ache,and other symptoms.The symptoms continued until he was hospitalized on 30 January.Coronavirus conventional polymerase chain reaction assay was positive for the throat swab sample.The patient,along with his wife and son,drove from Nanchang to back to Honghu City,Hubei Province,on 23 January 2020.After staying with his parents and brother’s family for 3 days,the patient drove back to Nanchang and arrived on 25 January.On the way back home,they stopped by Tongshan service area,Hubei Province,without any close contact with other people.After arriving home in Nanchang City,Jiangxi Province,none of them left their residence.In addition,his parents stayed at home for 20 days with his younger brother’s family before they got back.His younger brother and one of his brother’s children visited Wuhan on 5 January and came home on 6 January 2020.Conclusions:This report suggested that,in the early phase of COVID-19 pneumonia,routine screening could miss patients who were virus carriers.Highlighting travel history is of paramount importance for the early detection and isolation of severe acute respiratory syndrome coronavirus 2 cases. 展开更多

Background The interaction of adropin,glucagon-like peptide-2(GLP2),angiopoietin-like protein 4(ANGPTL4),and with childhood obesity and glucose metabolism is inconsistent.This study is to evaluate the association of the three cytokines and glucose homeostasis.Methods This was a cross-sectional study of children with obesity ranging from 5 to 14 years compared to age-and sex-matched children of normal weight.Fasting plasma glucose(FPG),oral glucose tolerance test 2-hour plasma glucose(OGT-T2hPG),and insulin(INS)were measured,and serum adropin,GLP2,and ANGPTL41evels were measured by enzyme-linked immunosorbent assay.The body mass index(BMI),BMI-Z scores,waist-to-hip ratio(WHR),and homeostasis model assessment of insulin resistance(HOMA-IR)were calculated.Results Thirty-nine children(9.70±1.71 years,18 females)with obesity and 29 normal weight children(8.98±1.98 years,16 females)were assessed.The levels of INS,HOMA-IR and GLP2 of the obesity group were significantly higher than the controls(P<0.05).Pearson correlation analysis showed that serum GLP2 was positively associated with WHR,FPG,and OGTT2hPG,and adropin was negatively associated with BMI,BMI-Z,WHR,INS,and HOMA-IR(all P<0.05).Further-more,GLP2 were negatively associated with adropin and ANGPTL4(both P<0.05).By binary logistic regression,adropin and GLP2 were found to be independent markers of obesity.Multiple linear regression showed that GLP2 was associated with OGTT2hPG,and adropin was associated with INS and HOMA-IR(all P<0.05).Conclusions Obese children had elevated GLP2 concentrations,and adropin and GLP2 associated with both childhood obesity and glucose homeostasis.Furthermore,there may be a physiologic interplay between adropin and GLP2 in obese children. 展开更多
