However, the antitumour activity depends on the efficient delivery of si-Stat3. The effects on the growth of mouse prostate cancer cells of si-Stat3 delivered by hydroxyapatite were determined in this study. RM-1 tumour blocks were transplanted into C57BIJ6 mice. CaCl2-modified hydroxyapatite carrying si-Stat3 plasmids were injected into tumours, and tumour growth and histology were determined. The expression levels of Star3, pTyr-Stat3, Bcl-2, Bax, Caspase3, VEGFand cyclin D1 were measured by western blot analysis. Amounts of apoptosis in cancer cells were analysed with immunohistochemistry and the terminal deoxyribonucleotidyl transferase-mediated dUTP-digoxigenin nick end-labelling (TUNEL) assay. The results showed that hydroxyapatite-delivered si-Stat3 significantly suppressed tumour growth up to 74% (P〈0.01). Stat3 expression was dramatically downregulated in the tumours. The immunohistochemistry and TUNEL results showed that si-Stat3-induced apoptosis (up to 42%, P〈0.01). The Stat3 downstream genes Bcl-2, VEGFand cyclin DI were also strongly downregulated in the tumour tissues that also displayed significant increases in Bax expression and Caspase3 activity. These results suggest that hydroxyapatite can be used for the in vivo delivery of plasmid-based siRNAs into tumours.展开更多
Carrot(Daucus carota)is an Apiaceae plant with multi-colored fleshy roots that provides a model system for carotenoid research.In this study,we assembled a 430.40 Mb high-quality gapless genome to the telomere-to-telo...Carrot(Daucus carota)is an Apiaceae plant with multi-colored fleshy roots that provides a model system for carotenoid research.In this study,we assembled a 430.40 Mb high-quality gapless genome to the telomere-to-telomere(T2T)level of“Kurodagosun”carrot.In total,36268 genes were identified and 34961 of them were functionally annotated.The proportion of repeat sequences in the genome was 55.3%,mainly long terminal repeats.Depending on the coverage of the repeats,14 telomeres and 9 centromeric regions on the chromosomes were predicted.A phylogenetic analysis showed that carrots evolved early in the family Apiaceae.Based on the T2T genome,we reconstructed the carotenoid metabolic pathway and identified the structural genes that regulate carotenoid biosynthesis.Among the 65 genes that were screened,9 were newly identified.Additionally,some gene sequences overlapped with transposons,suggesting replication and functional differentiation of carotenoid-related genes during carrot evolution.Given that some gene copies were barely expressed during development,they might be functionally redundant.Comparison of 24 cytochrome P450 genes associated with carotenoid biosynthesis revealed the tandem or proximal duplication resulting in expansion of CYP gene family.These results provided molecular information for carrot carotenoid accumulation and contributed to a new genetic resource.展开更多

BACKGROUND Klebsiella pneumoniae(K.pneumoniae)used to affect mainly people with compromised immunity or weakened by other infections,but recent emergence of hypervirulent strains has increased infections even in healt...BACKGROUND Klebsiella pneumoniae(K.pneumoniae)used to affect mainly people with compromised immunity or weakened by other infections,but recent emergence of hypervirulent strains has increased infections even in healthy individuals.These infections include liver abscess,pneumonia,bacteremia,meningitis,necrotizing fasciitis,and endophthalmitis.Although metastatic infection by hypervirulent K.pneumoniae(hvKP)is increasingly recognized,co-infection with Cryptococcus neoformans(C.neoformans)meningitis in immunocompetent hosts is rare but fatal.So,it is necessary to determine the risk factors,complications,and comorbidity of this disease.CASE SUMMARY This report describes a 58-year-old man with hvKP pulmonary abscess,bacteremia,and meningitis,accompanied by fatal Cryptococcus meningitis.This patient presented with fever for 1 wk and drowsiness for 3 d.Laboratory findings revealed pulmonary abscess and bacteremia of K.pneumoniae.He was given intravenous antibiotic therapy,and the infection was under control for about 1 wk.However,his condition deteriorated rapidly because of metastatic purulent meningitis.Although hvKP and C.neoformans were isolated and confirmed,the patient died of spontaneous respiratory and cardiac arrest caused by cerebral hernia.CONCLUSION HvKP has emerged as a cause of metastatic infections in immunocompetent hosts.polymicrobial co-infections should be taken into consideration when metastatic infection is present.展开更多

In the present study, we aimed to investigate the ex...Objective: Human carbonic anhydrases II (CAII) gene plays an important role in different cancer. However, its relevance to gastric cancer (GC) remains unclear. In the present study, we aimed to investigate the expression of CAII in GC and explore its correlation with some clinicopathologic characteristics of GC. Methods: The expression of CAII in 20 specimens of normal gastric mucosa, 38 specimens of intraepithelial neoplasia and 112 specimens of gastric carcinoma were detected by immunohistochemical techniques. Survival in GC with CAII expression was studied. Results: The positive rate of CAII protein in normal gastric mucosa was significantly higher than that in intraepithelial neoplasia and gastric carcinoma (100% vs. 63.16% and 28.57%, P0.001). The positive rate of CAII protein was significantly higher in gastric carcinoma at early stages than that at advanced stages (70.0% vs. 19.57%, P0.001). The positive rate of CAII protein was significantly lower in gastric carcinoma with lymph node metastases than that without lymph node metastases (10.81% vs. 37.33%, P0.05). Furthermore, the positive rate of CAII protein was significantly lower in poorly-differentiated gastric carcinoma than in moderately- or well-differentiated gastric carcinoma (15.94% vs. 31.03% or 60.00%, P0.05). Moreover, CAII expression was not related with sex, age and tumor size. The patients with CAII-positive tumors showed a better survival rate than those with CAII-negative tumors (P=0.024, log-rank test). Conclusion: CAII expression was related with stages and lymph node metastases in gastric carcinoma. The reduction of CAII expression in GC might promote tumor cell motility and contribute to tumor growth and metastasis.展开更多

Autoantibody workup revealed that the patient waspositive for c-anti-neutrophil cytoplasmic antibodies and proteinase-3 antineutrophilcytoplasmic antibodies. Pulmonary biopsies revealed a localgranulomatous structure. BACKGROUND Acute superior mesenteric venous thrombosis(MVT)is a rare condition associated with a high mortality rate.The treatment strategy for MVT is clinically challenging due to its insidious onset and rapid develo...BACKGROUND Acute superior mesenteric venous thrombosis(MVT)is a rare condition associated with a high mortality rate.The treatment strategy for MVT is clinically challenging due to its insidious onset and rapid development,especially when accompanied by kidney transplantation.CASE SUMMARY Here we present a rare case of acute MVT developed 3 years after renal transplantation.A 49-year-old patient was admitted with acute abdominal pain and diagnosed as MVT with intestinal necrosis.An emergency exploratory laparotomy was performed to remove the infarcted segment of the bowel.Immediate systemic anticoagulation was also initiated.During the treatment,the patient experienced bleeding,anastomotic leakage,and sepsis.However,after aggressive treatment was administered,all thrombi were completely resolved,and the patient recovered with his renal graft function unimpaired.CONCLUSION The present case suggests that accurate diagnosis and timely surgical treatment are important to improve the survival rate of MVT patients.Bleeding with anastomotic fistula needs to be treated with caution because of grafts.Also,previously published cases of mesenteric thrombosis after renal transplantation were reviewed.展开更多
The variation of 90 Laminaria gametophyte clones representing the introduced Laminaria japonica (Group 1) and Laminaria Iongissima (Group 2), the varieties of L. japonica (Group 3) and the varieties derived from...The variation of 90 Laminaria gametophyte clones representing the introduced Laminaria japonica (Group 1) and Laminaria Iongissima (Group 2), the varieties of L. japonica (Group 3) and the varieties derived from interspecific hybrids (Group 4) was determined with 18 microsatellite markers. The allelic diversity and Nei's gene diversity of Group 1 were significantly higher than those of Group 2 (2.9 vs. 1.8 and 0.414 vs. 0.161, respectively), demonstrating that the variation of the introduced L. japonica is richer than that of L. Iongissima. Both allelic diversity and Nei's gene diversity of Group 3 were lower than those of Group 1, indicating that only a portion of variation of L. japonica was incorporated into the varieties of L. japonica. Significant genetic differentiation was detected between four groups and between female (Population 1 ) and male (Population 2) gametophyte clones in each group. The variation among groups accounted for 39.95%, while that among populations accounted for 21.65% of the total. The genetic distance between Group 1 and Group 4 was obviously longer than that between Group 2 and Group 4 (0.686 vs. 0.291), indicating that maternal gametophyte clone contributed more variation to the hybrids than the paternal gametophyte clone did.展开更多
The structural engineering of hydrated ammonium vanadate as a cathode for aqueous Zn-ion batteries has attracted significant research interest because of its ability to suppress vanadium dissolution and accelerate the...The structural engineering of hydrated ammonium vanadate as a cathode for aqueous Zn-ion batteries has attracted significant research interest because of its ability to suppress vanadium dissolution and accelerate the electrochemical dynamics.Herein,a feasible fabrication strategy for oxygen-deficient(NH_(4))_(2)V_(10)O_(25)·xH_(2)O/GO(NVOH@GO)composites was proposed,and the charge storage mechanism was discussed.The results of characterization analysis showed that the introduction of graphene oxide(GO)not only enlarged the layer spacing and improved electrical conductivity,providing spacious channels for Zn^(2+)(de)intercalation and accelerating the ion diffusion dynamics,but also induced more oxygen vacancies,inhibited the dissolution of vanadium,and reduced self-discharging,offering additional and stable active sites for ion storage.The optimized NVOH@GO electrode delivered extraordinarily stable capacities of 334 mAh·g^(-1)after 2000 cycles at 5 A·g^(-1)and 238 mAh·g^(-1)after 10,000cycles at 20 A·g^(-1).Furthermore,ex-situ X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS),and Raman results systematically revealed the electrochemical mechanism,including a phase transition reaction and subsequent Zn^(2+)/H_(2)O co-(de)intercalation process.This study provides an effective strategy for expanding the interlayer spacing,inducing defect engineering,and enhancing the structural stability of vanadium-based cathodes for Zn-ion batteries and other multivalent aqueous ion batteries.展开更多
