目的:探讨翼状胬肉切除联合游离结膜瓣移植术治疗原发性翼状胬肉的疗效及对泪膜功能的影响。方法:选择2018-01/2019-10我院收治的120例126眼原发性翼状胬肉患者作为研究对象,按照随机数字表法分为2组,结膜移植组62例64眼行翼状胬肉切除...目的:探讨翼状胬肉切除联合游离结膜瓣移植术治疗原发性翼状胬肉的疗效及对泪膜功能的影响。方法:选择2018-01/2019-10我院收治的120例126眼原发性翼状胬肉患者作为研究对象,按照随机数字表法分为2组,结膜移植组62例64眼行翼状胬肉切除联合游离结膜瓣移植术治疗,羊膜移植组58例62眼行翼状胬肉切除联合羊膜移植治疗。测定术前,术后3、6mo时泪液分泌长度(SⅠt)、泪膜破裂时间(BUT)、角膜荧光素染色(FL)评分、非侵入性首次泪膜破裂时间(NIBUT)、泪河高度(TMH)、结膜杯状细胞数量,采用眼表疾病指数(OSDI)评估患者生活质量,于术后6mo时评估治疗效果及复发情况。结果:结膜移植组及羊膜移植组患者角膜上皮平均修复时间、症状持续时间比较均无差异(P>0.05)。术后6mo结膜移植组患者BUT(11.11±2.77s vs 10.01±2.41s)、NIBUT(10.01±1.52s vs 9.52±0.98s)高于羊膜移植组(P<0.05),FL评分(0.44±0.10分vs 0.50±0.11分)低于羊膜移植组(P<0.05)。术后两组患者结膜杯状细胞数量均明显增加(P<0.05),术后3、6mo时结膜移植组患者结膜杯状细胞数量多于羊膜移植组(311.41±58.45/mm2vs 285.46±68.96/mm2,342.41±66.89/mm2vs 314.41±70.12/mm2,均P<0.05)。术后两组患者OSDI评分均显著降低(P<0.05),结膜移植组、羊膜移植组患者OSDI评分组间无差异(P>0.05)。两组患者总体疗效无差异(P>0.05);结膜移植组及羊膜移植组患者复发率分别为6.3%、6.5%,组间无差异(P>0.05)。结论:翼状胬肉切除联合自体结膜瓣移植在术后恢复及预防翼状胬肉复发方面与羊膜移植相当,对患者泪膜功能稳定性有更好的改善效果。展开更多

METHODS: The carcinoembryonic ...AIM: To establish a more sensitive method for detection of free cancer cells in peritoneal washes from gastric cancer patients during surgery and to evaluate its clinical significance. METHODS: The carcinoembryonic antigen (CEA) mRNA levels in peritoneal washes from 65 cases of gastric cancer were detected by real-time RT-PCR. Peritoneal lavage cytology (PLC) was applied simultaneously to detection of free cancer cells. Negative controls included peritoneal washes from 5 cases of benign gastric disease and blood samples from 5 adult healthy volunteers. RESULTS: There was no CEA mRNA in peritoneal washes from benign gastric disease patients and in blood of adult healthy volunteers. The positive percentage of free cancer cells detected by real-time RT-PCR was 47.7% and only 22.3% by PLC. The positive rate of CEA mRNA was significantly related with serosa invasion between peritoneal metastasis and stage of gastric cancer. CONCLUSION: Real-time RT-PCR is a sensitive and rapid method for the detection of free cancer cells in peritoneal washes. The presence of free cancer cells in peritoneal washes is related to the Pathologic stage of gastric cancer.展开更多
Singing,as a method of combining respiratory function exercise and vocal intonation therapy,provides a new direction for respiratory function exercise in patients with spinal cord injury.This randomized controlled tri...Singing,as a method of combining respiratory function exercise and vocal intonation therapy,provides a new direction for respiratory function exercise in patients with spinal cord injury.This randomized controlled trial investigated the effects of oral motor respiratory exercise and vocal intonation therapy on respiratory function and vocal quality in patients with spinal cord injury.Among 31 included patients with spinal cord injury,18 completed the treatment.These 18 patients were randomly assigned to undergo music therapy(intervention group,30 min/d,5 times a week,for a total of 12 weeks;n=9,7 males and 2 females;30.33±11.74 years old)or normal respiratory training(control group,n=9;8 males and 1 female;34.78±11.13 years old).Both patient groups received routine treatment concurrently.Before and at 6 and 12 weeks after intervention,a standard respiratory function test,a voice test,the St.George's Respiratory Questionnaire,and a quality of life questionnaire were administered.The results showed that the inspiratory capacity,forced expiratory volume in 1 second,forced vital capacity,maximal mid-expiratory flow rate,sing-loud pressure level,and sustained note length were significantly increased in the intervention group compared with the control group.The St.George's Respiratory Questionnaire and quality of life results of patients in the intervention group were significantly superior to those in the control group.These findings suggest that oral motor respiratory exercise and vocal intonation therapy,as respiratory training methods in music therapy,are effective and valuable for improving respiratory dysfunction and vocal quality in patients with spinal cord injury.This study was approved by the Ethics Committee of China Rehabilitation Research Center(approval No.2019-78-1)on May 27,2019 and was registered with the Chinese Clinical Trial Registry(registration number:Chi CTR1900026922)on October 26,2019.展开更多
The current randomized controlled trial was performed at the China Rehabilitation Science Institute, China to test the hypothesis that musical auditory stimulation has positive effects on the autonomic nervous system ...The current randomized controlled trial was performed at the China Rehabilitation Science Institute, China to test the hypothesis that musical auditory stimulation has positive effects on the autonomic nervous system of patients with disorder of consciousness.Although past studies have recommended that patients with disorder of consciousness listen to patient-preferred music, this practice is not universally accepted by researchers.Twenty patients with severe disorder of consciousness listened to either therapist-selected(n = 10, 6 males and 4 females;43.33 ± 18.76 years old) or patient-preferred(n = 10, 5 males and 5 females, 48.83 ± 18.79 years old) musical therapy, 30 minutes/day, 5 times/week for 6 weeks.The results showed no obvious differences in heart rate variability-related parameters including heart rate, standard deviation of normal-to-normal R-R intervals, and the root-mean-square of successive heartbeat interval differences of successive heartbeat intervals between the two groups of patients.However, percentage of differences exceeding 50 ms between adjacent normal number of intervals, low-frequency power/high-frequency power, high-frequency power norm, low-frequency power norm, and total power were higher in patients receiving therapist-selected music than in patients receiving their own preferred music.In contrast, this relationship was reversed for the high-frequency power and very-low-frequency band.These results suggest that compared with preferred musical stimulation, therapist-selected musical stimulation resulted in higher interactive activity of the autonomic nervous system.Therefore, therapist-selected musical stimulation should be used to arouse the autonomic nervous system of patients with disorder of consciousness.This study was approved by the Institutional Ethics Committee of China Rehabilitation Research Center, China(approval No.2018-022-1) on March 12, 2018 and registered with the Chinese Clinical Trial Registry(registration number Chi CTR1800017809) on August 15, 2018.展开更多
In previous studies,researchers have used singing to treat respiratory function in patients with spinal cord injury.However,few studies have examined the way in which vocal training affects respiratory neural plastici...In previous studies,researchers have used singing to treat respiratory function in patients with spinal cord injury.However,few studies have examined the way in which vocal training affects respiratory neural plasticity in patients with spinal cord injury.Vocal respiratory training(VRT)is a type of vocal muscle-related treatment that is often a component of music therapy(MT)and focuses on strengthening respiratory muscles and improving lung function.In this randomized controlled study,we analyzed the therapeutic effects of VRT on respiratory dysfunction at 3 months after cervical spinal cord injury.Of an initial group of 37 patients,26 completed the music therapy intervention,which comprised five 30-minute sessions per week for 12 weeks.The intervention group(n=13)received VRT training delivered by professional certified music therapists.The control group(n=13)received respiratory physical therapy delivered by professional physical therapists.Compared with the control group,we observed a substantial increase in respiratory function in the intervention group after the 12-week intervention.Further,the nerve fiber bundles in the respiratory center in the medulla exhibited a trend towards increased diversification,with an increased number,path length,thickness,and density of nerve fiber bundles.These findings provide strong evidence for the effect of music therapeutic VRT on neural plasticity.This study was approved by the Ethics Committee of China Rehabilitation Research Center(approval No.2020-013-1)on April 1,2020,and was registered with the Chinese Clinical Trial Registry(registration No.Chi CTR2000037871)on September 2,2020.展开更多
OBJECTIVE Cistanche deserticola Y.C.Ma is an official plant that grows in arid or semi-arid areas.Our early work demonstrated that Cistanche extracts protect against sperm damage in mice under bisphenol A induced repr...OBJECTIVE Cistanche deserticola Y.C.Ma is an official plant that grows in arid or semi-arid areas.Our early work demonstrated that Cistanche extracts protect against sperm damage in mice under bisphenol A induced reproductive damage.Echinacoside(ECH) is one of the major active components.The present study investigated the mechanisms behind the possible protective effects of ECH against oligoasthenospermia in rat and identified the interaction between ECH and AR.METH.ODS The distribution of ECH was assayed by HPLC,the quantity and quality of sperm was evaluated and hormone levels were determined by radioimmunosorbent assay.The levels of androgen receptor(AR) and key steroidogenic-related genes were reduced as determined by Western blotting and qPCR analysis.The interaction between ECH and AR were evaluated by fluorescence localization assay,indirect ELISA and molecular docking.RESULTS ECH significantly increased the quantity of sperm and secretions of luteinizing hormone and testosterone.ECH was distributed to the hypothalamus but not in the testes.ECH combined with hypothalamic AR in the pocket of Met-894 and Val-713 to inhibit transfer of AR from the cytoplasm to nuclei in the hypothalamus.While negative feedback of sex hormone regula.tion was inhibited,positive feedback was stimulated to increase the secretion of luteinizing hormone and testosterone subsequently enhancing the quantity of sperm.C.militaris significantly alleviated the BPA-induced reproductive damage by increasing testicular superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) and glutathione(GSH);as well as by reducing serum malondialdehyde(MDA).C.militaris not only obviously enhanced the levels of serumLH and T,but it also improved the sperm count and motility compared to the BPA-treated group.CONCLUSION C.militaris and ECH protect the BPA induced reproductive damage.ECH blocks AR activity in the hypothalamus to increase the quantity of sperm and protect against oligoasthenospermia in rats.展开更多

This study assessed the effects of the new translocator protein ligand, YL-IPA08, on alleviating inflammation-induced depression-like behavior in mice and investigated its mechanism of action. Mice were intracerebroventricularly injected with 1, 10, 100 or 1000 ng lipopolysaccharide. The tail-suspension test and the forced swimming test confirmed that 100 ng lipopolysaccharide induced depression-like behavior. A mouse model was then established by intraventricular injection of 100 ng lipopolysaccharide. On days 16-24 after model establishment, mice were intragastrically administered 3 mg/kg YL-IPA08 daily. Immunohistochemistry was used to determine BrdU and NeuN expression in the hippocampus. YL-IPA08 effectively reversed the depression-like behavior of lipopolysaccharide-treated mice, restored body mass, increased the number of BrdU-positive cells, and the number and proportion of BrdU and NeuN double-positive cells. These findings indicate that YL-IPA08 can attenuate lipopolysaccharide-induced depression-like behavior in mice by promoting the formation of hippocampal neurons.展开更多

METHODS: The study group consisted of 172 participants with symptoms of AMD who were examined and diagnosed between January 2019 and June 2020. The control group was composed of 120 healthy individuals. Each participant was required to provide two milliliters of peripheral blood for DNA extraction. Two single nucleotide polymorphisms(SNPs) of CFH(rs1061170 and rs800292) and two SNPs of MAP1LC3B(rs8044820 and rs9903) were genotyped. The genotypes and allele frequencies of the SNPs in the study and control groups were further compared using Chi-square and Fisher’s exact tests. RESULTS: In a high-altitude population, the nominally significant differences of rs800292 and rs9903’s genotype AG frequencies were observed in the AMD group(P=0.034 and 0.004, respectively). The frequencies of allele G of rs800292 and allele A of rs9903 were also significantly dif ferent in the AMD group compared to the control [(P=0.034, OR=0.70, 95%CI: 0.50-0.98) and(P=0.004, OR=1.60, 95%CI: 1.15-2.22), respectively]. No significant differences in the genotype distributions(P=0.16 and 0.40, respectively) and allele frequencies(P>0.05) of rs1061170 and rs8044820 were observed in the AMD group.CONCLUSION: Genotype AG of rs800292 may be a protective factor for AMD. Conversely, rs9903 seems to be a risk factor for AMD. Therefore, allele G of rs800292 may be a protective factor, and allele A of rs9903, a risk factor for AMD in Qinghai high-altitude population.展开更多
BACKGROUND Nonfunctional pancreatic neuroendocrine tumours are difficult to diagnose in the early stage of disease due to a lack of clinical symptoms,but they can rarely manifest as autoimmune pancreatitis.Autoimmune ...BACKGROUND Nonfunctional pancreatic neuroendocrine tumours are difficult to diagnose in the early stage of disease due to a lack of clinical symptoms,but they can rarely manifest as autoimmune pancreatitis.Autoimmune pancreatitis is an uncommon disease that may cause recurrent acute pancreatitis and is therefore often regarded as a special type of chronic pancreatitis.CASE SUMMARY We report a case of a 42-year-old female who had nonspecific upper abdominal pain for 4 years and radiological abnormalities of the pancreas that mimicked autoimmune pancreatitis.The symptoms and pancreatic imaging did not improve following 1 year of steroid therapy.Finally,pancreatic biopsy was performed through endoscopic ultrasonography-guided fine-needle aspiration biopsy,and nonfunctional pancreatic neuroendocrine tumours were ultimately diagnosed.Pancreatectomy has resolved her symptoms.CONCLUSION Therefore,the differentiation of nonfunctional pancreatic neuroendocrine tumours from autoimmune pancreatitis is very important,although it is rare.We propose that endoscopic ultrasonography-guided fine-needle aspiration biopsy should be performed if imaging characteristics are equivocal or the diagnosis is in question.展开更多
OBJECTIVE To study the bioactive phytochemicals in the leaves of A.ilicifolius against Hepatocellular carcinoma(HCC) through in silico,in vitro and in vivo studies.METHODS A.ilicifolius leaves were collected from Cudd...OBJECTIVE To study the bioactive phytochemicals in the leaves of A.ilicifolius against Hepatocellular carcinoma(HCC) through in silico,in vitro and in vivo studies.METHODS A.ilicifolius leaves were collected from Cuddalore District,Tamil Nadu,India.Authenticated by the Botanical Survey of India.The fresh leaves of A.ilicifolius were washed and shade dried at room temperature(28 ± 2)℃.The dried leaves were powdered by electrical blender.25 gms of A.ilicifolius leaf powder was used for methanol extraction in the Soxhlet apparatus.The Phytochemical compounds were analyzed by GC-MS and the structure was retrieved from PubChem.Totally,seven HCC target proteins were collected from literature,ligand and proteins were prepared for in silico molecular docking.HepG2 cell lines were used for in vitro(MTT assay).BALB/c mice were used for in vivo studies,the biochemical parameters and histopathological studies were carried out with standard procedure.RESULTS The phytochemical26.27-Di(nor)-cholest-5,7,23-trien-22-ol,3-methoxymethoxy exhibited maximum docking score against the HCC target protein C-Jun N-terminal kinase 1(JNK 1)(-6.839798).MTT assay revealed that the extract at a concentration of 200 μg·mL-1,caused 60% cell death in HepG2 cell lines.Further animal studies using to injecting HCC induced BALB/c mice,restoration of haematological parameters and cells to normal was observed after 15 days of oral administration of the extract.These findings suggest the possibility of using A.ilicifolius leaves in the treatment of HCC.CONCLUSION The in silico,in vitro and in vivo studies indicated that the A.ilicifolius leaves had anticancereous activity against Hepatocellular carcinoma.There can be a possibility of synergistic activity of phytochemicals together against HCC.展开更多
OBJECTIVE Bisphenol A(BPA)is a commonly used phenolic environmental estrogen that impacts male reproductive system.Cistanchetubulosa(CT)is a traditional Chinese medicine used to boost male sexual activity.It has been ...OBJECTIVE Bisphenol A(BPA)is a commonly used phenolic environmental estrogen that impacts male reproductive system.Cistanchetubulosa(CT)is a traditional Chinese medicine used to boost male sexual activity.It has been proved for increasing the expression of steroidogenic enzyme and remarkable antioxidant activity.This study aimed to investigate the mechanisms behind the possible protective effects of CT against BPA-induced testicular and sperm toxicity in rats.METHODS CT was identified by 5.8sgene sequencing.The major compositions(echinacosideand acteoside)of CT were quantified by HPLC method.CT(200or 400mg·kg-1)or VE(positive control,300mg·kg-1)and BPA(200mg·kg-1)were administered orally(one time per day)to adult male ratsfor 6weeks.The sperm parameters were observed by dark-field microscope;serum hormone levels(FSH,LH and T)were tested by radio immunosorbent;LDH-x activity,redox balance status(content of MDA,GSH,H2O2 and OH-,and activity of SOD)were evaluated using commercial kits;the expression of the key steroidogenic enzymes were evaluated by qRT-PCR,heat map,immunofluorescence and Western blotting.RESULTS The CT treatment reversed BPA-induced abnormal in sperm characteristics,testicular structure and LDH-x activity,as well as normalized serum testosterone.CT increased both mRNA and protein expressions of key steroidogenic enzymes including StAR,CYP11A1,3β-HSD,17β-HSD and CYP17A1,suggesting that CT enhanced sex hormone synthesis.Moreover,the remarkable antioxidant effect of CT contributed to its protection against BPA-induced testicular toxicity,which was evidenced by the normalization of testicular,redox markers(MDA,H2O2,OH-and GSH contents,and SOD activity)after CT treatment.CONCLUSION CT effectively attenuated BPA-induced poor sperm quality and testicular toxicity in rats through steroidogenesis and redox pathways.展开更多
BACKGROUND During surgery for gastric cancer,peritoneal lavage using warm distilled water can cause temporary hemodynamic changes.AIM To examine the associations between changes in heart rate and single nucleotide pol...BACKGROUND During surgery for gastric cancer,peritoneal lavage using warm distilled water can cause temporary hemodynamic changes.AIM To examine the associations between changes in heart rate and single nucleotide polymorphisms(SNPs).METHODS This was a prospective observational study of patients with gastric cancer who underwent gastrectomy and peritoneal hypotonic lavage at the Third Afliated Hospital of Soochow University from March 2018 to March 2019.Related SNPs were selected,and the verified exons were analyzed.Heart rate and blood pressure(BP)were measured before and after lavage.The patients were grouped as heart rate change≥30%vs<30%.Comparison and regression analyses of the selected SNPs were performed between the two groups.RESULTS According to the inclusion/exclusion criteria,194 patients were included in the analysis.Of these patients,138 were male,with a mean age of 65.9±0.8 years,and 56 were female,with a mean age of 65.0±1.3 years.Heart rate dropped by 0%-10%in 65 participants,by 10%-15%in 29,by 15%-20%in 23,by 20%-50%in 39,by 50%-100%in four,six had a cardiac arrest,and 28 had an increase in heart rate.Considering the possible impact of exonic SNPs on the phenotypes,TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)were analyzed.The haplotype analysis suggested that the haplotypes CTT[odds ratio(OR)=2.018,95%confidence interval(CI):1.012-4.025,P=0.0430]and GCC(OR=2.293,95%CI:1.174-4.477,P=0.0131)of TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)increased the risk of a drop in heart rate>30%.CONCLUSION The TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)SNPs were associated with changes in heart rate≥30%during intraperitoneal lavage using distilled water after gastrectomy for gastric cancer.展开更多

Patients were grouped according to contact lens wear condition. The corneal endothelium was examined preoperatively and at 1d, 1wk and 1mo postoperatively.The corneal endothelium was analyzed for endothelial cell density(ECD), percentage of hexagonal cells, and coefficient of variation(CV) of cell size.·RESULTS: There were no significant decrease in the ECD, percentage of hexagonal cells or increase in CV at1 d, 1wk and 1mo postoperatively(P 〉0.05). However,there was a small increase of ECD by 2.88% in contact lens wearers(78.26±113.62 cell/mm2, P 〈0.05).· CONCLUSION: SMILE has no significant adverse effects on the corneal ECD and morphology during 1mo follow-up time.展开更多
In recent years,postpartum depression has gained increasing attention around the world due to its high prevalence and severe health consequences.Meanwhile,a considerable number of postpartum depression patients seek h...In recent years,postpartum depression has gained increasing attention around the world due to its high prevalence and severe health consequences.Meanwhile,a considerable number of postpartum depression patients seek help from alternative therapies especially acupuncture,despite the limited evidence available in terms of the effectiveness of acupuncture on postpartum depression.This article summarizes the Chinese and English randomized controlled trials available on acupuncture treatment for postpartum depression over the past ten years and provides a detailed review.10 randomized controlled trials were identified in this review.The treatment regimens described in these studies included acupuncture mono-therapy,electro-acupuncture mono-therapy,or acupuncture combined with other therapies;and the treatment duration ranged from one month to three months.The Hamilton Depression Scale,Edinburgh Postpartum Depression Scale,and estradiol levels in serum were commonly used to assess the efficacy of acupuncture on postpartum depression.The published data suggests that acupuncture,including manual and electrical stimulation,can significantly reduce Hamilton Depression scores and Edinburgh Postpartum Depression scores,and can increase serum estradiol levels compared with different controls.Furthermore,fewer adverse effects were reported following acupuncture treatment than in the control treatment group.Conversely,there have also been some studies reporting inconsistent results in the effectiveness of acupuncture on postpartum depression.Therefore,acupuncture appears to be a potentially effective and safe therapy for postpartum depression,but its outcomes require validation by more rigorously designed studies.展开更多
Accumulating evidence indicates that the alternative splicing program undergoes extensive changes during cancer develop-ment and progression.The RNA-binding protein QKI-5 is frequently downregulated and exhibits anti-...Accumulating evidence indicates that the alternative splicing program undergoes extensive changes during cancer develop-ment and progression.The RNA-binding protein QKI-5 is frequently downregulated and exhibits anti-tumor activity in lung cancer.Howeve-r,little is known about the functional targets and regulatory mechanism of QKI-5.Here,we report that upregulation of exon 14 inclusion of cytoskeletal gene Adducin 3(ADD3)significantly correlates with a poor prognosis in lung cancer.QKI-5 inhibits cell proliferation and migration in part through suppressing the splicing of ADD3 exon 14.Through genome-wide mapping of QKI-5 binding sites in vivo at nucleotide resolution by iCLIP-seq analysis,we found that QKI-5 regulates alternative splicing of its target mRNAs in a binding position-dependent manner.By binding to multiple sites in an upstream intron region,QKI-5 represses the splicing of ADD3 exon 14.We also identified several QKI mutations in tumors,which cause dysregulation of the splicing of QKI targets ADD3 and NUMB.Taken together,our results reveal that QKI-mediated alternative splicing of ADD3 is a key lung cancer-associated splicing event,which underlies in part the tumor suppressor function of QKI.展开更多
Colorectal cancer(CRC)is a common malignant tumor that affects people worldwide.Metagenomic analyses have shown an enrichment of Fusobacterium nucleatum(F.nucleatum)in colorectal carcinoma tissue;many studies have ind...Colorectal cancer(CRC)is a common malignant tumor that affects people worldwide.Metagenomic analyses have shown an enrichment of Fusobacterium nucleatum(F.nucleatum)in colorectal carcinoma tissue;many studies have indicated that F.nucleatum is closely related to the colorectal carcinogenesis.In this review,we provide the latest information to reveal the related molecular mechanisms.The known virulence factors of F.nucleatum promote adhesion to intestinal epithelial cells via FadA and Fap2.Besides,Fap2 also binds to immune cells causing immunosuppression.Furthermore,F.nucleatum recruits tumor-infiltrating immune cells,thus yielding a pro-inflammatory microenvironment,which promotes colorectal neoplasia progression.F.nucleatum was also found to potentiate CRC development through toll-like receptor 2(TLR2)/toll-like receptor 4(TLR4)signaling and microRNA(miRNA)-21 expression.In addition,F.nucleatum increases CRC recurrence along with chemoresistance by mediating a molecular network of miRNA-18a*,miRNA-4802,and autophagy components.Moreover,viable F.nucleatum was detected in mouse xenografts of human primary colorectal adenocarcinomas through successive passages.These findings indicated that an increased number o f F.nucleatum in the tissues is a biomarker for the diagnosis and prognosis of CRC,and the underlying molecular mechanism can probably provide a potential intervention treatment strategy for patients with F.nucleatum-associaied CRC.展开更多

It is involved in the pathogenesis of rheumatoid arthritis (RA). This study aimed to investigate the role oflL1F7 gene polymorphism in RA susceptibility in a large cohort of patients. Methods: Five selected single-nucleotide polynaorphisms in IL 1F7 genes (rs2723186, rs3811046, rs4241122, rs4364030, and rs4392270) were genotyped by TaqMan Allelic Discrimination in Northern Chinese Han population. The allele and the genotype were compared between patients with RA and healthy controls. Association analyses were performed on the entire data set and on different RA subsets based on the status of the anti-cyclic citrullinated peptide antibody and the rheumatoid factor by logistic regression, adjusting for age and gender. Results: Trend associations were detected between rs2723186, rs4241122, rs4392270, and RA in Stage I (160 patients with RA: 252 healthy controls). Further validation in Stage II comprised 730 unrelated patients with RA (mean age: 54.9 ± 12.6 years; 81.6% females) and 778 unrelated healthy individuals (mean age: 53.5 ± 15.7 years; 79.5% females). No significant differences in the distributions of alleles and genotypes were observed between the case and control groups in both the entire set and the different RA subsets. Disease activity and age of RA onset were also not associated with genotype distributions. Conclusion: 1L1F7 gene polymorphism does not significantly influence RA susceptibility in the Northern Chinese Hart population.展开更多
Background:Information about clinical outcomes of very preterm(VPT)infants in tertiary neonatal intensive care unit(NICU)setting is scant in China.This study aimed to investigate the mortality and morbidity of VPT inf...Background:Information about clinical outcomes of very preterm(VPT)infants in tertiary neonatal intensive care unit(NICU)setting is scant in China.This study aimed to investigate the mortality and morbidity of VPT infants admitted to BaYi Children’s Hospital,which serves as a NICU referral center for the city of Beijing,China.Methods:Retrospectively collected perinatal/neonatal data on all admissions of infants born at<32 weeks of gestational age and subsequently admitted to the VPT-NICU from clinical records between October 2010 and September 2011.Results:Totally 729 infants were identified.90%of VPT infants were outborn.The overall survival of the infants to discharge was 92%,which increased with increasing gestational age(range from 69%at<28 weeks to 99%at 31 weeks).The incidence of bronchopulmonary dysplasia was 4%,retinopathy of prematurity requiring treatment 2%,intraventricular hemorrhage III-IV 6%,and periventricular leukomalacia 2%.10%of the VPT infants had a major morbidity at discharge.Conclusions:The outcomes of the VTP infants at this referral NICU were comparable to those in tertiary centers in developed countries.The most common complications were lower than those in other cohorts.Accordingly,high-volume NICU may minimize the adverse effects of VPT infants’transport.展开更多

The aim of this study was to explore whether serum...Background:Rheumatoid arthritis (RA), a systemic autoimmune disease characterized by synovial inflammation, can cause cartilage and bone damage as well as disability. The aim of this study was to explore whether serum glucose-6-phosphate isomerase (GPI) is correlated with disease activity and the value of GPI in the evaluation of infliximab treatment in patients with RA.Methods:Sixty-two patients with RA who had an inadequate response to methotrexate (MTX) were enrolled in Peking University People’s Hospital from July 1, 2016 to July 31, 2018. Infliximab (3 mg/kg, intravenous at weeks 0, 2, and 6 and then every 8 weeks) was administered to patients with stable background MTX therapy. Serum samples were obtained at baseline and week 18. Serum GPI levels were determined using enzyme-linked immunosorbent assay. The associations between serum GPI levels and clinical features were analyzed.Results:Serum GPI was positively correlated with Disease Activity Score in 28 joints (DAS28), swollen joint count, tender joint count and C-reactive protein level ( P < 0.001, P < 0.001, P < 0.001, and P = 0.033, respectively). The change of DAS28 in GPI-positive patients was greater than that in GPI-negative patients ( P < 0.001). Compared with those for patients receiving MTX monotherapy at baseline, the GPI levels were significantly declined when MTX was combined with infliximab ( P < 0.001). Conclusion:Serum GPI is related to disease activity and clinical response to infliximab treatment.展开更多
文摘目的:探讨翼状胬肉切除联合游离结膜瓣移植术治疗原发性翼状胬肉的疗效及对泪膜功能的影响。方法:选择2018-01/2019-10我院收治的120例126眼原发性翼状胬肉患者作为研究对象,按照随机数字表法分为2组,结膜移植组62例64眼行翼状胬肉切除联合游离结膜瓣移植术治疗,羊膜移植组58例62眼行翼状胬肉切除联合羊膜移植治疗。测定术前,术后3、6mo时泪液分泌长度(SⅠt)、泪膜破裂时间(BUT)、角膜荧光素染色(FL)评分、非侵入性首次泪膜破裂时间(NIBUT)、泪河高度(TMH)、结膜杯状细胞数量,采用眼表疾病指数(OSDI)评估患者生活质量,于术后6mo时评估治疗效果及复发情况。结果:结膜移植组及羊膜移植组患者角膜上皮平均修复时间、症状持续时间比较均无差异(P>0.05)。术后6mo结膜移植组患者BUT(11.11±2.77s vs 10.01±2.41s)、NIBUT(10.01±1.52s vs 9.52±0.98s)高于羊膜移植组(P<0.05),FL评分(0.44±0.10分vs 0.50±0.11分)低于羊膜移植组(P<0.05)。术后两组患者结膜杯状细胞数量均明显增加(P<0.05),术后3、6mo时结膜移植组患者结膜杯状细胞数量多于羊膜移植组(311.41±58.45/mm2vs 285.46±68.96/mm2,342.41±66.89/mm2vs 314.41±70.12/mm2,均P<0.05)。术后两组患者OSDI评分均显著降低(P<0.05),结膜移植组、羊膜移植组患者OSDI评分组间无差异(P>0.05)。两组患者总体疗效无差异(P>0.05);结膜移植组及羊膜移植组患者复发率分别为6.3%、6.5%,组间无差异(P>0.05)。结论:翼状胬肉切除联合自体结膜瓣移植在术后恢复及预防翼状胬肉复发方面与羊膜移植相当,对患者泪膜功能稳定性有更好的改善效果。
文摘AIM: To establish a more sensitive method for detection of free cancer cells in peritoneal washes from gastric cancer patients during surgery and to evaluate its clinical significance. METHODS: The carcinoembryonic antigen (CEA) mRNA levels in peritoneal washes from 65 cases of gastric cancer were detected by real-time RT-PCR. Peritoneal lavage cytology (PLC) was applied simultaneously to detection of free cancer cells. Negative controls included peritoneal washes from 5 cases of benign gastric disease and blood samples from 5 adult healthy volunteers. RESULTS: There was no CEA mRNA in peritoneal washes from benign gastric disease patients and in blood of adult healthy volunteers. The positive percentage of free cancer cells detected by real-time RT-PCR was 47.7% and only 22.3% by PLC. The positive rate of CEA mRNA was significantly related with serosa invasion between peritoneal metastasis and stage of gastric cancer. CONCLUSION: Real-time RT-PCR is a sensitive and rapid method for the detection of free cancer cells in peritoneal washes. The presence of free cancer cells in peritoneal washes is related to the Pathologic stage of gastric cancer.

文摘Singing,as a method of combining respiratory function exercise and vocal intonation therapy,provides a new direction for respiratory function exercise in patients with spinal cord injury.This randomized controlled trial investigated the effects of oral motor respiratory exercise and vocal intonation therapy on respiratory function and vocal quality in patients with spinal cord injury.Among 31 included patients with spinal cord injury,18 completed the treatment.These 18 patients were randomly assigned to undergo music therapy(intervention group,30 min/d,5 times a week,for a total of 12 weeks;n=9,7 males and 2 females;30.33±11.74 years old)or normal respiratory training(control group,n=9;8 males and 1 female;34.78±11.13 years old).Both patient groups received routine treatment concurrently.Before and at 6 and 12 weeks after intervention,a standard respiratory function test,a voice test,the St.George's Respiratory Questionnaire,and a quality of life questionnaire were administered.The results showed that the inspiratory capacity,forced expiratory volume in 1 second,forced vital capacity,maximal mid-expiratory flow rate,sing-loud pressure level,and sustained note length were significantly increased in the intervention group compared with the control group.The St.George's Respiratory Questionnaire and quality of life results of patients in the intervention group were significantly superior to those in the control group.These findings suggest that oral motor respiratory exercise and vocal intonation therapy,as respiratory training methods in music therapy,are effective and valuable for improving respiratory dysfunction and vocal quality in patients with spinal cord injury.This study was approved by the Ethics Committee of China Rehabilitation Research Center(approval No.2019-78-1)on May 27,2019 and was registered with the Chinese Clinical Trial Registry(registration number:Chi CTR1900026922)on October 26,2019.

文摘The current randomized controlled trial was performed at the China Rehabilitation Science Institute, China to test the hypothesis that musical auditory stimulation has positive effects on the autonomic nervous system of patients with disorder of consciousness.Although past studies have recommended that patients with disorder of consciousness listen to patient-preferred music, this practice is not universally accepted by researchers.Twenty patients with severe disorder of consciousness listened to either therapist-selected(n = 10, 6 males and 4 females;43.33 ± 18.76 years old) or patient-preferred(n = 10, 5 males and 5 females, 48.83 ± 18.79 years old) musical therapy, 30 minutes/day, 5 times/week for 6 weeks.The results showed no obvious differences in heart rate variability-related parameters including heart rate, standard deviation of normal-to-normal R-R intervals, and the root-mean-square of successive heartbeat interval differences of successive heartbeat intervals between the two groups of patients.However, percentage of differences exceeding 50 ms between adjacent normal number of intervals, low-frequency power/high-frequency power, high-frequency power norm, low-frequency power norm, and total power were higher in patients receiving therapist-selected music than in patients receiving their own preferred music.In contrast, this relationship was reversed for the high-frequency power and very-low-frequency band.These results suggest that compared with preferred musical stimulation, therapist-selected musical stimulation resulted in higher interactive activity of the autonomic nervous system.Therefore, therapist-selected musical stimulation should be used to arouse the autonomic nervous system of patients with disorder of consciousness.This study was approved by the Institutional Ethics Committee of China Rehabilitation Research Center, China(approval No.2018-022-1) on March 12, 2018 and registered with the Chinese Clinical Trial Registry(registration number Chi CTR1800017809) on August 15, 2018.

文摘In previous studies,researchers have used singing to treat respiratory function in patients with spinal cord injury.However,few studies have examined the way in which vocal training affects respiratory neural plasticity in patients with spinal cord injury.Vocal respiratory training(VRT)is a type of vocal muscle-related treatment that is often a component of music therapy(MT)and focuses on strengthening respiratory muscles and improving lung function.In this randomized controlled study,we analyzed the therapeutic effects of VRT on respiratory dysfunction at 3 months after cervical spinal cord injury.Of an initial group of 37 patients,26 completed the music therapy intervention,which comprised five 30-minute sessions per week for 12 weeks.The intervention group(n=13)received VRT training delivered by professional certified music therapists.The control group(n=13)received respiratory physical therapy delivered by professional physical therapists.Compared with the control group,we observed a substantial increase in respiratory function in the intervention group after the 12-week intervention.Further,the nerve fiber bundles in the respiratory center in the medulla exhibited a trend towards increased diversification,with an increased number,path length,thickness,and density of nerve fiber bundles.These findings provide strong evidence for the effect of music therapeutic VRT on neural plasticity.This study was approved by the Ethics Committee of China Rehabilitation Research Center(approval No.2020-013-1)on April 1,2020,and was registered with the Chinese Clinical Trial Registry(registration No.Chi CTR2000037871)on September 2,2020.

文摘OBJECTIVE Cistanche deserticola Y.C.Ma is an official plant that grows in arid or semi-arid areas.Our early work demonstrated that Cistanche extracts protect against sperm damage in mice under bisphenol A induced reproductive damage.Echinacoside(ECH) is one of the major active components.The present study investigated the mechanisms behind the possible protective effects of ECH against oligoasthenospermia in rat and identified the interaction between ECH and AR.METH.ODS The distribution of ECH was assayed by HPLC,the quantity and quality of sperm was evaluated and hormone levels were determined by radioimmunosorbent assay.The levels of androgen receptor(AR) and key steroidogenic-related genes were reduced as determined by Western blotting and qPCR analysis.The interaction between ECH and AR were evaluated by fluorescence localization assay,indirect ELISA and molecular docking.RESULTS ECH significantly increased the quantity of sperm and secretions of luteinizing hormone and testosterone.ECH was distributed to the hypothalamus but not in the testes.ECH combined with hypothalamic AR in the pocket of Met-894 and Val-713 to inhibit transfer of AR from the cytoplasm to nuclei in the hypothalamus.While negative feedback of sex hormone regula.tion was inhibited,positive feedback was stimulated to increase the secretion of luteinizing hormone and testosterone subsequently enhancing the quantity of sperm.C.militaris significantly alleviated the BPA-induced reproductive damage by increasing testicular superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) and glutathione(GSH);as well as by reducing serum malondialdehyde(MDA).C.militaris not only obviously enhanced the levels of serumLH and T,but it also improved the sperm count and motility compared to the BPA-treated group.CONCLUSION C.militaris and ECH protect the BPA induced reproductive damage.ECH blocks AR activity in the hypothalamus to increase the quantity of sperm and protect against oligoasthenospermia in rats.

文摘Translocator protein has received attention for its involvement in the pathogenesis of depression. This study assessed the effects of the new translocator protein ligand, YL-IPA08, on alleviating inflammation-induced depression-like behavior in mice and investigated its mechanism of action. Mice were intracerebroventricularly injected with 1, 10, 100 or 1000 ng lipopolysaccharide. The tail-suspension test and the forced swimming test confirmed that 100 ng lipopolysaccharide induced depression-like behavior. A mouse model was then established by intraventricular injection of 100 ng lipopolysaccharide. On days 16-24 after model establishment, mice were intragastrically administered 3 mg/kg YL-IPA08 daily. Immunohistochemistry was used to determine BrdU and NeuN expression in the hippocampus. YL-IPA08 effectively reversed the depression-like behavior of lipopolysaccharide-treated mice, restored body mass, increased the number of BrdU-positive cells, and the number and proportion of BrdU and NeuN double-positive cells. These findings indicate that YL-IPA08 can attenuate lipopolysaccharide-induced depression-like behavior in mice by promoting the formation of hippocampal neurons.
文摘AIM: To evaluate the association of complement factor H(CFH) and microtubule-associated protein 1 light chain 3 beta(MAP1LC3B) gene polymorphisms with the risk of age-related macular degeneration(AMD) in a high-altitude population. METHODS: The study group consisted of 172 participants with symptoms of AMD who were examined and diagnosed between January 2019 and June 2020. The control group was composed of 120 healthy individuals. Each participant was required to provide two milliliters of peripheral blood for DNA extraction. Two single nucleotide polymorphisms(SNPs) of CFH(rs1061170 and rs800292) and two SNPs of MAP1LC3B(rs8044820 and rs9903) were genotyped. The genotypes and allele frequencies of the SNPs in the study and control groups were further compared using Chi-square and Fisher’s exact tests. RESULTS: In a high-altitude population, the nominally significant differences of rs800292 and rs9903’s genotype AG frequencies were observed in the AMD group(P=0.034 and 0.004, respectively). The frequencies of allele G of rs800292 and allele A of rs9903 were also significantly dif ferent in the AMD group compared to the control [(P=0.034, OR=0.70, 95%CI: 0.50-0.98) and(P=0.004, OR=1.60, 95%CI: 1.15-2.22), respectively]. No significant differences in the genotype distributions(P=0.16 and 0.40, respectively) and allele frequencies(P>0.05) of rs1061170 and rs8044820 were observed in the AMD group.CONCLUSION: Genotype AG of rs800292 may be a protective factor for AMD. Conversely, rs9903 seems to be a risk factor for AMD. Therefore, allele G of rs800292 may be a protective factor, and allele A of rs9903, a risk factor for AMD in Qinghai high-altitude population.

文摘BACKGROUND Nonfunctional pancreatic neuroendocrine tumours are difficult to diagnose in the early stage of disease due to a lack of clinical symptoms,but they can rarely manifest as autoimmune pancreatitis.Autoimmune pancreatitis is an uncommon disease that may cause recurrent acute pancreatitis and is therefore often regarded as a special type of chronic pancreatitis.CASE SUMMARY We report a case of a 42-year-old female who had nonspecific upper abdominal pain for 4 years and radiological abnormalities of the pancreas that mimicked autoimmune pancreatitis.The symptoms and pancreatic imaging did not improve following 1 year of steroid therapy.Finally,pancreatic biopsy was performed through endoscopic ultrasonography-guided fine-needle aspiration biopsy,and nonfunctional pancreatic neuroendocrine tumours were ultimately diagnosed.Pancreatectomy has resolved her symptoms.CONCLUSION Therefore,the differentiation of nonfunctional pancreatic neuroendocrine tumours from autoimmune pancreatitis is very important,although it is rare.We propose that endoscopic ultrasonography-guided fine-needle aspiration biopsy should be performed if imaging characteristics are equivocal or the diagnosis is in question.

文摘OBJECTIVE To study the bioactive phytochemicals in the leaves of A.ilicifolius against Hepatocellular carcinoma(HCC) through in silico,in vitro and in vivo studies.METHODS A.ilicifolius leaves were collected from Cuddalore District,Tamil Nadu,India.Authenticated by the Botanical Survey of India.The fresh leaves of A.ilicifolius were washed and shade dried at room temperature(28 ± 2)℃.The dried leaves were powdered by electrical blender.25 gms of A.ilicifolius leaf powder was used for methanol extraction in the Soxhlet apparatus.The Phytochemical compounds were analyzed by GC-MS and the structure was retrieved from PubChem.Totally,seven HCC target proteins were collected from literature,ligand and proteins were prepared for in silico molecular docking.HepG2 cell lines were used for in vitro(MTT assay).BALB/c mice were used for in vivo studies,the biochemical parameters and histopathological studies were carried out with standard procedure.RESULTS The phytochemical26.27-Di(nor)-cholest-5,7,23-trien-22-ol,3-methoxymethoxy exhibited maximum docking score against the HCC target protein C-Jun N-terminal kinase 1(JNK 1)(-6.839798).MTT assay revealed that the extract at a concentration of 200 μg·mL-1,caused 60% cell death in HepG2 cell lines.Further animal studies using to injecting HCC induced BALB/c mice,restoration of haematological parameters and cells to normal was observed after 15 days of oral administration of the extract.These findings suggest the possibility of using A.ilicifolius leaves in the treatment of HCC.CONCLUSION The in silico,in vitro and in vivo studies indicated that the A.ilicifolius leaves had anticancereous activity against Hepatocellular carcinoma.There can be a possibility of synergistic activity of phytochemicals together against HCC.

文摘OBJECTIVE Bisphenol A(BPA)is a commonly used phenolic environmental estrogen that impacts male reproductive system.Cistanchetubulosa(CT)is a traditional Chinese medicine used to boost male sexual activity.It has been proved for increasing the expression of steroidogenic enzyme and remarkable antioxidant activity.This study aimed to investigate the mechanisms behind the possible protective effects of CT against BPA-induced testicular and sperm toxicity in rats.METHODS CT was identified by 5.8sgene sequencing.The major compositions(echinacosideand acteoside)of CT were quantified by HPLC method.CT(200or 400mg·kg-1)or VE(positive control,300mg·kg-1)and BPA(200mg·kg-1)were administered orally(one time per day)to adult male ratsfor 6weeks.The sperm parameters were observed by dark-field microscope;serum hormone levels(FSH,LH and T)were tested by radio immunosorbent;LDH-x activity,redox balance status(content of MDA,GSH,H2O2 and OH-,and activity of SOD)were evaluated using commercial kits;the expression of the key steroidogenic enzymes were evaluated by qRT-PCR,heat map,immunofluorescence and Western blotting.RESULTS The CT treatment reversed BPA-induced abnormal in sperm characteristics,testicular structure and LDH-x activity,as well as normalized serum testosterone.CT increased both mRNA and protein expressions of key steroidogenic enzymes including StAR,CYP11A1,3β-HSD,17β-HSD and CYP17A1,suggesting that CT enhanced sex hormone synthesis.Moreover,the remarkable antioxidant effect of CT contributed to its protection against BPA-induced testicular toxicity,which was evidenced by the normalization of testicular,redox markers(MDA,H2O2,OH-and GSH contents,and SOD activity)after CT treatment.CONCLUSION CT effectively attenuated BPA-induced poor sperm quality and testicular toxicity in rats through steroidogenesis and redox pathways.
文摘BACKGROUND During surgery for gastric cancer,peritoneal lavage using warm distilled water can cause temporary hemodynamic changes.AIM To examine the associations between changes in heart rate and single nucleotide polymorphisms(SNPs).METHODS This was a prospective observational study of patients with gastric cancer who underwent gastrectomy and peritoneal hypotonic lavage at the Third Afliated Hospital of Soochow University from March 2018 to March 2019.Related SNPs were selected,and the verified exons were analyzed.Heart rate and blood pressure(BP)were measured before and after lavage.The patients were grouped as heart rate change≥30%vs<30%.Comparison and regression analyses of the selected SNPs were performed between the two groups.RESULTS According to the inclusion/exclusion criteria,194 patients were included in the analysis.Of these patients,138 were male,with a mean age of 65.9±0.8 years,and 56 were female,with a mean age of 65.0±1.3 years.Heart rate dropped by 0%-10%in 65 participants,by 10%-15%in 29,by 15%-20%in 23,by 20%-50%in 39,by 50%-100%in four,six had a cardiac arrest,and 28 had an increase in heart rate.Considering the possible impact of exonic SNPs on the phenotypes,TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)were analyzed.The haplotype analysis suggested that the haplotypes CTT[odds ratio(OR)=2.018,95%confidence interval(CI):1.012-4.025,P=0.0430]and GCC(OR=2.293,95%CI:1.174-4.477,P=0.0131)of TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)increased the risk of a drop in heart rate>30%.CONCLUSION The TEP1(rs938886),TEP1(rs1713449),and RECQL5(rs820196)SNPs were associated with changes in heart rate≥30%during intraperitoneal lavage using distilled water after gastrectomy for gastric cancer.

文摘AIM: To assess the effects of small incision lenticule extraction(SMILE) surgery on the corneal endothelium at1 d to 1mo postoperatively.·METHODS: A retrospective, observational study was conducted on 47 patients(47 eyes) who received SMILE surgery. Patients were grouped according to contact lens wear condition. The corneal endothelium was examined preoperatively and at 1d, 1wk and 1mo postoperatively.The corneal endothelium was analyzed for endothelial cell density(ECD), percentage of hexagonal cells, and coefficient of variation(CV) of cell size.·RESULTS: There were no significant decrease in the ECD, percentage of hexagonal cells or increase in CV at1 d, 1wk and 1mo postoperatively(P 〉0.05). However,there was a small increase of ECD by 2.88% in contact lens wearers(78.26±113.62 cell/mm2, P 〈0.05).· CONCLUSION: SMILE has no significant adverse effects on the corneal ECD and morphology during 1mo follow-up time.
文摘In recent years,postpartum depression has gained increasing attention around the world due to its high prevalence and severe health consequences.Meanwhile,a considerable number of postpartum depression patients seek help from alternative therapies especially acupuncture,despite the limited evidence available in terms of the effectiveness of acupuncture on postpartum depression.This article summarizes the Chinese and English randomized controlled trials available on acupuncture treatment for postpartum depression over the past ten years and provides a detailed review.10 randomized controlled trials were identified in this review.The treatment regimens described in these studies included acupuncture mono-therapy,electro-acupuncture mono-therapy,or acupuncture combined with other therapies;and the treatment duration ranged from one month to three months.The Hamilton Depression Scale,Edinburgh Postpartum Depression Scale,and estradiol levels in serum were commonly used to assess the efficacy of acupuncture on postpartum depression.The published data suggests that acupuncture,including manual and electrical stimulation,can significantly reduce Hamilton Depression scores and Edinburgh Postpartum Depression scores,and can increase serum estradiol levels compared with different controls.Furthermore,fewer adverse effects were reported following acupuncture treatment than in the control treatment group.Conversely,there have also been some studies reporting inconsistent results in the effectiveness of acupuncture on postpartum depression.Therefore,acupuncture appears to be a potentially effective and safe therapy for postpartum depression,but its outcomes require validation by more rigorously designed studies.

文摘Accumulating evidence indicates that the alternative splicing program undergoes extensive changes during cancer develop-ment and progression.The RNA-binding protein QKI-5 is frequently downregulated and exhibits anti-tumor activity in lung cancer.Howeve-r,little is known about the functional targets and regulatory mechanism of QKI-5.Here,we report that upregulation of exon 14 inclusion of cytoskeletal gene Adducin 3(ADD3)significantly correlates with a poor prognosis in lung cancer.QKI-5 inhibits cell proliferation and migration in part through suppressing the splicing of ADD3 exon 14.Through genome-wide mapping of QKI-5 binding sites in vivo at nucleotide resolution by iCLIP-seq analysis,we found that QKI-5 regulates alternative splicing of its target mRNAs in a binding position-dependent manner.By binding to multiple sites in an upstream intron region,QKI-5 represses the splicing of ADD3 exon 14.We also identified several QKI mutations in tumors,which cause dysregulation of the splicing of QKI targets ADD3 and NUMB.Taken together,our results reveal that QKI-mediated alternative splicing of ADD3 is a key lung cancer-associated splicing event,which underlies in part the tumor suppressor function of QKI.
文摘Colorectal cancer(CRC)is a common malignant tumor that affects people worldwide.Metagenomic analyses have shown an enrichment of Fusobacterium nucleatum(F.nucleatum)in colorectal carcinoma tissue;many studies have indicated that F.nucleatum is closely related to the colorectal carcinogenesis.In this review,we provide the latest information to reveal the related molecular mechanisms.The known virulence factors of F.nucleatum promote adhesion to intestinal epithelial cells via FadA and Fap2.Besides,Fap2 also binds to immune cells causing immunosuppression.Furthermore,F.nucleatum recruits tumor-infiltrating immune cells,thus yielding a pro-inflammatory microenvironment,which promotes colorectal neoplasia progression.F.nucleatum was also found to potentiate CRC development through toll-like receptor 2(TLR2)/toll-like receptor 4(TLR4)signaling and microRNA(miRNA)-21 expression.In addition,F.nucleatum increases CRC recurrence along with chemoresistance by mediating a molecular network of miRNA-18a*,miRNA-4802,and autophagy components.Moreover,viable F.nucleatum was detected in mouse xenografts of human primary colorectal adenocarcinomas through successive passages.These findings indicated that an increased number o f F.nucleatum in the tissues is a biomarker for the diagnosis and prognosis of CRC,and the underlying molecular mechanism can probably provide a potential intervention treatment strategy for patients with F.nucleatum-associaied CRC.
文摘Background: Interleukin (IL)-37, also called ILl F7, is a natural inhibitor of inflammatory and immune responses. It is involved in the pathogenesis of rheumatoid arthritis (RA). This study aimed to investigate the role oflL1F7 gene polymorphism in RA susceptibility in a large cohort of patients. Methods: Five selected single-nucleotide polynaorphisms in IL 1F7 genes (rs2723186, rs3811046, rs4241122, rs4364030, and rs4392270) were genotyped by TaqMan Allelic Discrimination in Northern Chinese Han population. The allele and the genotype were compared between patients with RA and healthy controls. Association analyses were performed on the entire data set and on different RA subsets based on the status of the anti-cyclic citrullinated peptide antibody and the rheumatoid factor by logistic regression, adjusting for age and gender. Results: Trend associations were detected between rs2723186, rs4241122, rs4392270, and RA in Stage I (160 patients with RA: 252 healthy controls). Further validation in Stage II comprised 730 unrelated patients with RA (mean age: 54.9 ± 12.6 years; 81.6% females) and 778 unrelated healthy individuals (mean age: 53.5 ± 15.7 years; 79.5% females). No significant differences in the distributions of alleles and genotypes were observed between the case and control groups in both the entire set and the different RA subsets. Disease activity and age of RA onset were also not associated with genotype distributions. Conclusion: 1L1F7 gene polymorphism does not significantly influence RA susceptibility in the Northern Chinese Hart population.

文摘Background:Information about clinical outcomes of very preterm(VPT)infants in tertiary neonatal intensive care unit(NICU)setting is scant in China.This study aimed to investigate the mortality and morbidity of VPT infants admitted to BaYi Children’s Hospital,which serves as a NICU referral center for the city of Beijing,China.Methods:Retrospectively collected perinatal/neonatal data on all admissions of infants born at<32 weeks of gestational age and subsequently admitted to the VPT-NICU from clinical records between October 2010 and September 2011.Results:Totally 729 infants were identified.90%of VPT infants were outborn.The overall survival of the infants to discharge was 92%,which increased with increasing gestational age(range from 69%at<28 weeks to 99%at 31 weeks).The incidence of bronchopulmonary dysplasia was 4%,retinopathy of prematurity requiring treatment 2%,intraventricular hemorrhage III-IV 6%,and periventricular leukomalacia 2%.10%of the VPT infants had a major morbidity at discharge.Conclusions:The outcomes of the VTP infants at this referral NICU were comparable to those in tertiary centers in developed countries.The most common complications were lower than those in other cohorts.Accordingly,high-volume NICU may minimize the adverse effects of VPT infants’transport.

文摘Background:Rheumatoid arthritis (RA), a systemic autoimmune disease characterized by synovial inflammation, can cause cartilage and bone damage as well as disability. The aim of this study was to explore whether serum glucose-6-phosphate isomerase (GPI) is correlated with disease activity and the value of GPI in the evaluation of infliximab treatment in patients with RA.Methods:Sixty-two patients with RA who had an inadequate response to methotrexate (MTX) were enrolled in Peking University People’s Hospital from July 1, 2016 to July 31, 2018. Infliximab (3 mg/kg, intravenous at weeks 0, 2, and 6 and then every 8 weeks) was administered to patients with stable background MTX therapy. Serum samples were obtained at baseline and week 18. Serum GPI levels were determined using enzyme-linked immunosorbent assay. The associations between serum GPI levels and clinical features were analyzed.Results:Serum GPI was positively correlated with Disease Activity Score in 28 joints (DAS28), swollen joint count, tender joint count and C-reactive protein level ( P < 0.001, P < 0.001, P < 0.001, and P = 0.033, respectively). The change of DAS28 in GPI-positive patients was greater than that in GPI-negative patients ( P < 0.001). Compared with those for patients receiving MTX monotherapy at baseline, the GPI levels were significantly declined when MTX was combined with infliximab ( P < 0.001). Conclusion:Serum GPI is related to disease activity and clinical response to infliximab treatment.
