This study focuses on the reform and development of physical education in colleges and universities in the post-epidemic era,beginning from the design of an innovative“Internet Plus Swimming”teaching mode to enhance the quality of swimming lessons in colleges and universities,and subsequently applying it to the teaching of technical movements in swimming,so as to promote the teaching reform of swimming lessons in colleges and universities,as well as improve the teaching quality.This study showed that the students in the experimental group were better than those in the control group in four aspects:theoretical knowledge of swimming,speed,technique,and students’independent learning ability.It can be concluded that it is both,effective and feasible to integrate multimedia learning resources,using digital information technology and network mobile devices,to construct the“Internet Plus Swimming”teaching mode and apply it to general swimming courses in colleges and universities in the post-epidemic era. 展开更多

SONG-HD量表评估血液透析患者疲劳程度的应用价值及影响疲劳的相关因素分析

目的探讨使用肾脏病血液透析标准化结局(standardized outcomes in nephrology-hemodialysis,SONG-HD)量表评估维持性血液透析(MHD)患者疲劳程度的应用价值及影响疲劳的相关因素。方法选取2021年4月在厦门大学附属第一医院肾内科血液透... 目的探讨使用肾脏病血液透析标准化结局(standardized outcomes in nephrology-hemodialysis,SONG-HD)量表评估维持性血液透析(MHD)患者疲劳程度的应用价值及影响疲劳的相关因素。方法选取2021年4月在厦门大学附属第一医院肾内科血液透析室规律MHD患者201例,通过使用SONG-HD疲劳测量工具评估疲劳程度。参与者均以面对面访谈问卷进行疲劳评估,分析各种临床和实验室指标,采用单因素及多因素logistic回归分析维持性血液透析患者发生疲劳的影响因素。结果201例MHD患者中,疲劳发生率为73.1%(147/201)。疲劳组超敏C反应蛋白(hs-CRP)、甲状旁腺素(PTH)水平分别为0.91(0.30,3.63)mg/L、216.00(141.00,347.00)ng/L,均显著高于非疲劳组的0.40(0.30,2.01)mg/L、153.00(96.73,308.50)ng/L(Z=2.12、2.17,均P<0.05),而疲劳组透析前二氧化碳结合力、血白蛋白水平分别为(21.03±2.65)mmol/L、(36.76±3.20)g/L,均低于非疲劳组的(22.68±3.01)mmol/L、(38.61±2.85)g/L(t=3.77、3.73,均P<0.05)。单因素logistic回归分析显示,hs-CRP、透析前二氧化碳结合力、血白蛋白及PTH与血液透析患者出现疲乏相关(Wald值分别为4.32、12.39、12.23、4.66,均P<0.05),同时,在调整混杂因素的多变量模型中,血液透析患者疲劳的独立危险因素为较低的透析前二氧化碳结合力及血白蛋白(Wald值分别为12.41、11.67,均P<0.05)。结论维持性血液透析患者疲劳发生率较高。SONG-HD量表评估MHD患者疲劳程度使用方便。在调整混杂因素后,维持性血液透析患者出现疲劳和较低的透析前二氧化碳结合力及血白蛋白之间存在关联。 展开更多

The expression of linear DNA sequence is precisely regulated by the three-dimensional(3D)architecture of chromatin.Morphine-induced aberrant gene networks of neurons have been extensively investigated;however,how morphine impacts the 3D genomic architecture of neurons is still unknown.Here,we applied digestion-ligation-only high-throughput chromosome conformation capture(DLO Hi-C)technology to investigate the effects of morphine on the 3D chromatin architecture of primate cortical neurons.After receiving continuous morphine administration for 90 days on rhesus monkeys,we discovered that morphine re-arranged chromosome territories,with a total of 391 segmented compartments being switched.Morphine altered over half of the detected topologically associated domains(TADs),most of which exhibited a variety of shifts,followed by separating and fusing types.Analysis of the looping events at kilobase-scale resolution revealed that morphine increased not only the number but also the length of differential loops.Moreover,all identified differentially expressed genes from the RNA sequencing data were mapped to the specific TAD boundaries or differential loops,and were further validated for changed expression.Collectively,an altered 3D genomic architecture of cortical neurons may regulate the gene networks associated with morphine effects.Our finding provides critical hubs connecting chromosome spatial organization and gene networks associated with the morphine effects in humans. 展开更多

An Improved Method for Preparation of Uniform and Functional Mitochondria from Fresh Liver

Background and Aims:As the major energy source for mammalian cells,mitochondria have been the subject of numerous studies.However,the isolation and purification of healthy mitochondria,especially from fresh tissue,remains challenging.The most popular methods and kits involve various centrifugation steps which require substantial time and equipment but do not consistently provide pure preparations of functional mitochondria.The aim of this study was to determine whether methods could be devised to improve the purity and yield of functional mitochondria from fresh tissue.Methods:Fresh mouse liver was homogenized,and cells lysed.Particle size analysis,quantitation of mitochondrial DNA,mitochondrial oxygen consumption,and purity of mitochondria(by electron microscopy)were measured in samples after various purification steps and significant differences determined.Results:A two-step procedure consisting of centrifugation followed by filtration through 1.2μand 0.8μfilters resulted in uniform mitochondrial preparations with diameters between 520-540 nm,and approximately 5-times more pure samples.The mitochondria thus obtained had oxygen consumption and sensitivities to mitochondrial inhibitors that were indistinguishable from those purified by centrifugation alone.Electron microscopy confirmed the presence of more uniform and 4-5 times greater concentrations of mitochondria compared to centrifugation alone.Conclusions:A two-step procedure consisting of sequential centrifugation followed by filtration is a rapid method for the production of highly purified,uniform and functional mitochondria. 展开更多

Defects in mitochondria are responsible for various genetic and acquired diseases.Mitochondrial transplantation,a method that involves introduction of healthy donor mitochondria into cells with dysfunctional mitochondria,could offer a novel approach to treat such diseases.Some studies have demonstrated the therapeutic benefit of mitochondrial transplantation and targeted delivery in vivo and in vitro within hepatocytes and the liver.This review discusses the issues regarding isolation and delivery of mitochondria to hepatocytes and the liver,and examines the existing literature in order to elucidate the utility and practicality of mito-chondrial transplantation in the treatment of liver disease.Studies reviewed demonstrate that mitochondrial uptake could specifically target hepatocytes,address the challenge of non-specific localization of donor mitochondria,and provide evidence of changes in liver function following injection of mitochondria into mouse and rat disease models.While potential benefits and advantages of mitochondrial trans-plantation are evident,more research is needed to determine the practicality of mitochondrial transplantation for the treatment of genetic and acquired liver diseases. 展开更多
