circRNA3669 promotes goat endometrial epithelial cells proliferation via miR-26a/RCN2 to activate PI3K/AKT-mTOR and MAPK pathways

The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats. 展开更多

Due to the complex natures of dietary food components,it is difficult to elucidate how the compounds affect host health.Dietary food often selectively presents its mechanism of action on different cell types,and participates in the modulation of targeted cells and their microenvironments within organs.However,the limitations of traditional in vitro assays or in vivo animal experiments cannot comprehensively examine cellular heterogeneity and the tissue-biased influences.Single-cell RNA sequencing(sc RNA-seq)has emerged as an indispensable methodology to decompose tissues into different cell types for the demonstration of transcriptional profiles of individual cells.Sc RNA-seq applications has been summarized on three typical organs(brain,liver,kidney),and two representative immune-and tumor related health problems.The everincreasing role of sc RNA-seq in dietary food research with further improvement can provide sub-cellular information and the coupling between other cellular modalities.In this review,we propose utilizing sc RNAseq to more effectively capture the subtle and complex effects of food chemicals,and how they may lead to health problems at single-cell resolution.This novel technique will be valuable to elucidate the underlying mechanism of both the health benefits of food nutrients and the detrimental consequences food toxicants at the cellular level. 展开更多

Structural damage in heterogeneousmaterials typically originates frommicrostructures where stress concentration occurs.Therefore,evaluating the magnitude and location of localized stress distributions within microstructures under external loading is crucial.Repeating unit cells(RUCs)are commonly used to represent microstructural details and homogenize the effective response of composites.This work develops a machine learning-based micromechanics tool to accurately predict the stress distributions of extracted RUCs.The locally exact homogenization theory efficiently generates the microstructural stresses of RUCs with a wide range of parameters,including volume fraction,fiber/matrix property ratio,fiber shapes,and loading direction.Subsequently,the conditional generative adversarial network(cGAN)is employed and constructed as a surrogate model to establish the statistical correlation between these parameters and the corresponding localized stresses.The stresses predicted by cGAN are validated against the remaining true data not used for training,showing good agreement.This work demonstrates that the cGAN-based micromechanics tool effectively captures the local responses of composite RUCs.It can be used for predicting potential crack initiations starting from microstructures and evaluating the effective behavior of periodic composites. 展开更多

When the traditional drill and blast method is applied to rock crushing projects,it has strong vibration,loud noise and dust pollution,so it cannot be used in densely populated areas such as urban public works.We developed a supercritical CO_(2)true triaxial pneumatic rock-breaking experimental system,and conducted laboratory and field tests of dry ice powder pneumatic rock-breaking.The characteristics of the blast-induced vibration velocity waveform and the evolution of the vibration velocity and frequency with the focal distance were analyzed and discussed.The fracturing mechanism of dry ice powder pneumatic rock breaking is studied.The research results show that:(1)The vibration velocity induced by dry ice powder pneumatic rock breaking decays as a power function with the increase of the focal distance;(2)The vibration frequency caused by dry ice powder pneumatic rock breaking is mainly distributed in 1–120 Hz.Due to the dispersion effect,the dominant frequency of 10–30 Hz appears abnormally attenuated;(3)The traditional CO_(2)phase change fracturing energy calculation formula is also applicable to dry ice pneumatic rock breaking technology,and the trinitrotoluene(TNT)equivalent of fracturing energy is applicable to the Sadovsky formula;(4)Dry ice powder pneumatic rock breaking is shock wave and highenergy gas acting together to fracture rock,which can be divided into three stages,among which the gas wedge action of high-energy gas plays a dominant role in rock mass damage. 展开更多

The state of charge(SOC)estimation of lithium-ion battery is an important function in the battery management system(BMS)of electric vehicles.The long short term memory(LSTM)model can be employed for SOC estimation,which is capable of estimating the future changing states of a nonlinear system.Since the BMS usually works under complicated operating conditions,i.e the real measurement data used for model training may be corrupted by non-Gaussian noise,and thus the performance of the original LSTM with the mean square error(MSE)loss may deteriorate.Therefore,a novel LSTM with mixture kernel mean p-power error(MKMPE)loss,called MKMPE-LSTM,is developed by using the MKMPE loss to replace the MSE as the learning criterion in LSTM framework,which can achieve robust SOC estimation under the measurement data contaminated with non-Gaussian noises(or outliers)because of the MKMPE containing the p-order moments of the error distribution.In addition,a meta-heuristic algorithm,called heap-based-optimizer(HBO),is employed to optimize the hyper-parameters(mainly including learning rate,number of hidden layer neuron and value of p in MKMPE)of the proposed MKMPE-LSTM model to further improve its flexibility and generalization performance,and a novel hybrid model(HBO-MKMPE-LSTM)is established for SOC estimation under non-Gaussian noise cases.Finally,several tests are performed under various cases through a benchmark to evaluate the performance of the proposed HBO-MKMPE-LSTM model,and the results demonstrate that the proposed hybrid method can provide a good robustness and accuracy under different non-Gaussian measurement noises,and the SOC estimation results in terms of mean square error(MSE),root MSE(RMSE),mean absolute relative error(MARE),and determination coefficient R2are less than 0.05%,3%,3%,and above 99.8%at 25℃,respectively. 展开更多

Lignin is a renewable carbon resource to produce arenes due to its abundant aromatic structures.For the liquid-phase hydrodeoxygenation(HDO)based on metallic catalysts,the preservation of aromatic rings in lignin or its derivatives remains a challenge.Herein,we synthesized Mndoped Cu/Al_(2)O_(3) catalysts from layered double hydroxides(LDHs)for liquid-phase HDO of lignin-derived anisole.Mn doping significantly enhanced the selective deoxygenation of anisole to arenes and inhibited the saturated hydrogenation on Cu/Al_(2)O_(3).With Mn doping increasing,the surface of Cu particles was modified with MnO_(x) along with enhanced generation of oxygen vacancies(Ov).The evolution of active sites structure led to a controllable adsorption geometry of anisole,which was beneficial for increasing arenes selectivity.As a result,the arenes selectivity obtained on 4Cu/8Mn4AlO_(x) was increased to be more than 6 folds of that value on 4Cu/4Al_(2)O_(3) over the synergistic sites between metal Cu and Ov generated on MnO_(x). 展开更多

The auxin receptor(TIR1/AFBs)family encodes the F-box protein subunit,which is involved in the formation of the E3 ubiquitin ligase SCFTIR1/AFBs complex,a key component of the auxin signaling pathway.However,there are few studies on the auxin receptor family in apple(Malus×domestica).In this study,eight MdAFBs were identified,and phylogenetic analysis showed that they were classified into four groups and distributed on eight chromosomes.Herein,a comprehensive analysis of the MdAFB gene family was conducted to identify cis-acting elements,gene structures,protein structures,aligned sequences,conserved motifs,conserved amino acids,and the protein–protein interaction network.The results of yeast two-hybrid assays showed that MdAFB1 interacted with three auxin repressor proteins.The results of qRT-PCR showed that MdAFB1 responded to osmotic and salt stress.The overexpression of MdAFB1 increased osmotic and salt resistance in apple calli,and the ectopic expression of MdAFB1 enhanced osmotic and salt tolerance in Arabidopsis.This study provided a basis for the identification of auxin receptor genes in apple and their functions in mediating osmotic and salt stress. 展开更多

Aggregation-induced thermally activated delayed fluorescence(TADF)phenomena have attracted extensive attention recently.In this paper,several theoretical models including monomer,dimer,and complex are used for the explanation of the luminescent properties of(R)-5-(9H-carbazol-9-yl)-2-(1,2,3,4-tetrahydronaphthalen-1-yl)isoindoline-1,3-dione((R)-ImNCz),which was recently reported[Chemical Engineering Journal 418129167(2021)].The polarizable continuum model(PCM)and the combined quantum mechanics and molecular mechanics(QM/MM)method are adopted in simulation of the property of the molecule in the gas phase,solvated in acetonitrile and in aggregation states.It is found that large spin–orbit coupling(SOC)constants and a smaller energy gap between the first singlet excited state and the first triplet excited state(△E_(st))in prism-like single crystals(SC_(p)-form)are responsible for the TADF of(R)-lmNCz,while no TADF is found in block-like single crystals(SC_(b)-form)with a larger △E_(st).The multiple ultralong phosphorescence(UOP)peaks in the spectrum are of complex origins,and they are related not only to ImNCz but also to a minor amount of impurities(ImNBd)in the crystal prepared in the laboratory.The dimer has similar phosphorescence emission wavelengths to the(R)-lmNCz-SC_(p) monomers.The complex composed of(R)-lmNCz and(R)-lmNBd contributes to the phosphorescent emission peak at about 600 nm,and the phosphorescent emission peak at about 650 nm is generated by(R)-lmNBd.This indicates that the impurity could also contribute to emission in molecular crystals.The present calculations clarify the relationship between the molecular aggregation and the light-emitting properties of the TADF emitters and will therefore be helpful for the design of potentially more useful TADF emitters. 展开更多

A PGPR strain AMCC100017 of Brevibacillus laterosporus synthesizes the plant hormone IAA in a tryptophan-dependent manner. In this study, the AMCC100017 strain was used to treat Malus robusta, an excellent natural rootstock for apple production, and assess its ability to promote growth. The fresh weight, dry weight, plant height, and lateral root growth of M. robusta were significantly increased with treatment. The presence of the AMCC100017 strain increased IAA content in M. robusta and promoted root secretion of tryptophan. Colonization of the strain in the roots allowed continuous synthesis of IAA and promoted plant growth. In addition, the photosynthetic efficiency in leaves increased after microbial treatment, and the utilization of nitrogen, zinc, iron, copper and magnesium in leaves was increased, which was conducive to photosynthesis.Interestingly, the activities of CAT and SOD, as well as the contents of ROS in plants colonized by AMCC100017 were increased compared to control plants, but the activities of POD and MDA contents were decreased. AMCC100017 strain affected the antioxidant capacity and stress resistance of plants. AMCC100017 strain increased the content of soluble protein and soluble sugar in plants, improved plant metabolic activity and stress resistance. Therefore, AMCC100017 not only increased IAA content and photosynthetic efficiency to promote M. robusta growth, but also affected plant through multiple metabolic pathways. 展开更多

In the present study, stem sap flow characteristics and responses to ambient meteo- rological factors of three tree species, Albizzia kalkora (native), Azadirachta indica (exotic), and Acacia auriculaeformis (exotic), in a dry-hot valley (Yuanmou, Yunnan Province, China) were investigated using thermal dissipation probes. The diurnal dynamics of sap flow in three studied species displayed an obvious circadian rhythm during the wet and dry seasons, with the exception of A. indica during the dry season. The sap flow velocity (SFV) in A. kalkora and A. auriculaeformis was significantly positively correlated with photosynthetically active radiation (PAR), air temperature, vapour pressure deficit (VPD) and wind speed, but negatively correlated with atmospheric relative humidity over the two seasons. The cross-corre- lation analysis also revealed that the SFV of the three species was significantly correlated with PAR and VPD (P 〈 0.001). Additionally, stem sap flow lagged behind PAR but ahead of VPD, and the diurnal sap flow was more dependent on PAR than on VPD. However, we found that the dominant climatic factor influencing the stem sap flow differed between daytime and nighttime. PAR was more influential than other meteorological factors during the daytime, while VPD or other factors were more influential overnight. When the nighttime refilling ability of the three tree species was compared, our results suggest that A. indica has higher drought resistance and better for afforestation of the studied region. 展开更多

Objective:Death receptor 4(DR4;TRAIL-R1)critically mediates extrinsic apoptosis cascades via binding to TNF-related apoptosis-inducing ligand(TRAIL).However,intrinsic and/or acquired resistance are observed in the clinical application of TRAIL.The aim of this study was to investigate the function and molecular mechanism of CD13 in the TRAIL/DR4 pathway against tumor cells,and provide a new strategy for improving therapeutic efficacy or overcoming TRAIL-resistance.Methods:TRAIL protein was expressed as a secretory protein in a Pichia pastoris expression system and was isolated and purified by affinity chromatography.The cell viability and apoptosis were evaluated with MTT(thiazolyl blue tetrazolium bromide)assays and annexin V-FITC/PI staining with flow cytometry analysis,respectively.Western blot analysis was used to detect the levels of the indicated proteins in tumor cells.DR4 degradation or stability was examined with cycloheximide chase assays,and cell surface DR4 was assessed with flow cytometric analysis after staining with a FITC-conjugated antibody.The effects of cell migration were determined with Transwell and gelatin zymography assays.A xenograft nude mouse model was used to detect the anti-tumor effect in vivo,and the proliferation in tumor tissues was examined with immunohistochemical staining.Results:CD13 inhibition potently sensitized tumor cells to TRAIL-induced killing,including proliferation inhibition,increased apoptosis,and migration suppression.In addition,the inhibition of CD13 elevated both total cellular expression and cell surface DR4 through stabilizing DR4 by suppressing its degradation.DR4 si RNA attenuated the enhanced anti-tumor effects of TRAIL plus CD13 inhibition.Interestingly,these phenomena were p-ERK1/2 independent,although p-ERK1/2 down-regulation was tightly correlated with the cooperation of TRAIL and CD13 inhibition.Moreover,a synergistic decrease in tumor growth was surprisingly achieved in the xenograft model by treatment of TRAIL with a CD13 inhibitor(**P<0.01,CDI=0.47).Conclusions:CD13 inhibition cooperates with TRAIL in enhancing DR4-mediated cell death,through the up-regulation and stabilization of DR4 in a p-ERK1/2-independent manner.Thus CD13 inhibition has emerged as an effective strategy for TRAIL/DR4-based therapy. 展开更多
