A Potential Role for GLUT4 in Predicting Sepsis in Critically Ill Children

Background: This study investigated serum Glucose transporter (GLUT) 4 levels and examined the relationship between serum GLUT4 levels and sepsis in critically ill children. Methods: This was a retrospective study of 77 critically ill children and 33 non-diabetic healthy children (controls) who were admitted between 07/2015 and 05/2016. Patient data, clinical information, and blood samples were collected on admission, alongside a large number of laboratory parameters that were routinely assessed. Critically ill patients were divided into sepsis and non-sepsis/systemic inflammatory response syndrome (SIRS). Serum GLUT4 was measured using western blotting and enzyme-linked immunosorbent assays. Insulin resistance indexes, clinical data, laboratory parameters, and inflammatory cytokines were assessed. Results: GLUT4 serum levels were higher in critically ill children than in healthy children (90.5 vs. 30.3 μg/L, P 0.05). Compared to healthy children, hyperglycemic patients (n = 48) had elevated GLUT4 serum levels (30.3 vs. 103.7 g/L, P Conclusions: GLUT4 serum levels might be significantly increased in critically ill children compared with healthy children, particularly those in septic shock. Peanut(Arachis hypogaea L.)is a globally important oil crop.Web blotch is one of the most important foliar diseases affecting peanut,which results in serious yield losses worldwide.Breeding web blotch-resistant peanut varieties is the most effective and economically viable method for minimizing yield losses due to web blotch.In the current study,a bulked segregant analysis with next-generation sequencing was used to analyze an F2:3 segregating population and identify candidate loci related to web blotch resistance.Based on the fine-mapping of the candidate genomic interval using kompetitive allele-specific PCR(KASP)markers,we identified a novel web blotch resistance-related locus spanning approximately 169 kb on chromosome 16.This region included four annotated genes,of which only Arahy.35VVQ3 had a non-synonymous single nucleotide polymorphism in the coding region between the two parents.Two markers(Chr.16.12872635 and Chr.16.12966357)linked to this gene were shown to be co-segregated with the resistance of peanut web blotch by 72 randomly selected recombinant inbred lines(RIL),which could be used in marker-assisted breeding of resistant peanut varieties. 展开更多

Breast cancer has become a killer of women's health nowadays.In order to exploit the potential representational capabilities of the models more comprehensively,we propose a multi-model fusion strategy.Specifically,we combine two differently structured deep learning models,ResNet101 and Swin Transformer(SwinT),with the addition of the Convolutional Block Attention Module(CBAM)attention mechanism,which makes full use of SwinT's global context information modeling ability and ResNet101's local feature extraction ability,and additionally the cross entropy loss function is replaced by the focus loss function to solve the problem of unbalanced allocation of breast cancer data sets.The multi-classification recognition accuracies of the proposed fusion model under 40X,100X,200X and 400X BreakHis datasets are 97.50%,96.60%,96.30 and 96.10%,respectively.Compared with a single SwinT model and ResNet 101 model,the fusion model has higher accuracy and better generalization ability,which provides a more effective method for screening,diagnosis and pathological classification of female breast cancer. 展开更多

Bimetallic compounds such as hydrotalcite-type layered double hydroxides(LDHs)are promising electrocatalysts owing to their unique electronic structures.However,their abilities toward nitrogen adsorption and reduction are undermined since the surface-mantled,electronegative-OH groups hinder the charge transfer between transition metal atoms and nitrogen molecules.Herein,a smart interfacing strategy is proposed to construct a coupled heterointerface between LDH and 2D g-C_(3)N_(4),which is proven by density functional theory(DFT)investigations to be favorable for nitrogen adsorption and ammonia desorption compared with neat LDH surface.The interfaced LDH and g-C_(3)N_(4) is further hybridized with a self-standing TiO_(2) nanofibrous membrane(NM)to maximize the interfacial effect owing to its high porosity and large surface area.Profited from the synergistic superiorities of the three components,the LDH@C_(3)N_(4)@TiO_(2) NM delivers superior ammonia yield(2.07×10^(−9) mol s^(−1) cm^(−2))and Faradaic efficiency(25.3%),making it a high-efficiency,noble-metal-free catalyst system toward electrocatalytic nitrogen reduction. 展开更多

This paper proposed a method of injection-production system adjustment to solve the problem that the water flooding effect was restricted because of the horizontal and vertical contradictions during the development process of fault block reservoirs. Considering the heterogeneity of reservoir, the Buckley-Leverett water flooding theory was applied to establish the relationship between the recovery and cumulative water injection. In order to achieve the goal of vertically balanced recovery of each section, the calculation method of vertical sectional injection allocation was proposed. The planar triangular seepage unit was assumed and sweep coefficients of different oil-water distribution patterns were characterized using multi-flow tube method. In order to balance and maximize the plane sweep coefficient, the calculation method of plane production system optimization was obtained. Then the injection-production system stereoscopic adjustment method based on equilibrium displacement was proposed with vertical sectional injection allocation and plane production system optimization. This method was applied to injection and production adjustment of BZ oilfield in southern Bohai. The effect of water control and oil increase was obvious. This method can greatly improve the effect of water flooding of offshore fault block reservoirs with the adjustment of injection-production system. 展开更多

Background The association between inflammation and major depressive disorder(MDD)remains poorly understood,given the heterogeneity of patients with MDD.Aims We investigated inflammatory markers,such as interleukin(IL)-6,high-sensitivity C reactive protein(hsCRP)and tumour necrosis factor-α.(TNF-α)in melancholic,atypical and anxious depression and explored whether baseline inflammatory protein levels could indicate prognosis.Methods The sample consisted of participants(aged 18-55 years)from a previously reported multicentre randomised controlled trial with a parallel-group design registered with ClinicalTrials.gov,including melancholic(n=44),atypical(n=37)and anxious(n=44)patients with depression and healthy controls(HCs)(n=33).Subtypes of MDD were classified according to the 30-item Inventory of Depressive Symptomatology,Self-Rated Version and the.17-item Hamilton Depression Rating Scale.Blood levels.of TNF-α,IL-6 and hsCRP were assessed using antibody array analysis.Results Patients with MDD,classified according to melancholic,atypical and anxious depression subtypes,and HCs did not differ significantly in baseline TNF-α,IL-6 and hsCRP levels after adjustment.In patients with anxious depression,hsCRP levels increased significantly if they experienced no pain(adjusted(adj.)p=0.010)or mild to moderate pain(adj.p=0.038)compared with those with severe pain.However,the patients with anxious depression and severe pain showed a lower trend in hsCRP levels than patients with atypical depression who experienced severe pain(p=0.022;adj.p=0.155).Baseline TNF-α(adj.p=0.038)and IL-6(adj.p=0.006)levels in patients in remission were significantly lower than those in patients with no remission among the participants with the atypical depression subtype at the eighth-week follow-up.Conclusions This study provides evidence of differences in inflammatory proteins in patients with varied symptoms among melancholic,atypical and anxious depression subtypes.Further studies on the immunoinflammatory mechanism underlying different subtypes of depression are expected for improved individualised therapy. 展开更多

We designed this study to identify potential key protein interaction networks,genes,and correlated pathways in dilated cardiomyopathy(DCM)via bioinformatics methods.We selected the GSE3586 microarray dataset,consisting of 15 dilated cardiomyopathic heart biopsy samples and 13 nonfailing heart biopsy samples.Initially,the GSE3586 dataset was downloaded and was analyzed with the limma package to identify differentially expressed genes(DEGs).A total of 172 DEGs consisting of 162 upregulated genes and ten downregulated genes in DCM were selected by the criterion of adjusted P values less than 0.01 and the log2-fold change of 0.6 or greater.Gene Ontology functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed to view the biological processes,cellular components,molecular function,and KEGG pathways of the DEGs.Next,protein-protein interactions were constructed,and the hub protein modules were identifi ed.Then we selected the key genes DLD,UQCRC2,DLAT,SUCLA2,ATP5A1,PRDX3,FH,SDHD,and NDUFV1,which are involved in a wide range of biological activities,such as the citrate cycle,oxidation-reduction processes and cellular respiration,and energy derivation by oxidation of organic compounds in mitochondria.Finally,we found that currently there are no related gene-targeting drugs after exploring the predicted interactions between key genes and drugs,and transcription factors.In conclusion,our study provides greater understanding of the pathogenesis and underlying molecular mechanisms in DCM.This contributes to the exploration of potential gene therapy targets. 展开更多

The Antibacterial Effect of CMCTS-Containing Chewing Gum

Objective: This paper was designed to confirm the efficacy of chewing carboxymethyl chitosan(CMCTS)-containing gum in suppressing the growth of oral bacteria when compared to a CMCTS-containing mouth rinse. Methods:Fourteen healthy subjects were recruited from among the staff and students of Qingdao University Dentistry Department. Before the experiments saliva was collected from all subjects and bacteria counts determined. For the gum study, the subjects chewed CMCTS-containing gum for 5 min and then rested for 5 min. When testing the CMCTS mouth rinse, the subjects gargled with 10 mL of solution for 30 s, followed by resting for 9rain 30 s. These protocols were repeated five times over a 50 min period on the same day. Post-experiment saliva samples were then collected at the following times: 0, 30 and 60 min. Results: Chewing gum containing CMCTS or rinsing with a CMCTS-containing rinse significantly decreased oral bacteria counts. The total bacteria counts, total Streptococci counts, and mutans streptococci counts of saliva from subjects who chewed CMCTS-containing gum were significantly lower than saliva from subjects in the rinse group in all three sampling periods, except in the case of the total bacteria count in the 60 min samples. Conclusion:CMCTS-containing gum chewing has a greater antibac- terial effect than using a CMCTS-containing mouth rinse. The present findings strongly indicate that the application of natural materials such as chitosan and its derivatives is useful for better oral health. 展开更多

Electrostatic and hydrophobic interaction cooperative nanochaperone regulates protein folding

Natural molecular chaperones utilize spatially ordered multiple molecular forces to effectively regulate protein folding.However,synthesis of such molecules is a big challenge.The concept of“aggregate science”provides insights to construct chemical entities(aggregates)beyond molecular levels to mimic both the structure and function of natural chaperone.Inspired by this concept,herein we fabricate a novel multi-interaction(i.e.,electrostatic and hydrophobic interaction)cooperative nanochaperone(multi-co-nChap)to regulating protein folding.This multi-co-nChap is fabricated by rationally introducing electrostatic interactions to the surface(corona)and confined hydrophobic microdomains(shell)of traditional single-hydrophobic interaction nanochaperone.We demonstrate that the corona electrostatic attraction facilitates the diffusion of clients into the hydrophobic microdomains,while the shell electrostatic interaction balances the capture and release of clients.By finely synergizing corona electrostatic attraction with shell electrostatic repulsion and hydrophobic interaction,the optimized multi-co-nChap effectively facilitated de novo folding of nascent polypeptides.Moreover,the synergy between corona electrostatic attraction,shell electrostatic attraction and shell hydrophobic interaction significantly enhanced the capability of multi-co-nChap to protect native proteins from denaturation at harsh temperatures.This work provides important insights for understanding and design of nanochaperone,which is a kind of ordered aggregate with chaperone-like activity that beyond the level of single molecule. 展开更多

Alternative polyadenylation(APA)contributes to transcriptome complexity and gene expression regulation and has been implicated in various cellular processes and diseases.Singlecell RNA sequencing(scRNA-seq)has enabled the profiling of APA at the single-cell level;however,the spatial information of cells is not preserved in scRNA-seq.Alternatively,spatial transcriptomics(ST)technologies provide opportunities to decipher the spatial context of the transcriptomic landscape.Pioneering studies have revealed potential spatially variable genes and/or splice isoforms;however,the pattern of APA usage in spatial contexts remains unappreciated.In this study,we developed a toolkit called stAPAminer for mining spatial patterns of APA from spatially barcoded ST data.APA sites were identified and quantified from the ST data.In particular,an imputation model based on the k-nearest neighbors algorithm was designed to recover APA signals,and then APA genes with spatial patterns of APA usage variation were identified.By analyzing wellestablished ST data of the mouse olfactory bulb(MOB),we presented a detailed view of spatial APA usage across morphological layers of the MOB.We compiled a comprehensive list of genes with spatial APA dynamics and obtained several major spatial expression patterns that represent spatial APA dynamics in different morphological layers.By extending this analysis to two additional replicates of the MOB ST data,we observed that the spatial APA patterns of several genes were reproducible among replicates.stAPAminer employs the power of ST to explore the transcriptional atlas of spatial APA patterns with spatial resolution.This toolkit is available at https://github.com/BMILAB/stAPAminer and https://ngdc.cncb.ac.cn/biocode/tools/BT007320. 展开更多

Alternative polyadenylation(APA)plays important roles in modulating mRNA stability,translation,and subcellular localization,and contributes extensively to shaping eukaryotic transcriptome complexity and proteome diversity.Identification of poly(A)sites(pAs)on a genomewide scale is a critical step toward understanding the underlying mechanism of APA-mediated gene regulation.A number of established computational tools have been proposed to predict pAs from diverse genomic data.Here we provided an exhaustive overview of computational approaches for predicting pAs from DNA sequences,bulk RNA sequencing(RNA-seq)data,and single-cell RNA sequencing(scRNA-seq)data.Particularly,we examined several representative tools using bulk RNA-seq and scRNA-seq data from peripheral blood mononuclear cells and put forward operable suggestions on how to assess the reliability of pAs predicted by different tools.We also proposed practical guidelines on choosing appropriate methods applicable to diverse scenarios.Moreover,we discussed in depth the challenges in improving the performance of pA prediction and benchmarking different methods.Additionally,we highlighted outstanding challenges and opportunities using new machine learning and integrative multi-omics techniques,and provided our perspective on how computational methodologies might evolve in the future for non-30 untranslated region,tissuespecific,cross-species,and single-cell pA prediction. 展开更多

Activation of(bi)sulfite(S(IV))by metal oxides is strongly limited by low electrons utilization.In this study,two carbon-supported cobalt ferrites spinels(CoFe^(2)O_(4) QDs-GO and CoFe^(2)O_(4) MOFs-CNTs)have been successfully synthesized by one-step solvothermal method.It was found that both catalysts could efficiently activate S(IV),with rapid reductive dechlorination and then oxidative degradation of a recalcitrant antibiotic chloramphenicol(CAP).Characterizations revealed that CoFe^(2)O_(4) spinels were tightly coated on the carbon bases(GO and CNTs),with effectiveness of the internal transfer of electrons.O_(2)˙−was identified for the reductive dechlorination of CAP,with simultaneously detection of both•OH and SO_(4)^(˙−)responsible for further oxidative degradation.The sulfur oxygen radical conversion reactions and molecular oxygen activation would occur together upon the carbon-based spinels.Spatial-separated interfacial reductive-oxidation of CAP would occur with dechlorination of CAP by O_(2)^(˙−)on the carbon bases,and oxidative degradation of intermediates by SO_(4)^(˙−/•)OH upon the CoFe^(2)O_(4) catalysts. 展开更多

DNA nanotechnology has been widely employed for biomedical applications.However,most DNA nanomaterials rely on noncovalent complementary base pairing of short single-stranded DNA oligonucleotides.Herein,we describe a general strategy to construct a long and covalently conjugated branched DNA structure for fast and in situ gelation in vivo.In our design,a short and covalently conjugated branched DNA structure can normally be employed as the DNA primer in the terminal deoxynucleotidyl transferase-dependent enzymatic polymerization system.After enzymatic extension,the DNA aptamer-modified branched DNA structures with the sequences of poly T or poly A can immediately coassemble for in situ encapsulation of the target protein and tumor cell.The fast and in situ gelation system can function in a murine model of local tumor recurrence for targeting residual tumor cells to achieve long-term drug release for efficient tumor inhibition in vivo.This rationally developed DNA self-assembly strategy provides a new avenue for the development of multifunctional DNA nanomaterials. 展开更多

本文通过常规溶剂挥发法,合成了钬的三元配合物:[Ho(2,5-DClBA)3(5,5′-DM-2,2′-bipy)]2(2,5-DClBA=2,5-二氯苯甲酸根;5,5′-DM-2,2′-bipy=5,5′-二甲基-2,2′-联吡啶)。通过EDTA络合滴定分析、元素分析、红外光谱以及X射线粉末衍射... 本文通过常规溶剂挥发法,合成了钬的三元配合物:[Ho(2,5-DClBA)3(5,5′-DM-2,2′-bipy)]2(2,5-DClBA=2,5-二氯苯甲酸根;5,5′-DM-2,2′-bipy=5,5′-二甲基-2,2′-联吡啶)。通过EDTA络合滴定分析、元素分析、红外光谱以及X射线粉末衍射等手段对配合物进行了表征,利用X射线单晶衍射仪测定了钬配合物的单晶结构。晶体结构分析表明,该晶体是一个双核分子,属于三斜晶系, Pī空间群。每个双核分子包含两个Ho(Ⅲ)离子、6个2,5-二氯苯甲酸根配体和两个5,5′-二甲基-2,2′-联吡啶配体。相邻的双核单元通过C–H···Cl氢键以及Cl-π作用组装成一维和二维超分子结构。利用TG-DTG/FTIR技术研究了钬配合物的热分解机理。用非线性等转化率法得到钬配合物第二步分解反应的活化能E随转化率a的变化关系,结果表明该步是一个简单反应,因此,进一步采用改进的双等双步法来确定钬配合物第二步分解反应的机理函数。通过计算得到了钬配合物的动力学参数(活化能E和指前因子A)和热力学参数(ΔH≠、ΔG≠和ΔS≠)。利用差示扫描量热(DSC)技术对钬配合物的摩尔热容值进行了测定。将所得的平均摩尔热容值与折合温度利用最小二乘法进行拟合得到了多项式方程。将多项式方程结合热力学方程进行计算得到了配合物的舒平摩尔热容值以及焓变、熵变等热力学函数值。 展开更多

Neuroimmune system may be involved in the pathological process of bipolar disorder(BD),but the essential association is not fully understood.Accumulating evidence has shown that BD involves the activation of immune cells and the release of inflammatory substances in the central nerve system(CNS).Meanwhile,neuroimmune responses also interact with other hypothesis of the etiology of BD that are widely recognized,such as neurotransmitter systems,neuroendocrine systems,neurotrophic factors,and oxidative stress.Simultaneously,related genes and immune changes in peripheral blood vary with it.Overall,neuroimmunity may play an important role in the pathogenesis of BD,and the inflammatory cytokines,especially interleukin-6 and tumor necrosis factor-alpha,have potential value for the clinical diagnosis and prognosis of BD,as well as predicting the therapeutic effects of drugs.Large-scale studies are needed to extend the evidence on neuroimmunity in BD,and to examine its clinical value for applications such as early prediction and treatment. 展开更多

The majority of terrestrial vascular plants are capable of forming mutualistic associations with obligate biotrophic arbuscular mycorrhizal (AM) fungi from the phylum Glomeromycota. This mutualistic symbiosis provides carbohydrates to the fungus, and reciprocally improves plant phosphate uptake. AM fungal trans- porters can acquire phosphate from the soil through the hyphal networks. Nevertheless, the precise func- tions of AM fungal phosphate transporters, and whether they act as sensors or as nutrient transporters, in fungal signal transduction remain unclear. Here, we report a high-affinity phosphate transporter GigmPT from Gigaspora margarita that is required for AM symbiosis. Host-induced gene silencing of GigmPT ham- pers the development of G. margarita during AM symbiosis. Most importantly, GigmPT functions as a phos- phate transceptor in G. margarita regarding the activation of the phosphate signaling pathway as well as the protein kinase A signaling cascade. Using the substituted-cysteine accessibility method, we identified residues A146 (in transmembrane domain [TNID] IV) and Va1357 (in TMD VIII) of GigmPT, both of which are critical for phosphate signaling and transport in yeast during growth induction. Collectively, our results pro- vide significant insights into the molecular functions of a phosphate transceptor from the AM fungus G. margarita. 展开更多
