Treated...This study aimed to investigate the synergism of the TAT PTD ( protein transduction domain in HIV-1 transactivator of transcription protein) to antibacte- rial peptide tachyplesin from Tachp/eus tr/dentatus. Treated with Pichia pastoris preferred codon optimization, using the eDNA sequence of tuchyplesin mature pep- tide (54 aa) harboring TAT FFD sequence (11 aa) as reference template, six single-stranded oligonueleotides were designed, the sequences of restriction sites EcoR I and Xba I were introduced to the 5' end of primers P1 and P6, respectively. TAT PTD + Tachyplesin fusion gene with a full length of 219 bp was artificially synthesized by overlap extension PCR, which laid the preliminary foundation for subsequent functional and synergism studies.展开更多
Objective This study aimed to improve the antitumor immunocompetence of a tumor lysate-pulsed dendritic cell(DC)vaccine through differential centrifugation and provide a theoretical basis for its clinical application ...Objective This study aimed to improve the antitumor immunocompetence of a tumor lysate-pulsed dendritic cell(DC)vaccine through differential centrifugation and provide a theoretical basis for its clinical application in glioblastoma.Methods Peripheral blood mononuclear cells were extracted using Ficoll-Paque PLUS and induced into mature DCs in vitro with a cytokine cocktail.The modified tumor lysate was generated by differential centrifugation.The maturity markers of DCs in each group,namely the modified tumor lysate,tumor lysate,and negative and positive control groups,were assessed using flow cytometry.Furthermore,their ability to stimulate lymphocyte proliferation and in vitro antitumor effects were assessed using Cell Trace TM CFSE.IFN-γsecretion levels were measured with ELISA.Intracellular reactive oxygen species were measured using 2’,7’-dichlorofluorescein diacetate(DCFDA)staining.The results were statistically analyzed using an unpaired Student’s t-test and were considered significant at P<0.05.Results Compared with tumor lysate-pulsed DCs,modified tumor lysate-pulsed DCs had a higher expression of maturity markers:CD1a(7.38±0.53%vs.4.47±0.75%)and CD83(19.81±4.09%vs.9.64±1.50%),were better capable of stimulating lymphocyte proliferation[proliferation index(PI):8.54±0.16 vs.7.35±0.05],secreting IFN-γ,and inducing stronger in-vitro cytotoxic T lymphocyte(CTL)cytotoxicity against glioblastoma cells.In addition,we found that the level of ROS in modified tumor lysate-pulsed DCs was lower than that in tumor lysate-pulsed DCs.Conclusion Differential centrifugation of tumor lysates can improve the antitumor immunocompetence of DC vaccines,and reactive oxygen species may be the key to affecting DC function in the whole tumor lysate.展开更多
The human striatum is essential for both lowand high-level functions and has been implicated in the pathophysiology of various prevalent disorders,including Parkinson's disease(PD)and schizophrenia(SCZ).It is know...The human striatum is essential for both lowand high-level functions and has been implicated in the pathophysiology of various prevalent disorders,including Parkinson's disease(PD)and schizophrenia(SCZ).It is known to consist of structurally and functionally divergent subdivisions.However,previous parcellations are based on a single neuroimaging modality,leaving the extent of the multi-modal organization of the striatum unknown.Here,we investigated the organization of the striatum across three modalities—resting-state functional connectivity,probabilistic diffusion tractography,and structural covariance—to provide a holistic convergent view of its structure and function.We found convergent clusters in the dorsal,dorsolateral,rostral,ventral,and caudal striatum.Functional characterization revealed the anterior striatum to be mainly associated with cognitive and emotional functions,while the caudal striatum was related to action execution.Interestingly,significant structural atrophy in the rostral and ventral striatum was common to both PD and SCZ,but atrophy in the dorsolateral striatum was specifically attributable to PD.Our study revealed a cross-modal convergent organization of the striatum,representing a fundamental topographical model that can be useful for investigating structural and functional variability in aging and in clinical conditions.展开更多
By analyzing block share transfers in China during 1999-2006,this study provides estimates of the private benefits of corporate control in China.We find that those controlling blocks are usually priced at a significan...By analyzing block share transfers in China during 1999-2006,this study provides estimates of the private benefits of corporate control in China.We find that those controlling blocks are usually priced at a significant positive premium compared to those comparable non-controlling blocks.The benefits of corporate control vary with shareholders structure,firm characteristics and institutional variables.In particular,targets with a high intangible asset ratio have less benefits of control,cross-listing in B or H shares curbs the extract of private benefits and companies acquired by private firms are associated with higher private benefits.Moreover,it is found that benefits of control are negatively associated with market economy and factor market development index.We argue that firms with higher private benefits experience more tunneling activities by their controlling shareholders.展开更多
