MicroRNAs (miRNAs) are endogenous 22-nt RNAs, which play important regulatory roles by post-transcriptional gene silencing. A computational strategy has been developed for the identification of conserved miRNAs based on features of known metazoan miRNAs in red flour beetle (Tribolium castaneum), which is regarded as one of the major laboratory models of arthropods. Among 118 putative miRNAs, 47% and 53% of the predicted miRNAs from the red flour beetle are harbored by known protein-coding genes (intronic) and genes located outside (intergenic miRNA), respectively. There are 31 intronic miRNAs in the same transcriptional orientation as the host genes, which may share RNA polymerase II and spliceosomal machinery with their host genes for their biogenesis. A hypothetical feed-back model has been proposed based on the analysis of the relationship between intronic miRNAs and their host genes in the development of red flour beetle. 展开更多

[Objectives]This study was conducted to select spring radish varieties with better growth,higher yield and better nutrition.[Methods]A new spring radish combination(2019MUA×Lixiang)was obtained through traditional hybridization methods,and various characters of this line were compared with its parents and commercial spring radish.The radish varieties such as Lixiang radish,2019MUA×Lixiang,2019MUA,commercial variety Zhongluo No.1 and Huangzhou radish were respectively compared and studied in detail in terms of botanical characters,entire leaf bolting ratios,nutritional physiological indexes and so on.[Results]The new radish combination 2019MUA×Lixiang has better indexes,and is more suitable for spring planting.[Conclusions]The study provides a reference basis for the selection on radish planted in Spring. 展开更多

以万寿菊(Tagetes erecta)里程碑·黄色的小叶为外植体,采用农杆菌介导法探究抗生素浓度、菌株类型、菌液浓度、侵染时间、共培养时间、乙酰丁香酮浓度和抗褐化剂种类对万寿菊遗传转化效率的影响。结果表明,头孢霉素(Cef)和硫酸卡... 以万寿菊(Tagetes erecta)里程碑·黄色的小叶为外植体,采用农杆菌介导法探究抗生素浓度、菌株类型、菌液浓度、侵染时间、共培养时间、乙酰丁香酮浓度和抗褐化剂种类对万寿菊遗传转化效率的影响。结果表明,头孢霉素(Cef)和硫酸卡那霉素(Kan)的适宜浓度分别为100 mg·L^(-1)和10 mg·L^(-1);EHA105菌株的稳定转化效率最高;菌液浓度OD600=0.1、侵染5分钟及共培养1天为最佳侵染条件。此外,在侵染过程中添加100μmol·L^(-1)乙酰丁香酮(AS)和筛选培养基中添加0.2 g·L^(-1)聚乙烯吡咯烷酮(PVP)均能提高出芽率。经GUS染色、PCR检测以及Southern blot检测证明转化成功,转化效率达到4%左右。研究结果为万寿菊基因功能研究和转基因育种奠定了基础。 展开更多

万寿菊(Tagetes erecta)是菊科万寿菊属重要的观赏花卉,其舌状花花冠边缘的类型有平滑(smooth)、波浪(undulate)及不同程度的缺刻(incision)等。以第1轮小花花冠筒边缘五裂且雄蕊发育正常的万寿菊突变体JH和舌状花花冠边缘平滑且雄蕊败... 万寿菊(Tagetes erecta)是菊科万寿菊属重要的观赏花卉,其舌状花花冠边缘的类型有平滑(smooth)、波浪(undulate)及不同程度的缺刻(incision)等。以第1轮小花花冠筒边缘五裂且雄蕊发育正常的万寿菊突变体JH和舌状花花冠边缘平滑且雄蕊败育的万寿菊自交系S5为亲本构建F_(2)代分离群体。遗传分析表明,JH突变性状由显性单基因控制,将其命名为Telf。组织学和细胞学观察发现JH第1轮小花为舌状花管瓣化,其花冠筒顶部具有5个裂片,中下部融合呈管状,且花药和花粉粒发育恢复正常。利用BSR-seq和比较基因组学方法,将控制舌状花花冠裂片形成的基因Telf定位于37003-SCAR标记和34032-CAPS标记之间,2个标记距Telf的遗传距离分别为3.684 cM和3.517 cM。该研究为后续精细定位目的基因Telf奠定了基础,也为万寿菊分子标记开发提供了新方法。 展开更多

With the intersection and convergence of multiple disciplines and technologies,more and more researchers are actively exploring interdisciplinary cooperation outside their main research fields.Facing a new research field,researchers often hope to quickly learn what is being studied in this field,which research points are receiving high attention,which researchers are studying these research points,and then consider the possibility of collaborating with core researchers on these research points.In addition,students who are preparing for academic further education usually conduct research on mentors and mentors’research platforms,including academic connections,employment opportunities,etc.In order to satisfy these requirements,we(1)design a research point state map based on a science map to help researchers and students understand the development state of a new research field;(2)design a bar-link author-affiliation information graph to help researchers and students clarify academic networks of scholars and find suitable collaborators or mentors;(3)designs citation pattern histogram to quickly discover research achievements with high research value,such as the Sleeping Beauty papers,recently hot papers,classic papers and so on.Finally,an interactive analytical system named PubExplorer was implemented with IEEE VIS publication data,and its effectiveness is verified through case studies. 展开更多

Although significant breakthroughs have been achieved in tuberculosis management, we still encounter numerous difficulties in diagnosis and treatment of the disease. Additionally, a new challenge, multidrug-resistant tuberculosis (MDR-TB) with unspecific clinical presentation, often results in delayed diagnosis. In this paper, we reported a case of disseminated tuberculosis with rare presentation of ventricular fibrillation, which proved resistant to both isoniazid and rifampicin. A review of literature showed that ventricular fibrillation or tachycardia in tuberculosis patients with pericarditis or myocarditis has been sporadically reported in the past, but none has been conducted involving patients with MDR-TB infections. 展开更多

Rare and undiagnosed diseases substantially decrease patient quality of life and have increasingly become a heavy burden on healthcare systems.Because of the challenges in disease-causing gene identification and mechanism elucidation,patients are often confronted with difficulty obtaining a precise diagnosis and treatment.Due to advances in sequencing and multiomics analysis approaches combined with patient-derived iPSC models and gene-editing platforms,substantial progress has been made in the diagnosis and treatment of rare and undiagnosed diseases.The aforementioned techniques also provide an operational basis for future precision medicine studies.In this review,we summarize recent progress in identifying disease-causing genes based on GWAS/WES/WGS-guided multiomics analysis approaches.In addition,we discuss recent advances in the elucidation of pathogenic mechanisms and treatment of diseases with state-of-the-art iPSC and organoid models,which are improved by cell maturation level and gene editing technology.The comprehensive strategies described above will generate a new paradigm of disease classification that will significantly promote the precision and efficiency of diagnosis and treatment for rare and undiagnosed diseases. 展开更多

A novel core-shell rifampicin/isoniazid electrospun nanofiber membrane for long time drug dissolution

Rifampicin(RIF)and isoniazid(INH)are commonly applied jointly in clinical to improve the treatment efficacy of tuberculosis.Due to the metabolism of the kidneys,most of RIF and INH would be excreted by human bodies after reaching a high drug concentration,which causes serious waste of drugs and does harm to our health.In this study,polylactic acid(PLLA)was chosen as the carrier to prepare core-shell drug-loaded nanofibers with RIF in the shell and INH in the core by coaxial electrospinning.The results showed that the average diameter of the core-shell drug-loaded fibers with an obvious core-shell structure was about 650 nm.Parts of RIF and INH in the fibers became amorphous;the rest maintained crystalline.The combination of PLLA and RIF made the fibers obvious hydrophobic and exhibited a slowly phased sustained-dissolve property during in vitro dissolution studies.The in vitro antibacterial experiments confirmed that the core-shell drug-loaded nanofibers had a favorite inhibitory effect on Staphylococcus aureus,which endowed practical medical value to the fibers.The core-shell drug-loaded nanofibers effectively separated RIF and INH,preventing the degradation of RIF caused by the direct contact of the two drugs.The slow-dissolve characteristics can maintain a relatively stable drug concentration and avoid the damage to the human body caused by the quick dissolve and rapid metabolism of drugs.The combination with coaxial electrospinning fills the gap in the core-shell system with two drugs and has great significance in the future. 展开更多
