The fully developed near-wall turbulent flow fields with and without polymer additives at the same Reynolds number were measured by TRPIV in a water channel. The compar- isons of turbulent statistics confirm that due to viscoelastic structure of long-chain polymers, the wall-normal velocity fluctuation and Reynolds shear stress in the near-wall region are suppressed significantly. Furthermore, it is noted that such a behavior of polymers is closely related to the decease of the motion of the second and forth quadrants, i.e., the ejection and sweep events, in the near-wall region. The spa- tial topological mode of coherent structures during bursts has been extracted by the new mu-level criteria based on locally averaged velocity structure function. Although the general shapes of coherent structures are unchanged by polymer additives, the fluctuating velocity, velocity gradient, velocity strain rate and vorticity of coherent structures during burst events are suppressed in the polymer additive solution com- pared with that in water. The results show that due to the polymer additives the occurrence and intensity of coherent structures are suppressed, leading to drag reduction.展开更多

A turbulent boundary layer flow at Reo = 2460 is measured by ...The present experimental work is devoted to in- vestigate a new space-time correlation model for the turbulent boundary layer over a flat and a wavy walls. A turbulent boundary layer flow at Reo = 2460 is measured by tomographic time-resolved particle image velocimetry (Tomo-TRPIV). The space-time correlations of instantaneous streamwise fluctuation velocity are calculated at 3 different wall-normal locations in logarithmic layer. It is found that the scales of coherent structure increase with moving far away from the wall. The growth of scales is a manifestation of the growth of prevalent coherent structures in the turbulent boundary layer like hairpin vortex or hairpin packets when they lift up. The resulting contours of the space-time correlation exhibit elliptic-like shapes rather than straight lines. It is suggested that, instead of Taylor hypothesis, the elliptic model of the space-time correlation is valid for the wallbounded turbulent flow over either a flat wall or a wavy wall. The elliptic iso-correlation curves have a uniform preferred orientation whose slope is determined by the convection velocity. The convection velocity derived from the space-time correlation represents the velocity at which the large-scale eddies carry small-scale eddies. The sweep velocity rep- resents the distortions of the small-scale eddies and is intimately associated with the fluctuation velocity in the logarithmic layer of turbulent boundary layers. The nondimensionalized correlation curves confirm that the elliptic model is more proper for approximating the space-time correlation than Taylor hypothesis, because the latter can not embody the small-scale motions which have non-negligible distortions. A second flow over a wavy wall is also recorded using TRPIV. Due to the combined effect of shear layers and the adverse pressure gradient, the space-time correlation does not show an elliptic-like shape at some specific heights over the wavy wall, but in the outer region of the wavy wallbounded flow, the elliptic model remains valid.展开更多
Mitochondrial superoxide overproduction is believed to be responsible for the neurotoxicity associated with neurodegeneration.Mitochondria-targeted antioxidants,such as MitoQ,have emerged as potentially effective anti...Mitochondrial superoxide overproduction is believed to be responsible for the neurotoxicity associated with neurodegeneration.Mitochondria-targeted antioxidants,such as MitoQ,have emerged as potentially effective antioxidant therapies.Methionine sulfoxide reductase A(MsrA)is a key mitochondrial-localized endogenous antioxidative enzyme and it can scavenge oxidizing species by catalyzing the methionine(Met)-centered redox cycle(MCRC).In this study,we observed that the natural L-Met acted as a good scavenger for antimycin A-induced mitochondrial superoxide overproduction in PC12 cells.This antioxidation was largely dependent on the Met oxidase activity of MsrA.S-methyl-L-cysteine(SMLC),a natural analogue of Met that is abundantly found in garlic and cabbage,could activate the Met oxidase activity of MsrA to scavenge free radicals.Furthermore,SMLC protected against antimycin A-induced mitochondrial membrane depolarization and alleviated 1-methyl-4-phenylpyridinium(MPP+)-induced neurotoxicity.Thus,our data highlighted the possibility for SMLC supplement in the detoxication of mitochondrial damage by activating the Met oxidase activity of MsrA.展开更多
Dimethyl sulfide(DMS)has been historically recognized as a metabolite of the marine microorganism or a disgusting component for the smell of halitosis patients.In our recent study,DMS has been identified as a cytoprot...Dimethyl sulfide(DMS)has been historically recognized as a metabolite of the marine microorganism or a disgusting component for the smell of halitosis patients.In our recent study,DMS has been identified as a cytoprotectant that protects against oxidative-stress induced cell death and aging.We found that at near-physiological concentrations,DMS reduced reactive oxygen species(ROS)in cultured PC12 cells and alleviated oxidative stress.The radical-scavenging capacity of DMS at near-physiological concentration was equivalent to endogenous methionine(Met)-centered antioxidant defense.Methionine sulfoxidereductase A(MsrA),the key antioxidant enzyme in Met-centered defense,bound to DMS and promoted its antioxidant capacity via facilitating the reaction of DMS with ROS through a sulfonium intermediate at residues Cys72,Tyr103,Glu115,followed by the release of dimethyl sulfoxide(DMSO).MTT assay and trypan blue test indicated that supplement of DMS exhibited cytoprotection against 6-hydroxydopamine and MPP+induced cell apoptosis.Furthermore,Msr A knockdown abolished the cytoprotective effect of DMS at near-physiological concentrations.The present study reveals new insight into the potential therapeutic value of DMS in Parkinson disease.展开更多
