SSR Molecular Marker for Late Blight Resistance Gene in Tomato CLN2037

[Methods] ... [Objectives] The research aimed to investigate the genetic rule of the disease resistance of tomato CLN2037,find molecular markers linked with disease-resistant genes and locate the disease-resistant genes. [Methods] BCr,BCs and F2 were constructed from tomato CLN2037 and susceptible cultivar T2-03,and their resistance to late blight was identified and analyzed by inoculating the disease in the seedling stage of tomatoes. The genetic linkage analysis was made based on the genetic linkage map constructed by ICu GI using group segregation method and 90 pairs of tomato SSR primers. [Results]The resistance of CLN2037 to late blight was controlled by a single recessive gene. The polymorphic bands amplified by primer DM0231 had a linkage relationship with resistance genes. The size of the polymorphism fragment was 226 bp with the genetic linkage distance of 2. 67 c M,and the disease resistance gene was localized on CCPB272-03741.[Conclusions]DM0231 could be used as a molecular marker for molecular breeding of tomato against late blight,which laid the foundation for further fine localization of late blight resistance genes. 展开更多

This paper studies the influence of three different pretreatments( blanching,citric acid soaking,and Na Cl soaking) on the moisture content,degree of puffing,color,hardness and crispness of Hami melon under explosion puffing drying at modified temperature and pressure in Xinjiang. The results show that using 60 s blanching pretreatment can help to reduce the moisture content of puffed product while maintaining product color; 2. 5% citric acid pretreatment can make products maintain high degree of puffing and crispness,but reduce product color; 2%Na Cl penetrating fluid plays a significant role in maintaining product color,while maintaining crispness. 展开更多

With reported pathogenic genes of Botrytis cirterea Pers. and unknown proteins related to pathogencity as experimental materials, the pathogenic genes were studied by bioinformatics methods, and these unknown proteins were analyzed with bioinforrnaties tools and softwares, so as to lay a theoretical basis for the control of tomato grey mould at molecular level in future. The results showed that through the alignment analysis by DNAMAN software, three pairs of unknown pro- teins with higher comprehensive matching degrees were obtained, and these proteins were predicted to be coded by hrp gene cluster of type Ili secretion system; through Bioedit and ProtParam, the amino acid composition, size, molecular weight, isoelectrie point and hydrophobicity of the each of the six unknown proteins were obtained; and corresponding secondary structures were obtained using DNAMAN. The results of this study could guide deep research of such genes. 展开更多

Genetic Analysis of Resistance to Tomato Gray Mold( Botrytis cinerea)

The study used four tomato varieties,03786( susceptible),05237( susceptible),C2985( resistant) and Wva536( resistant) with different resistance,to formulate combinations according to Griffing( I) complete diallel cross method. And Botrytis cinerea was inoculated to investigate the incidence of gray mold. The results showed that:(1) there were significant differences in disease resistance between different varieties and different combinations.(2) There were significant differences in general combining ability and special combining ability between the tested varieties,and Wva536 and C2985 had a higher general combining ability effect,and were both relatively good parents when formulating resistant combinations. Combinations Wva536 × 03786 and Wva536 × 05237 had the highest special combining ability,and thus have the value of further research and utilization.(3) The additive effect in resistance inheritance was located at the primary position,and partial dominance also existed as well as cytoplasmic effect.(4) The broad-sense heritability and narrow-sense heritability were,respectively,89. 73% and82. 15%,which were higher,indicating that the parents had a greater effect on offsprings. The resistance genes can be expressed in offsprings through gene accumulation,and should be selected in the early generation. 展开更多

In order to explore the fresh-keeping methods for zander fillet,we use vacuum packaging and partial freezing to process zander fillet,conduct a comparative analysis of sensory characteristics,percentage of water loss,stiffness period,muscle stiffness,TVB-N and water activity during the storage,and assess the fresh-keeping effect of zander fillet under the conditions of vacuum packaging and partial freezing. The results show that zander can be dead after being placed into the 2℃ water for 30 min,and the stiffness period can be extended about 100 min compared with natural death; the percentage of water loss under the conditions of vacuum packaging before partial freezing is 2. 6% lower than under the conditions of partial freezing before vacuum packaging; after being stored at- 7℃ and- 4℃ for 15 d,TVB- N of fillet ≤15 mg /kg; TVB- N of fillet at- 7℃ is 25% lower than at- 4℃. 展开更多

This study was conducted to screen tomato resources resistant to gray mold for the first time by in-vitro stem inoculation method.The results showed that Solanum habrochaites T2-07-316 was highly resistant to gray mold,and had the relative stem infection rate and relative stem rot expansion rate of 0 on the 6^(th) day after inoculation.There were also other seven transgenic common tomato materials which showed higher resistance to gray mold,with a relative stem infection rate in the range of 15.00%-38.33% and a relative stem rot expansion rate in the range of 10.22%-23.57%.Among them,T2-07-337 had the best resistance. 展开更多
