[Methods] BCr,BCs and F2 were constructed from tomato CLN2037 and susceptible cultivar T2-03,and their resistance to late blight was identified and analyzed by inoculating the disease in the seedling stage of tomatoes. The genetic linkage analysis was made based on the genetic linkage map constructed by ICu GI using group segregation method and 90 pairs of tomato SSR primers. [Results]The resistance of CLN2037 to late blight was controlled by a single recessive gene. The polymorphic bands amplified by primer DM0231 had a linkage relationship with resistance genes. The size of the polymorphism fragment was 226 bp with the genetic linkage distance of 2. 67 c M,and the disease resistance gene was localized on CCPB272-03741.[Conclusions]DM0231 could be used as a molecular marker for molecular breeding of tomato against late blight,which laid the foundation for further fine localization of late blight resistance genes.展开更多

The results showed that through the alignment analysis by DNAMAN software, three pairs of unknown pro- teins with higher comprehensive matching degrees were obtained, and these proteins were predicted to be coded by hrp gene cluster of type Ili secretion system; through Bioedit and ProtParam, the amino acid composition, size, molecular weight, isoelectrie point and hydrophobicity of the each of the six unknown proteins were obtained; and corresponding secondary structures were obtained using DNAMAN. And Botrytis cinerea was inoculated to investigate the incidence of gray mold. The results showed that:(1) there were significant differences in disease resistance between different varieties and different combinations.(2) There were significant differences in general combining ability and special combining ability between the tested varieties,and Wva536 and C2985 had a higher general combining ability effect,and were both relatively good parents when formulating resistant combinations. Combinations Wva536 × 03786 and Wva536 × 05237 had the highest special combining ability,and thus have the value of further research and utilization.(3) The additive effect in resistance inheritance was located at the primary position,and partial dominance also existed as well as cytoplasmic effect.(4) The broad-sense heritability and narrow-sense heritability were,respectively,89. 73% and82. 15%,which were higher,indicating that the parents had a greater effect on offsprings.
